Green fluorescent protein (GFP)-marked Fusarium oxysporum f. sp. melonis and nonmarked F. oxysporum f. sp. fragariae were stained with neutral red. The neutral red stained vacuoles of the fungi without disturbing GFP fluorescence in the cytoplasm.
GFP-marked fungi showed fluorescent hyphae with dark-stained vacuoles, whereas nonmarked fungi were detected as nonfluorescent
hyphae with dark-dotted vacuoles. Root colonization by these two fungi was monitored using this method. Microconidia attached
similarly to the root surface and elongated vegetative hyphae. Only the pathogenic fungi invaded, causing necrosis at the
inoculation site. Thus, the present method enabled us to track simultaneously the various formae speciales of F. oxysporum colonizing the root surface.

Extractive components were determined for the mantle muscle and liver of three species of Loliginidae squid, Sepioteuthis lessoniana, Loligo bleekeri, and L. edulis, and one species of Ommastrephidae, Todarodes pacificus, as a control. Total free amino acid levels and the major free amino acids in muscle, taurine, proline, glycine, alanine,
and arginine, were significantly higher in Loliginidae squids than in T. pacificus. The main nucleotide was adenosine 5′-monophosphate, which did not differ significantly between species. Malate was the organic
acid found in muscle in highest concentrations. The muscle of these species contained a large amount of trimethylamine oxide.
A large amount of glycine betaine was also detected in the muscle, but showed no large species difference. From these results,
the muscle of Loliginidae species is considered to have a much sweeter taste than that of T. pacificus. Compared with muscle, squid liver was characteristic with high contents of taurine, glutamate, bitter amino acids, succinate,
propionate, trimethylamine, and glycine betaine, and with low contents of sweet amino acids, arginine, nucleotides, malate,
and trimethylamine oxide. These results suggest that squid liver is characterized by a complicated taste containing umami,
bitterness, sourness, fishy flavor, and less sweetness. 相似文献

Japanese cypress (Chamaecyparis obtusa) is one of the most common plantation species in Japan. In most Japanese cypress plantations, closure of the forest canopy hinders understory growth because of insufficient management practices. Thus leaf litter barely covers the soil surface. Such plantations are vulnerable to surface erosion triggered by rainfalls, and could yield large amounts of suspended sediment (SS). However, few studies have investigated the annual SS yield. This study aimed to develop a modified model of SS yields, and to accurately estimate and characterize the annual SS yield from a Japanese cypress plantation. For this, hydrological surveys were conducted for 5 years, and stream water was sampled weekly as well as sequentially at 15–60 min intervals in each of 14 rain events in a small forested watershed of Japanese cypress. The root mean square error (RMSE) and the annual SS yield estimates were obtained using three different equations: the rating curve derived from instantaneous SS concentration versus stream discharge relationship (SSC − Q equation); the rating curve derived from cumulative SS yield versus cumulative specific discharge relationship (∑SS − ∑Q equation); and the multiple regression of cumulative SS yield expressed as functions of cumulative specific discharge and antecedent rainfalls (∑SS − ∑Q × AR equation). The ∑SS − ∑Q × AR equation is a modified model considering the effects of antecedent rain history on the SS yield. The parameters in each equation were determined by a nonlinear least square method. Of the three equations, the RMSE was the highest using the SSC − Q equation. Although the ∑SS − ∑Q equation and the ∑SS − ∑Q × AR equation gave similar RMSE values, the SS yields extrapolated by the ∑SS − ∑Q equation would be overestimated. Thus, the 5-year average of annual SS yield estimated by the ∑SS − ∑Q equation was three times larger than that estimated by the ∑SS − ∑Q × AR equation. These results indicate that the ∑SS − ∑Q × AR equation more accurately estimates the annual SS yield, compared with the other two equations. The results also suggest that antecedent rain history, as well as stream discharge, should be considered for appropriately estimating the annual SS yield. The annual SS yield estimated by the ∑SS − ∑Q × AR equation was the largest among 25 undisturbed forested watersheds, except for two watersheds dominated by sandstone. Zuiki, a stalk of taro (Colocasia esculenta), is a traditional vegetable in Japan. Raw zuiki is often boiled and vinegared to eat. The surface color of zuiki is reddish. Here, we isolated a red pigment from zuiki and identified it as cyanidin 3-rutinoside using instrumental analyses. The color of zuiki disappeared by boiling, but the zuiki turned red again in an acetic acid solution. It seems that the cyanidin 3-rutinoside that exists on the surface of zuiki elutes in boiling water and then, the pigment that seeps out from the inside of the zuiki is exposed to an acid solution, and its surface turns red again. The radical scavenging activity of purified zuiki anthocyanin was 114 mg equivalent to BHT/g. About half of the anthocyanin in fresh zuiki was washed out by boiling, and the radical scavenging activity of zuiki was definitely reduced. 相似文献

Four new abscisic acid related compounds (1-4), together with (+)-abscisic acid (5), (+)-beta-D-glucopyranosyl abscisate (6), (6S,9R)-roseoside (7), and two lignan glucosides ((+)-pinoresinol mono-beta-D-glucopyranoside (8) and 3-(beta-D-glucopyranosyloxymethyl)-2- (4-hydroxy-3-methoxyphenyl)-5-(3-hydroxypropyl)-7-methoxy-(2R,3S)-dihydrobenzofuran (9)) were isolated from the antioxidative ethanol extract of prunes (Prunus domestica L.). The structures of 1-4 were elucidated on the basis of NMR and MS spectrometric data to be rel-5-(3S,8S-dihydroxy-1R,5S-dimethyl-7-oxa-6-oxobicyclo[3,2,1]oct-8-yl)-3-methyl-2Z,4E-pentadienoic acid (1), rel-5-(3S,8S-dihydroxy-1R,5S-dimethyl-7-oxa-6-oxobicyclo[3,2,1]oct-8-yl)-3-methyl-2Z,4E-pentadienoic acid 3'-O-beta-d-glucopyranoside (2), rel-5-(1R,5S-dimethyl-3R,4R,8S-trihydroxy-7-oxa-6-oxobicyclo[3,2,1]oct-8-yl)-3-methyl-2Z,4E-pentadienoic acid (3), and rel-5-(1R,5S-dimethyl-3R,4R,8S-trihydroxy-7-oxabicyclo[3,2,1]- oct-8-yl)-3-methyl-2Z,4E-pentadienoic acid (4). The antioxidant activities of these isolated compounds were evaluated on the basis of oxygen radical absorbance capacity (ORAC). The ORAC values of abscisic acid related compounds (1-7) were very low. Two lignans (8 and 9) were more effective antioxidants whose ORAC values were 1.09 and 2.33 micromol of Trolox equiv/micromol, respectively. 相似文献
