Overseeded winter annuals in bermudagrass [Cynodon dactylon (L.) Pers.] improve annual dry‐matter (DM) yield and capture nutrients in fields receiving manure application. This study determined the DM and nutrient uptake responses of annual ryegrass (Lolium multiflorum L.), cereal rye (Secale cereale), berseem clover (Trifolium alexandrinum L.) and bermudagrass‐winter fallow to 0, 50, 100 and 150 kg N ha?1 applied approximately 2 months before a single spring harvest, and in addition to swine‐effluent N (258 and 533 kg ha?1 in summer 2000 and 2001, respectively). Under drought conditions in 2000, DM yield at the spring harvest was highest in ryegrass, and summer DM yield of bermudagrass was greater at 100 and 150 kg N ha?1 than 50 kg N ha?1(P < 0·05). The concentration and uptake of N at the spring harvest increased linearly across N rates in both years (P < 0.05). Cover crops differed in N uptake in 2000 (P < 0.01) and values ranged from approximately 141 kg N ha?1 in berseem clover to 86 kg N ha?1 in rye. Per unit of N applied, uptake of N increased by approximately 0·409 kg ha?1 in 2000 and 0·267 kg ha?1 in 2001; uptake of P increased by 0·029 and 0·014 kg ha?1 respectively. In 2000, uptake of P was responsive to N rate and this relationship was significant (P < 0·01) in winter fallow (slope = 0·032) and ryegrass (slope = 0·057). Foot-and-mouth disease virus (FMDV) is the causative agent of a highly contagious acute vesicular disease affecting cloven-hoofed animals, including cattle, sheep and pigs. The current vaccine induces a rapid humoral response, but the duration of the protective antibody response is variable, possibly associated with a variable specific CD4+ T cell response. We investigated the use of heat shock protein 70 (Hsp70) as a molecular chaperone to target viral antigen to the Major Histocompatibility Complex (MHC) class II pathway of antigen presenting cells and generate enhanced MHC II-restricted CD4+ T cell responses in cattle. Monocytes and CD4+ T cells from FMDV vaccinated cattle were stimulated in vitro with complexes of Hsp70 and FMDV peptide, or peptide alone. Hsp70 was found to consistently improve the presentation of a 25-mer FMDV peptide to CD4+ T cells, as measured by T cell proliferation. Complex formation was required for the enhanced effects and Hsp70 alone did not stimulate proliferation. This study provides further evidence that Hsp70:peptide complexes can enhance antigen-specific CD4+ T cell responses in vitro for an important pathogen of livestock. 相似文献

Background: Hyperuricosuria is a condition that predisposes dogs to urate urolithiasis. A mutation that causes canine hyperuricosuria was previously identified in 3 unrelated dog breeds. The occurrence of the mutation in additional breeds was not determined. Hypothesis/Objectives: Identify additional breeds that have the hyperuricosuria mutation and estimate the mutant allele frequency in those breeds. Animals: Three thousand five hundred and thirty dogs from 127 different breeds were screened for the hyperuricosuria mutation. Methods: DNA samples were genotyped by pyrosequencing and allele‐specific polymerase chain reaction methods. Results: Mutant allele frequencies that range from 0.001 to 0.15 were identified in the American Staffordshire Terrier, Australian Shepherd, German Shepherd Dog, Giant Schnauzer, Parson (Jack) Russell Terrier, Labrador Retriever, Large Munsterlander, Pomeranian, South African Boerboel, and Weimaraner breeds. Conclusions and Clinical Importance: The hyperuricosuria mutation has been identified in several unrelated dog breeds. The mutant allele frequencies vary among breeds and can be used to determine an appropriate breeding plan for each breed. A DNA test is available and may be used by breeders to decrease the mutant allele frequency in breeds that carry the mutation. Clusterin (CLU), also known as apolipoprotein J, is a widely expressed, heterodimeric, glycoprotein, important in tumourigenesis, apoptosis and immunoregulation. In humans, CLU expression has been associated with anaplastic large cell and Hodgkin's lymphoma. In this study, serum CLU levels in dogs with multicentric lymphoma (MLSA) were compared with healthy control dogs, using both western blot and enzyme‐linked immunosorbent assay (ELISA). Western blot confirmed the presence of CLU in dog sera at the predicted molecular weight and the relative levels detected correlated with the levels detected by ELISA. CLU level analysis by ELISA found treatment naïve dogs with MLSA had a significantly (P < .001) lower serum CLU level compared with healthy controls. However, there was no significant difference between MLSA dogs prior to treatment and in complete remission. The wide variation in serum CLU levels may limit its potential as a single candidate biomarker for MLSA, although any prognostic predictive value of serum CLU concentrations has yet to be assessed. 相似文献

Lamellar, autogenous, corneal grafts were performed on 9 clinically normal dogs (1 eye in each of 2 dogs and both eyes in each of 7 dogs) and 7 dogs with corneal disease (1 eye in each dog). Nylon, polyglycolic acid, polyglactin 910, and polydioxanone suture materials were used in 8-0 to 10-0 sizes. The eyes of the normal dogs were examined weekly for up to 16 weeks; corneal endothelium was reestablished in all eyes by 6 weeks after surgery. The eyes of the dogs with corneal disease were reexamined periodically for up to 9 months. Twenty-two of 23 (95.7%) grafts were considered successful in that they were translucent, and the dogs were able to see after surgery. The suture materials that caused the least tissue reaction were nylon and polyglactin 910. This procedure is indicated for chronic descemetoceles and other perforating corneal disease. It has the advantage of rapidly resolving severe corneal disease, with a high degree of success. 相似文献

Killed Brucella abortus strain 1119-3 cells were treated with hot phenol/water or dimethyl sulfoxide to extract soluble crude lipopolysaccharide antigen. Antigen preparations were characterized with respect to protein and lipopolysaccharide content and were compared for efficacy in the hemolysis-in-gel test (HIGT) for detecting anti-Brucella antibodies. All antigens were equally effective in sensitizing bovine RBC in the HIGT and in detecting the presence of anti-Brucella antibodies. Abstract Soil aggregate-size distribution and soil aggregate stability are used to characterize soil structure. Quantifying the changes of structural stability of soil is an important element in assessing soil and crop management practices. A 5-year tillage experiment consisting of no till (NT), moldboard plow (MP) and ridge tillage (RT), was used to study soil water-stable aggregate size distribution, aggregate stability and aggregate-associated soil organic carbon (SOC) at four soil depths (0–5, 5–10, 10–20 and 20–30 cm) of a clay loam soil in northeast China. Nonlinear fractal dimension (Dm) was used to characterize soil aggregate stability. No tillage led to a significantly greater aggregation for >1 mm aggregate and significant SOC changes in this fraction at 0–5 cm depth. There were significant positive relationships between SOC and >1 mm aggregate, SOC in each aggregate fraction, but there was no relationship between soil aggregate parameters (the proportion of soil aggregates, aggregate-associated SOC and soil stability) and soil bulk density. After 5 years, there was no difference in Dm of soil aggregate size distribution among tillage treatments, which suggested that Dm could not be used as an indicator to assess short-term effects of tillage practices on soil aggregation. In the short term, > 1 mm soil aggregate was a better indicator to characterize the impacts of tillage practices on quality of a Chinese Mollisol, particularly in the near-surface layer of the soil. 相似文献

Seasonal changes in water relations of current-year shoots of red spruce (Picea rubens Sarg.) were examined in relation to climatic conditions in trees growing at elevations of 1720 and 1935 m on Clingman's Dome, Tennessee, USA, where increment core data have shown that red spruce decline increases with elevation. Relative height growth of trees at 1720 m was 68% greater than in trees at 1935 m. Following two weeks in July with only traces of precipitation, trees at both sites showed decreased saturated osmotic potentials. The magnitude of the reduction was greater in trees at the high elevation site than in trees at the low elevation site. However, during August and September, shoot water relations of trees at both sites were similar. Precipitation patterns and water relations measurements suggested that, at both sites, trees experienced water stress only briefly during the growing season and to a degree that could not account for the lower growth rates of trees at the high elevation site. During the period of cold hardening in October and November, trees at the low elevation site exhibited saturated osmotic potentials that were lower by 0.2 MPa and solute accumulation (osmol kg(dw) (-1)) that was 48% greater than in trees at the high elevation site. 相似文献
