OBJECTIVE: To assess the validity and reliability of the most recent adaptation of Block's full-diet food-frequency questionnaire (FFQ) among a sample of Canadian women. DESIGN: Participants completed a self-administered FFQ (FFQ1), two unannounced 24-hour recalls (weekday and weekend) and a second FFQ (FFQ2) between October 2003 and February 2004. FFQs and recalls were analysed for 32 nutrients using Block Dietary Data Systems and the University of Minnesota's Nutrient Data System. Mean and median intakes were computed, along with crude and deattenuated Pearson correlation coefficients between FFQ1 and the average of two recalls (validity) and between FFQ1 and FFQ2 (reliability). SETTING: Ontario, Canada. SUBJECTS: A random population-based sample (n = 166) of women aged 25 to 74 years. RESULTS: One hundred and fifteen (69%) women completed FFQ1, 96 completed FFQ1 and both recalls, and 93 completed both FFQs, about 56 days apart. Mean intakes were similar for most nutrients. FFQ reliability was high, with Pearson correlation coefficients having a median of 0.75, ranging from 0.57 to 0.90 (macronutrients) and from 0.65 to 0.88 (micronutrients from supplements and food). FFQ validity was moderate to high, with deattenuated Pearson correlation coefficients having a median of 0.59, ranging from 0.11 to 0.73 (macronutrients) and from 0.50 to 0.76 (micronutrients from supplements and food). Our micronutrient correlations were similar to or higher than those of other studies that included supplements. Two correlations <0.40 were associated with fats. CONCLUSIONS: The validity and reliability of this full-diet version of the Block FFQ were moderate to high, supporting its use in future studies among Canadian women.  相似文献

In this study three different approaches were employed to identify key odorants in Sauvignon blanc wines. First, the concentrations of the odorants were compared to their respective aroma detection thresholds. The resulting odor activity values (OAV) were transformed into a normalized and weighted measure that allows the aroma profiles of different wines to be compared and the contribution of a single aroma in a complex mixture to be evaluated. Based on their OAV, 3-mercaptohexanol and 3-mercaptohexyl acetate were the two most important aroma compounds in many Marlborough Sauvignon blanc wines. Due to limitations with the OAV approach, the study was extended to include aroma extract dilution analysis (AEDA), which revealed that β-damascenone, together with the varietal thiols, esters, and higher alcohols, are key odorants in Sauvignon blanc wines. The final approach undertaken was aroma reconstitution and omission tests using a deodorized wine base and the creation of a model Marlborough Sauvignon blanc. Single compounds and groups of compounds were omitted from the model to study their impact on the sensory properties of the model wine. Reconstitution and omission confirmed that varietal thiols, esters, terpenes, and β-damascenone are all important contributors to Sauvignon blanc aroma. The methoxypyrazines showed an important but relatively low impact in all three of the approaches undertaken in this study.  相似文献

In this study, in-house isolated laccase isoforms, i.e., Lac-I and Lac-II of the basidiomycete Pycnoporus sanguineus (CS43), were evaluated in relation to their Remazol Brilliant Blue R (RBBR) dye degradation capacity. A modified Dhouib medium additionally supplemented with 3% ethanol as a secondary inducer was used to propagate P. sanguineus CS43 for enhanced production of laccase under liquid state fermentation. The crude laccase extract was purified by passing through ion exchange diethylaminoethanol (DEAE)-Sepharose and gel filtration-based Sephadex G-200 column chromatography. The purified laccase fractions were subjected to the electrophoresis, and sodium dodecyl sulfate-polyacrylamide gel electrophoresis analysis revealed two laccase isoforms Lac-I and Lac-II with 66 and 68 kDa, respectively. To explore the industrial applicability, for RBBR dye, degradation efficiencies ranged from 82 to 88% after 3 h of incubation for both; Lac-I and Lac-II at both concentrations were recorded. However, with 8 U/mL, the degradation ranged between 70 to 80% during the first 5 min of incubation. Enhanced degradation of RBBR dye was obtained in the presence of violuric acid and N-hydroxypthalamide as laccase mediators. Finally, using RBBR as a substrate kinetic characterization of both Lac-I and Lac-II isoforms was performed that revealed K m (0.243 and 0.117 mM for Lac-I and Lac-II) and V max (1.233 and 1.012 mM/Sec for Lac-I and Lac-II) values, respectively.  相似文献

The composting of organic waste materials using earthworms yields the value-added product vermicompost (VC). The use of VC as a growing substrate amendment for containerized horticultural crop production is a sustainable nutrient management approach which can benefit crop production by providing plant nutrients and improving crop growth. In order to provide nutrient management guidelines for users of VC amended container substrates, a study was conducted to quantify nutrients supplied to substrates solution and to develop nutrient release kinetic models to predict plant available nutrient based on total initial loading rates. Experimental treatments and results were expressed on a volume basis (4-L container) for easy interpretation and application by growers of crops in containers.  相似文献

The selective fermentation by human gut bacteria of gluco-oligosaccharides obtained from the reaction between the glucosyl group of sucrose and cellobiose, catalyzed by dextransucrases (DSR) from Leuconostoc mesenteroides , has been evaluated. Oligosaccharides were fractionated according to their molecular weight, and their effect on the growth of different bacterial groups was studied. To determine the structure (position and configuration of glycosidic linkages)-function relationship, their properties were compared to those of DSR maltose acceptor products (DSRMal) and of recognized prebiotic carbohydrates (fructo-oligosaccharides, FOS). Cellobiose acceptor products (DSRCel) showed bifidogenic properties similar to those of FOS. However, no significant differences related to molecular weight or isomeric configurations were found for DSRCel and DSRMal products.  相似文献

Relatively little is known about soil organic carbon (SOC) dynamics in montane ecosystems of the semi-arid western U.S. or the stability of current SOC pools under future climate change scenarios. We measured the distribution and quality of SOC in a mosaic of rangeland-forest vegetation types that occurs under similar climatic conditions on non-calcareous soils at Utah State University's T.W. Daniel Experimental Forest in northern Utah: the forest types were aspen [Populus tremuloides] and conifer (mixture of fir [Abies lasiocarpa] and spruce [Picea engelmannii]); the rangeland types were sagebrush steppe [Artemisia tridentata], grass-forb meadow, and a meadow-conifer ecotone. Total SOC was calculated from OC concentrations, estimates of bulk density by texture and rock-free soil volume in five pedons. The SOC quality was expressed in terms of leaching potential and decomposability. Amount and aromaticity of water-soluble organic carbon (DOC) was determined by water extraction and specific ultra violet absorbance at 254 nm (SUVA) of leached DOC. Decomposability of SOC and DOC was derived from laboratory incubation of soil samples and water extracts, respectively.

Although there was little difference in total SOC between soils sampled under different vegetation types, vertical distribution, and quality of SOC appeared to be influenced by vegetation. Forest soils had a distinct O horizon and higher SOC concentration in near-surface mineral horizons that declined sharply with depth. Rangeland soils lacked O horizons and SOC concentration declined more gradually. Quality of SOC under rangelands was more uniform with depth and SOC was less soluble and less decomposable (i.e., more stable) than under forests. However, DOC in grass-forb meadow soils was less aromatic and more bioavailable, likely promoting C retention through cycling. The SOC in forest soils was notably more leachable and decomposable, especially near the soil surface, with stability increasing with soil depth. Across the entire dataset, there was a weak inverse relationship between the decomposability and the aromaticity of DOC. Our data indicate that despite similar SOC pools, vegetation type may affect SOC retention capacity under future climate projections by influencing potential SOC losses via leaching and decomposition.  相似文献

The empirical calibration of Granier-type heat dissipation sap flow probes that relate temperature difference (DeltaT) to sap velocity (v) was reevaluated in stems of three tropical tree species. The original calibration was confirmed when the entire heated probe was in contact with conducting xylem, but mean v was underestimated when part of the probe was in contact with nonconducting xylem or bark. Analysis of the effects of nonuniform sap velocity profiles on heat dissipation estimates showed that errors increased as v and the proportion of the probe in nonconducting wood increased. If half of a 20-mm probe is in sapwood with a v of 0.15 mm s(-1) and the other half is in nonconducting wood, then mean v for the whole probe can be underestimated by as much as 50%. A correction was developed that can be used if the proportion of the probe in nonconducting wood is known. Even with the entire heated probe in contact with conducting xylem, v would be underestimated when radial velocity gradients are present. In this case, the error would be smaller except when velocity gradients are very steep, as can occur in species with ring-porous wood anatomy. Errors occur because the relationship between DeltaT and v is nonlinear. Mean DeltaT along the probe is therefore not a measure of mean v, and users of heat dissipation probes should not assume that v is integrated along the length of the probe. The same type of error can occur when DeltaT is averaged through time while v is changing, but the error is small unless there are sudden, step changes between zero and high sap velocity. It is recommended that relatively short probes (20 mm or less) be used and that probes longer than the depth of conducting sapwood be avoided. Multiple probes inserted to a range of depths should be used in situations where steep gradients in v are expected. If these conditions are met, heat dissipation probes remain useful and widely applicable for measuring sap flow in woody stems.  相似文献

The seeds of peas (Pisum sativum) and chickpeas (Cicer arietinum) expressing a gene for α-amylase inhibitor-1 (αAI) from the common bean (Phaseolus vulgaris) are protected from damage by old world bruchids (pea and cowpea weevils). Here, we used electrospray ionization time-of-flight mass spectrometry to compare the post-translational modifications of αAI from transgenic sources with the processed forms of the protein from several bean varieties. All sources showed microheterogeneity with differences in the relative abundance of particular variants due to differences in the frequency of addition of glycans, variable processing of glycans, and differences of C-terminal exopeptidase activity. The structural variation among the transgenics was generally within the range of the bean varieties. Previously, mice showed allergic reactions following ingestion of transgenic pea αAI but not bean αAI. Here, only minor differences were observed following intraperitoneal sensitization. Both of the transgenic pea and bean forms of αAI elicited Th1 and Th2 antibody isotype responses, suggesting that both proteins are immunogenic and could potentially be allergenic.  相似文献

Concentrations of Cl, total ammonia (TNH3), NO3 plus NO2, total P (TP), and soluble reactive P (SRP) were measured at two sites, located 5 km apart, on Ninemile Creek, New York, for a period of more than 8 mo. The sites bound the most recently formed Solvay waste beds, associated with the production of soda ash, that adjoin the creek. Concentrations of Cl and T-NH3 increased on average by factors of 16.1 and 7.6, respectively, over the monitored stream reach. The estimated average loadings of these materials to the stream over this reach were 2.3 × 105 and 1.2 × 102 kg d?1, respectively. These inputs are attributable to the Solvay waste beds. The loading of Cl from this source has not changed significantly over a 4 yr period since the closure of the soda ash manufacturing facility. This is the single largest source of Cl, and the second largest source of T-NH3, to polluted Onondaga Lake. Profiles of Cl in the lake indicated that at times the creek inflow plunges to subsurface layers as a result of its elevated density. This is at least in part a result of the creek's ionic enrichment. The concentration of SRP decreased by a factor of 2.0 on average over the study reach, probably due to adsorption to the CaCO3 deposits that cover the stream bed in this area. However, the TP load from the creek to the lake is not significantly affected by this phenomenon.  相似文献
