The persistence of plant-derived recombinant DNA in sheep and pigs fed genetically modified (Roundup Ready) canola was assessed by PCR and Southern hybridization analysis of DNA extracted from digesta, gastrointestinal (GI) tract tissues, and visceral organs. Sheep (n = 11) and pigs (n = 36) were fed to slaughter on diets containing 6.5 or 15% Roundup Ready canola. Native plant DNA (high- and low-copy-number gene fragments) and the cp4 epsps transgene that encodes 5-enolpyruvyl shikimate-3-phosphate synthase were tracked in ruminal, abomasal, and large intestinal digesta and in tissue from the esophagus, rumen, abomasum, small and large intestine, liver, and kidney of sheep and in cecal content and tissue from the duodenum, cecum, liver, spleen, and kidney of pigs. High-copy chloroplast-specific DNA (a 520-bp fragment) was detected in all digesta samples, the majority (89-100%) of intestinal tissues, and at least one of each visceral organ sample (frequencies of 3-27%) from sheep and swine. Low-copy rubisco fragments (186- and 540-bp sequences from the small subunit) were present at slightly lower, variable frequencies in digesta (18-82%) and intestinal tissues (9-27% of ovine and 17-25% of porcine samples) and infrequently in visceral organs (1 of 88 ovine samples; 3 of 216 porcine samples). Each of the five cp4 epsps transgene fragments (179-527 bp) surveyed was present in at least 27% of ovine large intestinal content samples (maximum = 64%) and at least 33% of porcine cecal content samples (maximum = 75%). In sheep, transgene fragments were more common in intestinal digesta than in ruminal or abomasal content. Transgene fragments were detected in 0 (esophagus) to 3 (large intestine) GI tract tissues from the 11 sheep and in 0-10 of the duodenal and cecal tissues collected from 36 pigs. The feed-ingested recombinant DNA was not detected in visceral tissues (liver, kidney) of lambs or in the spleen from pigs. Of note, however, one liver and one kidney sample from the pigs (different animals) were positive for a 278-bp fragment of the transgenic cp4 epsps (denoted F3). Examination of genomic libraries from these tissues yielded no conclusive information regarding integration of the fragment into porcine DNA. This study confirms that feed-ingested DNA fragments (endogenous and transgenic) do survive to the terminal GI tract and that uptake into gut epithelial tissues does occur. A very low frequency of transmittance to visceral tissue was confirmed in pigs, but not in sheep. It is recognized that the low copy number of transgenes in GM feeds is a challenge to their detection in tissues, but there was no evidence to suggest that recombinant DNA would be processed in the gut in any manner different from endogenous feed-ingested genetic material.  相似文献

Ginger oil, obtained by steam distillation of the rhizome of Zingiber officinale Roscoe, is used in the beverage and fragrance industries. Ginger oil displays considerable compositional diversity, but is typically characterized by a high content of sesquiterpene hydrocarbons, including zingiberene, ar-curcumene, beta-bisabolene, and beta-sesquiphellandrene. Australian ginger oil has a reputation for possessing a particular "lemony" aroma, due to its high content of the isomers neral and geranial, often collectively referred to as citral. Fresh rhizomes of 17 clones of Australian ginger, including commercial cultivars and experimental tetraploid clones, were steam distilled 7 weeks post-harvest, and the resulting oils were analyzed by GC-MS. The essential oils of 16 of the 17 clones, including the tetraploid clones and their parent cultivar, were found to be of substantially similar composition. These oils were characterized by very high citral levels (51-71%) and relatively low levels of the sesquiterpene hydrocarbons typical of ginger oil. The citral levels of most of these oils exceeded those previously reported for ginger oils. The neral-to-geranial ratio was shown to be remarkably constant (0.61 +/- 0.01) across all 17 clones. One clone, the cultivar "Jamaican", yielded oil with a substantially different composition, lower citral content and higher levels of sesquiterpene hydrocarbons. Because this cultivar also contains significantly higher concentrations of pungent gingerols, it possesses unique aroma and flavor characteristics, which should be of commercial interest.  相似文献

Water droplets falling onto finished fabrics can create spots which can spoil the appearance of the fabric. This study compares the propensity of softened filament polylactic acid and polyester fabrics to exhibit the adverse affects of ‘water-spotting’, and to identify suitable softeners and methods for their application to minimise and eliminate the problem. The degree of water spotting was greater on softened polylactic acid fabrics than on softened polyester fabrics. Polylactic acid and polyester fabrics with hydrophobic properties did not exhibit any water spotting. Softeners applied by an exhaustion process resulted in a finished fabric which exhibited no water spotting for either dyed polylactic acid or dyed polyester fabrics. The softening active agent was not responsible for the water spotting. The other components in the softener formulation (such as emulsifier, wetting agent), which carries the unfixed dyes with water, were found to be responsible. The water spot halo disappeared after a single machine laundering process.  相似文献

Loss of zoospores has happened independently several times in different phylogenic lines and has, it is claimed, no major phylogenetic significance. But whether or not, how, and under which conditions plant pathogens retain the ability to produce motile asexual spores has fundamental importance from an ecological and epidemiological perspective. Recent molecular investigations of the early evolution of fungi and oomycetes are shedding light on the issue of zoospore loss in organisms able to cause plant diseases. Zoospore loss may have accompanied the development of new forms of dispersal adapted to the terrestrial environment, or the simplification processes which often follow the shift to parasitic or biotrophic life-forms. In this review we consider hybridisation events between Phytophthora species, long distance dispersal of oomycetes, sporangia and zoospore survival, direct and indirect infection processes and newly observed sporulating structures. These aspects are all relevant features for an understanding of the epidemiology of zoosporic plant pathogens. Disease management should not be based on the presumption that the zoosporic stage is a weak link in the life cycle. Oomycete plant pathogens show remarkable flexibility in their life cycles and ability to adapt to changing environmental circumstances.

The giant freshwater prawn Macrobrachium rosenbergii was first introduced into mainland China by the Chinese Academy of Agricultural Sciences from Japan in 1976. It was cultured at the Guangdong Province Fishery Institute (now as Pearl River Fishery Institute, Chinese Academy of Fishery Sciences). Following these early efforts, giant freshwater prawn culture has gradually spread to northern and inland areas. At this time (2011), China has become the largest producer of giant freshwater prawn in the world. Due to a preference of cooking with fresh aquatic products, Chinese retail markets have developed which have special live‐holding counters. From small rural shops to urban supermarkets and specialty restaurants, live seafoods are commonly available whereby customers pick out the products they wish to purchase and consume. Giant freshwater prawns are produced in over 35 countries throughout the world. They are usually sold either fresh, on‐ice, or frozen. What makes the Chinese prawn industry unique is its focus on a live transportation and marketing model. As a result, numerous innovative live‐transport technologies have been developed which have been key to the success of this expanding domestic industry. In this article, we would like to give a general overview of the giant freshwater prawn industry, its status in China, and the special role live‐transportation has for expanding production of giant freshwater prawns.  相似文献

Numerous examples exist of successful mammalian invasive alien species (IAS) eradications from small islands (<10 km2), but few from more extensive areas. We review 15 large‐scale removals (mean area 2627 km2) from Northern Europe since 1900, including edible dormouse, muskrat, coypu, Himalayan porcupine, Pallas' and grey squirrels and American mink, each primarily based on daily checking of static traps. Objectives included true eradication or complete removal to a buffer zone, as distinct from other programmes that involved local control to limit damage or spread. Twelve eradication/removal programmes (80%) were successful. Cost increased with and was best predicted by area, while the cost per unit area decreased; the number of individual animals removed did not add significantly to the model. Doubling the area controlled reduced cost per unit area by 10%, but there was no evidence that cost effectiveness had increased through time. Compared with small islands, larger‐scale programmes followed similar patterns of effort in relation to area. However, they brought challenges when defining boundaries and consequent uncertainties around costs, the definition of their objectives, confirmation of success and different considerations for managing recolonisation. Novel technologies or increased use of volunteers may reduce costs. Rapid response to new incursions is recommended as best practice rather than large‐scale control to reduce the environmental, financial and welfare costs. © 2016 Crown copyright. The horticultural sector has seen much structural change both nationally and internationally over the last decades, but the implications for plant health have been neglected. We review in the context of the risk of emerging plant diseases recent developments including the movement towards a global horticultural market, the rise of the horticultural industry of many developing countries, and the economic integration of the European Union. North America is typically well ahead of other regions in economic developments, and in horticulture this is shown for example by the growing importance of Mexican growers. Asia is rapidly catching up also in horticulture, with China and India becoming key producers. Australia and New Zealand show the impact of change in horticulture extension services. The Eastern enlargement of the EU is having profound influences on fruit and vegetable growers both in the new and in the old member countries. Similar developments are taking place in South America and Africa. In all continents, there is a general trend towards fewer and larger horticultural growers, an increasing role of supermarkets and a concentration of the retail pathways. These developments have consequences for the control of plant pathogens and invasive species. Technical issues seem to be of lesser consequence in terms of structural change compared with labour and trade aspects. However, examples can be found where technical innovations have opened up new opportunities or provided solutions to pressing problems, as can be seen in the hardy nursery stock and ornamental industry in the UK. Future technical, economic and social impacts on the sector are likely to play a key role for securing a diverse and reliable food supply for the still expanding world's population. Recent advances in modelling disease spread in complex networks representing trade pathways should be used to target control of introductions of new plant pathogens. There is a need for more long-term research on how structural change in the horticultural sector will affect and be affected by climate change.  相似文献
