Presence of Mycobacterium avium subsp. paratuberculosis (MAP) in milk for human consumption is a concern due to its possible relationship with Crohn's disease in humans. Pasteurization effectively reduces the MAP load by four to five logs, but the efficacy depends on the MAP concentration, which depends on the prevalence among contributing herds and individuals. Considerable variation of MAP in bulk tank milk (BTM) and individual cow's milk (IM) is reported, but factors associated with MAP occurrence in milk at farm level have not been described. This study systematically reviewed published studies aiming at estimating the occurrence of MAP in on-farm BTM and IM by meta-analysis. A total of 692 articles were identified through electronic databases and initially screened using title and abstract. The quality of the 61 potentially relevant articles was assessed using full text and 31 articles were eventually included in the meta-analysis. The apparent prevalence (AP) of MAP in BTM and IM on farm were summarized in relation to strata defined by the test used to identify MAP and the infection status of the herds/animals. There was considerable inconsistency in the reporting, resulting in missing information potentially explaining the dispersion in the estimated AP. The overall AP and 95% confidence intervals based on PCR and culture of MAP were summarized to 0.10 (0.04-0.22) in BTM and 0.20 (0.12-0.32) in IM. Quantifying the MAP load in test-positive milk samples was not possible because very few articles provided quantitative information on individual samples.  相似文献

The present study describes the occurrence of various gastrointestinal helminths, lungworms and liver flukes in Norwegian sheep and goats as assessed from faecal samples and post mortem examinations performed between 2007 and 2010. Faecal samples for gastrointestinal nematode egg counts were collected from 77 sheep flocks and 30 dairy goat flocks from three geographical regions in Norway. Additionally, thirty-two lambs and 16 adult goats were euthanized for necropsy examination and for identification of adult gastrointestinal nematodes and tapeworms, lungworms and liver flukes. The survey showed that there was a higher mean excretion of trichostrongyle eggs in sheep than in goats at the individual level (392 EPG vs. 154 EPG, p < 0.001). For both host species, the mean prevalence and intensity of excreted trichostrongyle eggs were significantly higher in the southern coastal region compared with the inland and northern regions (p < 0.001). Third stage larvae of Trichostrongylus/Teladorsagia, Haemonchus and Nematodirus type were the most prevalent ones in the coprocultures from sheep, whereas larvae of Trichostrongylus/Teladorsagia and Nematodirus type dominated in goats. The most prevalent gastrointestinal nematode species found at necropsy was Teladorsagia circumcincta (75.0 and 81.2% respectively in sheep and goats), while the largest mean worm burdens were recorded for Haemonchus contortus in sheep (724 ± 623) and T. circumcincta in goats (377 ± 529). Infections with Mycobacterium avium subsp. paratuberculosis (MAP) can be latent for years without affecting the animal, but the animal may become infectious or clinical at some point. Diagnosis of paratuberculosis can be a challenge primarily in latent stages of the infection, and different diagnosis interpretations are usually required by the variety of decision makers. The objective of this paper was to provide a critical review of reported accuracies of ELISA tests, interferon-gamma assays (IFN-gamma) and faecal culture (FC) techniques used for diagnosis of three defined target conditions: MAP infected, MAP infectious and MAP affected animals. For each animal species, target condition and diagnostic test-type, sensitivities (Se) and specificities (Sp) were summarised based on a systematic, critical review of information in literature databases. The diagnostic test information often varied substantially for tests of the same type and make, particularly ELISA, which was the most frequently reported test-type. Comparison of the various tests accuracies was generally not possible, but stratification of test-evaluations by target condition improved the interpretation of the test accuracies. Infectious and affected animals can often be detected, but Se for infected animals is generally low. A main conclusion of the review was that the quality of design, implementation and reporting of evaluations of tests for paratuberculosis is generally poor. Particularly, there is a need for better correspondence between the study population and target population, i.e. the subjects chosen for test evaluation should reflect the distribution of animals in the population where the test is intended to be used.  相似文献

Background

The quality of the floor is essential to the welfare of piglets as abrasions often are recorded in newborn piglets, and such lesions may lead to lameness. Apart from animal suffering, lameness contributes to losses in form of dead piglets, decreased growth, and increased use of antibiotics and manual labour.

Methods

In a herd with three different farrowing systems, 37 litters (390 piglets) were studied until the age of 3 weeks with respect to presence of skin wounds and abrasions. Lameness was registered until the age of 7 weeks. Eight lame piglets were sacrificed before medical treatment and subjected to necropsy including histopathological and microbiological examinations. Isolates of streptococci, staphylococci and E. coli were tested with respect to antimicrobial resistance. Mastitis was observed in ten sows.

Results

The most severe abrasions at carpus and soles were seen in the system with a new solid concrete floor with a slatted floor over the dunging area. The lowest magnitude was observed in the deep litter system with peat. Sole bruising was more common in the systems with concrete floor compared to the deep litter system with peat, and the differce in prevalence was significant at all examination days. The lesions decreased with time and about 75% of the treatments for lameness were performed during the first three weeks of life. The overall prevalence of lameness was highest in the system with new solid concrete floor with a slatted floor over the dunging area (9.4%) followed by the old solid concrete floor (7.5%). A lower (p < 0.05) prevalence was seen in the deep litters system with peat (3.3%). No significant relationship between mastitis and abrasions or lameness in the offspring was observed.

Conclusion

There were large differences in the prevalence of abrasions and lameness between the floor types. The deep litter system with peat provided a soft and good floor for piglets. The overall prevalence of lameness was only diagnosed in every fourth litter in that system compared to in every second litter in the systems with concrete floor. In contrast, the incidence of mastitis in the sows during the first week after farrowing was higher than in the systems with concrete floor.  相似文献

A bottleneck of crustacean larval culture concerns nutrition and associated cannibalism in communal rearing systems, which impact on larval survival, development and growth. For early‐stage European lobster, Homarus gammarus larvae, feeding ecology and body composition are largely unknown. We initiated four progressive feeding experiments (novel feed types, feeding regime and feed size and cannibalism effects) on growth and survival, to inform and update husbandry protocols. Performance of larvae offered a dry commercial feed was not significantly different compared with a conventional wet plankton feed of the same ration and size grade (both within 600–1,000 μm). Further experiments found that the same ration of dry feed offered six times daily improved development and growth, over the conventional regime of three times daily. Small‐grade dry feed (particles: 250–360 μm) improved larval performance compared with a larger feed (360–650 μm). Larvae were also fed different proportions of dry feed and/or conspecifics in both communal and individual rearing systems (the latter preventing cannibalism via segregation). Individually reared larvae, fed only dead conspecifics, displayed the greatest survival (80%) to postlarvae. This underlines the impact of cannibalism on survival and nutrition in H. gammarus larviculture. A final experiment analysed H. gammarus zoea 1 composition, identifying deficiencies in ash and carbohydrate in lobster feeds. This suggests a need for a species‐specific, formulated dry feed for H. gammarus larviculture. Our research represents the first investigation of H. gammarus larval composition and dietary requirements and highlights decreased growth potential associated with providing nutrition solely from generic commercial feed.  相似文献

Three problems need to be addressed in networks of Infratec Grain Analysers: 1) the networks are not interconnected, 2) the partial least squares (PLS) calibrations used so far have to be individually adjusted for bias when transferred to the slave instruments, and 3) the calibrations are not entirely stable over time. Nonlinear artificial neural network (ANN) calibrations based on a large common European data set (≈4,000 samples in the training sets and ≈1,000 samples in the stop sets) were introduced to overcome these constraints. The performance of these ANN calibrations was compared with Danish PLS models for protein and moisture in cereals during the 1998 harvest in Denmark, and subsequently with PLS models based on the same European data set. ANN models were more accurate than PLS and, unlike PLS, were linear and transferable up to 25% moisture. It is suggested that the improved performance of the ANN models is attributable to the modeling technique rather than the size and nature of the European data set. In most cases, ANN models could be applied directly and without bias adjustment to slave instruments. The ANN models were also more stable, they required fewer bias adjustments or remodeling over time compared with Danish PLS models. ANN calibrations using shared data have been adopted for commercial use in several European countries and work is in progress to develop global ANN models for determination of protein in wheat and barley.  相似文献

The survival of fish eggs and larvae, and therefore recruitment success, can be critically affected by transport in ocean currents. Combining a model of early‐life stage dispersal with statistical stock–recruitment models, we investigated the role of larval transport for recruitment variability across spatial scales for the population complex of North Sea cod (Gadus morhua). By using a coupled physical–biological model, we estimated the egg and larval transport over a 44‐year period. The oceanographic component of the model, capable of capturing the interannual variability of temperature and ocean current patterns, was coupled to the biological component, an individual‐based model (IBM) that simulated the cod eggs and larvae development and mortality. This study proposes a novel method to account for larval transport and success in stock–recruitment models: weighting the spawning stock biomass by retention rate and, in the case of multiple populations, their connectivity. Our method provides an estimate of the stock biomass contributing to recruitment and the effect of larval transport on recruitment variability. Our results indicate an effect, albeit small, in some populations at the local level. Including transport anomaly as an environmental covariate in traditional stock–recruitment models in turn captures recruitment variability at larger scales. Our study aims to quantify the role of larval transport for recruitment across spatial scales, and disentangle the roles of temperature and larval transport on effective connectivity between populations, thus informing about the potential impacts of climate change on the cod population structure in the North Sea.  相似文献
