Glutenins, which form the network of gluten protein, are of great importance for the quality of flour products. Glutenins can be divided into HMW and LMW subunits according to molecular weight. Three genes for LMW glutenin subunits (LMW‐GS), named lmw‐cnd1, lmw‐cnd2, and lmw‐cnd3 with open reading frames of 1,053, 903, and 969 bp, respectively, were cloned from wheat cultivar Cheyenne. Heterologous expression vectors of the three LMW‐GS were constructed, and the recombinant proteins LMW‐CND1, LMW‐CND2, and LMW‐CND3 were overexpressed in Escherichia coli. After cell disruption with ultrasound, target proteins of high purity were obtained by using Ni2+ affinity chromatography. Farinograph and TAPlus measurements were used to investigate the effects of the three LMW‐GS on the characteristics of flour and dough. The results showed that the addition of each LMW‐GS can lead to an increase in the elasticity of the dough. Moreover, LMW‐CND2 and LMW‐CND3 promoted the strength of the dough. All three LMW‐GS caused a decrease of hardness and increase of springiness and cohesiveness of dough according to texture profiling results. Sulfonamides are the second most used antibiotic class in veterinary medicine and applied to livestock to treat bacterial infections. Subsequently, they are spread onto agricultural soils together with the contaminated manure used as fertilizer. Both manure and antibiotics affect the soil microbial community. However, the influence of different liquid manure loads on effects of antibiotics to soil microorganisms is not well understood. Therefore, we performed a microcosm experiment for up to 32 d to clarify whether the function and structure of the soil microbial community is differently affected by interactions of manure and the antibiotic sulfadiazine (SDZ). To this end selected concentrations of pig liquid manure (0, 20, 40, 80 g kg–1) and SDZ (0, 10, 100 mg kg–1) were combined. We hypothesized that incremental manure amendment might reduce the effect of SDZ in soils, due to an increasing sorption capacity of SDZ to organic compounds. Clear dose‐dependent effects of SDZ on microbial biomass and PLFA pattern were determined, and SDZ effects interacted with the liquid manure application rate. Soil microbial biomass increased with incremental liquid manure addition, whereas this effect was absent in the presence of additional SDZ. However, activities of enzymes such as urease and protease were only slightly affected and basal respiration was not affected by SDZ application, while differences mostly depended on the concentration of liquid manure. These results illustrated that the microbial biomass and structural composition react more sensitive to SDZ contamination than functional processes. Furthermore, effects disproportionally increased with incremental liquid manure addition, although extractable amounts of SDZ declined with increasing liquid manure application. 相似文献

The relationship between nitrate reductase activity and ferredoxin levels in lemon tree leaves was studied. The experiments were carried out on leaves from full‐nutrient sufficient trees as the reference, and on leaves from trees with several nutritional stresses, mainly iron chlorosis from trees growing under Fe‐stressed conditions.
Iron deficiency reduced leaf ferredoxin concentration and consequently decreased nitrate reductase activity. Fe(II) infiltration treatments of intact leaves, as well as several incubation assays, permit to deduce the dependence of the enzymatic nitrate reduction of the leaf ferredoxin levels. 相似文献

Homogenates were prepared from the leaves of hydroponically cultivated sunflowers (Helianthus annuus, L.) under deficient, normal and toxic B conditions and the polyphenol oxidase activity together with the ortho‐diphenolic and micronutrient content was measured every week from one month to flowering. B deficiency slightly depressed polyphenol oxidase activity and B toxicity decreased it significantly at all times, the response fitting a line versus the logarithm of the B concentration in solution. We found a positive correlation between polyphenol oxidase activity and the B content in leaves. B action on enzymatic activity seemed not to be caused by direct interaction with substrates as the infiltration of B into deficient leaves did not restore the normal activity According to our results we suggest that polyphenol oxidase activity may be a valid functional parameter to indicate the nutritional status of B in sunflower plants, if its specificity for B can be demonstrated. The ortho‐diphenolic content did not change with the B levels used in our experiments. Neither did we find any correlation between enzymatic activity and the o‐diphenolic content of the leaves. 相似文献

Development of high‐protein digestibility (HPD)/high‐lysine (hl) sorghum mutant germplasm with good grain quality (i.e., hard endosperm texture) has been a major research objective at Purdue University. Progress toward achieving this objective, however, has been slow due to challenges posed by a combination of genetic and environmental factors. In this article, we report on the identification of a sorghum grain phenotype with a unique modified endosperm texture that has near‐normal hardness and possesses superior nutritional quality traits of high digestibility and enhanced lysine content. These modified endosperm lines were identified among F6 families developed from crosses between hard endosperm, normal nutritional quality sorghum lines, and improved HPD/hl sorghum mutant P721Q‐derived lines. A novel vitreous endosperm formation originated in the central portion of the kernel endosperm with opaque portions appearing both centrally and peripherally surrounding the vitreous portion. Kernels exhibiting modification showed a range of vitreous content from a slight interior section to one that filled out to the kernel periphery. Microstructure of the vitreous endosperm fraction was dramatically different from that of vitreous normal kernels in sorghum and in other cereals, in that polygonal starch granules were densely packed but without the typically associated continuous protein matrix. We speculate that, due to the lack of protein matrix, such vitreous endosperm may have more available starch for animal nutrition, and possibly have improved wet‐milling and dry‐grind ethanol processing properties. The new modified endosperm selections produce a range that approaches the density of the vitreous parent, and have lysine content and protein digestibility comparable to the HPD/hl opaque mutant parent. 相似文献

There is lack of information available concerning the effect of humic substances (HS) applied via fertigation on plant growth in sandy soils. Therefore, a field experiment was carried out at El‐Saff district (20 km southwest of Cairo), Egypt, to investigate the role of HS fertigation on water retention of a sandy soil, yield and quality of broccoli (Broccoli oleracea L.) as well as on soil nutrient concentration retained after harvest. The experiment consisted of six fertigation treatments (50%, 75%, and 100% of the recommended NPK‐fertilizer rate for broccoli combined with and without HS application at 120 L ha–1) in a complete randomized block design with three replicates. Humic substances affected spatial water distribution and improved water retention in the root zone. Furthermore, application of HS increased total marketable yield and head diameter of broccoli as well as quality parameters (i.e., total soluble solids, protein, and vitamin C). Higher nutrient concentrations were found in the broccoli heads and concentrations of plant‐available nutrients in soil after harvesting were also higher, indicating an improvement in soil fertility. In conclusion, HS fertigation can be judged as an interesting option to improve soil water and nutrient status leading to better plant growth. 相似文献

Eight Scandinavian oat cultivars were studied as grains, groats before and after kiln drying, rolled oats, and oatmeal for 62 physical, chemical, and sensory parameters over three consecutive crop years. The objectives were to study cultivar and year differences, and the relationships between parameters to find out the cultivars most suitable for rolled oats production and to understand how grain parameters are reflected on groat, rolled oat, and oatmeal characteristics. The cultivars and crop years differed (P < 0.05) in most of the parameters. In addition, many groat, rolled oats, and oatmeal variables showed a significant year by crop interaction. Several grain variables were significantly interrelated but only two, moisture and amount of dark and damaged grains, had a strong correlation (r > 0.70) to rolled oat parameters, the former correlating negatively to crude fat and the latter positively with maximum viscosity value. However, some weaker (r < 0.70) negative associations were found such as those between kernel size value and β‐glucan content and water binding capacity of rolled oats. In general, the variables analyzed at different processing stages correlated strongly, indicating good retention of groat properties during processing. Various associations were found between the physicochemical parameters of rolled oats and sensory properties of oatmeal. In principal component analysis, factor 1 mostly represented rolled oat parameters such as crude fat, color values L and a, and oatmeal parameters such as toasted aromatics, coarseness, and size of swollen particles. Factor 2 represented properties that pertain to structure and water and rolled oats interactions. The cultivars were also grouped according to these properties. The results suggest that the rolled oat process deserves optimization on a cultivar basis, particularly when specific product properties such as good water binding capacity are required. 相似文献

Pasting profiles of selected starches were compared by using a Micro Visco‐Amylo‐Graph (MVA) and a Rapid Visco Analyser (RVA). Effects of cooking (heating/cooling) rate and stirring speed on starch pasting properties were examined. The pasting viscosity of a starch suspension (8%, w/w, dsb) was measured at a fast (6°C/min) and slow (1.5°C/min) cooking rate while being stirred at either 75 rpm or 160 rpm. The pasting temperatures (PT) of all starches were higher when measured at the fast cooking rate than those at the slow cooking rate, except for wheat measured by using the RVA. PT was also higher when measured at the slow stirring speed (75 rpm) than at the fast stirring speed (160 rpm) in both RVA and MVA. When stirring speed increased from 75 rpm to 160 rpm, peak viscosity of all starch pastes except potato decreased measured by using the RVA, but increased by using the MVA. In general, amylograms of these starches obtained by using the MVA showed less breakdown, but greater setback viscosity than did that obtained by using the RVA. Differences in starch pasting properties between MVA and RVA, measured at the same cooking and stirring rates, were attributed mainly to the difference in spindle structure. 相似文献
