Methods used to prepare antigens from caprine syncytial retrovirus (CSR) for use in the agarose gel immunodiffusion test (AGID) or an indirect enzyme-linked immunosorbent assay (ELISA) are described. Caprine and ovine sera were tested for antibody to CSR using the AGID test and ELISA incorporating a caprine system (CSR antigen and rabbit anti-goat IgG) or an ovine system (maedi-visna virus antigen and rabbit anti-sheep IgG). Good correlation was achieved in the results of the 3 tests when sera were devoid of antibody or were strongly positive. Variations in the results on weakly positive sera were considered to be more a matter of interpretation than due to basic differences in the reagents employed. 相似文献
The effects of extensive sward management and patch size on the persistence and colonization of gaps in sown swards was examined by creating gaps of five different sizes (2·3, 7, 10, 14 and 19 cm in diameter) in four different sward treatments: a fertilized sward grazed to 4 cm, i.e. relatively intensive management, and three extensively managed unfertilized swards, which were not grazed or grazed to 4 cm or 8 cm. The swards were originally sown with ryegrass (Lolium perenne L.) and white clover (Trifolium repens L.), but had developed differences in species composition as a result of the management treatments imposed 2 years before and during the experiment. Light quality measurements, i.e. red-far red (R/FR) ratio, were used to determine when the light environment in the gaps no longer differed from that in control, uncut patches and this was used as an estimate of gap persistence. Persistence of gaps depended on both sward management and gap size. Gaps disappeared most rapidly in the ungrazed sward and fertilized 4-cm sward, and most slowly in the unfertilized 8-cm sward. Small gaps persisted for up to 2 weeks in all but 8-cm swards, whereas larger gaps were estimated to persist for up to 20–25 weeks in unfertilized, grazed swards. There was no evidence that the number of grass or dicotyledonous species increased in the gaps compared with the control areas. There were significant positive linear relationships between the vegetation that developed in gaps and that in the control, uncut patches, reflecting the different species composition of the established sward of the grazed (grass-dominant) and ungrazed (Ranunculus repens-dominant) treatments. For total grass dry matter and tiller numbers, as well as L. perenne tiller numbers, there was a small, but significant, effect of both patch size and sward management on the slopes of the regressions between the controls and gaps. Traditional methods of teaching intracellular biological processes and pathways use figures or flowcharts with the names of molecules linked with arrows. Many veterinary students, presented with such material, simply memorize the names or chemical structures of the molecules and are then likely to forget the material once the examination is completed. To address this problem, the authors designed, created, and field-tested new teaching media that incorporate realistic three-dimensional (3D) animations depicting the dynamic changes that occur in intracellular molecules during cellular activation. Testing found that veterinary students taught using traditional teaching media (e.g., lectures, handouts, textbooks) are proficient in memorizing the names and order of intracellular molecules but unable to appreciate the interactions between these elements or their spatial relationships within cells. In contrast, more than 90% of veterinary students taught using 3D animations not only recall the facts about the intracellular elements but also develop accurate mental images of the interactions among these molecules and their spatial relationships. These findings strongly suggest that the comprehension of complex biological processes by veterinary students can be enhanced by the use of dynamic 3D depictions of these processes in the classroom. 相似文献
The effects of the addition of fatty acids, monoglycerides (MG), and wheat germ oil (WGO) on the level of crystallinity and the crystalline structure of extrusion-cooked wheat starch have been studied using twin-screw extruders. Measurements of water solubility and water absorption indices were made on the extrudates, together with specific mechanical energy (SME) consumption and die pressure for the extruder. MG and the fatty acids added to a level of 4% caused an increase in Vhydrate type crystallinity. WGO addition to a level of 8% caused no change in crystallinity, although the Ehydrate type was favored at lower moisture contents. All additives caused a decrease in SME and an increase or maximum in die pressure. WGO behaved differently than MG and fatty acids in that its addition caused the water solubility index and expansion to increase, as previously observed for other oils added to flours. 相似文献

Test weight and groat proportion are two very important quality characteristics of oat grain. In this study, we pose the hypothesis that these two characteristics are related through characteristics of grain density. Test weight is defined as the product of kernel density and packing proportion. Groat proportion, in theory, is the ratio of the groat mass to the kernel mass. We present two theoretical constructions expressing test weight in terms of groat proportion, packing proportion and kernel density components. To test these, we have applied measurements of test weight, groat proportion, kernel density components, and packing proportion of 18 oat cultivars grown at six environments. Whereas the groat proportion alone accounted for only 34% of the variation in test weight, our theoretical constructions that included groat proportion could account for ≤82% of variation in test weight. Also, we present previously undescribed variation in oat kernel density components across genotypes and environments. Although the kernel density alone could account for most of the variation in test weight across genotypes, packing proportion appeared to be more important in describing variation in test weight of a genotype across different environments. We observed significant variation in both groat and hull density which, together with groat proportion, described most of the variation in kernel density. 相似文献
We evaluated a group-based, participatory training intervention to improve food safety among meat processors and retailers in Bodija Market, Ibadan, Nigeria. An interactive training workshop was held for Butchers Associations’ representatives who were selected to pass on information and training to their group. Meat hygiene knowledge, attitude and practice was assessed before attending the workshop and afterwards (n = 63). It was also assessed for those who did not attend the workshop (n = 68) but were intended recipients of training through their association. Microbiological quality of meat was assessed before and after the workshop (n = 400 samples). After the workshop, participants significantly improved knowledge, attitude and practice in key food safety aspects; specifically, understanding sources of contamination and food-borne diseases, use of bleach and disinfectant and hand washing. Participants also shared information with an average of 18 other group members and improvements were seen in group members who did not attend the workshop but received training through their Butchers Association. Microbiological quality of meat sold also significantly improved after the intervention. In conclusion, participatory, group-based methods targeted at meat processors and retailers can improve meat hygiene knowledge, attitude and practice as well as the microbiological quality and hence safety of meat.
A 3-year-old sexually intact male Bull Mastiff underwent splenectomy for splenic thrombosis; prior to and after splenectomy, multiple blood transfusions were administered. Two weeks after the procedure, T-cell lymphoproliferative disease was diagnosed. Treatment with prednisone and chlorambucil was initiated, and 2 weeks later, cytologic examination of a blood smear revealed small (0.3 microm), coccoid basophilic bodies on the surface of approximately 70% of the RBCs. Morphologically, these resembled "Candidatus Mycoplasma haemominutum." A polymerase chain reaction assay was used to amplify a partial 16S rRNA sequence in blood obtained from the dog; the product was sequenced and compared with 16S rRNA gene sequences of other hemotropic mycoplasmas. The sequence was 98% homologous to that of "Candidatus M haemominutum", but only 77% homologous to that of M haemocanis and M haemofelis. 相似文献
