An orthoreovirus was isolated from an Ostrich (Struthio camelus) and rapidly identified as orthoreovirus by the rapid determination of viral RNA sequences (RDV) system and electron microscopy. Phylogenetic analysis of the sigma A protein indicated that the isolate belonged to avian species and was closely related to chicken orthoreovirus strain 138. The results of the present study indicated that an ostrich orthoreovirus is slight different from other chicken orthoreoviruses and provided evidence of diversity among avian orthoreoviruses. To our knowledge, this is the first genetic report of an orthoreovirus isolated from an ostrich. 相似文献

Borna disease virus (BDV) infection has been suggested to cause spontaneous neurological disease in cats referred to as staggering disease. However the evaluation of BDV infection in neurologically asymptomatic cats remained unclear. In the present study, BDV infected, asymptomatic cats in Tokyo were surveyed both by the presence of plasma antibodies against BDV-p24 and -p40 and by RNA detection in peripheral blood mononuclear cells. Seven of 32 domestic cats (21.9%) were serologically or genetically judged to be BDV-infected. Six cats were positive for anti-BDV antibody and two cats were positive for BDV RNA. Within the 2 RNA-positive cats, only one was positive for anti-BDV antibodies. Furthermore, the findings of anti-BDV-p40 and anti-BDV-p24 antibody-positive cats did not completely overlap. These results suggest that there are neurologically asymptomatic domestic cats infected with BDV present in the Tokyo area. 相似文献

Invasion of apple fruit by Erwinia amylovora from fruit-bearing twigs through the abscission layer at fruit maturation was examined. Erwinia amylovora (ca. 105 cfu) tagged with bioluminescence genes from Vibrio fischeri was deposited in artificial wounds on fruit-bearing twigs of apple trees grown in a containment greenhouse on September 22,
27, or October 5, 2004. On October 22, 176 apples were harvested and cut horizontally in half. The upper halves were stamped
on plates of selective medium, and the lower halves were flooded with iodine solution to assess maturity. All fruit were symptomless
and fully mature. The pathogen was recovered from 19 (10.8%) apples. The result showed that if at least ca. 105 cfu of E. amylovora are present in fruit-bearing twigs at the time of fruit maturation, the bacteria can pass through the abscission layer into

There is an urgent need for the international community to address not only the issues of preserving the global environment and reducing poverty in developing countries, but to also face up to new challenges, such as providing assistance to the reconstruction in Afghanistan. Furthermore, it is necessary to address these issues and challenges by means of mutual collaboration. This latter point has been the subject of lively discussion.In this paper, we describe a new direction in international cooperation taken by the Rural Development Bureau of the Ministry of Agriculture, Forestry and Fisheries, Japan in response to the trend of international discussions. This shoulb be achieved by utilizing Japans experience and expertise gained in the development of its own paddy rice agriculture.Specifically, based on the philosophy that sustainable agricultural and rural development can conserve a regional environment, the Bureau intends to undertake: (1) extension of the results from a cooperation project to other regions, by modifying it and converting it to multi-purpose use; (2) collaboration with aid-related entities in Japan and abroad, and dissemination of related information to them; and (3) promotion of cooperation focused on a specific topic.Furthermore, the Bureau will promote the following matters as the basis for international cooperation in each major task. (1) Contribution towards sustainable water use. (2) Extension of the participatory integrated rural development, and its modification for reconstruction assistance. To determine the feasibility of preserving oocytes without freezing, we stored mouse oocytes in several media at different temperatures for one day. Confocal microscopy of the metaphase-II spindle in these stored oocytes revealed gross abnormalities in both the spindle and the arrangement of chromosomes. The abnormal spindles could not be rescued by transplanting the aged spindle-chromosome complex into a fresh enucleated oocyte. A diploid parthenogenetic development showed that some of the oocytes stored at room temperature could still develop into blastocysts (10-57%). However, oocytes stored in a refrigerator (5%) or incubator (0%) lost the potential almost entirely. Fertilization of room-temperature-preserved oocytes with fresh spermatozoa by ICSI or IVF resulted in, respectively, 4 and 10%, full-term births. These results suggest that when oocytes are stored at room temperature for one day, most have irreversible damage not only to their cytoplasm but also to the spindle. However, since at least a few percent of stored oocytes retained the potential for full-term development, it may be possible to overcome these problems and develop a simple method for preserving mammalian oocytes without freezing. 相似文献

Although cats are induced ovulators, the relationship between the day of breeding, the number of matings and the likelihood of ovulation and conception have not been extensively investigated. In this experiment, cats were mated either once or three times on day 1 or day 5 of oestrus to study the incidence of the LH surge, ovulation and conception rates. The percentage ovulating and the conception rates after a single mating on day 1 of oestrus were 60% (6/10) and 33.3% (2/6), respectively, and for cats mated once on day 5 of oestrus were 83.3% (10/12) and 40% (4/10), respectively. When cats were mated three times on day 1 of oestrus, the ovulation rates and conception rates were 70% (7/10) and 85.7% (6/7), respectively, and for those mated three times on day 5 of oestrus were 100% (10/10) and 100% (10/10), respectively. The concentration of LH did not increase in non-ovulating cats, and cats that were mated three times had LH concentrations that were numerically higher than those that were mated once. Litter size was neither related to the day of mating nor to the number of matings. Although an increase in the number of matings on day 1 of oestrus produced a numerically larger LH surge, it did not increase the ovulation rate, suggesting that plasma oestradiol concentrations were not sufficiently elevated to induce a high pituitary response to mating stimulation. The conception rate after a single mating was low, suggesting that the number of sperm per mating was not sufficient. These results suggest that mating more than once in the middle of oestrus is required to improve ovulation rates and conception rates in cats. 相似文献
