Green tea has antidiabetic, antiobesity, and anti-inflammatory activities in animal models, but the molecular mechanisms of these effects have not been fully understood. Quantitative real-time polymerase chain reaction (PCR) was used to investigate the relative expression levels and the effects of green tea (1 and 2 g solid extract/kg diet) on the expression of glucose transporter family genes (Glut1/Slc2a1, Glut2/Slc2a2, Glut3/Slc2a3, and Glut4/Slc2a4) and insulin signaling pathway genes (Ins1, Ins2, Insr, Irs1, Irs2, Akt1, Grb2, Igf1, Igf2, Igf1r, Igf2r, Gsk3b, Gys1, Pik3cb, Pik3r1, Shc1, and Sos1) in liver and muscle of rats fed a high-fructose diet known to induce insulin resistance and oxidative stress. Glut2 and Glut4 were the major Glut mRNAs in rat liver and muscle, respectively. Green tea extract (1 g) increased Glut1, Glut4, Gsk3b, and Irs2 mRNA levels by 110, 160, 30, and 60% in the liver, respectively, and increased Irs1 by 80% in the muscle. Green tea extract (2 g) increased Glut4, Gsk3b, and Pik3cb mRNA levels by 90, 30, and 30% but decreased Shc1 by 60% in the liver and increased Glut2, Glut4, Shc1, and Sos1 by 80, 40, 60, and 50% in the muscle. This study shows that green tea extract at 1 or 2 g/kg diet regulates gene expression in the glucose uptake and insulin signaling pathway in rats fed a fructose-rich diet.  相似文献

The trend in wildlife management over the last two decades has been to develop locally based approaches for responsiveness to local conditions, but some state wildlife agencies are finding the amount of staff time required to service this approach prohibitive. Although local engagement strategies have been lauded as assuring that public trust obligations of state government to citizens are met, we can expect that states with a local focus as their operational level of stakeholder engagement may opt to change their approach to reflect their resource limitations. We argue for comprehensive regional level effort to understand stakeholders augmented with local engagement processes where needed to deal with special circumstances in smaller areas within a region. Such an approach can be anticipated to have implications for stakeholder engagement and human dimensions research needs, which we discuss in the context of public trust resource administration and good governance of wildlife resources.  相似文献

The structure of the ion conduction pathway or pore of voltage-gated ion channels is unknown, although the linker between the membrane spanning segments S5 and S6 has been suggested to form part of the pore in potassium channels. To test whether this region controls potassium channel conduction, a 21-amino acid segment of the S5-S6 linker was transplanted from the voltage-activated potassium channel NGK2 to another potassium channel DRK1, which has very different pore properties. In the resulting chimeric channel, the single channel conductance and blockade by external and internal tetraethylammonium (TEA) ion were characteristic of the donor NGK2 channel. Thus, this 21-amino acid segment controls the essential biophysical properties of the pore and may form the conduction pathway of these potassium channels.  相似文献

Voltage-gated sodium channels are transmembrane proteins of approximately 2000 amino acids and consist of four homologous domains (I through IV). In current topographical models, domains III and IV are linked by a highly conserved cytoplasmic sequence of amino acids. Disruptions of the III-IV linker by cleavage or antibody binding slow inactivation, the depolarization-induced closed state characteristic of sodium channels. This linker might be the positively charged "ball" that is thought to cause inactivation by occluding the open channel. Therefore, groups of two or three contiguous lysines were neutralized or a glutamate was substituted for an arginine in the III-IV linker of type III rat brain sodium channels. In all cases, inactivation occurred more rapidly rather than more slowly, contrary to predictions. Furthermore, activation was delayed in the arginine to glutamate mutation. Hence, the III-IV linker does not simply act as a charged blocker of the channel but instead influences all aspects of sodium channel gating.  相似文献

The concept of an ecoregion, a discrete spatial area where species composition is presumed to be relatively homogeneous compared to that between areas, has become an increasingly common conservation tool. We test the effectiveness of one ecoregion delineation (World Wildlife Fund) in capturing patterns of change in species composition of birds, mammals, and trees across the United States (excluding Hawaii) and Canada, and describe the extent to which each ecoregion boundary is concurrent with relatively large changes in species composition. Digitized range maps were used to record presence/absence in 50 × 50 km equal-area grid cells covering the study area. Jaccard’s index of similarity was calculated for all pairs of cells in the same or adjacent ecoregions. The average rate at which similarity declined with geographic distance was calculated using pairs of cells within the same ecoregion (intra-ecoregion turnover) or using pairs of cells in adjacent ecoregions (inter-ecoregion turnover). The intra-ecoregion rate varies widely among ecoregions and between taxa, with trees having a faster rate of similarity decline than mammals or birds. For all three species groups, most ecoregion boundaries have similar rates across them (inter-ecoregion) than that within each adjacent ecoregion (intra-ecoregion), with the exception of zones of transition between biomes and major geographical features. Although the ecoregion concept is useful for many other conservation applications, the lack of systematic, high turnover rates along ecoregion boundaries suggests that ecoregions should not be used as a quantitative basis for delineating geographic areas of a particular taxonomic group.  相似文献

Isolated hepatocytes were prepared from 100- to 125-kg Holstein male calves (n = 10) by perfusion of the caudate process of the caudate lobe of the liver. The 11th or 12th rib on the right side was resected to provide exposure of the caudate process. Complete postsurgical recovery of the donor from partial lobectomy was confirmed by growth data and serum chemical and hematologic criteria. Hepatocytes were isolated under aseptic conditions, using a 2-step collagenase vascular perfusion procedure. Hepatocyte preparations averaged 85% viability, and the yield averaged 1.2 X 10(7) viable hepatocytes/g of (wet weight) liver. Morphologic characteristics of hepatocytes examined under light and scanning electron microscopy were considered normal, except for occasional surface blebs. Freshly isolated hepatocytes in suspension rapidly decreased in viability and xenobiotic metabolizing capacity (aldrin epoxidation and ethoxycoumarin 0-deethylation and 7-hydroxycoumarin glucuronidation and sulfation), and hepatocytes surviving the initial 2 to 3 hours appeared to undergo repair. As an alternative, primary monolayer cultures on collagen-coated plates were evaluated. Hepatocytes attached to the collagen surface within 4 hours and appeared flattened by 12 hours. Although metabolic activity decreased about 30% over 8 hours in culture, the pattern of ethoxycoumarin metabolites was relatively constant. It was not determined to what extent the apparent loss of metabolic capacity was caused by hepatocyte detachment from the collagen surface. Although complicated by the requirement for asepsis, primary cultures were superior to suspensions for xenobiotic metabolism studies in cattle.  相似文献

Administration of 100 ppm fenbendazole to 661 pheasants (Phasianus colchicus colchicus) and 25 partridges (Perdix perdix) for 4 days reduced natural infection with Syngamus trachea and/or Capillaria obsignata and in some cases with Heterakis spp. by more than 90%. A 5-day administration of feed medicated with 30 or 60 ppm fenbendazole resulted in almost 100% efficacy against natural C. obsignata and Heterakis gallinae infection in 343 pullets. The treated pullets were kept under poor hygienic conditions, resulting in slight reinfection, which manifested itself in immature nematode stages at autopsy and in a slight excretion of nematode eggs in individual cases. Fenbendazole treatment effected the clinical recovery of the stocks within a few days and was very well tolerated.  相似文献
