Alternative pre-messenger RNA (pre-mRNA) splicing plays important roles in development, physiology, and disease, and more than half of human genes are alternatively spliced. To understand the biological roles and regulation of alternative splicing across different tissues and stages of development, systematic methods are needed. Here, we demonstrate the use of microarrays to monitor splicing at every exon-exon junction in more than 10,000 multi-exon human genes in 52 tissues and cell lines. These genome-wide data provide experimental evidence and tissue distributions for thousands of known and novel alternative splicing events. Adding to previous studies, the results indicate that at least 74% of human multi-exon genes are alternatively spliced.  相似文献

Ostertagia circumcincta larvae were administered to adult immune ewes either during the breeding period or around parturition to investigate whether reproductive performance was affected. Two groups of 25 ewes were given water or 12,000 ostertagia larvae three times weekly for five weeks before and five weeks after breeding to synchronised oestrus. Plasma pepsinogen levels were raised in the larval-challenged ewes during the treatment period but no significant effect of treatment was observed on ovulation rate or plasma progesterone concentration. Neither ewe live-weight nor body condition score differed between groups throughout gestation or for four weeks post partum and there was no effect from larval challenge during the breeding period on lambing rate, lamb birthweight or liveweight at four weeks old. A second experiment involved 20 twin-bearing immune ewes which were administered 17,000 ostertagia larvae three times weekly for three weeks before and three weeks after parturition. Plasma pepsinogen concentration was elevated during the larval challenge period and nematode eggs were detected in the faeces after parturition but there was no effect from treatment either on ewe liveweight and body condition score or on lamb birthweight and lamb liveweight at four weeks old. These results indicate that larval challenge with ostertagia larvae at dose rates comparable to larval intake from heavily contaminated pasture did not affect the general well being and reproductive performance of immune Greyface ewes.  相似文献

In May 1985 four groups of 10 calves, aged between four and five months, were turned out on to separate, permanent pastures of equal area which had been seeded during the previous few days with larvae of Dictyocaulus viviparus. One group acted as a control, the second was vaccinated with lungworm vaccine before turnout and treated with thiabendazole three, eight and 13 weeks after turnout, while the third and fourth groups were given ivermectin three times (three, eight and 13 weeks after turnout) and twice (three and eight weeks after turnout), respectively. A severe outbreak of parasitic bronchitis resulted in the death of three control calves within five weeks of turnout and parasitic bronchitis and gastroenteritis affected the second group of calves after approximately four months at pasture. The calves given ivermectin excreted no lungworm larvae and remained free of clinical parasitism throughout the trial.  相似文献

Calves which had received daily infections of Cooperia oncophora over a six week period showed inappetence, weight loss, impaired nitrogen retention and a loss of plasma proteins into the gut. During this period the infections occurred in the duodenum, jejunum and ileum and many larval stages were present in the mucosa of the small intestine. At necropsy, on weeks 3, 6 and 10 after the first infection, stunting and thickening of the villi were noted with excessive mucus production. Prior administration of a morantel slow release bolus prevented these changes. By week 12 most of the worms had been expelled yet a loss of plasma proteins to the gut was still occurring.  相似文献
