This study investigated the hypothesis that feeding broilers over-processed meat and bone meal (MBM) would impair gut health in the absence of phytase and in turn, affect inositol phosphate (inositol x-phosphate, IPx: IP3, IP4, IP5 and IP6) ester hydrolysis, intestinal permeability, hematology, jejunal gene expression and intestinal morphology during necrotic enteritis (NE). Ross 308 male broilers (n = 768) were assigned to one of 8 dietary treatments in a 2 × 2 × 2 factorial arrangement, with 6 replicate pens per diet and 16 birds per pen in a completely randomized design. Factors were: NE challenge (no or yes), phytase level (500 or 5,000 FTU/kg) and MBM processing (as-received or over-processed). For the NE challenge, half of the birds were challenged with field strains of Eimeria spp. on d 9 and 108 CFU/mL of Clostridium perfringens strain EHE-NE18 on d 14 and 15. A 3-way challenge, phytase and MBM processing interaction was detected for IP5 (P < 0.05) and IP6 (P < 0.05) levels in the ileum. Birds fed low phytase had increased IP5 and IP6 in unchallenged birds only when diets contained over-processed MBM. Challenge with NE increased intestinal permeability as measured by serum fluorescein isothiocyanate dextran (FITC-d; P < 0.001), increased white blood cells (WBC; P < 0.001), decreased mean corpuscular volume (MCV; P < 0.001) and mean corpuscular hemoglobin (MCH; P < 0.05), and decreased crypt-to-villi ratio (P < 0.05). The over-processed MBM reduced the villi-to-crypt ratio (P < 0.05). A 3-way challenge × phytase × MBM processing interaction was detected for mucin 2 (MUC-2) expression (P < 0.05) where only in unchallenged birds fed over-processed MBM did high phytase reduce MUC-2 expression. A lower expression of aminopeptidase N (APN; P < 0.001) and vitamin D receptor (VDR; P < 0.001) were recorded in NE challenged birds. In conclusion, NE has a negative impact on the gut and hematology of broilers, but its effect on phytate hydrolysis is minimal.  相似文献

The goal of this study was to assess the feasibility of across-country genomic predictions in Norwegian White Sheep (NWS) and New Zealand Composite (NZC) sheep populations with similar development history. Different training populations were evaluated (i.e., including only NWS or NZC, or combining both populations). Predictions were performed using the actual phenotypes (normalized) and the single-step GBLUP via Bayesian inference. Genotyped NWS animals born in 2016 (N = 267) were used to assess the accuracy and bias of genomic estimated breeding values (GEBVs) predicted for birth weight (BW), weaning weight (WW), carcass weight (CW), EUROP carcass classification (EUC), and EUROP fat grading (EUF). The accuracy and bias of GEBVs differed across traits and training population used. For instance, the GEBV accuracies ranged from 0.13 (BW) to 0.44 (EUC) for GEBVs predicted including only NWS, from 0.06 (BW) to 0.15 (CW) when including only NZC, and from 0.10 (BW) to 0.41 (EUC) when including both NWS and NZC animals in the training population. The regression coefficients used to assess the spread of GEBVs (bias) ranged from 0.26 (BW) to 0.64 (EUF) for only NWS, 0.10 (EUC) to 0.52 (CW) for only NZC, and from 0.42 (WW) to 2.23 (EUC) for both NWS and NZC in the training population. Our findings suggest that across-country genomic predictions based on ssGBLUP might be possible for NWS and NZC, especially for novel traits.  相似文献

ObjectiveTo assess the potential of a thermal carbon dioxide (CO2) laser to explore antinociception in pain-free cats.Study designExperimental, prospective, blinded, randomized study.AnimalsSixty healthy adult female cats with a (mean ± standard deviation) weight of 3.3 ± 0.6 kg.MethodsCats were systematically allocated to one of six treatments: saline 0.2 mL per cat; morphine 0.5 mg kg−1; buprenorphine 20 μg kg−1; medetomidine 2 μg kg−1; tramadol 2 mg kg−1, and ketoprofen 2 mg kg−1. Latency to respond to thermal stimulation was assessed at baseline and at intervals of 15–30, 30–45, 45–60, 60–75, 90–105 and 120–135 minutes. Thermal thresholds were assessed using time to respond behaviourally to stimulation with a 500 mW CO2 laser. Within-treatment differences in response latency were assessed using Friedman’s test. Differences amongst treatments were assessed using independent Kruskal–Wallis tests. Where significant effects were identified, pairwise comparisons were conducted to elucidate the direction of the effect.ResultsCats treated with morphine (X2 = 12.90, df = 6, p = 0.045) and tramadol (X2 = 20.28, df = 6, p = 0.002) showed significant increases in latency to respond. However, subsequent pairwise comparisons indicated that differences in latencies at specific time-points were significant (p < 0.05) only for tramadol at 60–75 and 90–105 minutes after administration (21.9 and 43.6 seconds, respectively) in comparison with baseline (11.0 seconds). No significant pairwise comparisons were found within the morphine treatment. Injections of saline, ketoprofen, medetomidine or buprenorphine showed no significant effect on latency to respond.Conclusions and clinical relevanceThe CO2 laser technique may have utility in the assessment of thermal nociceptive thresholds in pain-free cats after analgesic administration and may provide a simpler alternative to existing systems. Further exploration is required to examine its sensitivity and comparative utility.  相似文献

ObjectiveTo describe the effects of presurgical preparation with an isopropyl alcohol or water rinse on the perioperative rectal temperature (RT) of puppies and kittens.Study designRandomized clinical trial.AnimalsA total of 48 intact female mixed breed puppies and 43 intact female Domestic Short Hair kittens aged 8–18 weeks.MethodsAll animals were premedicated with intramuscular buprenorphine (0.02 mg kg–1) and acepromazine (0.05 mg kg–1). Anesthesia was induced with intravenous propofol (4 mg kg–1 to effect) for puppies or ketamine (5 mg kg–1) and midazolam (0.25 mg kg–1) for kittens. RT was measured every minute for the first 15 minutes at the beginning of hair/fur removal, then every 5 minutes for 45 minutes (dogs) and 35 minutes (cats). All animals were prepared for surgery using a 1.6% chlorhexidine solution, then rinsed with either isopropyl alcohol (group CA) or water (group CW).ResultsMean RT difference between the groups was not significant at any time point. The mean RT at 45 minutes for dogs was 35.9 °C and 36.0 °C in groups CA and CW, respectively (p = 0.74). The mean RT at 35 minutes for cats was 35.1 °C in both groups (p = 0.84).Conclusions and clinical relevanceThe use of either water or alcohol as a rinsing agent results in the same degree of perioperative temperature change. Other factors that contribute to perioperative hypothermia should be considered when choosing between these rinsing agents in surgical preparation of pediatric and small animals.  相似文献

Two studies were conducted to investigate the effect of Bacillus amyloliquefaciens CECT 5940 (BA) as a probiotic on growth performance, amino acid digestibility and bacteria population in broiler chickens under a subclinical necrotic enteritis (NE) challenge and/or fed diets with different levels of crude protein (CP). Both studies consisted of a 2 × 2 factorial arrangement of treatments with 480 Ross 308 mix-sexed broiler chickens. In study 1, treatments included 1) NE challenge (+/−), and 2) BA (1.0 × 106 CFU/g of feed) supplementation (+/−). In study 2, all birds were under NE challenge, and treatments were 1) CP level (Standard/Reduced [2% less than standard]) and 2) BA (1.0 × 106 CFU/g of feed) supplementation (+/−). After inducing NE infection, blood samples were taken on d 16 for uric acid evaluation, and cecal samples were collected for bacterial enumeration. In both studies, ileal digesta was collected on d 35 for nutrient digestibility evaluation. In study 1, the NE challenge reduced body weight gain (BWG), supressed feed conversion ratio (FCR) and serum uric acid levels (P < 0.001). Supplementation of BA increased BWG (P < 0.001) and reduced FCR (P = 0.043) across dietary treatments, regardless of challenge. Bacillus (P = 0.030) and Ruminococcus (P = 0.029) genomic DNA copy numbers and concentration of butyrate (P = 0.017) were higher in birds fed the diets supplemented with BA. In study 2, reduced protein (RCP) diets decreased BWG (P = 0.010) and uric acid levels in serum (P < 0.001). Supplementation of BA improved BWG (P = 0.001) and FCR (P = 0.005) and increased Ruminococcus numbers (P = 0.018) and butyrate concentration (P = 0.033) in the ceca, regardless of dietary CP level. Further, addition of BA reduced Clostridium perfringens numbers only in birds fed with RCP diets (P = 0.039). At d 35, BA supplemented diets showed higher apparent ileal digestibility of cystine (P = 0.013), valine (P = 0.020), and lysine (P = 0.014). In conclusion, this study suggests positive effects of BA supplementation in broiler diets via modulating gut microflora and improving nutrient uptake.  相似文献

Biochar application to arable soils could be effective for soil C sequestration and mitigation of greenhouse gas (GHG) emissions. Soil microorganisms and fauna are the major contributors to GHG emissions from soil, but their interactions with biochar are poorly understood. We investigated the effects of biochar and its interaction with earthworms on soil microbial activity, abundance, and community composition in an incubation experiment with an arable soil with and without N-rich litter addition. After 37 days of incubation, biochar significantly reduced CO2 (up to 43 %) and N2O (up to 42 %), as well as NH4 +-N and NO3 ?-N concentrations, compared to the control soils. Concurrently, in the treatments with litter, biochar increased microbial biomass and the soil microbial community composition shifted to higher fungal-to-bacterial ratios. Without litter, all microbial groups were positively affected by biochar × earthworm interactions suggesting better living conditions for soil microorganisms in biochar-containing cast aggregates after the earthworm gut passage. However, assimilation of biochar-C by earthworms was negligible, indicating no direct benefit for the earthworms from biochar uptake. Biochar strongly reduced the metabolic quotient qCO2 and suppressed the degradation of native SOC, resulting in large negative priming effects (up to 68 %). We conclude that the biochar amendment altered microbial activity, abundance, and community composition, inducing a more efficient microbial community with reduced emissions of CO2 and N2O. Earthworms affected soil microorganisms only in the presence of biochar, highlighting the need for further research on the interactions of biochar with soil fauna.  相似文献

The net effect of anthropogenic aerosols on climate is usually considered the sum of the direct radiative effect of anthropogenic aerosols, plus the indirect effect of these aerosols through aerosol-cloud interactions. However, an additional impact of aerosols on a longer time scale is their indirect effect on climate through biogeochemical feedbacks, largely due to changes in the atmospheric concentration of CO(2). Aerosols can affect land and ocean biogeochemical cycles by physical forcing or by adding nutrients and pollutants to ecosystems. The net biogeochemical effect of aerosols is estimated to be equivalent to a radiative forcing of -0.5 ± 0.4 watts per square meter, which suggests that reaching lower carbon targets will be even costlier than previously estimated.  相似文献

Assessing the impact of future anthropogenic carbon emissions is currently impeded by uncertainties in our knowledge of equilibrium climate sensitivity to atmospheric carbon dioxide doubling. Previous studies suggest 3 kelvin (K) as the best estimate, 2 to 4.5 K as the 66% probability range, and nonzero probabilities for much higher values, the latter implying a small chance of high-impact climate changes that would be difficult to avoid. Here, combining extensive sea and land surface temperature reconstructions from the Last Glacial Maximum with climate model simulations, we estimate a lower median (2.3 K) and reduced uncertainty (1.7 to 2.6 K as the 66% probability range, which can be widened using alternate assumptions or data subsets). Assuming that paleoclimatic constraints apply to the future, as predicted by our model, these results imply a lower probability of imminent extreme climatic change than previously thought.  相似文献
