To determine the most effective dose of arabinogalactan-protein (AGP) in regeneration medium, mature embryos of genotypes in three different ploidy levels (Triticum aestivum L. cv. Ikizce-96, Triticum durum Desf. cv. Mirzabey and Hordeum vulgare L. cv. Tokak) were used to establish an efficient plant regeneration system for cereals. Percentage of callus production, capacity of regeneration were calculated, and also culture effect, root, stem, and total plant length of regenerant plants were observed in six different regeneration media (MS control, MS+2, 5, 7, 10, 12 mg L-1 AGP) in these three different genotypes. According to the results, the highest amount of callus production was found in Ikizce-96 as 93.75% using 5 mg L-1 dicamba and 1 mg L-1 kinetin in induction medium. However, the most improved callus was observed in diploid barley Tokak as 179.95 mg in weight and 6.18 mm in diameter, respectively. The highest regeneration capacity was observed in the dose of 5 mg L-1 AGP in MS of all the genotypes and hexaploid wheat Ikizce-96 gave the best results with the highest regeneration capacity and culture effects (94.86 and 92.5%) in the same dose of AGP. These results indicated that effective dose of AGP in regeneration medium increase plant regeneration in calli derived from cereal mature embryos.  相似文献

A total of 101Verticillium dahliae isolates were recovered from cotton plants at 57 sites in the Aegean region of Turkey between 2003 and 2004. Isolates were tested for vegetative compatibility by observing heterokaryon formation among complementary nitrate-nonutilizing (nit) mutants. Forty-six isolates were assigned to VCG 1, 12 to VCG 2A, 33 to VCG 2B and four to VCG 4B. The remaining six isolates could not be tested for vegetative compatibility because of their inability to yieldnit mutants. All isolates recovered were tested for pathogenicity on cotton cultivars Acala SJ-1 and Deltapine 15-21 by the stem-injection method. The isolates of VCG 2 and 4B, irrespective of their origin, induced weak to severe symptoms on cotton and were similar to the previously described cotton non-defoliating pathotype. In contrast, all cotton isolates of VCG1 caused severe foliar symptoms, stunting, defoliation and often death. The development and reproductive rates ofTetranychus cinnabarinus Boisduval (Acari: Tetranychidae), the carmine spider mite, were studied on two strawberry cultivars (Fragaria ×ananassa Duch.; ‘Camarosa’ and ‘Sweet Charlie’) at three NaCl concentrations, 1760, 2400 and 3040 mgl −1, and control. The effects of NaCl application on the contents of plant nutrients, chlorophyll, proline, peroxidase activity (POX) and proteins were assessed. On Camarosa,T. cinnabarinus had a faster development rate with applications of NaCl than without, except for female development at the highest salinity level. The total development time of females increased with the NaCl concentration, whereas total development time of males decreased. The oviposition period and female longevity on Camarosa was significantly longer in the control than with NaCl, whereas daily and total fecundity were significantly higher at all NaCl concentrations than in the control, except for total fecundity at the lowest salinity level. The total development time, oviposition period, female longevity, daily and total fecundity on Sweet Charlie did not differ significantly between NaCl salinity levels and the control. The intrinsic rate of natural increase (r m ) on Camarosa and Sweet Charlie were significantly higher with NaCl than in the control. However, the contents of Na, Cl, P, chlorophyll, proline, POX and protein in the two strawberry cultivars changed depending on NaCl concentration. Eight different pepper genotypes inoculated by two different arbuscular mycorrhizal fungi (AMF) [Glomus intraradices (Gi) and Gigaspora margarita (Gm)] in a growth chamber experiment under normal seedling growing conditions were evaluated for seedling traits, colonization and relative mycorrhizal dependency (RMD). In general, inoculated plants had greater dry weights compared to non-inoculated plants. Five cultivars responded positively to inoculation with AM fungi and three responded negatively. A great variation in mycorrhizal colonization dependency was observed among the pepper genotypes, with the N52 genotype showing the highest RMD and the Karaisali genotype the lowest. RMD and dry weights of pepper genotypes were inversely correlated.  相似文献

The aim of this study was to investigate effects of an organophosphate insecticide, diazinon (DI), on pancreas, and possible ameliorating role of vitamins E and C. We examined both in vivo and in vitro effects of DI on serum activities of alkaline phosphatase (ALP), γ-glutamyltransferase (GGT), amylase, and lipase enzymes. We also evaluated possible ameliorating effects of vitamins E and C combination against DI toxicity and blood levels of thiobarbituric acid reactive substances (TBARS) only in vivo. In vivo experimental groups were: control group, DI-treated group, and DI + vitamins E plus C-treated group. In vitro study groups were: control group and DI-treated group. The biochemical analyses were determined in in vitro experiments at both hour 0 and 24 while in in vivo experiments were determined only at hour 24. Lipase activity and TBARS level were found increased by DI in in vivo experiments while lipase activity was found decreased in in vitro experiments. Amylase and ALP activities were found decreased by DI in both in vivo and in vitro experiments. Also, the combination of vitamins E and C was found to partially improve these disorders. These results suggest that DI treatment causes pancreas damage via increasing oxidative stress in rats, and a combination vitamins E and C can reduce this lipoperoxidative effect.  相似文献

In this study, the prevalence and distribution of various Arcobacter spp. were investigated in samples taken from the cloacae of healthy domestic geese raised in Turkey. A membrane filtration technique with a non-selective blood agar was employed after enrichment in Arcobacter enrichment broth (AEB) to isolate a wide range of Arcobacter spp. In addition, the isolates were characterized phenotypically and identified at species level using a multiplex-PCR assay. A total of 90 cloacal swab samples taken from geese, collected on three farms (18, 25, 47 samples, respectively), were examined. Of the samples examined, 16 (18%) were found positive for Arcobacter. One Arcobacter species was isolated from each bird. Of the 16 Arcobacter isolates, 7 (44%), 7 (44%) and 2 (12.5%) were identified by m-PCR as A. cryaerophilus, A. skirrowii and A. butzleri, respectively. The present study indicates that domestic geese can harbour a variety of Arcobacter spp. in their cloacae. The presence of Arcobacter in geese may be of significance as reservoirs in their dissemination. Detailed research is needed for better understanding of the epidemiology and zoonotic potential of this emerging pathogen.  相似文献

Fig tree mainly spread in regions where Mediterranean climate prevails can be grown over a wide range of soils without any significant deficiency or toxicity symptoms of plant nutrients. Surveys revealed that, however, quality is highly affected by nutrition especially N, K and Ca in fig production for commercial drying. This research work was initiated based on few studies performed on fertilization of fig. The experiment was performed between 2000 and 2002 in three fig orchards of cv. Sar?lop (syn. Calimyrna) in Ayd?n-Turkey. Trees received seven types of fertilization as (i) untreated control; (ii) NPK (430 g N, 200 g P2O5 and 430 g K2O per tree); (iii) NPK + 70 g Ca; (iv) NPK + 140 g Ca; (v) NPK + 280 g Ca; (vi) NPK + 420 g Ca and (vii) N + 420 g Ca, Ca(NO3)2 used as the Ca source. Tested fertilizer applications showed significant differences in respect to yield and quality of fig. Basic NPK fertilization with additional 280 g Ca treatment increased overall quality by reducing the number of fruit with ostiole-end crack and sunscald. Results showed that applied fertilizers exerted significantly in reduction of cull ratio and could alleviate the negative impact of yearly drought conditions.  相似文献

Orthopaedic diseases are common in the pelvic limbs of dogs, and reference values for large muscle groups of the pelvic limb may aid in diagnosis such diseases. As such, the objective of this study was to compare the large muscle groups of the pelvic limb in seven breeds of dogs. A total of 126 dogs from different breeds were included, and the widths of the quadriceps, hamstring and gastrocnemius muscles were measured from images of the lateral radiographies. The width of the quadriceps was not different between the breeds, but the widths of the hamstring and gastrocnemius muscles were significantly different between the breeds. The widest hamstring and gastrocnemius muscles were seen in the Rottweilers and the Boxers, respectively. The narrowest hamstring and gastrocnemius muscles were seen in the Belgian Malinois and the Golden retrievers, respectively. All ratios between the measured muscles differed significantly between the breeds. Doberman pinschers and Belgian Malinois had the highest ratio of gastrocnemius width:hamstring width. Doberman pinschers had also the highest ratio of quadriceps width:hamstring width. German shepherds had the highest ratio of gastrocnemius width:quadriceps width. The lowest ratios of quadriceps width:hamstring width were determined in the German shepherds. The ratios of the muscle widths may be used as reference values to assess muscular atrophy or hypertrophy in cases of bilateral or unilateral orthopaedic diseases of the pelvic limbs. Further studies are required to determine the widths and ratios of the large muscle groups of the pelvic limbs in other dog breeds.  相似文献

This study was conducted in 2007–2008 to determine the effects of different irrigation regimes on yield, quality and water-yield relationships of young dwarf ‘Gala, Galaxy’ apple trees in Isparta-Turkey. Irrigation water was applied based on a ratio of class A pan evaporation (rates of 0.0 (kcp0), 0.25 (kcp1), 0.50 (kcp2), 0.75 c (kcp3), 1.00 (kcp4) and 1.25 (kcp5)) with 5-day intervals. The effects of irrigation regimes on yield and fruit weight were statistically significant. Seasonal evapotranspiration was measured as 246.5–608.2 mm and 289.3–631.9 mm in 2007 and 2008, respectively. Water use efficiency lead to a yield ranging from 0.0023 to 0.0055 t ha?1mm?1 and from 0.0033 to 0.0111 t ha?1mm?1 and irrigation water use efficiency varies from 0.0009 to 0.0073 t ha?1mm?1 and from 0.0045 to 0.0186 t ha?1mm?1 in 2007 and 2008, respectively. The annual average yield response factor, ky, was 1.22. As a result of the research, it was concluded that the irrigation schedule for the 0.75 kcp3 treatment, applying 0.75 of evaporation from class A pan, could be used in the irrigation of young dwarf apple trees without evident reduction in yield and that high water use efficiencies could be obtained.  相似文献
