Fresh citrus honey was stored at 10, 20, and 40 degrees C for 12 months. The effect of storage on the quality of honey was evaluated using physicochemical parameters, volatile compounds, mono-, di-, and trisaccharides, and sensory analysis. Diastase activity and HMF were out of the legal limit in honey stored 12 months at 40 degrees C. Volatile compounds (especially terpenes and terpene derivatives), monosaccharides, and disaccharides presented important losses during honey storage at any temperature. Honey storage at 10 or 20 degrees C maintained their floral, fresh, citric, and fresh fruit aroma, while the intensities of these attributes were diminished. Storage at 40 degrees C during 12 months resulted in the appearance of attributes such as "medicinal, smoked, toasted, cooked vegetable, and ripened fruit", associated with compounds formed during the Maillard reaction or through degradation of sugars such as volatile pyrroles, furanones, pyranones, and pyrazines, which appeared or increased in concentration during honey storage mainly at high temperature. 相似文献

A new method was developed to simultaneously measure 2 acute-phase proteins (APPs) by time-resolved immunofluorometry. The assay, based on double-label quantification of haptoglobin (Hp) and C-reactive protein (CRP) in meat juice samples from pigs, was constructed by use of a combination of europium and samarium chelate lanthanides as labels. Meat juice samples from 154 pigs were used for analytic and clinical validation of the assay through determination of precision, accuracy, limit of detection, and quantification. The analytic performance of the assay was satisfactory, with good intra-assay and interassay precision and accuracy. The levels of Hp and CRP were increased in the meat juice samples of diseased animals compared with healthy ones. According to the results, higher sensitivity could be achieved if the cut-off values of both proteins were taken into account for clinical relevance rather than used individually. Since the dual assay saved both time and sample, it could be used as a rapid and sensitive screening test in porcine production. 相似文献

A field trial was conducted to study Mycoplasma hyopneumoniae (Mh) infection dynamics by nested polymerase chain reaction (nPCR) and serology in pigs of a farm affected by enzootic pneumonia (EP). Moreover, correlation of Mh detection at different respiratory tract sites with presence of EP gross and microscopic lung lesions was assessed. These parameters were studied and compared between vaccinated (two doses at 1 and 3 weeks of age versus one dose at 6 weeks of age) and non-vaccinated pigs. Animals were monitored from birth to slaughter by nPCR from nasal swabs and by serology. From 3 to 22 weeks of age, an average of three pigs per treatment and per batch were necropsied (n = 302). The remaining pigs were sent to the slaughter (n = 103). Nasal, bronchial and tonsillar swabs were taken from the necropsied/slaughtered pigs; gross and microscopic EP-suggestive lung lesions were also assessed. Single and double vaccination resulted in earlier seroconversion and higher percentage of Mh seropositive pigs compared to control group. At slaughter, double vaccinated pigs showed lower percentage of EP-compatible gross lung lesions and lower Mh prevalence at upper respiratory tract sites (nasal cavity and tonsil) than control pigs. 相似文献

The effects of feeding two fibre-rich diets with contrasting solubility and a concentrated low dietary fibre on the rate of gastric emptying were examined in six gastric cannulated pregnant sows. Additionally, it was examined whether any effect could be related to the physico-chemical properties of digesta, i.e. viscosity and/or water binding capacity. The sows were fed each diet for one week in a 3 x 3 Latin Square design and the samples were taken in a randomised order 0.5, 1, 2, 3, 5 and 15.5 h after the morning meal. The stomach contents were evacuated through the gastric cannula once daily. The evacuated gastric digesta was quantified and a representative sample was taken to determine its viscosity, water binding capacity and its content of dry matter, dietary components and solid (Cr2O3) and liquid (polyethylene glycol) phase markers. The flow of liquid digesta was calculated as the difference between digesta and dry matter. Increasing the content of dietary fibre in the diet led to higher recovery of liquid digesta but did not have any significant effect on the gastric emptying of dry matter and dietary components. The effect of dietary fibre could not be attributed to the viscosity of the liquid phase of digesta but might be related to the ability of the increased gastric dietary fibre content to hold water. The stomach selectively retained the insoluble dietary fibre components most noticeably seen with the bran-supplemented diet where the concentration of insoluble NSP in digesta increased significantly from 2 hours and onward. 相似文献

Fusarium oxysporum and Fusarium proliferatum are important causal agents of crown and root rot of asparagus. In order to detect differences in pathogenicity and aggressiveness, two F. proliferatum and five F. oxysporum single spore isolates from asparagus spears from plantings in Austria and Germany, 55 pure cultures of F. oxysporum from asparagus roots from a planting in Hesse, Germany, and a single F. oxysporum isolate from an asparagus shoot collected in Austria were evaluated in a 28-day quick test on Hoagland??s agar in glass culture tubes. Plantlets were inoculated with spore suspensions from each respective isolate after 14 days of growth under sterile, controlled conditions in a growth chamber. A severity scale was used to assess symptoms on roots two weeks after inoculation. The effects of the single-spore isolates on root and shoot fresh weights of the plantlets were also determined. The pathogenicity of the majority of the F. proliferatum and F. oxysporum isolates included in this study was confirmed. Inoculation with pure and single-spore cultures resulted in elevated disease severity in comparison to non-inoculated controls. In particular, the two F. proliferatum isolates were found to be more aggressive than the F. oxysporum isolates. Moreover, all single spore isolates caused a reduction in fresh weight of roots and shoots in comparison to the controls. With respect to differences among asparagus cultivars, ??Ramos??, was found to be more susceptible than ??Ravel??. Overall, the quick test method was found to be capable of evaluating the pathogenicity and aggressiveness of the tested F. oxysporum and F. proliferatum isolates towards asparagus within 28 days. 相似文献

The effect of different types of fibre on the intestinal digesta and microbial diversity was evaluated in growing pigs. The animals were fed during forty-two days with one of four experimental diets including a control group, a diet enriched with resistant starch type I, by coarse ground corn, and other two diets containing sugar beet pulp (8%) and wheat bran (10%) (as sources of soluble and insoluble non-starch polysaccharides respectively). Body weight (BW) and average daily feed intake (ADFI) were assessed at day 7, 21 and 42. Luminal samples of digesta from proximal colon were collected to analyse water retention capacity (WRC), concentration of short-chain fatty acids (SCFA) and the microbial diversity. Overall, animals fed with higher amounts of non-digestible carbohydrates had lower ADFI and BW. Sugar beet pulp provoked an increase of WRC, a higher concentration of SCFA, and a more stable microbial diversity throughout the experimental period. The aim of this research was to confirm the efficacy of the yeast antagonist Candida sake CPA‐1 in suppressing botrytis bunch rot development, in an organic vineyard under Mediterranean conditions for two seasons, and compare its performance with that of two biologically based products currently registered for botrytis bunch rot control in New Zealand. In 2009, treatments applied were: commercial formulations of Ulocladium oudemansii (BOTRY‐Zen®) and chitosan (ARMOUR‐Zen®), C. sake CPA‐1 combined with the fatty acid‐based additive Fungicover® and combinations of these products. All treatments were applied six times between early flowering and harvest and compared with an unsprayed control. In 2010, the treatments focused on C. sake and Fungicover and the number of applications was reduced from six to four. The population dynamics of U. oudemansii and C. sake were measured and wine quality tests were carried out in both seasons. Disease control achieved by C. sake treatments in 2009 were comparable to those achieved by BOTRY‐Zen and ARMOUR‐Zen. Applications of C. sake plus Fungicover between flowering and harvest significantly (P < 0·05) reduced botrytis bunch rot incidence and severity by 64% and 90%, respectively, compared with the untreated control in 2009, and by 67% and 89%, respectively, in 2010. Treatments did not adversely affect wine quality parameters after treated grapes were processed. Candida sake consistently provided effective control of botrytis bunch rot in grapes under different meteorological and disease pressure conditions, thereby improving its potential for future commercial applications. 相似文献
