Deep brain stimulation (DBS) of the subthalamic nucleus markedly improves the motor symptoms of Parkinson's disease, but causes cognitive side effects such as impulsivity. We showed that DBS selectively interferes with the normal ability to slow down when faced with decision conflict. While on DBS, patients actually sped up their decisions under high-conflict conditions. This form of impulsivity was not affected by dopaminergic medication status. Instead, medication impaired patients' ability to learn from negative decision outcomes. These findings implicate independent mechanisms leading to impulsivity in treated Parkinson's patients and were predicted by a single neurocomputational model of the basal ganglia.  相似文献

Euclinostomum heterostomum and Euclinostomum ardeolae, both encysted metacercariae (EMC), were found to infect farmed Oreochromis niloticus in the Sahary fish hatchery and wild O. niloticus, Sarotherodon galilaeus, and Tilapia zillii in Lake Nasser, Aswan Governorate, Egypt, at a prevalence of 25.25% and an infection intensity of 1–14 EMC/fish. Macroscopic and microscopic examinations were used to identify them morphologically. PCR amplification, sequencing of the 28S large ribosomal RNA region, and phylogenetic analysis were used for molecular characterization. E. heterostomum and E. ardeolae were isolated from farmed O. niloticus kidneys, peritoneum, abdominal cavity, and under the gills attached to the head bone at prevalence of 13.5% and 6%, respectively, and from wild O. niloticus, S. galilaeus, and T. zillii at a prevalence of 15% and 4.5%, 18% and 10%, and 22.5% and 11.5% respectively. The molecular analyses of rRNA-28S marker revealed that the two Euclinostomum species examined are related yet distinct. This is the first molecular and phylogenetic evidence linking E. ardeolae to Euclinostomum. The present study added two new sequences to the GenBank databases for the genus Eulinostomum from Egypt, with accession numbers MW604803 and MW604806. Euclinostomum spp. have been shown to have detrimental and destructive effects on the kidneys of infected fish; these effects may eventually result in the parasites’ cysts replacing the kidney’s tissues. The kidneys of tilapia spp. infected with Euclinostomum spp. displayed degenerative alterations, with dilated renal tubules associated with migratory tunnels composed primarily of leucocytic infiltrations. Euclinostomum spp. exhibited clear hemophagic characteristics.

Foot-and-mouth disease (FMD) is a highly contagious and transboundary viral disease of domesticated and wild cloven-hoofed animals. The present study summarizes the knowledge generated from the epidemiology, diagnosis, and surveillance of the disease in the Maghreb (2014–2015) in assessing its threat to southern European countries. Currently, two serotypes of the FMD virus (O and SAT-2) and four lineages are circulating in the Maghreb region. The introduction of serotype SAT-2 in Libya and Mauritania in 2012 and 2015 respectively sets their neighbors and subsequently south European countries at constant risk of FMD re-emergence. The potential pathways of FMD introduction to southern European countries from the Maghreb are the illegal introduction of infected animals and animal products, particularly meat or meat products carried by refugees.

The present study was carried out to evaluate the use of Biogen and sodium butyrate (SB) as feed additives in the diet of Nile tilapia (Oreochromis niloticus) fingerlings, in two parallel experiments. Biogen was incorporated in isonitrogenous (35% crude protein) and isocaloric (19 MJ kg?1) diets at four levels (0.0 (control), 0.5, 1.0, and 2.0%), while SB was included at five levels (0.0 (control), 0.5, 1.0, 2.0, or 3.0%). The diets were fed to fingerling Nile tilapia (10.5 ± 0.5 g) at a daily rate of 4% of their body weight, three times a day, for 60 days. Except the lymphocytes and monocyte numbers in fish fed SB, hematological parameters, including red blood cell (RBC) hematocrit (Ht), hemoglobin (Hb), and white blood cells (WBC) were not significantly (P > 0.05) affected by dietary Biogen and SB. The lymphocytes number in Nile tilapia fed on SB increased with increasing SB up to 2% level, and decreased afterwards. Monocyte numbers showed irregular patterns. The activities of serum enzymes alanine aminotransferase (ALT) and aspartate aminotransferase (AST) of fish fed diets containing Biogen or SB were not significantly affected by dietary treatments (P > 0.05). No structural differences of tilapia liver were detected between all Biogen treatments and sodium butyrate concentrations up to 1% with control. At concentrations of 2 and 3% sodium butyrate, liver steatosis increased leading to shrinked acentric nuclei. At a concentration of 2% Biogen, some glomerulus cells had fading cytoplasm. Concerning fish fed SB diets, the structure of kidney was the same as in control except at concentration of 3% SB, where the septum between cells disappeared. No changes in gill structure were noticed at all concentrations of Biogen and SB.  相似文献

The influence of trehalose on European mouflon spermatozoa cryopreservation during the non-breeding season was tested. Semen was frozen in two different extenders: (a) recommended Tris-based ram extender (CTR); (b) CTR extender supplemented with trehalose 0.147 mm (TRH). Sperm viability and acrosome integrity were assessed using propidium iodide and fluorescein isothiocynate labelled Pisum Sativum agglutinin. Trehalose significantly enhanced sperm viability after thawing compared with CTR extender (62.7% vs 51.8%; p < 0.05), whereas no differences were observed on acrosome integrity (42.9% vs 42.1%). Trehalose influence was also evidenced in the in vitro fertility test performed with sheep oocytes matured in vitro. Both fertilization rates (60.9% TRH vs 43.6% CTR; p < 0.05) and cleavage rates (58% TRH vs 39.8% CTR; p < 0.001) were higher for trehalose frozen semen compared with control extender frozen semen. A higher percentage of zygotes resulting from fertilization with trehalose cryopreserved semen presented the first cleavage earlier if compared with the group fertilized with control semen (48.7% vs 31.5%, respectively; p < 0.01). This result was confirmed by embryo kinetic development. Fertilization with trehalose cryopreserved semen leaded to an higher percentage of blastocysts (40.2% vs 27.8% CTR; p < 0.05), and enhanced in particular the number of blastocysts that developed on the day 6th of culture (28.6% vs 17% CTR; p < 0.05). Our data demonstrated that, during mouflon non-breeding season, trehalose extender enhances spermatozoa viability and its in vitro fertilizing capacity, allowing the production of an higher number of blastocysts.  相似文献

The larvicidal activity of sustained-release formulations ofBacillus thuringiensis H-14 (Bactimos briquets) and controlledrelease plastic formulations of Abate against mosquito larvae ofAedes aegypti (L.) has been evaluated. The test formulations provided continuous effective control with 90–100% larval mortality for several weeks. The presence of suspended soil constituents was associated with a lowering of larval mortality and tended to reduce the durations of effective control of Bactimos and Abate plastic pellets against mosquito larvae by about 1.8 and 2.2 times, respectively. Treatments with such formulations did not appear to prevent hatch of eggs which oviposited after application, but all newly hatched larvae failed to develop beyond the second instar.  相似文献

Objective To investigate wool organophosphorus concentrations resulting from a range of farm pesticide application methods.
Design Random sampling of wool for pesticide residues and on-farm interviews to determine associated treatments.
Procedure Tasmanian fleece wool lots were sampled at random and tested for organophosphorus residues. The grower was identified and the pesticide treatments applied to the sheep were ascertained by on-farm interview.
Results The residue concentrations showed a large variation that was not accounted for by differences in treatments by growers. Organophosphorus concentrations were proportional to the number of treatments applied, and inversely related to the time between pesticide application and the subsequent shearing, and were significantly influenced by the method of application. After allowing for the time of application, plunge dipping resulted in pesticide residue concentrations 2 to 2.5 times greater than shower dipping, using spray races or hand jetting, and the use of these methods caused larger residues than the use of jetting races.
Conclusions We recommend that plunge or shower dipping should not be used more than 7 weeks after shearing, nor at higher concentration than the standard dose rate used for lice control, whereas jetting may be satisfactory for up to 7 months after shearing, provided only one application is administered.  相似文献

In an attempt to explore the functioning of nitric oxide (NO) in pancreatic exocrine cells, we have recently obtained several lines of circumstantial evidence indicating that one of molecular targets of NO is phospholipase C (PLC), the activation of which leads to an increase in the cytosolic Ca2+ concentration ([Ca2+]i) via inositol 1, 4, 5-trisphosphate, IP3. However, whether IP3 is actually produced by NO has not yet been substantiated. The present study was therefore designed to directly measure the intracellular IP3, concentration ([IP3]i) for better understanding of the underlying mechanisms with the help of pharmacological tools. [IP3]i was measured using a fluorescence polarization technique (HitHunter). We obtained the following results: 1) varying concentrations of an NO donor, sodium nitroprusside (SNP), elevated [IP3]i, 2) this elevation was completely inhibited in the presence of the soluble guanylyl cyclase (sGC) inhibitor, 1H-[1, 2, 4] oxadiazolo [4, 3-a] quinoxalin-1-one (ODQ), 3) varying concentrations of the cGMP analogue, 8-Br-cGMP, also increased [IP3]i, 4) the cGMP analogue-induced IP3 production was abolished by pretreatment with either a PLC inhibitor, U73122, or a G-protein inhibitor, GP2A, and 5) KT5823, a potent and highly selective inhibitor of cGMP-dependent protein kinase G (PKG), also abolished the IP3 production induced by 8-Br-cGMP. These results suggest that the NO-induced [Ca2+]i increase is triggered by an increase in [IP3]i located downstream from intracellular cGMP elevation. In this intracellular pathway, each sGC, cGMP-dependent PKG, G-protein and PLC were suggested to be involved. The present work provides new insights into the intracellular signaling accelerated by NO. NO triggers a [Ca2+]I increase via cGMP and IP3 in pancreatic acinar cells.  相似文献
