Diploid and triploid intergeneric hybrids obtained by crosses among Gloriosa superba ‘Lutea’ (2n = 2x = 22), G. ‘Marron Gold’ (2n = 4x = 44), Littonia modesta (2n = 2x = 22), and Sandersonia aurantiaca (2n = 2x = 24) were analyzed for their meiotic chromosome pairing in pollen mother cells by genomic in situ hybridization (GISH) with digoxigenin-labeled total DNA of one parent as probe. Chromosomes from each parent could be clearly distinguished in pollen mother cells of all the five intergeneric hybrids by GISH. For three diploid hybrids, L. modesta × G. superba ‘Lutea’ (2n = 2x = 22), L. modesta × S. aurantiaca (2n = 2x = 23) and S. aurantiaca × G. superba ‘Lutea’ (2n = 2x = 23), 0.04?0.27 autosyndetic bivalents (intragenomic pairing of non-homologous chromosomes) and 0.13?0.36 allosyndetic bivalents (intergenomic chromosome pairing) were observed per pollen mother cell, indicating that there are some homologous chromosomal regions within each genome and among the genomes of Gloriosa, Littonia and Sandersonia. Differences in the average number of allosyndetic bivalents per pollen mother cell among different genome combinations may reflect the evolutionary distances among the three genera, and Gloriosa and Littonia may be closely related to each other, while Sandersonia may have relatively distant relationships with Gloriosa and Littonia. For two triploid hybrids, L. modesta × G. ‘Marron Gold’ (2n = 3x = 33) and S. aurantiaca × G. ‘Marron Gold’ (2n = 3x = 34), no allosyndetic bivalents were observed. Based on the results obtained in the present study, possible utilization of the diploid and triploid intergeneric hybrids for further breeding of colchicaceous ornamentals is discussed.  相似文献

An experiment was conducted to study the association between cold shock treatment (CST) (0 °C, 1 h) and heat shock treatment (HST) (55 °C, 30 s) on antioxidant enzymes of broccoli at different storage temperatures (20, 10, or 5 °C). There was no difference in antioxidant enzyme activities between CST and HST under storage at 20 °C; superoxide dismutase (SOD) and catalase (CAT) activities were increased in comparison with the controls. However, activities of antioxidant enzymes, except for ascorbate peroxidase (APX), with CST were higher than those with HST during storage at 10 °C, while there were opposite results except for CAT activity at 5 °C. These results suggest that CST and HST have the same effect on the antioxidant enzymes of broccoli during 20 °C storage, but have different effects at below 10 °C, which become more pronounced as the temperature is lowered.  相似文献

A unicellular coccolithophorid alga, Pleurochrysis carterae, contained 125.4 +/- 1.2 microg of vitamin B12 per 100 g dry cell weight of the lyophilized algal cells. A vitamin B12 compound was purified from the lyophilized algal cells and partially characterized. The silica gel 60 TLC and reversed-phase HPLC patterns of the purified pink-colored compound were identical to those of authentic vitamin B12, but not those of vitamin B12 analogues inactive for humans. When 22-week-old B12-deficient rats which excreted substantial amounts of methylmalonic acid (75.5 +/- 12.3 mg/day) in urine were fed the P. carterae (10 g per kg diet)-supplemented diet for 12 d, urinary methylmalonic acid excretion (as an index of vitamin B12 deficiency) of the rats became undetectable and hepatic vitamin B12 level of the rats was significantly increased.  相似文献

The vitamin B(12) concentration of an algal health food, spirulina (Spirulina sp.) tablets, was determined by both Lactobacillus leichmannii ATCC 7830 microbiological and intrinsic factor chemiluminescence methods. The values determined with the microbiological method were approximately 6-9-fold greater in the spirulina tablets than the values determined with the chemiluminescence method. Although most of the vitamin B(12) determined with the microbiological method was derived from various vitamin B(12) substitutive compounds and/or inactive vitamin B(12) analogues, the spirulina contained a small amount of vitamin B(12) active in the binding of the intrinsic factor. Two intrinsic factor active vitamin B(12) analogues (major and minor) were purified from the spirulina tablets and partially characterized. The major (83%) and minor (17%) analogues were identified as pseudovitamin B(12) and vitamin B(12), respectively, as judged from data of TLC, reversed-phase HPLC, (1)H NMR spectroscopy, ultraviolet-visible spectroscopy, and biological activity using L. leichmannii as a test organism and the binding of vitamin B(12) to the intrinsic factor.  相似文献

Three K silicates having SiO2/K2O ratios of 4, 5, and 6 were compared with single and split applications of KCI, (KPO3)n, and sulfur-coated KCI (SCK-34) for their availability to four cuttings of common bermudagrass. At the rate of 200 mg of K/pot, all silicates applied in fine particles gave the same pattern of yield and K uptake as readily soluble KCl; however, less response to these finely sized K silicates at applications from 200 to 500 mg of K/pot suggests that they supply K more slowly than do soluble sources. K2O.6SiO2, when granulated in the size of 0.41-1.2 or 1.65-2.36 rom gave typical yieiJ and K uptake patterns of slowrelease sources similar to those for SCK-34 at both rates of application. (KPO3) n supplied K in the same pattern as KCl and no effect of granulation was observed. A better balanced K supply through the whole period and the production of forage of more stable mineral composition is expected by the use of these slow-release K sources.  相似文献

In order to improve the efficiency of stock enhancement programs for the sea urchin Hemicentrotus pulcherrimus, information on food algal species, which affect growth and gonad production greatly, is necessary. Since it is difficult to identify species from the macroalgal fragments within the gut contents of the sea urchin by microscopic observation, we tried to apply a DNA barcoding method for gut contents analysis. We used a partial rbcL gene sequence for taxonomic section and newly designed primer sets, respectively, for brown algae and for red algae. Direct sequencing of the PCR products was carried out. Species identification was based on the phylogenetic relationship. We could objectively identify four species and two taxonomic groups (genus or family) in brown algae, and two species and four taxonomic groups in red algae from the gut contents. Sargassum hemiphyllum was the most abundant brown alga in the gut contents but was not dominant in the study site. The result showed the importance of identification to the species level. In addition, red algal epiphytes were detected with brown algal fragments. The DNA barcoding method will enable the researchers to verify the important role of epiphytes as a potential food source.  相似文献

The chrysomelid beetle Colaspis crinicornis Schaeffer (Coleoptera: Chrysomelidae) occurs primarily in the Great Plains region of the United States. Little is known about the biology and ecology of this species, but over the last decade, it has become increasingly common in the corn, Zea mays L., and soybean, Glycine max (L.) Merrill, agroecosystem of southeastern Nebraska. As part of a larger comprehensive project to understand the natural history and pest potential of this species, laboratory experiments were conducted to study the developmental biology, morphological characters of immature stages, and the effect of adult diet on consumption, longevity, and fecundity. Females readily deposited egg clusters in the soil, and percentage egg hatch was high under laboratory conditions. Larvae and pupae were confirmed to be soil-dwelling stages. C. crinicornis has relatively short egg, pupal, and adult stages with the majority of its life cycle spent in the larval stage. Results of choice and no-choice adult feeding experiments indicate that diets of corn or soybean leaves did not significantly affect consumption, longevity, or fecundity of adult C. crinicornis, suggesting that corn and soybean leaves are similarly suitable food sources for adults. The ability to effectively utilize tissues from very different plant families as adult food sources suggests that C. crinicornis is polyphagous in the field.  相似文献

Many corals establish symbiosis with Symbiodiniaceae cells from surrounding environments, but very few Symbiodiniaceae cells exist in the water column. Given that the N-acetyl-d-glucosamine-binding lectin ActL attracts Symbiodiniaceae cells, we hypothesized that corals must attract Symbiodiniaceae cells using ActL to acquire them. Anti-ActL antibody inhibited acquisition of Symbiodiniaceae cells, and rearing seawater for juvenile Acropora tenuis contained ActL, suggesting that juvenile A. tenuis discharge ActL to attract these cells. Among eight Symbiodiniaceae cultured strains, ActL attracted NBRC102920 (Symbiodinium tridacnidorum) most strongly followed by CS-161 (Symbiodinium tridacnidorum), CCMP2556 (Durusdinium trenchii), and CCMP1633 (Breviolum sp.); however, it did not attract GTP-A6-Sy (Symbiodinium natans), CCMP421 (Effrenium voratum), FKM0207 (Fugacium sp.), and CS-156 (Fugacium sp.). Juvenile polyps of A. tenuis acquired limited Symbiodiniaceae cell strains, and the number of acquired Symbiodiniaceae cells in a polyp also differed from each other. The number of Symbiodiniaceae cells acquired by juvenile polyps of A. tenuis was correlated with the ActL chemotactic activity. Thus, ActL could be used to attract select Symbiodiniaceae cells and help Symbiodiniaceae cell acquisition in juvenile polyps of A. tenuis, facilitating establishment of symbiosis between A. tenuis and Symbiodiniaceae cells.  相似文献

The purpose of this retrospective study was to describe pre‐ and postcontrast computed tomographic (CT) characteristics of confirmed nonparenchymal hemangiosarcoma in a group of dogs. Medical records were searched during the period of July 2003 and October 2011 and dogs with histologically confirmed nonparenchymal hemangiosarcoma and pre‐ and postcontrast CT images were recruited. Two observers recorded a consensus opinion for the following CT characteristics for each dog: largest transverse tumor diameter, number of masses, general tumor shape, character of the tumor margin, precontrast appearance, presence of dystrophic calcification, presence of postcontrast enhancement, pattern of postcontrast enhancement, presence of regional lymphadenopathy, and presence of associated cavitary fluid. A total of 17 dogs met inclusion criteria. Tumors were located in the nasal cavity, muscle, mandible, mesentery, subcutaneous tissue, and retroperitoneal space. Computed tomographic features of nonparenchymal hemangiosarcoma were similar to those of other soft tissue sarcomas, with most tumors being heterogeneous in precontrast images, invasive into adjacent tissue, and heterogeneously contrast enhancing. One unexpected finding was the presence of intense foci of contrast enhancement in 13 of the 17 tumors (76%). This appearance, which is not typical of other soft tissue sarcomas, was consistent with contrast medium residing in vascular channels. Findings indicated that there were no unique distinguishing CT characteristics for nonparenchymal hemangiosarcoma in dogs; however, the presence of highly attenuating foci of contrast enhancement may warrant further investigation in prospective diagnostic sensitivity and treatment outcome studies.  相似文献
