Verticillium wilt, caused by Verticillium dahliae, is the most serious disease in olive cultivation areas in western Turkey. Two hundred and eight isolates of V. dahliae from olive (Olea europea var. sativa) trees were taken for vegetative compatibility analysis using nitrate non-utilizing (nit) mutants. One isolate did not produce a nit mutant. Nit mutants of 207 isolates were tested against tester strains of internationally known vegetative compatibility groups (VCGs) 1A, 2A, 2B, 3, 4A and 4B, and also paired in many combinations among themselves. One hundred and eighty nine of the isolates (90.9%) were strongly compatible with T9, the tester strain of VCG1A, and thus were assigned to VCG1A. Eight isolates were assigned to VCG2A and four isolates to VCG4B. One isolate was heterokaryon self-incompatible (HSI) and five isolates could not be grouped to any of the VCGs tested. Pathogenicity assays were conducted on a susceptible olive cultivar (O. europea cv. Manzanilla) and a susceptible local cotton cultivar (Gossypium hirsutum cv. Çukurova 1518). Both cotton and olive inoculated with all VCG1A isolates showed defoliating symptoms in greenhouse tests. This is the first report on VCGs in V. dahliae from olive trees in Turkey which demonstrates that VCG1A of the cotton-defoliating type is the most commonly detected form from olive plants in the western part of Turkey.  相似文献

This study describes the immunohistochemical and histochemical phenotypes of inflammatory cells in sheep lungs infected with lungworms. A total of 20 naturally infected sheep lungs were used. Protostrongylus spp., Muellerius capillaris, Neostrongylus linearis, and Cystocaulus ocreatus were the chief organisms determined from such lesions, which were of a chronic nature. All the lungs had many developmental stages of the parasites and a similar inflammatory response, which included numerous mast cells, eosinophils, T cells, B cells, dendritic cells, and macrophages. In the bronchial and interstitial tissues, the inflammatory cells were dominated by MHCII, CD1, CD4, CD5, CD14, CD21, IgM, and CD172a positive cells, whereas CD2 and WC1 positive cells were detected less. The data provided additional evidence that subsets of inflammatory cells were included within ovine lungs infected with lungworms; however, understanding the entire immune-response process and development of resistance to lungworms in sheep remain to be clearly elucidated.  相似文献

In this study, pathological, serological and virological examinations were performed on 15 sheep from a flock of 250 sheep and lambs that suffer from simultaneous naturally occurring BTV, PPRV and SPV outbreaks. SPV was diagnosed macroscopically and histopathologically, BTV was diagnosed by ELISA, and PPRV was diagnosed pathologically and by ELISA. Clinically fever, diarrhea, depression, polypnea, conjunctivitis, lacrimation, rhinitis, erosive stomatitis, edema of eyelids, photophobia, cutaneous eruption with erythematous areas especially noticeable in wool-free parts of the body and axilla lesions evolving into papules were observed. At necropsy, the most effected organs were lungs and gut. Subepicardial hemorrhages were also commonly seen. While typical pox lesions were observed in some lambs, usually fibrinous pleuropneumonia was more prominent lung lesion. SPV and PPRV lesions were seen at the histopathological examination of the lesioned tissues, BT lesions were mild than SPV and PPRV microscopically. Serum and leukocyte samples of 15 animals were examined for PPRV and BTV by ELISA; 5 samples were positive for PPRV and 6 BTV, 4 were positive for both PPRV and BTV simultaneously. One hundred animals died, most were lambs. Mortality rates were 100% in lambs and 80% in the herd.  相似文献

In this report we examined the presence of specific antibodies against equine herpesvirus type 1 (EHV-1), and equine herpesvirus type 4 (EHV-4) in several equidae, including mules, donkeys, horses. The presence of EHV-1 and EHV-4 in respiratory diseases of equids, and ability of multiplex nested polymerase chain reaction (PCR) screening in simultaneous diagnosis of horses acutely infected by EHV-1 and EHV-4 were also investigated. Sera from 504 horses, mules and donkeys sampled were tested for the presence of EHV-1 and EHV-4 specific antibodies. Blood samples taken from 21 symptomatic horses and nasal swabs taken from 40 symptomatic horses were tested for the presence of EHV-1 and EHV-4 by a multiplex nested PCR. A total of 14.3% (3/21) of buffy coat samples and 32.5% (13/40) nasal swab samples were found to contain EHV-1 DNA, while 19% (4/21) buffy coat samples and 22.5% (9/40) nasal swab samples were found to be positive for EHV-4 DNA. By species, 14.5% of horses, 37.2% of mules and 24.2% of donkeys tested were EHV-1 seropositive. EHV-4 specific antibodies were detected in 237 (81.7%) of 290 horse sera tested. Results from this investigation demonstrate that EHV-1 and EHV-4 are prevalent throughout the equid population, and that donkeys and mules might also represent an important source of infection for other equids. We also showed that the multiplex nested PCR assay might be useful for diagnosis of mixed respiratory infections in horses due to EHV-1 and EHV-4.  相似文献

ABSTRACT

Boron (B) toxicity is an important disorder that can limit plant growth on soils of arid and semi arid environments throughout the world. Although of considerable agronomic importance, our understanding of B toxicity is rather fragmented and limited. The effects of increasing levels of B (0, 0.5, 5, 50 mg kg? 1) on plant growth, proline accumulation, membrane permeability, nitrate reductase activity (NRA), and mineral nutrient interactions of tomato and pepper plants were investigated in greenhouse conditions. Increasing levels of B increased the B contents of plants. Boron toxicity symptoms occurred at 5 and 50 mg kg? 1 levels. Fresh and dry weights of the plants clearly decreased with the application of the 50 mg kg? 1 level of B. Membrane permeability and proline accumulation were significantly increased by the 50 mg kg? 1 level of B. Nitrate reductase activity of tomato plants was increased with increasing levels of B. With the exception of potassium (K) and calcium (Ca) in pepper and magnesium (Mg) in tomato, B treatments significantly affected nutrient concentrations of tomato and pepper. Except for sulfur (S) and Ca in tomato, the highest rate of B applied increased the N, phosphorus (P), and K concentrations of tomato and N, P, Mg, and S concentrations of pepper.  相似文献

The objective of the study is to investigate the role of calcium (Ca) and boron (B) synergy in higher maintenance of Ca in fruit buds during senescence. To study the hypothesis, an experiment was conducted for two years in two ‘Starking Delicious’ apple (Malus domastica Borkh.) orchards established on seedling rootstocks in Banaz, Usak, Turkey. Treatments composed of postharvest foliar sprays of Ca at rate of 15.13 kg h?1, B at rate of 2.47 kg h?1 and their combination (Ca+B) compared with control trees sprayed with water. Fruiting spurs, flower ovaries, young and mature fruits were sampled and analyzed for their Ca, B, potassium (K) and magnesium (Mg) contents. Fruit were stored for six months and quality attributes were studied. Results put forth that Ca+B treatments significantly increase fruit Ca and B concentrations and firmness. The results displayed that postharvest B spray improves Ca nutrition of fruits through synergism of Ca+B on Ca translocation.  相似文献

Cells use both deterministic and stochastic mechanisms to generate cell-to-cell heterogeneity, which enables the population to better withstand environmental stress. Here we show that, within a clonal population of mycobacteria, there is deterministic heterogeneity in elongation rate that arises because mycobacteria grow in an unusual, unipolar fashion. Division of the asymmetrically growing mother cell gives rise to daughter cells that differ in elongation rate and size. Because the mycobacterial cell division cycle is governed by time, not cell size, rapidly elongating cells do not divide more frequently than slowly elongating cells. The physiologically distinct subpopulations of cells that arise through asymmetric growth and division are differentially susceptible to clinically important classes of antibiotics.  相似文献

Mucus clearance is the primary defense mechanism that protects airways from inhaled infectious and toxic agents. In the current gel-on-liquid mucus clearance model, a mucus gel is propelled on top of a "watery" periciliary layer surrounding the cilia. However, this model fails to explain the formation of a distinct mucus layer in health or why mucus clearance fails in disease. We propose a gel-on-brush model in which the periciliary layer is occupied by membrane-spanning mucins and mucopolysaccharides densely tethered to the airway surface. This brush prevents mucus penetration into the periciliary space and causes mucus to form a distinct layer. The relative osmotic moduli of the mucus and periciliary brush layers explain both the stability of mucus clearance in health and its failure in airway disease.  相似文献

Butyrylcholinesterase (BChE) and paraoxonase 1 (PON1) are two serum enzymes synthesized by the liver that are related with inflammation. The main objectives of this study were to determine changes in serum BChE and PON1 by using a canine model of endotoxemia, and to evaluate whether choline alters BChE and PON1 activities during inflammation. For this purpose, a total of 20 mongrel dogs were divided into four groups: control, choline (C), lipopolysaccharide (LPS), and LPS+C. Dogs in the control group were injected with 0.9% NaCl (0.2 ml/kg, i.v.). Dogs in C and LPS+C groups received choline chloride (20 mg/kg, i.v., three times with 4 h intervals). Endotoxin was injected (0.02 mg/kg, i.v., once) to the dogs of LPS and LPS+C groups. Statistically significant decreases in BChE and PON1 activities in LPS group were detected 24 and 48 h post injection, respectively. No statistically significant changes in BChE and PON1 activities at different times were detected in control, C, or LPS+C groups. In conclusion, the data obtained in present study revealed a decrease in serum BChE and PON1 activities in dogs during experimentally induced endotoxemia and that choline administration attenuates these changes.  相似文献
