Surveys of serum antibodies to enterovirus serotype 8 and parvovirus were conducted in pigs three to 21 weeks old using the virus neutralisation (VN), enzyme-linked immunosorbent assay (ELISA) and haemagglutination inhibition tests. Antibody levels to enterovirus were at their lowest at four to eight weeks old, increased at 14 to 16 weeks to high levels which were maintained until 21 weeks. Specific immunoglobulin G (IgG) values measured by ELISA paralleled VN values and were similar in two separate farm surveys. Measurement of immunoglobulin M (IgM) levels indicated that enterovirus infection occurred about four weeks old. Sera obtained from a large number of geographically separated farms, from abattoir-bled animals and from colostrum-deprived piglets raised for 33 weeks from birth in an isolated environment were tested by VN. Results revealed that porcine enterovirus serotype 8 is spread widely throughout northern Victorian piggeries and high antibody levels prevail. In contrast to the enterovirus antibody results, parvovirus antibody levels measured in piglets declined from high levels at three to four weeks to undetectable levels from 13 weeks old.  相似文献

A two-year-old, neutered female cross-bred labrador had multiple cutaneous nodules, biopsies of which revealed pathological changes consistent with cutaneous histiocytosis. During a period of one month the dog developed multicentric lymphadenopathy, a retrobulbar mass and masses within the quadriceps and cervical muscles. Fine needle aspiration cytology of the cutaneous nodules and lymph nodes and histological examination of the cutaneous nodules and muscle masses suggested the presence of lymphoblastic lymphoma. A definitive diagnosis of CD8+ T cell lymphoma was achieved by immunophenotyping the tumour cells by flow cytometry.  相似文献

We assessed the effect of feeding damage by pear thrips, Taeniothrips inconsequens Uzel (Thysanoptera:Thripidae), on gas exchange and water relations of sugar maple (Acer saccharum Marsh.) seedlings. Compared to undamaged seedlings, feeding punctures in the leaf epidermis of thrips-damaged seedlings decreased water use efficiency, increased leaf conductance to water vapor, and decreased predawn water potential. Under conditions of high soil water and high light intensity, carbon dioxide exchange rate (CER) was greater for thrips-damaged than undamaged seedlings because of greater CO(2) conductance through feeding punctures. Under conditions of low soil water, CER was lower for thrips-damaged than undamaged seedlings as a result of water stress. Carbon dioxide exchange rate at low light and low soil water was limited by non-stomatal factors, but no difference in non-stomatal limitation to CER was detected between thrips-damaged and undamaged seedlings. Leaf tissue water relations differed between thrips-damaged and undamaged seedlings and under high and low soil water conditions. The results suggest that the reduction in leaf area of thrips-damaged seedlings can be partially compensated by elevated CER under conditions of high light intensity and high soil water. However, high gas exchange rates through feeding punctures predisposes thrips-damaged seedlings to water stress that can reduce CER under conditions of low soil water.  相似文献

Skin test negative cattle from a herd containing an unusually high proportion (194/382) of tuberculin skin test positive cattle were investigated for remaining Mycobacterium bovis infected animals. Blood samples from the skin test negative cattle, analysed by an antibody ELISA and an interferon-gamma assay, were mostly test negative for M. bovis. Radiometric culture of nasal mucus samples from 48 of the cattle yielded 22 culture positives with acid-fast bacilli and cording in 6 of these. Subculture on solid media was successful for 7, including 2 with cording of the 22 radiometric culture positives. Mycobacterium tuberculosis complex DNA probe testing using the Accuprobe (Gen-Probe, Inc.) and M. tuberculosis complex-specific PCR amplification, performed on the solid media subcultures, were negative. 16S rRNA PCR and sequence analysis were successful for 6 of the 7 solid media subcultures obtained and revealed the presence of Mycobacterium nonchromogenicum in all 6 subcultures. This is the first report of M. nonchromogenicum in nasal mucus of cattle. The observation highlights the importance of integrating definitive tests such as the PCR for diagnosis of bovine tuberculosis and indicates a possible zoonotic risk.  相似文献

The objective was to evaluate chemical, mechanical, and sensory attributes associated with tenderness in divergent cattle breeds--Wagyu (W; n = 12), Limousin (L; n = 12) and F1-cross (WxL; n = 12)--fed two dietary treatments (0 or 6% sunflower oil (DM basis)). A randomized complete block repeated measures design in a 3 x 2 factorial arrangement of treatments was used, and effects of breed, diet, block, and associated interactions were tested. Cattle were fed barley-based diets for an average of 259 d. Twenty-four hours postmortem (PM), steaks from the longissimus muscle (LM) were sliced, vacuum-packaged, aged (1, 3, 7, 14, 28, and 56 d PM) at 2 degrees C, and frozen (-40 degrees C) until analyzed. Wagyu steaks had lower (P < 0.05) Warner-Bratzler shear force (WBSF) values than L steaks across all aging times. At 1 d PM, W steaks required slightly more (P > 0.05) force to shear than WxL or L (0.30 and 0.11 kg, respectively); however, by d 14 PM, W steaks required 0.77 kg less (P < 0.05) force to shear than L. Wagyu steaks received higher (P < 0.05) sensory panel sustained tenderness scores at d 14 PM than L. The pH decline was slower (P < 0.05), and temperature decline more (P < 0.05) rapid, in W carcasses than L or WxL carcasses. Breed and diet did not affect (P > 0.10) free calcium levels (FCL) over time (0, 1, 3, 7, and 14 d PM), 0-h calpastatin activity (CA), d-1 percent collagen (OH-PRO), or d-1 collagen cross-linking (HP). Western blot analysis for the presence of the troponin-T (TNT) 30-kDa fragment, conducted only on samples from steers fed the 0% sunflower oil diet, demonstrated more proteolysis by d 3 PM in L than W or WxL. Overall, breed differences in mechanical and sensory measures of tenderness were not explained by FCL, CA, OH-Pro, and HP. Even though the initial appearance of the TNT 30-kDa fragment was greater in L, linear slopes for appearance of TNT degradation product across aging time were greater for W and WxL (P < 0.01 and P = 0.056, respectively) than for L, suggesting that tenderness differences due to breed may have been facilitated by more-rapid proteolytic degradation over time.  相似文献

Sodium chloride, at rates up to 100 mg g?1, was added to a Sassafras sandy loam amended with finely-ground alfalfa to determine the effect of NaCl on CO2 evolution, ammonification, and nitrification in a 14-week study. A NaCl concentration of 0.25 mg g?1 significantly reduced CO2 evolution by 16% in unamended soil and 5% in alfalfa-amended soil. Increasing NaCl progressively reduced CO2 evolution, with no CO2 evolved from the soil receiving 100 mg NaCl g?1. A 0.50 mg NaCl g?1 rate was required before a significant reduction in decomposition of the alfalfa occurred. The NO?2-N + NO?3-N content of the soil was significantly reduced from 40 to 37 μg g?1 at 0 and 0.25 mg NaCl g?1, respectively in the unamended soil. In the alfalfa amended soil, nitrification was significantly reduced at 5 mg NaCl g?1. At 10 mg NaCl g?1, nitrification was completely inhibited, there being only 6 and 2 μg NO?2-N + NO?3-N g?1 in the alfalfa amended and unamended soil, respectively. In the alfalfa amended soil NH+4-N accumulated from 6 μg g?1 at the 0 NaCl rate to a maximum of 54 μg g?1 with 25 mg NaCl g?1. These higher NH+4-N values resulted in a 0.5 unit increase in the pHw over that of the 0 NaCl rate in the alfalfa amended soil. At NaCl concentrations above 25 mg g?1 there was a reduction in NH+4-N. The addition of alfalfa to the soil helped to alleviate the adverse affects of NaCl on CO2 evolution and nitrification.  相似文献

Thin zeolite films are attractive for a wide range of applications, including molecular sieve membranes, catalytic membrane reactors, permeation barriers, and low-dielectric-constant materials. Synthesis of thin zeolite films using high-aspect-ratio zeolite nanosheets is desirable because of the packing and processing advantages of the nanosheets over isotropic zeolite nanoparticles. Attempts to obtain a dispersed suspension of zeolite nanosheets via exfoliation of their lamellar precursors have been hampered because of their structure deterioration and morphological damage (fragmentation, curling, and aggregation). We demonstrated the synthesis and structure determination of highly crystalline nanosheets of zeolite frameworks MWW and MFI. The purity and morphological integrity of these nanosheets allow them to pack well on porous supports, facilitating the fabrication of molecular sieve membranes.  相似文献

A cytoplasmic protein that greatly enhances the guanosine triphosphatase (GTPase) activity of N-ras protein but does not affect the activity of oncogenic ras mutants has been recently described. This protein (GAP) is shown here to be ubiquitous in higher eukaryotes and to interact with H-ras as well as with N-ras proteins. To identify the region of ras p21 with which GAP interacts, 21 H-ras mutant proteins were purified and tested for their ability to undergo stimulation of GTPase activity by GAP. Mutations in nonessential regions of H-ras p21 as well as mutations in its carboxyl-terminal domain (residues 165-185) and purine binding region (residues 117 and 119) did not decrease the ability of the protein to respond to GAP. In addition, an antibody against the carboxyl-terminal domain did not block GAP activity, supporting the conclusion that GAP does not interact with this region. Transforming mutations at positions 12, 59, and 61 (the phosphoryl binding region) abolished GTPase stimulation by GAP. Point mutations in the putative effector region of ras p21 (amino acids 35, 36, and 38) were also insensitive to GAP. However, a point mutation at position 39, shown previously not to impair effector function, did not alter GAP-p21 interaction. These results indicate that GAP interaction may be essential for ras p21 biological activity and that it may be a ras effector protein.  相似文献

A model was developed for the structure of p21, the protein with a molecular weight of 21,000 that is produced by the ras genes. This model predicts that p21 consists of a central core of beta-sheet structure, connected by loops and alpha helices. Four of these loops comprise the guanine nucleotide binding site. The phosphoryl binding region is made up of amino acid sequences from 10 to 16 and from 57 to 63 of p21. The latter sequence may contain a site for magnesium binding. Amino acids defining guanine specificity are Asn-116 and Asp-119, and sequences around amino acid 145 may contribute to guanine binding. The model makes it possible to visualize how oncogenic mutations of p21 affect interaction with guanine nucleotides.  相似文献
