Cellular signal transduction pathways modify gene expression programs in response to changes in the environment, but the mechanisms by which these pathways regulate populations of genes under their control are not entirely understood. We present evidence that most mitogen-activated protein kinases and protein kinase A subunits become physically associated with the genes that they regulate in the yeast (Saccharomyces cerevisiae) genome. The ability to detect this interaction of signaling kinases with target genes can be used to more precisely and comprehensively map the regulatory circuitry that eukaryotic cells use to respond to their environment. 相似文献

Summary In earlier work on improvement of persistance in forage legumes, we selected genotypes from highly productive cultivars of alfalfa, Algonquin and Apica (Euphytica 45: 105–112, 1990) and cv. Florex red clover (Plant Cell Reports 8: 395–398, 1989) capable of in vitro regeneration from callus and cell culture. The alfalfa germplasm and its F1 progeny as well as an F2 red clover population were tested for cold stress tolerance. Plantlets were hardened in culture tubes at 2 or 5°C, 8h photoperiod, for at least four weeks and then subjected to freezing temperatures, –16 or –10°C for alfalfa and red clover, respectively. Survival of regenerative genotypes was significantly higher than of the non-regenerative ones in both species. A strong oositive correlation (r=0.78) between the regenerative trait and plant survival was found in alfalfa. The experiments indicate that in vitro selection for regenerative trait may improve cold stress tolerance of alfalfa and red clover. 相似文献

The pandemic influenza virus of 1918-1919 killed an estimated 20 to 50 million people worldwide. With the recent availability of the complete 1918 influenza virus coding sequence, we used reverse genetics to generate an influenza virus bearing all eight gene segments of the pandemic virus to study the properties associated with its extraordinary virulence. In stark contrast to contemporary human influenza H1N1 viruses, the 1918 pandemic virus had the ability to replicate in the absence of trypsin, caused death in mice and embryonated chicken eggs, and displayed a high-growth phenotype in human bronchial epithelial cells. Moreover, the coordinated expression of the 1918 virus genes most certainly confers the unique high-virulence phenotype observed with this pandemic virus. 相似文献

The objective of this research was to develop a methodology to describe the movement of the coffee berry borer Hypothenemus hampei (Coleoptera: Curculionidae) in the field through: (i) the evaluation of allele variation of a microsatellite marker on polymorphic Colombian H. hampei populations; (ii) the invention of a device for releasing H. hampei adults; (iii) the standardization of a release-recapture technique for H. hampei populations; (iv) the estimation of the flight distance of the insect; and (v) the calculation of a mathematical expression that describes the movement of H. hampei in space over time. The results indicated that: (i) the microsatellite molecular marker HHK.1.6 was exclusively present in a population from Guapotá-Santander, was dominant and allows the evaluation of H. hampei movement for several generations; (ii) a device that released 88.8% of H. hampei adults in 2 s was designed; (iii) this device was used as H. hampei populations containing HHK.1.6 marker release strategy, and coffee seeds as recapture strategy; (iv) it was estimated that H. hampei adults flew as far as 65 m, however, 90% were recovered in a radius of <40 m. Finally, (v) the mathematical expression that described the movement of H. hampei in space over time was , being the average number of borer beetles recaptured per tree, and x the distance in meters. Push–pull involves intercropping of cereals with Desmodium as a “push” crop and planting Napier grass/Brachiaria as the “pull” crop at the border. The technology has been reported to effectively control stemborers, striga weed, and fall armyworm (FAW), and to improve soil nutrition, resulting in increased grain yield. This study evaluated the impact of stemborer and FAW management using this technology on incidence of maize ear rots and preharvest contamination of grains with aflatoxin and fumonisin in western Kenya. The study was conducted during three cropping seasons on maize grown under the push–pull system and as a monocrop. Incidence of stemborer and FAW damage was determined as percentage of damaged plants, while incidence of ear rots was determined as percentage of ears with symptoms. At harvest, fungi were isolated from kernels and aflatoxin and fumonisin were quantified using enzyme-linked immunosorbent assay. Stemborer and FAW damage was significantly (p = .001) reduced by over 50% under the push–pull system. There was also a significant (p < .001) reduction in the incidence of Fusarium verticillioides (60%) and Aspergillus flavus (86%), which was reflected in a reduced incidence of ear rots (50%) with the push–pull system (p = .001). Fumonisin in maize from push-pull farms was significantly (p = .048) reduced (39%) but the technology had no significant (p > .05) effect on aflatoxin. The study showed that push–pull is an effective strategy for managing maize ear rots and fumonisins, and therefore could play a role in improving food safety among smallholder maize farmers in the region. 相似文献

Planting three rows of potatoes in a bed the width of two conventional rows offers an easily managed way to increase seed piece populations, with the potential of increasing tuber yield and enhancing tuber quality. A wide bed production system (3 rows of potatoes planted on a 1.9 m flat-topped raised bed) was compared to a conventional-ridged system (1 row of potatoes in sharply sloped ridges on 96 cm centers) in 1996 and 1997 on a Norfolk sandy loam soil and a Portsmouth fine sandy loam soil in eastern North Carolina. Potato plant stands, leaf area index at approximately 9 WAP, yield, and quality were measured. Soil temperature, soil moisture, and cone index, as a measure of soil penetration resistance, were also measured, wide beds were more moist than conventional ridges early in the season. Cone index was greater throughout the root profile in wide beds in two of three tests. The row on the west side of an individual wide bed was most similar to conventional ridges in daily soil temperature fluctuations between minimum and maximum temperatures, and had greater fluctuations than the middle and eastern rows of the wide bed. Total yield and yield of grade A potatoes were not significantly different between wide beds and conventional ridges at either site. At one site, yield of grade B potatoes was significantly less in the wide bed; among the three rows in the wide bed, the eastern row had significantly lower yield of grade B potatoes. Conventional ridges had a higher percent of green grade A potatoes than the wide beds in one of three trials. Under North Carolina conditions, changing production systems would be unadvisable for most growers because wide beds do not increase yield enough to justify spending the money for more seed and to change equipment. 相似文献

Highly pathogenic avian influenza (HPAI) is a devastating viral disease of poultry and quick control of outbreaks is vital. Airborne transmission has often been suggested as a route of transmission between flocks, but knowledge of the rate of transmission via this route is sparse. In the current study, we quantified the rate of airborne transmission of an HPAI H5N1 virus strain between chickens under experimental conditions. In addition, we quantified viral load in air and dust samples. Sixteen trials were done, comprising a total of 160 chickens housed in cages, with three treatment groups. The first group was inoculated with strain A/turkey/Turkey/1/2005 H5N1, the second and third group were not inoculated, but housed at 0.2 and 1.1m distance of the first group, respectively. Tracheal and cloacal swabs were collected daily of each chicken to monitor virus transmission. Air and dust samples were taken daily to quantify virus load in the immediate surroundings of the birds. Samples were tested by quantitative RRT-PCR and virus isolation. In 4 out of 16 trials virus was transmitted from the experimentally inoculated chickens to the non-inoculated chickens. The transmission rate was 0.13 and 0.10 new infections per infectious bird at 0.2m and 1.1m, respectively. The difference between these estimates was, however, not significant. Two air samples tested positive in virus isolation, but none of these samples originated from the trials with successful transmission. Five dust samples were confirmed positive in virus isolation. The results of this study demonstrate that the rate of airborne transmission between chickens over short distances is low, suggesting that airborne transmission over a long distance is an unlikely route of spread. Conjugated organic polymers generally must include large substituents for stability, either contained within or appended to the polymer chain. In polydiacetylenes, the substituents fulfill another important role: During topochemical polymerization, they control the spacing between the diyne monomers to produce an ordered polymer. By using a co-crystal scaffolding, we have prepared poly(diiododiacetylene), or PIDA, a nearly unadorned carbon chain substituted with only single-atom iodine side groups. The monomer, diiodobutadiyne, forms co-crystals with bis(nitrile) oxalamides, aligned by hydrogen bonds between oxalamide groups and weak Lewis acid-base interactions between nitriles and iodoalkynes. In co-crystals with one oxalamide host, the diyne undergoes spontaneous topochemical polymerization to form PIDA. The structure of the dark blue crystals, which look copper-colored under reflected light, has been confirmed by single-crystal x-ray diffraction, ultraviolet-visible absorption spectroscopy, and scanning electron microscopy. 相似文献
