Histological examination of the reproductive tract of the male dog failed to identify glandular tissue other than the prostate gland and its disseminated portion. A biochemical study of the three fractions of the ejaculate demonstrates similarities between the first and the third fractions. The composition of three ejaculates collected over 12 hours indicated that storage of prostatic secretion was a possible explanation for the biochemical differences observed. Interleukin-8 (IL-8), an in vitro and in vivo neutrophil chemoattractant, is expressed at high levels in the lesions observed in bovine pneumonic pasteurellosis. Because of the role of neutrophils in the pathogenesis of pneumonic pasteurellosis, we investigated the relative importance of IL-8 as a neutrophil chemoattractant in this disease. Bronchoalveolar lavage (BAL) fluid was harvested from calves experimentally infected with bovine herpesvirus-1 and challenged with Mannheimia haemolytica. Neutrophil chemotactic activity was measured in pneumonic BAL fluid samples treated with a neutralizing monoclonal antibody to ovine IL-8, and compared to the activity in samples treated with an isotype-matched control antibody. Bronchoalveolar lavage fluid was analyzed at a dilution which induced a half-maximal response, and the concentrations of antibody were optimized in a preliminary experiment. Following incubation of replicate samples of diluted pneumonic bovine BAL fluid with 70 microg/mL of IL-8-neutralizing antibody or control antibody, the neutrophil chemotactic activities of the samples were determined using an in vitro microchemotaxis assay. Overall, pretreatment of BAL fluid samples with neutralizing anti-IL-8 antibody reduced neutrophil chemotactic activity by 15% to 60%, compared to pretreatment with control antibody. This effect was highly significant (P < 0.001), and was present in 5 of 5 samples. These data indicate that IL-8 is an important neutrophil chemoattractant in calves with pneumonic pasteurellosis, but that mediators with actions redundant to those of IL-8 must also be present in the lesions. 相似文献

The present study was conducted to examine the effect of high heparin concentration on capacitation of buffalo spermatozoa with a short incubation time. Frozen thawed spermatozoa from three buffalo bulls were pooled and treated with either 50, 100 or 200 microg/ml heparin for 30 min. Capacitation was evaluated by acrosome reaction of spermatozoa and in vitro fertilization rate (per cent cleavage rate, per cent cleavage index). Acrosome reaction was induced in heparin treated spermatozoa with calcium ionophore A23187 and staining was carried out with Coomassie G-250 to evaluate the response as compared with control (0 heparin + calcium ionophore). Significantly higher percentage of acrosome reaction (AR) spermatozoa was noted after heparin treatment (36.8-48.2%) as compared with control (8.1% ; p < 0.05) but differences among the three heparin concentrations were non-significant. However, a significantly higher in vitro fertilization rate was recorded in spermatozoa capacitated by 50 and 100 microg/ml heparin (80.4 and 75.9% cleavage rate, respectively) as compared with 200 microg/ml heparin (47.2% cleavage rate; p < 0.001). It is concluded that buffalo spermatozoa capacitated with 50-100 microg/ml heparin had significantly higher ability to improve in vitro fertilization rate in buffalo. 相似文献

A number of monkey species, including African green monkeys and African vervet monkeys (Chlorocebus aethiops), are frequently infected in the wild and in captivity with a Simian immunodeficiency virus strain, SIVagm, a primate lentivirus. Up to 50% of African green monkeys are estimated to be infected with SIVagm. SIV strains are very closely related to HIV-2 strains, which are a cause of AIDS in humans, predominantly in western Africa, although cases in Australia have also been reported. It is generally thought that SIV is non-pathogenic in several natural hosts, including African green monkeys. Nevertheless many SIV strains induce a profound immunodeficiency virtually identical to HIV-1 induced AIDS in humans when administered to Asian macaque species such as rhesus (Macaca mulatta) or pigtailed macaques (M nemestrina). SIV infection of Asian macaque species is frequently employed as an animal model for AIDS vaccine studies. In November 1996 a group of 10 African vervet monkeys were imported from the USA for display at Victoria's Open Range Zoo in Werribee. Two animals in this group of monkeys later developed a fatal gastroenteric illness. These diagnoses led us to initiate SIV testing of the colony. 相似文献

The palatine tonsils are part of the mucosa-associated lymphoid tissue (MALT), strategically located in the oropharynx at the entrance of respiratory and gastrointestinal tracts, and are recognized portals of entry and sites of multiplication and persistence of several pathogens in pigs. As the tonsillar crypt epithelium is in close contact with external environment and the underlying lymphoid tissue, the characterization of the intra-epithelial lymphocyte subpopulations is essential for the understanding of initial steps of pathogenesis of several diseases. In this work we investigated specific lymphocyte subsets in the tonsillar crypt epithelium of 10 adult healthy pigs, using monoclonal antibodies against lymphocyte markers CD3, CD4, CD8, gammadelta T cell receptor and immunoglobulin light-chain in an avidin-biotin immunoperoxidase technique. The crypt epithelium was usually extensively infiltrated by a diverse population of T cells and by B cells. The degree of infiltration of each subset was variable among animals and within individual animals. In the T cell population CD4 cells and gammadelta TCR cells predominated over CD8 cells. These data suggest that the crypt lymphoepithelium is capable of participating in both cellular and humoral immune responses and that gammadelta T cells may play an important role in the defense of this mucosa. 相似文献

Extensive data on the trajectories of icebergs located off the coast of Labrador has yielded information on tidal currents, the effect of wind on iceberg motion, and the properties of the low-frequency eddies. Statistical properties of the data were shown to be related to theoretical ideas on two-dimensional turbulence and to the connection between Eulerian and Lagrangian velocity statistics. The results have led to an estimate of the cross-shelf mixing rate and a statistical prediction scheme for iceberg trajectories. 相似文献

Whole genome comparisons identified introgression from archaic to modern humans. Our analysis of highly polymorphic human leukocyte antigen (HLA) class I, vital immune system components subject to strong balancing selection, shows how modern humans acquired the HLA-B*73 allele in west Asia through admixture with archaic humans called Denisovans, a likely sister group to the Neandertals. Virtual genotyping of Denisovan and Neandertal genomes identified archaic HLA haplotypes carrying functionally distinctive alleles that have introgressed into modern Eurasian and Oceanian populations. These alleles, of which several encode unique or strong ligands for natural killer cell receptors, now represent more than half the HLA alleles of modern Eurasians and also appear to have been later introduced into Africans. The provision of analgesia at the time of marking has been adopted by the Australian sheep industry, but data on production benefits are lacking. In the current study, alternate lambs were provided with either meloxicam (non‐steroidal anti‐inflammatory drug [NSAID], n = 781) or no analgesia (NONE, n = 822) at the time of ring castration and tail docking. Six distinct management groups of lambs were studied. Lambs were weighed immediately before marking and then again at weaning. There was no significant effect of treatment on average daily gain between marking and weaning in cross‐bred lambs. In Merino lambs, average daily gain was 5 g/day lower (P < 0.005) in lambs receiving NSAID, but this may not be biologically significant. Lamb losses were significantly (P < 0.05) lower in the NSAID group (1.1%) than in the NONE group (2.7%). This observation is worth validating in larger studies, particularly considering that lamb mortality is a significant cost to production and welfare concern. 相似文献

Regional oxidant distributions produced under various atmospheric conditions and emission scenarios are investigated using the Regional Acid Deposition Model (RADM). RADM is a complex, evolving three-dimensional Eulerian model that describes the chemistry, transport and deposition of tropospheric trace species including SO2, sulfate, NO x and volatile organic compounds as well as O3, other major oxidants and acids. The model calculates the short-term temporal evolution of atmospheric trace gas concentrations and their deposition on the regional scale. This study is focused on oxidant production in the eastern United States and southeastern Canada. The influence of atmospheric conditions is explored by comparing three characteristic winter, summer and spring/fall cases. Base-case 1985 emissions of SO x , NO x , volatile organic compounds (VOCs), NH3 and CO are specified using the comprehensive pollutant emissions inventory developed as part of the National Acid Precipitation Assessment Program (NAPAP). The perturbed case, which represents projected anthropogenic emission changes for 2010, indicates changes in daily total 80 km grid average NO x emissions ranging from increases of 75% to decreases of 45% and VOC emission changes ranging from increases of 65% to decreases of 20%. The largest NO x emission changes occur in the northeast, and the largest VOC changes occur in the Gulf Coast area. Ground level grid average midday O3 concentrations for the 1985 emission cases are highest (on the order of 70 to 100 ppb) in the New York City and Houston metropolitan areas for the summer and spring cases; the summer case also indicates relatively high grid average O3 concentrations of greater than 80 ppb in the southeast. Winter case values are much lower than summer O3 values throughout the region, with highs of 40 to 50 ppb occurring in the southeast and the Great Lakes area. Changes in NO x and other emissions under the complex 2010 emissions scenario for the summer case result in maximum O3 concentration reductions of 10% in the Houston area and increases in O3 of a few percent in some rural areas of the southeast. This study underscores the need for more comprehensive assessment of the complex relationships among regional emission changes, oxidant production and atmospheric conditions. 相似文献
