Published studies of rhizobial populations, communities and other strain collections were analysed to identify trends in strain richness, strain dominance and genetic diversity within and between locations. For individual populations and communities, strain richness indices were calculated by dividing the number of strain types identified by the number of isolates recovered. Where possible, strain dominance (the proportion of rhizobial isolates represented by each strain type) was also calculated. Analysis of the genetic diversity of populations, communities and rhizobial strain collections originating from diverse legume hosts and locations, was confined to studies using multilocus enzyme electrophoresis (MLEE) so that diversity could be compared on the basis of published H values. Strain richness indices were highly variable (0.02-0.94) and were influenced by both the discriminatory power of the strain typing method and the type and number of legume species used to recover rhizobia from soil. The strain richness of populations recovered either directly from soil, or from the nodules of trap hosts inoculated with the same soil, was similar. Because the arithmetic relationship between the number of strain types and number of isolates varies between different populations and communities, strain richness curves are proposed as the most appropriate method for reporting rhizobial structural diversity. Comparison of over 50 rhizobial populations and communities from published studies showed that strain dominance patterns in nodules were often similar. Typically, a single strain type occupies more than 30% of nodules with the majority of strain types being recovered at low frequency (∼75% of published populations and communities). Rhizobial populations and communities characterised by MLEE varied in their genetic diversity, with H values ranging from 0.06 to 0.78. In several studies, most of the genetic diversity within a site could be recovered from the nodules of a single plant. When hierarchical analyses were performed on populations within and between sites, the genetic diversity within sites was similar to the genetic diversity between sites. Similarly, the genetic diversity of strain collections originating from multiple hosts and locations was no more diverse than some individual populations and communities. Strain richness and genetic diversity measures were not always correlated for rhizobial populations. Populations with low strain richness were sometimes genetically diverse, and the relationship between the diversity measures varied for different legume species at the same location. We suggest that both strain richness and genetic diversity measures are required to fully describe rhizobial population and community diversity.  相似文献

Precision irrigation utilizing soil moisture data and valve-in head sprinkler systems may be a viable solution for sustainable water management on complex turfgrass areas. There is currently no research investigating the factors that influence soil moisture and turfgrass quality variability within sand-capped golf course fairways to aid in precision irrigation-related management decisions. Therefore, the objective of this study was to measure several turfgrass and soil characteristics from two sand-capped fairways during dry down events from either rainfall or irrigation to determine their relationship and contribution to soil moisture and turfgrass quality variability. Considerable spatiotemporal variability was observed within the two fairways during the dry down periods. Factors that were found to have a significant influence on soil moisture and turfgrass quality were sand capping depth, elevation, and thatch depth, but these relationships were not consistent between rainfall versus irrigation events, days after dry down, or even the specific fairways. Also, the direction of many of the relationships were opposite of what was expected. These findings highlight the complexity of soil moisture and turfgrass quality variability on sand-capped golf course fairways. To incorporate soil moisture sensor technologies into large-scale precision irrigation practices, mapping soil moisture with an understanding of contributing factors is a necessary preliminary step. Although there are several current practical limitations, the information presented in this study provides a foundation for future research.

The terminal uranium nitride linkage is a fundamental target in the study of f-orbital participation in metal-ligand multiple bonding but has previously eluded characterization in an isolable molecule. Here, we report the preparation of the terminal uranium(V) nitride complex [UN(Tren(TIPS))][Na(12-crown-4)(2)] {in which Tren(TIPS) = [N(CH(2)CH(2)NSiPr(i)(3))(3)](3-) and Pr(i) = CH(CH(3))(2)} by reaction of the uranium(III) complex [U(Tren(TIPS))] with sodium azide followed by abstraction and encapsulation of the sodium cation by the polydentate crown ether 12-crown-4. Single-crystal x-ray diffraction reveals a uranium-terminal nitride bond length of 1.825(15) angstroms (where 15 is the standard uncertainty). The structural assignment is supported by means of (15)N-isotopic labeling, electronic absorption spectroscopy, magnetometry, electronic structure calculations, elemental analyses, and liberation of ammonia after treatment with water.  相似文献

Haematological and biochemical investigations were performed on 14 koalas with uncomplicated cystitis, 8 with complicated cystitis, 8 with conjunctivitis, 8 with lymphosarcoma, and 14 with miscellaneous diseases. Changes were limited and inconsistent in individual koalas with uncomplicated cystitis and conjunctivitis. In contrast, individual koalas with complicated cystitis were more likely to have anaemia, leukocytosis due to neutrophilia, hypoproteinaemia due to hypoalbuminaemia, and azotaemia due to elevated urea concentration. Although these changes were non-specific they did allow assessment of prognosis for survival and response to treatment. Koalas with lymphosarcoma were invariably anaemic, leukaemic, azotaemic and hypoalbuminaemic. Elevated enzymes (aspartate transaminase [AST]. lactate dehydrogenase [LD] and gamma glutamyl transferase [GGT]) were more common in koalas with lymphosarcoma. Koalas affected by miscellaneous conditions showed variable changes but once again anaemia, leukocytosis, azotaemia, elevated AST and LD, and hypoalbuminaemia were not uncommon. On the basis of these findings a minimal profile is suggested for the investigation of sick koalas and would include haematocrit, total and differential leukocyte counts, urea, total protein and albumin concentrations and AST, GGT and LD activities.  相似文献

In the present study, the occurrence and molecular phylogeny of trypanosome parasites were studied in both wild and captive marsupials from Western Australia and Queensland. Blood samples were screened by PCR at the 18S rDNA locus, and the glycosomal glyceraldehyde phosphate dehydrogenase gene. Overall, 5.3% of the blood samples were positive at the 18S rDNA locus. All positives belonged to wild-captured Western Australian individuals, where trypanosome-specific DNA was detected in 9.8% of the screened samples from wild marsupials, in common brushtail possums, and woylies. The detection rate of trypanosome DNA in these two host species was 12.5% and 20%, respectively. Phylogenetic analyses based on two loci, indicated that the possum-derived trypanosome isolates were genetically distinct, and most closely related to the Australian marsupial trypanosomes H25 from a kangaroo, and BRA2 from a bush rat. This is the first study to genetically characterise trypanosome isolates from possums. The analysis of the woylie-derived isolates demonstrated that this marsupial host can harbour multiple genotypes within the same geographical location and furthermore multiple genotypes within the same host, indicative of mixed infections. All the woylie-derived genotypes grouped with trypanosomes found in Australian marsupials, suggesting that they are more likely to belong to an endemic or Australasian trypanosome species. This is the first study to genetically characterise trypanosome isolates from possums (Trichosurus vulpecula). Although the clinical significance of these infections is currently unknown, the identification of these novel sequences may support future investigations on transmission, threats to endangered wildlife, and evolutionary history of the genus Trypanosoma.  相似文献

AIMS: To identify network measures with relevance to disease spread in a network of movements derived from the Department of Conservation (DOC) translocation records from 1970 to mid-2014, and to identify conservation sites that should be prioritised for surveillance activities and improvements to data collection to make the best use of network analysis techniques in the future.

METHODS: Data included the source and destination of translocated specimens, the species and the dates the translocations were expected to occur. The data were used to construct a directed, non-weighted network in which a translocation event represented a tie in the network. Network density, in-degree (movements entering a node of interest) and out-degree (movements leaving a node of interest) and reciprocity were calculated.

RESULTS: The data analysed consisted of 692 unique translocations between 307 sites, with the majority (518; 73%) being for birds. The constructed network for bird, reptile and frog translocations comprised 260 nodes, with 34/260 (13%) having two-way movements and 47/260 (18%) non-reciprocal movements. The median degree score (sum of in- and out-degree) was two (min 0, max 36) with a mean of 3.5 in a right skewed distribution. Most sites acted as receivers or senders of consignments with only a few having both high in- and high out-degree, and thus had characteristics that made them sites of interest for surveillance activities. These included the National Wildlife Centre at Mount Bruce, Tiritiri Matangi Island and Te Kakahu (Chalky Island).

CONCLUSIONS: The presence of linking sites that join larger clusters within the network creates the potential for rapid disease spread if a pathogen were to be introduced. The important sites that supply or receive specimens for translocations are already well recognised by those performing translocations in New Zealand, and this paper provides further information by quantifying their role within the network.  相似文献

A numerical model has been developed which produces periodic surging as a characteristic of some glaciers for a certain accumulation and bedrock distribution in contrast to the normal steady state for nonsurging glaciers. The epidemiology of 16 cases of infectious coryza, an upper respiratory tract disease of chickens caused by Haemophilus paragallinarum, was investigated in a retrospective study. The cases occurred over a 14-month period on 10 farms in northern New South Wales. The available field data indicated that the cases formed six unrelated outbreaks. The 16 isolates of H. paragallinarum were subjected to serotyping by the Page and Kume schemes and biotyping based on carbohydrate fermentation and antimicrobial drug-resistance patterns. As well, newer fingerprinting techniques--plasmid profiles, whole-cell protein profiles, immunoblots of whole-cell protein profiles and total DNA restriction endonuclease analysis (REA)--were evaluated. Antimicrobial biotyping and REA profile typing proved most useful, allowing the recognition of three groups among the isolates. The other techniques gave either limited or no subdivision among the isolates. The combined results of the laboratory study indicated that, rather than six unrelated outbreaks, the 16 isolates represented three pairs of related outbreaks. This study represents the first application of sensitive biotyping and fingerprinting techniques to outbreaks of infectious coryza. The results have established that farms can be repeatedly infected with a single strain of H. paragallinarum that re-emerges at intervals. This study also obtained the first detailed evidence that replacement stock are a major source of infectious coryza.  相似文献

In sheep infected with the parapoxvirus orf virus, primary infection orf skin lesions developed and resolved within 8 weeks. Reinfection lesions were smaller and resolved within 3 weeks. The host response in the skin was characterized by an accumulation of neutrophils, dendritic cells, CD4+ T cells, CD8+ T cells, B cells and T19+ gammadelta T cells. The magnitude of this accumulation paralleled orf virus replication in the skin. In situ hybridization was used to detect cells expressing interferon-gamma (IFN-gamma), tumor necrosis factor-alpha (TNF-alpha) and interleukin-4 (IL-4) mRNAs in orf skin. Cells expressing IL-4 mRNA were not detected at any time after infection. Cells expressing IFN-gamma mRNA were detected after reinfection but not after primary infection. Cells expressing TNF-alpha mRNA included epidermal cells, vascular endothelium and uncharacterized cells that increased more rapidly in the skin after reinfection compared to primary infection. The results are consistent with a prominent role for IFN-gamma in the host immune response controlling the severity of the disease.  相似文献
