High-resolution slow magic-angle spinning (150 Hz) 1H PASS NMR spectroscopy is performed on intact excised rat m. tibialis anterior. Untreated muscles and muscles in vitro incubated in Krebs-Ringers buffer based on deuterium oxide are investigated. In the high-frequency region of the 1H NMR spectra, resonances from H4 (approximately 7.1-7.2 ppm) and H2 (approximately 8.2-8.5 ppm) in histidine are observed. In addition, a resonance appears at 6.7 ppm for the untreated muscles. However, this resonance is absent in muscles following incubation in deuterium oxide. On the basis of its behavior in deuterium oxide combined with supplementary measurements for creatine solutions, the 6.7 ppm resonance is ascribed to the amino protons in creatine. Moreover, the present study demonstrates that the observation of the 6.7 ppm resonance depends on pH, which explains earlier reports stating its occasional appearance. Finally, measurements on solutions of ATP/AMP and histidine indicate that both ATP/AMP and histidine contribute to the resonances at approximately 8.2-8.5 ppm in the 1H NMR spectra of muscle tissue.  相似文献

Plant pathologists have traditionally worked in the area of clarifying and understanding the disease cycles of specific diseases, factors influencing epidemiology, yield loss potential and host-pathogen interactions in order to be able to minimise the disease risk, build warning systems or recommend specific control thresholds in relation to the application of fungicides. The decision support system Crop Protection Online (CPO) is an example of a threshold-based system that determines economically viable fungicide strategies. The system is based on using appropriate doses aimed at minimising the overall pesticide input. CPO is used widely by advisors and many of the thresholds are generally accepted and disseminated through newsletters. The national figures for the use of fungicides in cereals have shown a major reduction during the last 20 years and their use today is much in line with the level that can be achieved from using CPO as indicated from validation trials. The number of end-users among farmers has been stable at around 3% during the last 10 years (800–1,000 farmers). Major hurdles in increasing the number of users are believed to be: (1) the requirements for carrying out assessments in the field, (2) farm sizes getting larger, leaving less time for decision making for individual fields, (3) lack of economic incentives to change from standard treatments, (4) the failure of decision support systems to interact with other computer-based programmes on the farm, (5) the lack of compatibility of decision support systems with farmers’ ways of making decisions on crop protection in general, (6) the need for direct interactions with advisors. A sociological investigation into the farmers’ way of making decisions in the area of crop protection has shown that arable farmers can be divided into three major groups: (a) systems-orientated farmers, (b) experienced-based farmers and (c) advisory-orientated farmers. The information required by these three groups is different and has to be looked at individually from the end-user’s perspective rather than from the scientist’s perspective. New ways of entering the decision support system where specific field inspections are omitted and where regional disease data are relied on, have been investigated and tested in field trials. The results show possibilities for further developments in that direction, which might be one way of gaining more end-users.  相似文献

The effect of the use of different doses of the fungicide fenpropimorph on populations of barley powdery mildew Blumeria ( Erysiphe ) graminis f. sp. hordei was investigated in a laboratory selection experiment. A sample from the Danish aerial population of powdery mildew was split into populations, and these were kept separately for 31 generations on susceptible barley seedlings treated with fungicide at two concentrations, as well as on a control. Samples from these populations were tested for their resistance to fenpropimorph and their virulence spectra. There was a large amount of environmental variation in the ED50 values used to measure fungicide resistance. In both treated populations, the average level of fungicide resistance increased, this increase being faster and greater in the population treated with the high dose. The diversity of pathotypes of the treated populations decreased, with the decline being more rapid in the population treated with the high dose, where one pathotype dominated the population after 31 generations. This pathotype was apparently not the fittest in the population treated with the low dose. This implies that knowledge of ED50 is not sufficient to predict pathotype evolution under different fungicide treatments. The dominant pathotype in the high-dose treatment may not have been clonal, as there was evidence of two levels of fungicide resistance. The large environmental variation observed in estimated ED50 values for resistance towards fenpropimorph may help to explain why this resistance has evolved at a slower rate than resistance towards other fungicides.  相似文献

Over the last two decades, ash dieback has become a major problem in Europe, where the causative fungus has invaded the continent rapidly. The disease is caused by the invasive pathogenic fungus Hymenoscyphus pseudoalbidus (anamorph Chalara fraxinea), which causes severe symptoms and dieback in common ash, Fraxinus excelsior. It is becoming a significant threat to biodiversity in forest ecosystems and the economic and aesthetic impacts are immense. Despite the presence of the disease for at least 10 years in Scandinavia, a small fraction of F. excelsior trees have remained vigorous, and these trees exhibit no or low levels of symptoms even where neighbouring trees are very sick. This gives hope that a fraction of the ash trees will retain a sufficiently viable growth to survive. Following a period of high mortality in natural populations, selection and breeding of remaining viable ash trees could therefore provide a route for restoring the role of ash in the landscape. This paper reviews the available data on disease dissemination, and the consequences thereof in terms of symptom severity and mortality, and appraises studies that have tested the hypothesis that less‐affected trees have genetically based resistance. The implications of the results for the adaptive potential of common ash to respond to the disease through natural or assisted selection are discussed. The risks of adverse fitness effects of population fragmentation due to high mortality are considered. Finally, it is recommended that resistant trees (genotypes) should be selected to facilitate conservation of the species.  相似文献

Background

Laminitis and energy-related postpartum diseases share several risk factors, indicating a common etiology. Thus, a herd’s incidence rate of energy-related postpartum diseases, such as displaced abomasum and clinical ketosis, might reflect the likelihood of cows to suffer from laminitis-related claw lesions. The aim of this study was to investigate the association between herd-level incidence rate of displaced abomasum and clinical ketosis, general risk factors, and claw lesions in individual cows recorded at maintenance claw trimming.

Methods

The dataset consisted of 6773 trimmings, performed between 2004 and 2006 by professional trimmers, from 3607 Swedish Red and Swedish Holstein cows in 26 herds. The herds were classified as having a high, inconsistent-high or low incidence rate of energy-related postpartum diseases, based on the number of recorded cases of veterinary-diagnosed displaced abomasum and clinical ketosis in the Swedish national animal disease recording system during 2002 to 2006, and observations and interviews in connections with herd visits. Generalized linear mixed models were used to investigate the association between herd-level incidence rate of energy-related postpartum diseases and laminitis-related lesions including sole ulcer and sole hemorrhage; and hygiene-related lesions including interdigital dermatitis, digital dermatitis, heel-horn erosion, verrucose dermatitis, and interdigital hyperplasia; and absence of any claw lesion. Systematic effects, including first-order interactions, with P < 0.05 were included in the models. Herd classification was forced into the models, and a random effect of herd was included.

Results

In comparison to herds with a high incidence rate of energy-related postpartum diseases, low-incidence herds showed a lower odds ratio (OR; 0.2) for laminitis-related lesions in cows trimmed during the summer months. Low-incidence herds also showed numerically lower OR estimates for laminitis-related lesions in all parity classes and a numerically lower OR for hygiene-related lesions. In addition, low-incidence herds showed tendencies towards a numerically higher OR for absence of any lesion, irrespective of trimming season or parity.

Conclusions

Only a few statistically significant associations were found, but several tendencies pointed towards better claw health in herds with low as compared with high incidence rate of energy-related postpartum diseases.  相似文献

Crop yields and yield potentials on Danish coarse sandy soils are strongly limited due to restricted root growth and poor water and nutrient retention. We investigated if biochar amendment to subsoil can improve root development in barley and significantly increase soil water retention. Spring barley (Hordeum vulgare cv. Anakin) was grown in soil columns (diameter: 30 cm) prepared with 25 cm topsoil, 75 cm biochar‐amended subsoil, and 30 cm un‐amended subsoil lowermost placed on an impervious surface. Low‐temperature gasification straw‐biochar (at 0, 0.50, 1.0, 2.0, and 4.0 wt%) and slow pyrolysis hardwood‐biochar (at 2 wt%) were investigated. One wt% can be scaled up to 102 Mg/ha of char. After full irrigation and drainage, the in‐situ moisture content at 30‐80 cm depth increased linearly (R2 = 0.99) with straw‐biochar content at a rate corresponding to 0.029 m3/m3/%. The lab determined wilting point also increased linearly with char content (R2 = 0.99) but at a much lower rate (0.003 m3/m3/%). Biochar at concentrations up to 2% significantly increased the density of roots in the 40–80 cm depth interval. Addition of 1% straw‐biochar had the most positive effect on root penetration resulting in the highest average root density (54% coverage compared to 33% without biochar). This treatment also resulted in the greatest spring barley grain yield increase (22%). Improving the quality of sandy subsoils has global potentials, and incorporation of the right amount of correctly treated residues from bioenergy technologies such as straw‐biochar is a promising option.  相似文献

The sow at parturition is challenged with respect to energy status due to increases in energetic expenses associated with 1) nest building 2) uterine contractions, and 3) colostrum production. A previous study indicated that sows were depleted of glucogenic energy around farrowing. The aim was to investigate whether intravenous infusion of glucose from observed nest-building behavior to 24 h postpartum affected the farrowing kinetics and colostrum production in sows. Ten multiparous sows (DanBred landrace × DanBred Yorkshire) were fitted with a jugular vein catheter on each side (one for infusion and the other one for blood sampling). Sows were infused with either 0.9% saline (CON; n = 5) or 10% glucose (GLU; n = 5) solution at a constant rate of 125 mL/h. From day 108 of gestation, sows were fed once daily with 3.33 kg of a standard lactation diet. During farrowing, sows were monitored to register the onset of farrowing, time of birth, birth status (live or dead), sex, stillbirth rate (SR), and weight of newborn piglets. Farrowing assistance (FA) was provided when the birth interval exceeded 60 min. In late gestation, 1 mL of blood was collected every third hour for blood gas analysis and every sixth hour for harvesting plasma. During farrowing, 1 mL (for blood gas) and 9 mL of blood were collected at 0, 3, 6, 9, 12, 15, 18, 21, and 24 h in milk (HIM). Colostrum and milk samples were collected at 0, 6, 12, 18, 24, and 36 HIM and also at 3, 10, 17, and 24 d in milk. Compared with CON sows, GLU infusion decreased the SR (16.1% vs. 7.4%; P = 0.03), FA (21% vs. 9.0%; P = 0.01), and surprisingly also blood glucose at the onset of farrowing (5.53 vs. 5.09 mmol/L; P = 0.03), respectively. A tendency to higher plasma lactate at the onset of farrowing (P = 0.05) but decreased piglet survival from 0 to 24 h (P = 0.06) was also found for GLU sows. No effects of treatment on farrowing duration or mean birth intervals were found. Lactate in whole blood (P = 0.003) and plasma (P = 0.002) was increased for GLU sows as compared with CON sows during the colostrum period. No effect of GLU infusion was seen on colostrum and milk composition and yield. The increase in lactate was most likely due to a shift toward a greater proportion of glucose oxidation and insufficient O2 supply to fuel uterine contractions. In conclusion, infusion of glucose reduced the frequency of SR and FA, and improved energy status of the sow which seems to be crucial to enhance total piglet survival.  相似文献
