This paper describes the evolution of IPM in Cuba during three pre- and post-revolutionary periods. The state of the art of IPM after the collapse of Cuba's relations with the soviet block is analyzed in detail. During this 'special period' Cuba is undergoing a major conversion from conventional agriculture dependent on external inputs to a semi-organic agriculture dependent on local resources and low external inputs. The massive production and use of microbial pesticides complemented by cultural and habitat management techniques are at the heart of this new quest of agricultural sustainability in the midst of the economic crisis. BACKGROUND
Pyrethroids are among the most applied adulticides worldwide to control mosquito vectors for prevention of arboviral diseases transmission. However, pesticide resistance development in a mosquito population could lead to decreased control efficacy. While most studies investigate the resistant genotype (i.e. kdr, CYP450, etc.) as explanatory variables, few field efficacy studies have measured pesticide quantities deposited at different distances from the sprayer in association with observed mosquito mortality. The current study determined field delivered amounts of an applied ULV permethrin/PBO formulation (31% permethrin + 66% piperonyl butoxide) by GC/MS and estimated practical resistance ratios using caged mosquito females.RESULTS
For field samples, the extraction method recovered 78 ± 3.92–108 ± 8.97% of the permethrin/PBO formulation when utilizing the peaks of PBO from GC/MS to estimate the concentrations of adulticide deposited near the mosquito cages. The field bioassay showed that the spatial distribution of permethrin/PBO formulation was heterogeneous among three pseudo-replicates within the same distance. Within the quantifiable permethrin/PBO range of 15.7–51.4 ng/cm2, field-collected mosquito mortalities started at 64% and linearly increased reaching 100% only in two areas, while all Sebring susceptible mosquitoes died. The field LC95 resistance ratio (RR) of F0 Cx. quinquefasciatus ranged from 2.65–3.51, falling within the 95% CI of RR95 estimated by laboratory vial assays. Tests with and without PBO indicated P450's enzymes contributed to field resistance.CONCLUSION
Results showed the suitability of the collection and quantification method to estimate the field resistance ratio at the applied pesticide rate. Pesticide quantification would also allow the association of the known frequencies of resistance mechanisms (e.g. kdr, CYP450) with field mortalities to estimate the resistance level conferred by such mechanisms. © 2023 The Authors. BACKGROUND: Trifluoromethyl ketones (TFMKs), structurally related to the pheromones, are good inhibitors of pheromone communication in insects. To determine their activity on Zeuzera pyrina L. (Lepidoptera: Cossidae), a polyphagous pest, the authors have prepared two diunsaturated TFMK analogues of the major (3) and the minor (4) pheromone components, and two monounsaturated ones (5, 6). Their biological activity in electroantennogram (EAG), wind tunnel and field tests is presented. RESULTS: The synthetic strategy to obtain the allylic TFMKs 3 and 5 is based on the reactions of diene 10 and 1‐octadecene with trifluoroacetaldehyde ethyl hemiacetal, followed by Dess–Martin oxidation of the resulting homoallylic trifluoromethyl alcohols. In EAG, topical application of analogues 3 and 4 on male antennae significantly reduced the pheromone response. In the wind tunnel, compound 4 reduced the number of contacts with the pheromone source. In the field, traps baited with mixtures of pheromone and inhibitors captured significantly fewer males than the pheromone alone. CONCLUSION: An efficient synthesis of allylic TFMKs is reported, with good overall yield, regiospecificity and diastereoselectivity. These compounds are good inhibitors of the pheromone in electrophysiology, wind tunnel and field tests. The results show the importance of two unsaturations at positions 2 and 13 of the trifluoroacyl group in the structure of the analogues, the latter being critical for inhibitory activity. Trichoderma spp. are common soil fungi used as biocontrol agents due to their capacity to produce antibiotics, induce systemic resistance
in plants and parasitize phytopathogenic fungi of major agricultural importance. The present study investigated whether colonization
of Arabidopsis thaliana seedlings by Trichoderma atroviride affected plant growth and development. Here it is shown that T. atroviride promotes growth in Arabidopsis. Moreover, T. atroviride produced indole compounds in liquid cultures. These results suggest that indoleacetic acid-related indoles (IAA-related indoles)
produced by T. atroviride may have a stimulatory effect on plant growth. In addition, whether colonization of Arabidopsis roots by T. atroviride can induce systemic protection against foliar pathogens was tested. Arabidopsis roots inoculation with T. atroviride provided systemic protection to the leaves inoculated with bacterial and fungal pathogens. To investigate the possible pathway
involved in the systemic resistance induced by T. atroviride, the expression profile of salicylic acid, jasmonic acid/ethylene, oxidative burst and camalexin related genes was assessed
in Arabidopsis. T. atroviride induced an overlapped expression of defence-related genes of SA and JA/ET pathways, and of the gene involved in the synthesis
of the antimicrobial phytoalexin, camalexin, both locally and systemically. This is the first report where colonization of
Arabidopsis roots by T. atroviride induces the expression of SA and JA/ET pathways simultaneously to confer resistance against hemibiotrophic and necrotrophic

We describe a detailed study on the possibility of analyzing aminated pesticides and metabolites using pre-column derivatization with fluorescein isothiocyanate (FITC) and their subsequent separation and detection by cyclodextrin capillary electrophoresis-laser-induced fluorescence (CE-LIF) detection. Different variables affecting the derivatization reaction (pH, FITC concentration, reaction time and temperature) and those related with the separation itself (buffer concentration, addition of various organic modifiers, pH, applied voltage and injection time) were studied. The limit of detection obtained was between 0.45 and 3.48 microg litre(-1) showing a relative standard deviation between 0.26 and 2.08% at a concentration level of 50 microg litre(-1). 相似文献

Exposure to microbes during early childhood is associated with protection from immune-mediated diseases such as inflammatory bowel disease (IBD) and asthma. Here, we show that in germ-free (GF) mice, invariant natural killer T (iNKT) cells accumulate in the colonic lamina propria and lung, resulting in increased morbidity in models of IBD and allergic asthma as compared with that of specific pathogen-free mice. This was associated with increased intestinal and pulmonary expression of the chemokine ligand CXCL16, which was associated with increased mucosal iNKT cells. Colonization of neonatal-but not adult-GF mice with a conventional microbiota protected the animals from mucosal iNKT accumulation and related pathology. These results indicate that age-sensitive contact with commensal microbes is critical for establishing mucosal iNKT cell tolerance to later environmental exposures. 相似文献

The disrupted in schizophrenia 1 (DISC1) gene is a candidate susceptibility factor for schizophrenia, but its mechanistic role in the disorder is unknown. Here we report that the gene encoding phosphodiesterase 4B (PDE4B) is disrupted by a balanced translocation in a subject diagnosed with schizophrenia and a relative with chronic psychiatric illness. The PDEs inactivate adenosine 3',5'-monophosphate (cAMP), a second messenger implicated in learning, memory, and mood. We show that DISC1 interacts with the UCR2 domain of PDE4B and that elevation of cellular cAMP leads to dissociation of PDE4B from DISC1 and an increase in PDE4B activity. We propose a mechanistic model whereby DISC1 sequesters PDE4B in resting cells and releases it in an activated state in response to elevated cAMP. 相似文献

ABSTRACT: Comparative fishing trials were undertaken with two commercial collapsible pots for the crab Charybdis japonica . One pot was box-shaped with two slit entrances at the ends, while the other was dome-shaped with two open funnel entrances. Pots were placed at the bottom of a pond that is connected to Kagoshima Bay. Fishing trials were carried out using a 1-day soaking time; the bait used was mackerel. The box-shaped pot had higher catches of crabs and non-target organisms, but the number of large crabs (carapace width >8 cm) was almost half as many as that caught with the dome-shaped pot. The catch of non-target species was also greatly reduced in the dome-shaped pot, and fish caught were mostly enmeshed in the netting. The reduction in catches of small crabs and non-target organisms was due to the nature of the dome-shaped pot's larger mesh size and open entrances, which allowed for escape. The dome-shaped pot seemed more efficient for harvesting commercial size crabs, while the box-shaped pot seemed better for removing all crab sizes. 相似文献

Red drum (Sciaenops ocellatus) is a euryhaline fish commonly found in the Gulf of Mexico and along the Atlantic coast of North America. Because of high commercial demand and its euryhaline characteristics, aquaculture of this species has diversified from marine to low-salinity aquaculture systems. In recent years, interest in the feasibility of producing red drum in inland freshwater systems has grown and this prompted us to investigate its osmoregulatory capacity after rearing for 8 months in a freshwater aquaculture system. We compared the activities of several genes and enzymes involved in the osmoregulatory process in freshwater-acclimatized (FW) and seawater (SW) red drum. The gene expression profiles were variable: the expression of genes encoding Na+/K+-ATPase (NKA) and the cystic fibrosis transmembrane regulator (CFTR) was slightly higher in SW than FW fish, while phosphoenolpyruvate carboxykinase (PEPCK) and the glucocorticoid receptor messenger RNA (mRNA) levels were higher in FW red drum. The total plasma K concentration was 60.3% lower, and gill NKA activity was 63.5% lower in FW than in SW fish. PEPCK activity was twofold higher in FW than in SW red drum. Similarly, liver glycogen was 60% higher in FW fish. In summary, both gene expression and the enzyme activity data support the phenotypic plasticity of red drum and suggest that the limited capacity for ion homeostasis observed, in particular the low plasma K concentration, was due to the composition of freshwater and does not necessarily reflect a physiological inability to osmoregulate. 相似文献

- 1. The Strobel Meseta, a basaltic plateau of Patagonia (Santa Cruz Province, Argentina), holds thousands of shallow fishless lakes that are prime habitat for many species of waterbirds, including some considered ‘near threatened’. In recent years, several lakes have been stocked with trout which has created uncertainty about the potential effects on the recipient ecosystem.
- 2. Limnological and topographical analyses were performed in a group of 32 lakes of the Strobel Meseta in order to characterize and classify individual lakes of the meseta based on their limnological and topographic features, analyze the association between lake type and use by aquatic birds in general and by the endemic hooded grebe (Podiceps gallardoi) in particular, and evaluate the overlap between trout aquaculture and critical habitat for waterbirds.
- 3. The lakes were classified by multivariate analyses into four characteristic types: turbid, high conductivity lakes (T), small vegetated lakes (SV) and larger lakes which were subdivided into either vegetated (LV) or unvegetated (LU). In general, macrophyte cover was the main classificatory variable, whereas conductivity, pH, surface, and depth contributed moderately. Large vegetated lakes were generally found to be important for waterbirds and provided critical habitat for the hooded grebe, whereas trout farmers largely favoured large unvegetated lakes. However, since some large vegetated lakes have already been stocked, there is some level of geographical overlap between waterbird habitat and trout farming.
- 4. The existence of some level of spatial segregation between production and critical waterbird habitat affords opportunities for designing a spatially‐based management system for trout aquaculture.
