The color of Undaria pinnatifida after boiling is an important factor determining its marketable value. Our previous study showed that decreased nutrients and elevated irradiance resulted in increases of lightness and yellowness (i.e., discoloration) of this alga. However, little is known about the optimal levels of nutrients and irradiance required to decrease these color values and the combined effects of these factors and boiling. We conducted two culture experiments to test the effects of nutrients (non-enriched and 1.25, 5, and 25% PESI enriched treatments), irradiance (0, 10, 30, and 180 µmol m?2 s?1), and boiling on lightness L*, redness a*, and yellowness b* of this alga. L* and b* did not differ between non-enriched and 1.25–5% PESI treatments, but were lower in the 25% PESI treatment. L* and b* were lowest at 0–10 µmol m?2 s?1, although negative growth occurred at 0 µmol m?2 s?1. Decreased irradiance had a positive or little effect on a* before boiling, but had a negative effect after boiling. These results suggest that around 25% PESI and 10 µmol m?2 s?1 were the optimal levels to decrease the three color values of this species after boiling. 相似文献

To satisfy the world’s growing need for animal protein, it is important to develop or select fish species which are suitable for optimal cultivation in closed recirculating aquaculture systems (CRASs) as an environment-friendly aquaculture technology for providing safe and secure aquaculture food products. We have developed an ‘all fish’ growth-hormone (GH)-transgenic Nile tilapia Oreochromis niloticus (GHTi) which appears to be a predominant candidate for CRAS since it has been found to exhibit two important characteristics, namely, a shorter rearing duration to a market size and lowered N and P discharge load. However, these fish have undergone morphological changes, resulting in a short body which is less acceptable to consumers as an edible fish. One possible solution to reducing the prevalence of such morphological deformities is to determine the association between changes in the nutritional conditions of these fish. To this end, we first calculated the ideal total calcium (Ca) and phosphorous (P) intake (g/kg), ideal daily Ca and P intake and retention (g/kg/day) and ideal daily feeding rate (%) for the fourth filial (F4) offspring of GHTi. Our hypothesis was that GHTi would achieve the same total retention of Ca (14.6 g/kg) or P (7.69 g/kg) during a 69-day shorter rearing period as their production-matched pedigree common domestic strain (CTi). We estimated that when GHTi were fed conventional commercial diets to satiety, those with a body weight (BW) of more than approximately 230 g could not retain sufficient Ca, while those with a BW of more than approximately 429 g could not retain sufficient P. We then attempted to reduce the prevalence of morphological deformities in GHTi via dietary compound regulation by supplying a high amount of dietary Ca [6.82 % in the diet with Ca supplementation (Ca-S)] or P [4.75 % in the diet with P supplementation (P-S)]. Both the Ca-S and P-S diet improved mineral retention, while those fed on a Ca-S diet showed a lower prevalence of deformities, close to the size-matched CTi. These results suggest that the requirements of dietary Ca and P were significantly higher in GHTi than in CTi and, consequently, that the conventional levels of Ca and P in commercial diets for CTi cannot meet the mineral requirement of GHTi. We therefore propose to develop an optimal feed to meet the altered nutritional requirements of GH-transgenic Nile tilapia for growth-promoting optimization. ABSTRACT: Genetic polymorphisms were investigated to develop a simple and rapid method to differentiate between the two closely related species, Porphyra tenera and Porphyra yezoensis . Polymerase chain reaction (PCR) using the specific primer pair of the ARP4 gene gave length polymorphic single fragments of genomic DNAs from five strains of P. tenera (Japan T-8, JTW; Korea KTY1, KTY2, KTY3) and seven strains of P. yezoensis (Japan TU-1, TU-2, TUH-25, JHU, JA-1; Korea KGJ, KPH). All strains of P. yezoensis had introns 60 bp longer than that of P. tenera . Multiple cleaved amplified polymorphic sequence (CAPS) markers were also developed to differentiate P. tenera and P. yezoensis . This is the first report of length polymorphisms that can be used to differentiate between the two species using only PCR amplification with agarose gel electrophoresis. It is expected that the length polymorphism and plentiful CAPS profiles obtained in this study will be useful in the assessment of genetic diversity within P. tenera and P. yezoensis as well as in breeding science that requires the collection of various strains of the two species. 相似文献

Abstract Beets were grown on soils with various exchangeable sodium percentages (ESP). A saline non-sodic soil (SA, ESP = 3.2), a saline sodic soil (SO, ESP = 23), and a saline high sodic soil (HSO, ESP = 78) were prepared from Tottori sand dune soil (CO). K-type and Ca-type artificial zeolites (50 g kg?1) were applied to these soils in order to evaluate their effects on the chemical properties of saturation extracts of the soils, water deficit, cation uptake and transport, and cation balance of beet plants. In the zeolite-free treatments, beet growth was accelerated in SA and SO, but was suppressed in HSO compared with CO. The addition of both types of zeolites ameliorated plant growth in all the soils studied, especially HSO. The relative dry weight of the soils treated by the K-type zeolite to the zeolitefree soil was 189% for CO, 125% for SA, 130% for SO, and 222% for HSO. For the soils treated with the Ca-type zeolite, the values were 169, 116, 132, and 341%, respectively. In SA, SO, and HSO, the addition of the K-type zeolite increased the K uptake due to the increase of the K concentration of saturation extracts of soils. The addition of the Ca-type zeolite increased the Ca uptake due to the increase in the Ca concentration of the saturation extracts of soils which was accompanied by an increase in the K uptake. The increase in the uptake of K or Ca and decreased in the transport of Na by the addition of both types of zeolites improved the cation balance of the plants. The Ca-type zeolite did not increase the water deficit even though it increased the electric conductivity in all the soils. The results indicated that both types of artificial zeolites were able to improve the growth performance of beets in saline and sodic soils and that the K-type zeolite could be used as a K-fertilizer as well. 相似文献

We evaluated the ability of Cry1Aa9, Cry1Ab4, and Cry1Ac1 insecticidal toxins from Bacillus thuringiensis to destroy liposomes. Cry1A toxins are thought to form pores in midgut apical cell membranes (BBMV), thereby disrupting midgut cells. Liposomes containing fluorescent calcein were prepared using phosphatidylcholine (PC) and phosphatidylserine (PS) (PC/PS-Liposomes) or PC alone (PC-Liposomes). Cry1Ab (1.4 μM), but not Cry1Aa or Cry1Ac, disrupted PC/PS-Liposomes and PC-Liposomes. PC/PS-Liposomes containing cholesterol and oligosaccharylceramide from Plutella xylostella midgut were damaged even more extensively by Cry1Ab, but the inclusion of either lipid alone had no effect. The initial velocity of Cry1Ab-mediated liposome disruption increased 17-fold when liposomes were prepared with Triton X-100-soluble proteins from Bombyx mori BBMV and PC (PC/Proteo-Liposomes), and Cry1Aa and Cry1Ac also caused slight disruption. These data suggest that Cry1Ab achieves higher penetration into PC/PS-Liposomes, PC-Liposomes, and PC/Proteo-Liposomes compared with Cry1Aa or Cry1Ac and that Cry1Ab may interact with membrane proteins. 相似文献

Spontaneous renal tubule tumors (RTTs), with a distinctive morphological phenotype, were
present in three Sprague-Dawley rats, 1 male and 2 females, out a total of 120 animals of each
sex from untreated and placebo control groups in a 2-year carcinogenicity study. One female had
one carcinoma, adenoma and hyperplasia, and the other female had five adenomas and many
hyperplastic lesions; the male case had one carcinoma. From these cases, a biological continuum
of hyperplasia, adenoma and carcinoma could be recognized. The tumors were present in the renal
cortex and appeared as solid lobulated growths with occasional central necrosis. The lobules
were divided by a small amount of fibrovascular tissue. Occasionally the larger tumors
contained a cystic area. Tumor cells appeared distinctive and exhibited variable amounts of
eosinophilic/amphophilic and vacuolated cytoplasm. Nuclei were round to oval with a prominent
nucleolus. Mitotic figures were uncommon, and no distant metastasis was noted. The tumors were
seen as multiple and bilateral lesions in two animals and had no apparent relationship to
chronic progressive nephropathy (CPN). Foci of tubule hyperplasia were also noted to contain
the same type of cellular morphology. The morphological and biological features of these 3
cases resembled the amphophilic-vacuolar (AV) variant of RTT that has been posited to be of
familial origin. This is a report of spontaneous familial renal tumors in Sprague-Dawley rats
