As the body of research on the ecosystem service of pollination grows, our ability to tackle a range of agricultural, conservation, and land management issues is limited by our understanding of pollinator foraging patterns and requirements. In particular, better knowledge of which habitats bees utilize for foraging over their lifetime would inform a range of applied and theoretical questions. Traditional methods of studying foraging are either impractical for insects (e.g. radio tracking) or else are limited spatially or temporally (e.g. observations by researchers). Here we describe a method for using stable isotopes of carbon and nitrogen from bee tissues to gain an integrated signal of which habitats a bee has foraged in over its lifetime, using three species of social stingless bee (Apidae: Meliponini) in a fragmented tropical forest landscape in southern Costa Rica as a test case.  相似文献

Calcium uptake by bell pepper (Capsicum annuum L. cv. ‘California Wonder') varied by stage of plant development and N form supplied (NO3 NH4 + ratios: 1:0, 3:1, 1:1, 1:3, and 0:1) in a hydroponic study. Uptake of Ca++ was highest at bloom and during fruit expansion, making the fruit development stage the highest demand period. Calcium uptake declined with each increasing increment of NH4 + relative to NO3 supplied, although fruit yield was not significantly reduced until the ratio of NH4 + to NO3 exceeded 50%. Tissue Ca++ levels in the blossom‐end of the fruit were reduced whenever NH4 + was included with N supplied. Vegetative yield of plants followed the same trend as that observed for total fruit dry weights. Our results indicate that pepper yields are higher when NO3 is the predominant form of N. Also, these results strongly suggest that Ca++ fertilizer applications should precede the bloom period and continue during fruit development to ensure adequate Ca++ availability for fruit development.  相似文献

Abstract

The primary nitrogen forms utilized by plants are ammonium and nitrate. Although the importance of nutrients other than nitrogen for proper turfgrass growth is well established, the amounts of these nutrients in the plant tissue in relation to the use of different N‐forms has not been clearly documented. This study was conducted under greenhouse conditions to determine the effect of N‐form and cutting regime on growth, macronutrient, and micronutrient content of creeping bentgrass (Agrostis palustris Huds. ‘Penncross'). Treatments consisted of 100% NO3? (calcium nitrate), 100% NH4 + (ammonium sulfate), and a 50:50 ratio of NH4 +:NO3 ?. Half the turfgrass plants were maintained at a height of 1 cm (cut), while the other half of the plants were not cut until the end of the study (uncut). The uncut 50:50 treatment yielded the highest shoot, verdure, and total plant dry matter, while the uncut NO3 ? treatment produced the highest root dry matter. The uncut NH4 + treatment yielded the least shoot, root, and total plant dry matter. Plants of the uncut NO3 ? treatment had greater accumulation of macronutrients in the shoot and root tissue compared to plants of the NH4 + treatment. The uncut NO3 ? and 50:50 treatments had higher total accumulation of micronutrients compared to the uncut NH4 +‐treated plants. The cut NO3 ? treatment resulted in the highest macronutrient and micronutrient contents in the root tissue in comparison to other cut treatments. The cut treatments had the highest percentage accumulation of nutrients in the verdure tissue, while the uncut treatments had the highest percentage accumulation of nutrients in the shoot tissue.  相似文献

Brassica oleracea L. are important economic vegetables, and are capable of selenium (Se) enrichment to enhance human nutrition and health. Because Se enrichment may influence the nutrient balance of this crop, a study was done to test the effects of selenate‐Se on plant nutrients. Plants of a rapid‐cycling B. oleracea population were grown in nutrient solutions amended with Na2SeO4at 0.0, 3.0, 6.0, and 9.0 mg L‐1. Leaf tissue was then analyzed for nutrient content. Boron (B) (P=0.01), iron (Fe) (P=0.01), and phosphorus (P) (P=0.01) content decreased, while Se (P=0.01), sulfur (S) (P=0.01), and potassium (K) content (P=0.01) increased with increasing selenate‐Se treatments. Significant quadratic responses were found for magnesium (P=0.01) and molybdenum (P=0.01). No significant differences in leaf fresh or dry weight were detected. Changes in plant nutrient content can be expected when Brassicas are enhanced for delivery of beneficial organic Se.  相似文献

The gene for the porcine beta2-adrenergic receptor (pbeta2AR) was transfected into Chinese hamster ovary (CHO) cells for expression. Fourteen stable cell lines were obtained and exhibited receptor densities ranging from 12 to 2,371 fmol/mg membrane protein. The receptor density was not correlated with estimates of gene copy number obtained by Southern hybridization. The pbeta2AR in CHO cells exhibited saturable binding of [125I]CYP (Kd = 14.5 pM) and stereospecificity for (-)- and (+)-isoproterenol. The relative affinities for (-)-isoproterenol (ISO), (-)-epinephrine (EPI), and (-)-norepinephrine (NEPI) were ISO > EPI > NEPI, which are characteristic of beta2AR. The affinity values for these ligands were similar to those in other species. Binding of ISO, EPI, and NE revealed two affinity states of the betaAR; the high-affinity state was eliminated by adding Gpp(NH)p, a nonhydrolyzable GTP analogue. Binding of the antagonist propranolol modeled to only one affinity state, and Gpp(NH)p did not affect binding. Multiple affinity states are characteristic of agonist-induced coupling of betaAR with G-proteins, and the data suggest that the cloned pbetaAR is functionally competent. Data confirm that the pbeta2AR is the pig version of beta2AR. Stable CHO cell lines will be useful for characterization of pbeta2AR and screening and designing potential drugs that may be used to enhance pig production.  相似文献

Objective To review the breed, age, gender, clinical and laboratory findings, treatment and outcome of horses with caecal disease presented to a referral centre. Design Retrospective study of 96 cases. Procedure The breed, age, and gender of the study population were compared with the corresponding hospital population for the same period. The means (± SD) for clinical and laboratory findings were recorded for each caecal disorder. Treatment was categorised as medical or surgical, and outcome was recorded. Results Caecal diseases included impaction (40% of total cases), rupture associated with concurrent unrelated disease (13%), rupture with parturition (9%), rupture with no associated disease (5%), infarction (11%), torsion (9%), abscess or adhesion (7%), tumour (3%), and miscellaneous conditions (3%). The breed or gender of affected horses did not differ from the hospital population, although horses > 15 years were more frequently represented (P < 0.05). This age group was specifically more predisposed to caecal impaction (P < 0.05), as were Arabian, Morgan, and Appaloosa breeds (P < 0.05). In horses with caecal impaction transrectal examination was the most useful diagnostic procedure; 90% of affected horses treated medically were discharged while horses treated by typhlotomy alone, or typhlotomy and blind end ileocolostomy, had survival rates to discharge of 71% and 86%, respectively. Horses with caecal rupture associated with concurrent un-related disease showed no signs of impending rupture; all were receiving phenylbutazone, all were euthanased, and 50% had caecal ulceration at necropsy. Of horses with caecal rupture with parturition 56% had prior dystocia; in two-thirds the site of rupture was the ileocaecal junction and all were euthanased. Horses with caecal rupture with no associated disease died or were euthanased; rupture was idiopathic. Horses with caecal infarction usually had signs of abdominal pain and abdominal fluid changes consistent with peritonitis; transrectal examination was nonspecific, and typhlectomy was successful in seven of eight horses. Horses with caecal torsion had signs of severe, acute abdominal pain and typhlectomy was successful in three of five horses. Diagnosis of caecal adhesion or abscess was assisted by transrectal palpation in two of seven horses and surgical treatment was successful in two of five horses. A caecal tumour was diagnosed in three horses aged 20 years or older that presented with chronic weight loss. Other caecal diseases were uncommon. Conclusion Caecal disease is uncommon in equids but some specific features of the history and physical findings can alert the veterinarian to the possibility of caecal involvement in horses with gastrointestinal dysfunction. Medical or surgical therapy can be effective in horses where caecal rupture has not occurred.  相似文献

Two experiments were conducted to study gastric and small intestinal digestion of soybean glycinin and beta-conglycinin in preruminant calves fed milk replacers containing a mixture of skim milk powder and antigenic heated soybean flour. In experiment 1, duodenal passage of immunoreactive beta-conglycinin lasted for a much longer time after the morning meal than that of glycinin. Western blotting revealed the early abomasal outflow of glycinin subunits that associated nearly intact basic polypeptides to partially degraded acidic polypeptides. Intact beta-conglycinin was evidenced at most sampling times. In experiment 2, intact basic glycinin (M(r) = 21000) associated with partially digested acidic glycinin (7000 < M(r) < 25000) was demonstrated in ileal digesta up to 8-10 h after the meal. beta-Conglycinin immunoreactivity could not be evidenced by Western blotting in ileal digesta.  相似文献

The molecular basis by which human breast milk supports the development of a protective intestinal microbiome in infants is unknown. After lactose and lipids, human milk oligosaccharides (HMOs) are quantitatively the third largest and most diverse component of breast milk. In this work, glycomic profiling of HMO consumption by bifidobacteria using Fourier transform ion cyclotron resonance mass spectrometry reveals that one species, Bifidobacterium longum biovar infantis ATCC 15697, an isolate from the infant gut, preferentially consumes small mass oligosaccharides, representing 63.9% of the total HMOs available. These HMOs were detected in human breast milk at the onset and constantly through the first month of lactation by use of high performance liquid chromatography-chip time-of-flight mass spectrometry. Further characterization revealed that strain ATCC 15697 possesses both fucosidase and sialidase activities not present in the other tested strains. This work provides evidence that these small mass HMOs are selectively metabolized by select bifidobacterial strains and represent a potential new class of bioactive molecules functioning as prebiotics to facilitate a protective gut colonization in breast-fed newborns.  相似文献
