While developing a low-sulphate system combining indirect chromate-reduction by biologically-produced hydrogen sulphide and direct biological chromate-reduction to treat chromate-bearing waters, the aim of the present work was to evaluate the influence of sulphate and H2 starvation on chromate reduction. Chromate-reduction was performed under continuous-feed conditions in a fixed-film column bioreactor originally inoculated with a bacterial consortium containing Desulfomicrobium norvegicum, and fed with H2. With 500 mg l?1 of sulphate in the feed solution, total chromate-reduction was observed in the effluent whereas sulphate-reduction was strongly decreased, as also confirmed by measurements of isotopic ratios for sulphur. In the absence of sulphate, a chromate-reduction activity was still observed but was lower than in the presence of sulphate, and chromate-reduction was H2-dependent. Molecular biology techniques revealed the composition of the bacterial population in the effluent. D. norvegicum together with other micro-organisms of the Bacteria domain were detected. They include members related to the genera Acinetobacter, Acetobacterium and Rhodocyclus. Even when sulphate-reduction was strongly decreased, the presence of sulphate enhances the efficiency of the H2-dependent chromate-reduction. A H2- and CO2-consuming bacterial population may be used in a globally autotrophic process to reduce chromate at low sulphate concentration, thus avoiding excess sulphide production.  相似文献

One hundred and ninety-one sera from horses that recently were exposed to West Nile virus (WNV) by either vaccination or natural infection or that were not vaccinated and remained free of infection were used to evaluate fluorescent microsphere immunoassays (MIAs) incorporating recombinant WNV envelope protein (rE) and recombinant nonstructural proteins (rNS1, rNS3, and rNS5) for detection of equine antibodies to WNV. The rE MIA had a diagnostic sensitivity and specificity, respectively, of 99.3% and 97.4% for detection of WNV antibodies in the serum of horses that were recently vaccinated or naturally infected with WNV, as compared to the plaque reduction neutralization test (PRNT). The positive rE MIA results were assumed to be WNV-specific because of the close agreement between this assay and the PRNT and the fact that unvaccinated control horses included in this study were confirmed to be free of exposure to the related St Louis encephalitis virus. The NS protein-based MIA were all less sensitive than either the rE MIA or PRNT (sensitivity 0-48.0), although the rNSI MIA distinguished horses vaccinated with the recombinant WNV vaccine from those that were immunized with the inactivated WNV vaccine (P < 0.0001) or naturally infected with WNV (P < 0.0001). The rE MIA would appear to provide a rapid, convenient, inexpensive, and accurate test for the screening of equine sera for the presence of antibodies to WNV.  相似文献

A soil column experiment under outdoor conditions was performed to monitor the fate of 14C-ring-labelled sulcotrione, 2-(2-chloro-4-mesylbenzoyl)cyclohexane-1,3-dione and atrazine, 6-chloro-N2-ethyl-N4-isopropyl-1,3,5-triazine-2,4-diamine, in water leachates and in the ploughed horizon of a sandy loam soil. Two months after treatment, the cumulative amounts of herbicide residues leached from the soil were 14.5% and 7% of the applied radioactivity for sulcotrione and atrazine, respectively. Maximum leachate concentrations for each herbicide were observed during the first month following application: 120 and 95 microg litre(-1) for sulcotrione and atrazine respectively. After 2 weeks, 78% of the sulcotrione and atrazine was extractable from the soil, whereas after two months only 10 and 4%, respectively, could be extracted. The maximum sulcotrione content in the first 10 cm of soil was identical with that of atrazine. For both molecules, the content of non-extractable residues was low, being around 15%. Sulcotrione seems to be more mobile than atrazine but the consequences for water contamination are similar since lower doses are used.  相似文献

Two groups of eight honey bee colonies were fed with two different concentrations of imidacloprid in saccharose syrup during summer (each colony was given 1 litre of saccharose syrup containing 0.5 microg litre(-1) or 5 microg litre(-1) of imidacloprid on 13 occasions). Their development and survival were followed in parallel with control hives (unfed or fed with saccharose syrup) until the end of the following winter. The parameters followed were: adult bee activity (number of bee entering the hive and pollen carrying activity), adult bee population level, capped brood area, frequency of parasitic and other diseases, mortality, number of frames with brood after wintering and a global score of colonies after wintering. The only parameters linked to feeding with imidacloprid-supplemented saccharose syrup when compared with feeding with non-supplemented syrup were: a statistically non-significant higher activity index of adult bees, a significantly higher frequency of pollen carrying during the feeding period and a larger number of capped brood cells. When imidacloprid was no longer applied, activity and pollen carrying were re-established at a similar level for all groups. Repeated feeding with syrup supplemented with imidacloprid did not provoke any immediate or any delayed mortality before, during or following the next winter, whereas such severe effects are described by several French bee keepers as a consequence of imidacloprid use for seed dressing in neighbouring cultures. In any case, during the whole study, mortality was very low in all groups, with no difference between imidacloprid-fed and control colonies. Further research should now address several hypotheses: the troubles described by bee keepers have causes other than imidacloprid; if such troubles are really due to this insecticide, they may only be observed either when bees consume contaminated pollen, when no other sources of food are available, in the presence of synergic factors (that still need to be identified), with some particular races of bees or when colonies are not strong and healthy.  相似文献

No histochemical reaction is available to localize diflubenzuron in tissues. The insecticide can be localized in Pieris tissues by its fluoride atoms, used as tracers in secondary ion microanalysis, which is very sensitive for such light elements. In diflubenzuron-treated insects, especially in females, fluoride intensities are much higher than in controls; images demonstrate a wide distribution of the insecticide in the insect tissues. Thus, besides target organs, i.e., epidermis, diflubenzuron is present in other tissues, such as fat bodies, which may be related to the general metabolic effects and to a detoxification of the insecticide.  相似文献

Parturition was induced to 23 sows on the 112th day of pregnancy by intramuscular injection of 7.5 mg PGF2 alpha (Dinoprost-Upjohn Co.). Onset of parturition was achieved within 27 hours and 52 minutes +/- four hours and 54 minutes in 21 animals (91.3 per cent). Average litter size was 11.33 piglets, with parturition time per litter having been five hours and 45 minutes +/- two hours and 39 minutes. Stillbirths accounted for 6.3 per cent. The average birth weight, 1.245 g +/- 264 g did not differ with significance from that of the controls. The physiological process accompanying parturition is described in greater detail. Unobstructed re-intergration with further reproduction of animals which had received PGF2 alpha treatment is established with hard evidence on the basis of clinical examination of oestrus, ovulation, conception, and farrowing rates as well as by histological examination of the endometrium. Alterations in the blood state recorded from animals with PGF2 alpha treatment close to full term were identical with those recorded from untreated animals.  相似文献
