Bovine foamy virus (BFV) is distributed through worldwide cattle herds. Although the biological features of BFV are not well understood, appearance of clinical manifestation by superinfection with other microorganisms is inferred. In Japan, reports of genomic characterizations and epidemiology of this virus are limited. In this study, we performed whole genomic sequencing of BFV strains Ibaraki and No.43, which were isolated in this country. Additionally, we investigated BFV in geographically distant four daily farms in Japan, to estimate the distribution of BFV and its correlation to bovine leukemia virus (BLV). BFV was distributed throughout Japan; the average positive rate was 12.7%. The nucleotide sequence identities of the isolates were 99.6% when compared with BFV strain isolated in the USA. The phylogenetic tree using env gene sequence showed strains Ibaraki, No.43 and Kagoshima were sorted in the same cluster including the USA and Chinese strains, while Hokkaido strain was in the other cluster including European strains. Although no clear correlation between BFV and BLV could be found, BFV and BLV infections were likely to increase with ages. Our data on epidemiology and characteristics of BFV will provide important information to reveal biological features of BFV. 相似文献

Mammalian sexual fate is determined by the presence or absence of sex determining
region of the Y chromosome (Sry) in the “bipotential” gonads.
Recent studies have demonstrated that both male and female sexual development are induced
by distinct and active genetic pathways. Breeding the Y chromosome from Mus m.
domesticus poschiavinus (POS) strains into C57BL/6J (B6J) mice
(B6J-XYPOS) has been shown to induce sex reversal (75%: bilateral ovary, 25%:
true hermaphrodites). However, our B6N-XYPOS mice, which were generated by
backcrossing of B6J-XYPOS on an inbred B6N-XX, develop as males (36%: bilateral
testis with fertility as well as bilateral ovary (34%), and the remainder develop as true
hermaphrodites. Here, we investigated in detail the expressions of essential sex-related
genes and histological features in B6N-XYPOS mice from the fetal period to
adulthood. The onsets of both Sry and SRY-box 9 (Sox9) expressions as determined
spatiotemporally by whole-mount immunohistochemistry in the B6N-XYPOS gonads
occurred 2–3 tail somites later than those in B6N-XYB6 gonads, but earlier than
those in B6J-XYPOS, respectively. It is possible that such a small difference
in timing of the Sry expression underlies testicular development in our
B6N-XYPOS. Our study is the first to histologically show the expression and
ectopic localization of a female-related gene in the XYPOS testes and a
male-related gene in the XYPOS ovaries. The results from these and previous
experiments indicate that the interplay between genome variants, epigenetics and
developmental gene regulation is crucial for testis development. 相似文献

The present study evaluated the effects of single-dose filgrastim on hematology in 16
healthy horses after long-distance transportation. Horses were assigned to receive
filgrastim (0.23 µg/kg, SC, once; G-CSF group; n=8) or saline (0.9% NaCl)
solution (0.3 ml, SC, once; control group; n=8) ≤ 1 hr before
transportation. Horses were transported 2,530 km using commercial vans over the course of
approximately 44 hr. Clinical examinations and hematologic analyses were performed on all
horses before and after transportation. Because the post-transportation white blood cell
counts and bacillary neutrophil to segmented neutrophil ratio were significantly higher in
the G-CSF group, filgrastim may have promoted the mobilization of neutrophils from marrow.
Filgrastim deserves a further study for efficacy in preventing horse shipping fever. 相似文献

We investigated the effect of supplemental vitamin E on the peripheral blood leukocyte population in Japanese Black calves. Twenty-six calves kept at the same farm were studied. They were divided into two groups; thirteen calves received 300 IU/day of vitamin E orally from 1 to 3 months of age (VE group), and the other thirteen calves did not receive the vitamin E supplement (control group). The VE group showed a higher serum vitamin E concentration at 2 and 3 months of age compared with the control group (P<0.01). The numbers of CD3+ cells and CD4+ cells were higher in the VE group than in the control group, and the difference was statistically significant at 3 months of age (P<0.05). The numbers of CD21+ cells were higher in the VE group than in the control group, and the difference was statistically significant at 2 months of age (P<0.05). The numbers of CD335+ cells
tended to be higher in the VE group than in the control group. The numbers of CD8+ cells and CD14+ cells tended to be higher in the VE group than in the control group at 3 and 4 months of age. This study demonstrated that the supplementation of suckling Japanese Black calves with vitamin E might affect the numbers of some immune cell types in the peripheral blood. 相似文献

Transitional cell carcinoma
(TCC), a urinary bladder tumor with high mortality, is encountered commonly in dogs.
Whereas overexpression of epidermal growth factor receptor (EGFR) is associated with
development of human urinary bladder cancer, information on EGFR expression in canine TCC
is lacking. In this study, EGFR protein and mRNA expression in canine normal bladder
(n=5), polypoid cystitis (n=5) and TCC (n=25) were examined by immunohistochemistry and
real-time polymerase chain reaction. EGFR protein expression was significantly higher in
TCC than that in normal healthy bladder (P<0.001) and polypoid
cystitis (P<0.005). High EGFR protein expression was significantly
(P<0.01) associated with TCC with a sensitivity of 72% and
specificity of 100%. Comparative analysis of protein and mRNA expression levels in TCC
showed significant positive correlation (r=0.88, P<0.05) between mRNA
and protein expression. These findings suggest that intense expression of EGFR protein
could be used as a marker to help canine TCC diagnosis. 相似文献

In the mammalian testis, the ubiquitin-proteasome system plays important roles in the process that promotes the formation of
mature sperm. We recently identified zygote-specific proteasome
assembly phaperone (ZPAC), which is specifically expressed in the mouse
gonads and zygote. ZPAC mediates a unique proteasome assembly pathway in the zygote, but the expression profile and function
of ZPAC in the testis is not fully understood. In this study, we investigated the possible role of ZPAC during mouse
spermatogenesis. First, we analyzed the expression of ZPAC and 20S proteasome subunit α4/PSMA7 in the adult mouse testis.
ZPAC and α4 were expressed in spermatogonia, spermatocytes, and round spermatids. In elongating spermatids, ZPAC was
expressed until step 10, whereas expression of α4 persisted until step 12. We then examined the expression profile of ZPAC
and α4 in a mouse model of experimental unilateral cryptorchidism. Consistent with appearance of morphologically impaired
germ cells following cryptorchidism, the ZPAC protein level was significantly decreased at 4 days post induction of
experimental cryptorchidism (D4) compared with the intact testis, although the amount of α4 protein persisted at least until
D10. Moreover, intense ZPAC staining was co-localized with staining of annexin V, an early indicator of apoptosis in
mammalian cells, in germ cells of cryptorchid testis, but ZPAC was also expressed in germ cells showing no detectable
expression of annexin V. These results suggest that ZPAC plays a role during spermatogenesis and raises the possibility that
20S proteasome mediated by ZPAC may be involved in the regulation of germ cell survival during spermatogenesis. 相似文献

Antimicrobial activities of 139 Enterococcus isolates (48 E. faecium and 91 E. faecalis) obtained from canine feces, boiler meat samples, swine feces, wild waterfowl feces, and human feces were examined against respective bacteria, including Streptococcus pyogenes, Staphylococcus aureus, Bacillus subtilis, Listeria monocytogenes, Salmonella Enteritidis, and Escherichia coli. Bacteriocin (BAC) production assay revealed that the antimicrobial activity against at least one of 6 indicator strains (BAC+ phenotype) was found in 51 (37%) isolates (29 E. faecium and 22 E. faecalis). Twenty-four of 46 isolates positive for at least one of the enterocin structural genes (entA, entB, entL50AB, and cylL) showed a BAC+ phenotype. The existence of other enterocins or nonenterocin factors was implied because the BAC+ phenotype was detected in a total of 27 Enterococcus isolates that had none of the enterocin genes tested. The antimicrobial activity against Gram-negative strains (Salmonella Enteritidis and E. coli) was detected in the 6 Enterococcus isolates that had either the entA, entB, entL50AB or cylL genes. Moreover, the proportion of the antimicrobial activity against L. monocytogenes among the cylL-positive E. faecalis isolates showing beta-hemolysis (10/16) was significantly (p<0.01) higher than among those lacking beta-hemolysis (2/15). The results suggested that certain characteristics are likely to be associated with the antimicrobial activity against specific organisms. 相似文献

The relay intercropping system for cultivating direct-seeding rice and winter cereal is a low-cost method particularly for double cropping, because it eliminates the steps of raising seedlings, paddling, and transplanting. However, in this system, the seedling establishment rate (SER) of rice is low and unstable. The objective of this study was to identify the factors affecting SER to highlight the ways to improve SER. Experiments were conducted in experimental fields in Tsukuba City, Ibaraki Prefecture in 2015 and 2016. To determine the time of rice seed death and to calculate seed survival rates, ‘Akidawara’ seeds were embedded in soil (depth: 3 cm) and dug up after defined periods. We then analyzed the correlation between SER and meteorological factors at two sites (Ibaraki Prefecture and Gunma Prefecture) over the same two years. Based on mean air temperature (MT), we divided the period from seeding to June 20 (when irrigation had been initiated in almost all the fields) into four phases. In Phase 1, the number of days with rainfall (≥5 mm) and soil-wetting days (water potential ≥ ?100 kPa) were significantly and negatively correlated with SER. We found that most of the seed deaths occurred after germination, and the germination rates were presumably affected by water absorption during the low temperature phase. Further investigations are needed to understand the occurrences during the seedling emergence period. The reproductive performance of dairy cows has declined, along with an increase in their milk yield. First-service conception rates in lactating dairy cows are often lower than 50%. The precise detection of estrus is an important factor in the reproductive management of dairy cows for successful fertilization and pregnancy. However, estrus expression has been decreasing in modern dairy cows, affecting the detection rate of estrus. In addition to estrus, a high incidence of ovulation disorders affects the fertility of dairy cows. To address these problems, it is necessary to understand the changes in the endocrine functions that underlie estrus and ovulation disorders, and to develop effective treatment strategies. Recent studies have revealed that neurokinin B and neurokinin 3 receptor signaling play important roles in the regulation of the secretion of gonadotropin-releasing and luteinizing hormones, suggesting a potential clinical avenue for the stimulation of gonadal function. In this review, I have discussed the problems in estrus and ovulation disorders in modern dairy cows as well as the possible applications of neurokinin 3 receptor agonists in the treatment of these disorders. 相似文献
