Cylindrocarpon species are known to be a component of the pathogen/pest complex that incites apple replant disease. In South Africa, no
information is available on apple associated Cylindrocarpon species and their pathogenicity. Therefore, these aspects were investigated. Among the isolates recovered from apple roots
in South Africa, four species (C. destructans, C. liriodendri, C. macrodidymum and C. pauciseptatum) were identified using β-tubulin gene sequencing and phylogenetic analysis. This is the first report of C. liriodendri, C. macrodidymum and C. pauciseptatum on apple trees. Cylindrocarpon macrodidymum was the most prevalent. Isolates within each of the four species were pathogenic towards apple seedlings, but varied in their
virulence. With a single exception, all isolates were able to induce lesion development on seedling roots. Only 57% of the
isolates, which represented all four species, were able to cause a significant reduction in seedling weight and/or height.
The greatest seedling growth reductions were caused by two isolates of C. destructans, and one isolate each of C. liriodendri and C. macrodidymum. A quantitative real-time polymerase chain reaction (qPCR) method was developed for simultaneous detection of all four Cylindrocarpon species. qPCR analyses of Cylindrocarpon from the roots of inoculated seedlings showed that the amount of Cylindrocarpon DNA in roots was not correlated to seedling growth reductions (weight and height) or root rot. The qPCR method is, however,
very useful for the rapid identification of apple associated Cylindrocarpon species in roots. The technique may also hold potential for being indicative of Cylindrocarpon disease potential if rhizosphere soil rather than roots are used. 相似文献
Five rye lines exhibiting a wide range of extract viscosities were evaluated for the rheological and baking properties of their flours, individually and in blends with hard red spring wheat flour. Commercial cultivars of rye and triticale were included in the study as controls. Extract viscosities of rye flours were higher than those of corresponding wholemeals, indicating shifting of water-extractable arabinoxylan into flour during roller milling. Falling numbers of the rye flours correlated positively with their extract viscosities in the presence (r = 0.73, p < 0.05) or absence (r = 0.65, p < 0.05) of an enzyme inhibitor. Farinograms revealed the weakness of rye and triticale flours compared to wheat flour. Extract viscosities of rye flours were negatively correlated (r = -0.65, p < 0.05) with mixing tolerance index and positively correlated (r = 0.64, p < 0.05) with dough stability, suggesting a positive impact of extract viscosity on dough strength. Extract viscosity was negatively correlated (r = -0.74, p < 0.05) with loaf volume and specific volume (r = -0.73, p < 0.05) and positively correlated (r = 0.73, p < 0.05) with loaf weight of rye/wheat bread. Overall, the results indicated that 30% of flour from high or low extract viscosity rye could be incorporated into rye/wheat breads without seriously compromising bread quality. Inclusion of rye, particularly high extract viscosity rye, in chick diets seriously impeded growth performance and feed efficiency. Part of the arabinoxylan survived bread-making and exerted an effect on chicks, although substantially lower digesta viscosities were observed in chicks fed rye bread diets than in those fed rye wholemeals. 相似文献
In Uganda, control of vector-borne diseases is mainly in form of vector control, and chemotherapy. There have been reports
that acaricides are being misused in the pastoralist systems in Uganda. This is because of the belief by scientists that intensive
application of acaricide is uneconomical and unsustainable particularly in the indigenous cattle. The objective of this study
was to investigate the strategies, rationale and effectiveness of vector-borne disease control by pastoralists. To systematically
carry out these investigations, a combination of qualitative and quantitative research methods was used, in both the collection
and the analysis of data. Cattle keepers were found to control tick-borne diseases (TBDs) mainly through spraying, in contrast
with the control of trypanosomosis for which the main method of control was by chemotherapy. The majority of herders applied
acaricides weekly and used an acaricide of lower strength than recommended by the manufacturers. They used very little acaricide
wash, and spraying was preferred to dipping. Furthermore, pastoralists either treated sick animals themselves or did nothing
at all, rather than using veterinary personnel. Oxytetracycline (OTC) was the drug commonly used in the treatment of TBDs.
Nevertheless, although pastoralists may not have been following recommended practices in their control of ticks and tick-borne
diseases, they were neither wasteful nor uneconomical and their methods appeared to be effective. Trypanosomosis was not a
problem either in Sembabule or Mbarara district. Those who used trypanocides were found to use more drugs than were necessary. 相似文献
An observational longitudinal study was carried out on 92 randomly selected smallholder farms in two coastal lowland zones of Kwale District in Kenya between December 1997 and November 1999. The objective was to estimate the incidence of the main vector-transmitted diseases in pre-weaned calves. From an initial 41 pure or cross-bred Bos taurus calves which were less than 2 months and whose birth and disease histories were known, study calves were recruited progressively and monitored until they were weaned at around 146 days. Overall, 130 calves in 67 farms were monitored and these contributed a total risk period of 30,062 days.
Disease parameters were analysed and compared as true annual and age-specific incidence rates. The incidences of East Coast fever (ECF) (23.1%) and trypanosomosis (29.1%) were the highest among the vector-borne diseases. The corresponding mortality incidence rates of ECF and trypanosomosis were 10.9 and 3.6%, respectively. The annual incidence rates of anaplasmosis and babesiosis were 10.9 and 1.2%, respectively. There was no mortality arising specifically from anaplasmosis or babesiosis. Analysis of survival times to natural infection indicated that the field challenge resulting to cases of trypanosomosis was much higher compared to the risk of either ECF or anaplasmosis. It was concluded that these vector-borne diseases constrain production of replacement stock in this coastal lowlands region of Kenya. 相似文献
Most animals adapt readily to increased supplies of carbohydrate in the intestinal lumen by increasing enzymes for degradation and increasing glucose transporter activity. However, the extent of upregulation of Na+-dependent glucose cotransporter 1 (SGLT1) activity and content in response to increased delivery of carbohydrate to the small intestinal lumen of ruminants is unclear. Therefore, an experiment was conducted to determine the effect of glucose and starch hydrolysate on the activity and abundance of SGLT1 in the small intestine of steers. In a randomized complete block design, 40 crossbred beef steers (243+/-2 kg BW) were fed 0.163 Mcal of ME/(kg BW0.75(d; W), 0.215 Mcal of ME/(kg BW0.75 x d; 2M), or 0.163 Mcal ME/(kg BW0.75 x d) and infused for 35 d into the rumen (R) or abomasum (A) with 12.6 g/(kg BW0.75 x d) of starch hydrolysate (S) or into the abomasum with 14.4 g/(kg BW0.75 x d) of glucose (G). Steers were slaughtered, and brush-border membrane vesicles were prepared from the small intestinal samples obtained from five equidistant sites along the intestine. Maltase activity in vesicles and homogenates differed with intestinal sampling site (quadratic, P < 0.001). Steers on the AG treatment yielded a greater intestinal maltase activity (38 nmol glucose x mg protein(-1) x min(-1)) compared with the AS, RS, W, or 2M treatments (34, 26, 23, and 23 nmol glucose x mg protein(-1) x min(-1) respectively [SEM = 3; P = 0.02]). Sodium-dependent glucose uptake averaged 18.4+/-3.94 pmol glucose/(mg protein x s) and was not affected by treatment, but uptake decreased distally along the intestine (P < 0.001). There was no effect of treatment on SGLT1 protein abundance, but SGLT1 protein abundance increased linearly from the duodenum to the ileum (P = 0.05). The inverse relationship between glucose uptake and SGLT1 abundance suggests that the regulation of brush border Na+-dependent glucose transport capacity is complex, involving factors other than the presence of luminal carbohydrate. 相似文献
In this study we utilize field observations and data collected from 190 pedons from Wright and Taylor Valleys to search for evidence of high-water-level lakes proposed to have existed during the Last Glacial Maximum and early Holocene (2.7–25.7 ka) in the McMurdo Dry Valleys. We hypothesize that soils above the uppermost paleo-lake level should be more strongly developed and contain more salts than soils below. During detailed mapping of soils in the Dry Valleys, we found no evidence of former lake sediments nor did we find high-level strandlines except for strandlines on the north valley wall ca. 50 m above Lake Vanda, ice-shove features, or paleo-shore features. However, there may have been minor expansions of major lakes in the McMurdo Dry Valleys. In central Taylor and Wright Valleys, soils on equivalent-aged drifts above and below the conjectured upper limits of Glacial Lakes Washburn (336 m) and Wright (550 m), respectively, are all well developed with no appreciable differences in their properties. Moreover, there were no significant differences in the slopes of regression equations relating soil property to age of the parent materials above and below the high-water lake levels. 相似文献
This study aimed to establish the relationship between ME intake and energy and nutrient absorption across the portal-drained viscera (PDV) of forage-fed beef steers. Eight Angus (328 +/- 40 kg of BW) steers were surgically fitted with portal, mesenteric arterial, and mesenteric venous catheters, and were fed alfalfa cubes in a replicated 4 x 4 Latin square design with 4 levels of energy intake between 1 and 2 times maintenance energy requirements. On d 28 of each experimental period, p-aminohippuric acid was infused to measure blood and plasma flow across the PDV, and blood samples (1 every hour, for 6 h) were collected simultaneously from arterial and venous catheters for net absorption measurements. Oxygen utilization, and therefore energy utilization, increased (P < 0.05) linearly in relation to ME intake. Glucose net uptake was unaffected, but lactate net release increased linearly in response to ME intake (P < 0.05). Net absorption of all AA except tryptophan, glutamate, and glutamine increased linearly with ME intake (P < 0.05). The constant net absorption of glutamate and glutamine indicated increased net utilization of these AA when dietary supply was increased. These data provide quantitative measures of the PDV effects on energy and AA availability for productive tissues, and suggest that the greater net utilization of some AA when ME intake is increased could relate to their catabolism for energy production. Prediction estimates of small intestinal AA absorption, based on the Cornell Net Carbohydrate and Protein System (CNCPS), exceeded observed net AA PDV absorption. Mean bias represented the greatest proportion (87 to 96%) of the deviation between individual AA absorption and observed net AA PDV absorption, suggesting that the CNCPS model may be used to predict AA net absorption when factors describing AA utilization by the PDV are applied to model predictions. 相似文献
Research in control of tick-borne diseases and trypanosomosis, and their vectors, namely, ticks and tsetse flies respectively,
has been on going for decades. However, very little attention has been paid to the socio-economic factors that are likely
to influence the outcome of the interventions in the control of these diseases. Thus, this study was designed to investigate
these factors, mainly the intra-household factors influencing decision-making in the control of Vector-borne diseases in the
pastoralist areas of Uganda. These factors included: indigenous technical knowledge, household economic factors, and gender.
Both qualitative and quantitative methods were used in the collection and analysis of data. The tools used for data collection
included among others, participatory learning and action (PLA), and Case studies. The findings included the following: In
pastoralist households, a big proportion of the household budget was allocated to vector-borne diseases control. In the male-headed
households, men dominated decision-making on vector-borne diseases control, although the goals and priorities of men and women
in these households were not the same. Also, vector-borne disease control was predominantly by use of modern veterinary drugs,
and pastoralists treated sick cattle by themselves even in situations where there were veterinary personnel. 相似文献
The objective of this study was to determine the effect of a subtherapeutic level of chlortetracycline (CTC) fed to growing beef steers under conditions of limited and adequate dietary protein on plasma concentrations of GH, thyroid-stimulating hormone (TSH), and thyroid hormones before and after an injection of thyrotropin-releasing hormone (TRH) + GHRH. Young beef steers (n = 32; average BW = 285 kg) were assigned to a 2x2 factorial arrangement of treatments of either a 10 or 13% crude protein diet (70% concentrate, 15% wheat straw, and 15% cottonseed hulls) and either a corn meal carrier or carrier + 350 mg of CTC daily top dressed on the diet. Steers were fed ad libitum amounts of diet for 56 d, and a jugular catheter was then placed in each steer in four groups (two steers from each treatment combination per group) during four consecutive days (one group per day). Each steer was injected via the jugular catheter with 1.0 microg/kg BW TRH + .1 microg/kg BW GHRH in 10 mL of saline at 0800. Blood samples were collected at -30, -15, 0, 5, 10, 15, 20, 30, 45, 60, 120, 240, and 360 min after releasing hormone injection. Plasma samples were analyzed for GH, TSH, thyroxine (T4), and triiodothyronine (T3). After 84 d on trial, the steers were slaughtered and the pituitary and samples of liver were collected and analyzed for 5'-deiodinase activity. Feeding CTC attenuated the GH response to releasing hormone challenge by 26% for both area under the response curve (P<.03) and peak response (P<.10). Likewise, CTC attenuated the TSH response to releasing hormone challenge for area under the response curve by 16% (P<.10) and peak response by 33% (P<.02), and attenuated the T4 response for area under the curve by 12% (P<.08) and peak response by 14% (P<.04). Type II deiodinase activity in the pituitary was 36% less (P<.02) in CTC-fed steers than in steers not fed CTC. The results of this study are interpreted to suggest that feeding subtherapeutic levels of CTC to young growing beef cattle attenuates the release of GH and TSH in response to pituitary releasing hormones, suggesting a mechanism by which CTC may influence tissue deposition in cattle. 相似文献
Several Phytophthora spp. are known to cause a range of symptoms on citrus, resulting in significant crop losses worldwide. In South Africa, Phytophthora remains a destructive citrus disease, but the species and their distribution have not been well documented. A total of 162 Phytophthora isolates was collected from 60 citrus orchards in seven provinces of South Africa (Eastern Cape, Kwazulu-Natal, Limpopo, Mpumalanga, Northern Cape, North West and Western Cape). Isolates were identified to the species level through PCR-RFLP (restriction fragment length polymorphism) analyses of the internal transcribed spacer region. The identity of a subset of the isolates was confirmed using morphological and sequence analyses. Phytophthora nicotianae was the predominant species (76 % of isolates) and occurred in 80 % of the orchards in all of the provinces, followed by P. citrophthora (22 % of isolates in 28 % of orchards). The P. citrophthora isolates were further subdivided into two previously identified subgroups, G1 and G2, with most (69 %) of the isolates belonging to the G1 subgroup. Other Phytophthora species included P. multivora in the Western Cape Province, and an unknown species in the Eastern Cape Province with high sequence similarity (98 %) to a putative new species submitted to GenBank as Phytophthora taxon Sisuluriver. Phytophthora palmivora, a known citrus pathogen, was not identified. Most of the P. nicotianae isolates (79 %) were of the A1 mating type. The P. citrophthora isolates were mostly sterile (64 %), including most of the G1 isolates (81 %). The remaining G1 isolates (19 %) belonged to the A1 mating type, whereas almost all G2 isolates belonged to the A2 mating type except for one isolate that was sterile. 相似文献
