Dietary acidifiers have been recognized as beneficial in animal production including aquacultural production of fish where they confer such benefits as improved feed utilization, growth, and resistance to bacterial pathogens. If improvements in growth and immune responses by acidifier supplementation can be confirmed in shrimp, then mortalities due to diseases could be minimized, limiting the emergence of disease‐resistant bacterial pathogens as a potential result of antibiotic misuse. With this in mind, a 35‐d feeding trial was conducted to evaluate growth, enteric microbiota populations, and nonspecific immune responses of Litopenaeus vannamei fed diets containing the commercial acidifier Vitoxal, based on acidic calcium sulfate in an indoor temperature‐controlled, recirculating culture system without any natural productivity. Experimental diets were formulated to contain 0 (basal), 0.4, 1.2, 1.6, or 2.0% acidic calcium sulfate (ACS) by weight. Shrimp fed in excess, 15 times a day using automatic feeders. Weight gain and survival among treatments were excellent, but not significantly different (P > 0.05). Denaturing gradient gel electrophoresis analysis revealed that the enteric microbial community of shrimp fed the basal diet differed markedly from those fed the acidifier on the basis of 64.9% similarity coefficient. Shrimp fed the commercial acidifier at 1.2 and 2.0% responded significantly (P < 0.05) better to reduced stress and displayed enhanced immune responses including hemocyte phagocytic capacity, hemolymph protein concentration, hyaline cell counts, and hemolymph glucose, compared with shrimp fed the basal diet. These results point to an enhanced performance in terms of positive shifts in the composition of enteric microbial communities as well as improved immune performance, with no changes in growth or survival. 相似文献

While large grazers can often be excluded effectively from algal aquaculture operations, smaller herbivores such as small crustaceans and gastropods may be more difficult to control. The susceptibility of three Gracilaria species to herbivores was evaluated in multiple-choice experiments with the amphipod Ampithoe ramondi and the crab Acanthonyx lunulatus. Both mesograzers are common along the Mediterranean coast of Israel. When given a choice, the amphipod preferred to consume Gracilaria lemaneiformis significantly more than either G. conferta or G. cornea. The crab, however, consumed equivalent amounts of G. lemaneiformis and G. conferta, but did not consume G. cornea. Organic content of these algae, an important feeding cue for some mesograzers, could not account for these differences. We further assessed the susceptibility of a candidate species for aquaculture, G. lemaneiformis, against local algae, including common epiphytes. When given a choice of four algae, amphipods preferred the green alga Ulva lactuca over Jania rubens. However, consumption of U. lactuca was equivalent to those of G. lemaneiformis and Padina pavonica. In contrast, the crab showed a marked and significant preference for G. lemaneiformis above any of the other three algae offered. Our results suggest that G. cornea is more resistant to herbivory from common mesograzers and that, contrary to expectations, mixed cultures or epiphyte growth on G. lemaneiformis cannot reduce damage to this commercially appealing alga if small herbivores are capable of recruiting into culture ponds. Oat kernel size uniformity is important to the oat milling industry because of the size fractionations that occur in the milling process. We measured frequency of single, double, and triple kernel spikelets and kernel mass of primary, secondary, and tertiary kernels from each spikelet type (for a total of six kernel types) to determine relative influence of kernel type, panicle position, genotype, and environment on oat kernel size for 10 oat cultivars grown at four locations. Kernel type was the most important factor affecting kernel size. Primary kernels from triple kernel spikelets were larger than primary kernels from double kernel spikelets. Tertiary kernels were the smallest. Environments that produced larger kernels also produced higher frequencies of triple kernel spikelets. Some genotypes produced no triple kernel spikelets in any environment, whereas others produced varying proportions, depending on the environment. Kernels closer to the top of the panicle were larger than those near the base. The presence of tertiary kernels was neither associated with lower groat percentages nor with increased proportions of undersized kernels. Effect of corn degermination mill parameters (clearance between mill plates and rpm) were assessed on the broken germ and number of whole kernels in mash so as to optimize the cracking procedure for the intermittent milling and dynamic steeping (IMDS) process. The dynamic steep time and number of intermittent milling stages for the IMDS process were also optimized for maximum starch recovery. A comparison was made between the IMDS and the conventional steeping process for fraction yields. A clearance of 0.45–0.48 cm between the plates gave the most optimum processing conditions (minimum broken germ and least amount of whole kernels in mash after cracking). Effect of rpm on germ damage and kernel cracking was not significant when optimum clearance between the degermination plates was maintained. Two stages of intermittent milling with a dynamic steep time of 30 min or higher was recommended because it produced the highest yield of starch and germ. Comparison of the IMDS process with the conventional wet‐milling process showed that starch and gluten yield increased by 1.6 and 4.26%, respectively, in the IMDS process. Germ recovered from the IMDS process was 0.54% lower than that from the conventional steeping process. 相似文献

Influence of likelihood function choice for estimating crop model parameters using the generalized likelihood uncertainty estimation method 总被引:3,自引:0,他引:3
Proper estimation of model parameters is required for ensuring accurate model predictions and good model-based decisions. The generalized likelihood uncertainty estimation (GLUE) method is a Bayesian Monte Carlo parameter estimation technique that makes use of a likelihood function to measure the closeness-of-fit of modeled and observed data. Various likelihood functions and methods of combining likelihood values have been used in previous studies. This research was conducted to determine the effects of using previously reported likelihood functions in a GLUE procedure for estimating parameters in a widely-used crop simulation model. A factorial computer experiment was conducted with synthetic measurement data to compare four likelihood functions and three methods of combining likelihood values using the CERES-Maize model of the Decision Support System for Agrotechnology Transfer (DSSAT). The procedure used an arbitrarily-selected parameter set as the known “true parameter set” and the CERES-Maize model to generate true output values. Then synthetic observations of crop variables were randomly generated (four replicates) by using the simulated true output values (dry yield, anthesis date, maturity date, leaf nitrogen concentration, soil nitrate concentration, and soil moisture) and adding a random observation error based on the variances of corresponding field measurements. The environmental conditions were obtained from a sweet corn (Zea mays L.) experiment conducted in 2005 in northern Florida. Results showed that the method of combining likelihood values had a strong influence on parameter estimates. The combination method based on the product of the likelihoods associated with each set of observations reduced the uncertainties in posterior distributions of parameter estimates most significantly. It was also found that the likelihood function based on Gaussian probability density function was the best among those tested. This combination accurately estimated the true parameter values, suggesting that it can be used when estimating CERES-Maize model parameters for real experiments. 相似文献

The COVID-19 pandemic is a major human health concern. The pathogen responsible for COVID-19, severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), invades its host through the interaction of its spike (S) protein with a host cell receptor, angiotensin-converting enzyme 2 (ACE2). In addition to ACE2, heparan sulfate (HS) on the surface of host cells also plays a significant role as a co-receptor. Our previous studies demonstrated that sulfated glycans, such as heparin and fucoidans, show anti-COVID-19 activities. In the current study, rhamnan sulfate (RS), a polysaccharide with a rhamnose backbone from a green seaweed, Monostroma nitidum, was evaluated for binding to the S-protein from SARS-CoV-2 and inhibition of viral infectivity in vitro. The structural characteristics of RS were investigated by determining its monosaccharide composition and performing two-dimensional nuclear magnetic resonance. RS inhibition of the interaction of heparin, a highly sulfated HS, with the SARS-CoV-2 spike protein (from wild type and different mutant variants) was studied using surface plasmon resonance (SPR). In competitive binding studies, the IC50 of RS against the S-protein receptor binding domain (RBD) binding to immobilized heparin was 1.6 ng/mL, which is much lower than the IC50 for heparin (~750 ng/mL). RS showed stronger inhibition than heparin on the S-protein RBD or pseudoviral particles binding to immobilized heparin. Finally, in an in vitro cell-based assay, RS showed strong antiviral activities against wild type SARS-CoV-2 and the delta variant. 相似文献
