The serotonergic regulation of feeding behaviour has not so far been studied in ruminants. Therefore, the effects of some serotonin (5-HT) receptor agonists and antagonists on food intake and forestomach motility were studied in dwarf goats.Goats ate less food when treated intravenously (IV) with the 5-HT precursor 5-HTP (25 µg, 50 µg or 100 µg kg–1 min–1 over 15 min) than when they were treated with 5-HT (which does not pass the blood-brain barrier) or with saline. Accordingly, IV dexfenfluramine infusions (50 µg or 100 µg kg–1 min–1 over 15 min), which induces release of brain 5-HT, also led to dose-related reductions in food intake. In contrast, no anorectic effects were observed after IV infusions with the selective 5-HT reuptake inhibitor fluoxetine (100 µg kg–1 min–1 over 15 min), the selective 5-HT1A agonist 8-OH-DPAT (0.5 µg kg–1 min–1 over 15 min), or eltoprazine (4 or 8 µg kg–1 min–1 over 15 min), a mixed 5-HT1A/5HT1B receptor agonist. None of the 5-HT antagonists tested gave any increase in food consumption in this model. Interestingly, the non-selective 5-HT receptor antagonist methysergide (360 µg/kg IV) reduced food intake. This effect was most noticeable at 3 h after injection. The 5-HT3 receptor antagonist ondansetron (IV 10 µg kg–1 min–1 over 15 min) and the peripheral 5-HT2 receptor antagonist xylamidine (IV 100 µg kg–1 min–1 over 10 min) failed to modify food intake. These results provide evidence for central serotonergic involvement in the control of feeding. However, this control system differs markedly in goats and rodents.Dexfenfluramine, 5-HTP and eltoprazine administered at similar dose rates to those used in the food intake experiments induced some clinical signs including inhibition of forestomach contractions. These results, together with our earlierin vivo andin vitro observations, suggest that the inhibitory effects of serotonin receptor agonists on forestomach contractions are due to interactions with both peripheral and central serotonergic receptors. The change in smooth muscle tension, which leads to a change in the signals transmitted via vagal afferents to the central nervous system, appears not to modify feeding behaviour in dwarf goats.  相似文献

Cell-to-cell interaction via cell contact-dependent pathway is essentially important for maintenance and regulation of corpus luteum (CL) integrity and its physiological actions. The objective of the present study was to evaluate the mRNA expression of the cell adhesion molecules (CAMs) that are constituent factors of gap junctions [connexin (Cx) 43] and adherence junctions (VE-, E-, N-cadherin) in two types of endothelial cells from the mid CL and in CL tissue during the estrous cycle and PGF(2alpha)-induced luteolysis in the cow. Specific mRNA expression for Cx43 and N-cadherin was detected in cytokeratin-positive (CK+) and cytokeratin-negative (CK-) luteal endothelial cells (EC) and fully luteinized granulosa cells (LGC). E-cadherin mRNA was expressed in CK+EC and LGC, but not in CK-EC. VE-cadherin mRNA was expressed in both CK+ and CK-EC. During the estrous cycle, Cx43 mRNA expression was significantly lower in the regressing CL. VE-cadherin expression also tended to increase in the mid CL and increased significantly in the regressing CL. E-cadherin mRNA expression was higher in the early and late CL than in the mid- and regressing CL. N-cadherin mRNA expression gradually increased from the early to late CL followed by a decrease in the regressing CL. During PGF(2alpha)-induced luteolysis, Cx43 mRNA expression appeared to increase, and VE-cadherin and E-cadherin mRNA significantly increased at 24 h. N-cadherin mRNA expression decreased 2 and 4 h after PGF(2alpha) administration. Collectively, expression of the mRNAs for CAMs was different in the two types of luteal endothelial cells and fully luteinized granulosa cells and changed independently in the CL during the estrous cycle and PGF(2alpha)-induced luteolysis in the cow. The results suggest that CAMs play physiological roles in cell-to-cell communication to regulate both gap and adherence junctions during CL development and regression in the cow.  相似文献

We examined the effects of oral glucosamine hydrochloride (GlcN), N-acetyl-D-glucosamine (GlcNAc) and d-glucose (Glc) administration on plasma total free amino acid (PFAA) concentrations in dogs. The PFAA concentrations increased in the control group and the GlcNAc group at one hour after feeding, and each amino acid concentration increased. On the other hand, in the GlcN group and the Glc group PFAA concentrations decreased at one hour after feeding. A significant decrease in amino acid concentration was observed for glutamate, glycine and alanine. Our results suggest the existence of differences in PFAA dynamics after oral administration of GlcN and GlcNAc in dogs.  相似文献

Interactive effects of water stress and potassium (K) on some physiological attributes and nutritional status of melon (Cucumis melo L. cv. ‘Tempo F1’) plants were assessed in a pot experiment. Treatments used were: (1) control or well-watered (WW) + K1, (2) WW + K2, (3) WW + K3, (4) water stress (WS) + K1, (5) WS + K2, and (6) WS + K3. Water stress (WS) was imposed by maintaining the moisture level equivalent to 50% pot capacity, whereas well-watered (WW) pots (control) were maintained at full pot capacity (100% PC). Hoagland's nutrient solution was modified to supply K as potassium sulfate (K2SO4) at 6, 9, and 12 mM for K1, K2, and K3 treatments, respectively. Water stress reduced fruit yield, total dry matter, chlorophyll content and relative water content (RWC), but increased proline accumulation in the melon plants. However, additional supply of K as 3 or 6 mM significantly enhanced all the earlier mentioned physiological parameters, but the values were still not the same as the levels of the control treatment. Water stress also reduced leaf calcium (Ca) and K of the melon plants, but additional supply of K to the root zone increased the levels of both nutrients much higher than those at the control (C) treatment. Our study revealed that additional supply of K improved water stress tolerance in melon plants by enhancing chlorophyll, relative water content and concentrations of some essential nutrients in leaves.  相似文献

Abstract

An experiment was carried out in a controlled temperature (CT) room for five weeks with tomato cvs., Moneymaker, Liberto, and Calypso, to investigate possible relationships between zinc (Zn) deficiency or toxicity and electrolyte leakage in plant leaves. The concentrations of Zn in nutrient solution were 0.01, 0.5, and 5.0 mg L?1, respectively. There were significant reductions in the dry matter and chlorophyll content of all three cultivars grown both at 0.01 (low) and 5 mg L?1 (high) Zn compared to 0.5 mg L?1. The concentration of Zn at 0.01 mg L?1 was not sufficient to provide for optimal plant growth, while 5 mg L?1 in nutrient solution was detrimental to plant growth for all three cultivars. Dry matter production was generally lowest in the plants grown at low (0.01 mg L?1) Zn except for Moneymaker where the lowest biomass was in the high Zn treatment. Zinc concentration was increased in the leaves and roots with increasing Zn concentration in nutrient solution. Phosphorus concentration was toxic in the leaves of the plants grown at low (0.01 mg L?1) and was deficienct at high Zn (5 mg L?1). The electrolyte leakage (%) gradually increased in the plants grown at low and high Zn concentrations and these increases were greatest in the leaves of plants grown at low Zn (except for Moneymaker grown at high Zn where reduction in dry matter was less). The best results for all growth parameters tested were for the plants grown at 0.5 mg L?1 Zn. The results of this short‐term experiment show that electrolyte leakage which is relatively simple and easy to measure may be a good indicator of cultivar tolerance to Zn deficiency and toxicity.  相似文献

The raccoon (Procyon lotor), indigenous to North America, has naturalized in Japan as an invasive alien species, having been introduced into the country in the 1970s. In Hokkaido, the northernmost island of Japan, feral raccoons have been increasing in number and spreading throughout the island. The age at the onset of puberty for raccoons is important for estimating individual lifetime reproductive success and population growth. The present study investigated the timing of and potential factors affecting the onset of puberty in male raccoons in Hokkaido. External characteristics and histology of testes were studied in 151 male feral raccoons and in 1 captive juvenile. For the majority of feral yearling raccoons, prepubertal development began in May, and spermatozoa production began in October prior to their second mating season. However, some larger juveniles attained puberty during the juvenile period. The captive juvenile, which was fed throughout the winter, attained puberty only 11 months after birth. These results suggest that if male raccoons can achieve enough body growth before the first mating season, puberty can be attained early. In both juveniles and yearlings, spermatozoa production was only observed after autumn. This timing coincided with the recrudescence of seasonally active spermatogenesis in adult males. Therefore, attaining puberty in male raccoons appears to require both adequate body nutrient development and several environmental factors that control seasonal testicular changes.  相似文献

In carrot (Daucus carota L.), the taproot colors orange, yellow and white are determined mostly by the Y, Y2, and Or loci. One of the most severe issues in carrot seed production is contamination by wild white carrot. To evaluate the contamination ratio, easily detectable DNA markers for white carrot are desired. To develop PCR-based DNA markers for the Y2 locus, we have re-sequenced two orange-colored carrot cultivars at our company (Fujii Seed, Japan), as well as six white- and one light-orange-colored carrots that contaminated our seed products. Within the candidate region previously reported for the Y2 locus, only one DNA marker, Y2_7, clearly distinguished white carrots from orange ones in the re-sequenced samples. The Y2_7 marker was further examined in 12 of the most popular hybrid orange cultivars in Japan, as well as ‘Nantes’ and ‘Chantenay Red Cored 2’. The Y2_7 marker showed that all of the orange cultivars examined had the orange allele except for ‘Beta-441’. False white was detected in the orange-colored ‘Beta-441’. The Y2_7 marker detected white root carrot contamination in an old open-pollinated Japanese cultivar, ‘Nakamura Senkou Futo’. This marker would be a useful tool in a carrot seed quality control for some cultivars.  相似文献

This research investigated the proximate chemical composition of whiting roe from October to April. The highest water content (83.99%) was found in March, while the lowest (72.40%) was found in December. The highest protein (14.54%) and lipid contents (9.71%) were found in December, while the lowest protein content (7.35%) was found in January and the lowest lipid content (0.38%) in April. Crude ash content of whiting roe changed from 0.86 to 1.49%. The highest (154 kcal/100 g) and lowest energy values (64 kcal/100 g) were determined in December and April, respectively. Whiting roe can be used as a source of valuable nutrients for human nutrition.  相似文献
