A model for the tertiary structure of p21, the product of the ras oncogene   总被引:39,自引:0,他引:39  
A model was developed for the structure of p21, the protein with a molecular weight of 21,000 that is produced by the ras genes. This model predicts that p21 consists of a central core of beta-sheet structure, connected by loops and alpha helices. Four of these loops comprise the guanine nucleotide binding site. The phosphoryl binding region is made up of amino acid sequences from 10 to 16 and from 57 to 63 of p21. The latter sequence may contain a site for magnesium binding. Amino acids defining guanine specificity are Asn-116 and Asp-119, and sequences around amino acid 145 may contribute to guanine binding. The model makes it possible to visualize how oncogenic mutations of p21 affect interaction with guanine nucleotides.  相似文献

Climate variations influenced the agricultural productivity, health risk, and conflict level of preindustrial societies. Discrimination between environmental and anthropogenic impacts on past civilizations, however, remains difficult because of the paucity of high-resolution paleoclimatic evidence. We present tree ring-based reconstructions of central European summer precipitation and temperature variability over the past 2500 years. Recent warming is unprecedented, but modern hydroclimatic variations may have at times been exceeded in magnitude and duration. Wet and warm summers occurred during periods of Roman and medieval prosperity. Increased climate variability from ~250 to 600 C.E. coincided with the demise of the western Roman Empire and the turmoil of the Migration Period. Such historical data may provide a basis for counteracting the recent political and fiscal reluctance to mitigate projected climate change.  相似文献

We report classes of electronic systems that achieve thicknesses, effective elastic moduli, bending stiffnesses, and areal mass densities matched to the epidermis. Unlike traditional wafer-based technologies, laminating such devices onto the skin leads to conformal contact and adequate adhesion based on van der Waals interactions alone, in a manner that is mechanically invisible to the user. We describe systems incorporating electrophysiological, temperature, and strain sensors, as well as transistors, light-emitting diodes, photodetectors, radio frequency inductors, capacitors, oscillators, and rectifying diodes. Solar cells and wireless coils provide options for power supply. We used this type of technology to measure electrical activity produced by the heart, brain, and skeletal muscles and show that the resulting data contain sufficient information for an unusual type of computer game controller.  相似文献

Results of the first year of data collection by the SAM (Stratospheric Aerosol Measurement) II satellite system are presented. Almost 10,000 profiles of stratospheric aerosol extinction in the Arctic and Antarctic regions are used to construct plots of weekly averaged aerosol extinction versus altitude and time and stratospheric optical depth versus time. Corresponding temperature fields are presented. These data show striking similarities in the aerosol behavior for corresponding seasons. Wintertime polar stratospheric clouds that are strongly correlated with temperature are documented. They are much more prevalent in the Antarctic stratosphere during the cold austral winter and increase the stratospheric optical depths by as much as an order of magnitude for a period of about 2 months. These clouds might represent a sink for stratospheric water vapor and must be considered in the radiative budget for this region and time.  相似文献

Summary A field experiment was conducted on the west side of the San Joaquin Valley in California to determine water use, crop growth, yield and water use efficiency of Acala (SJ-2) cotton (Gossypium hirsutum L.) grown in 0.5 m spaced rows on a Panoche clay loam soil (Typic Torriorthents). Evapotranspiration was determined by water balance techniques utilizing neutron soil moisture measurements. All neutron measurements were made within a 3 m soil profile in 0.20 m increments. The measured evapotranspiration was compared to climatic estimates of potential evapotranspiration, and to calculations using a one-dimensional soil water balance model that separately computed soil water evaporation and plant transpiration. Crop growth was determined by weekly destructive plant sampling. Leaf area was determined along with dry matter components of leaves, stems, fruiting parts (flowers and squares) and bolls. Final yield was determined by machine harvesting (brush stripper) 720 m2 from each plot. Lint yields and fiber quality were determined by sample ginning and fiber analysis at the U.S. Cotton Research Station at Shafter, California. Three irrigation regimes were established that resulted in an evapotranspiration range from a high deficit condition to full irrigation at the calculated atmospheric demand.The measured evapotranspiration of narrow row cotton under a full irrigation regime was 778 mm, 594 mm under a limited irrigation regime and 441 mm under a regime with no post-plant irrigation. The evapotranspiration from these irrigation treatments was accurately simulated by a water balance model. that used inputs of potential evapotranspiration, leaf area index, soil water holding capacity and root development.The average lint yield from narrow row cotton with a full irrigation regime was 1583 kg/ha, the average lint yield from a limited irrigation regime was 1423 kg/ha and the average lint yield from a treatment with no postplant irrigation (fully recharged soil profile at planting) was 601 kg/ha. The full irrigation regime resulted in a dry matter production of approximately 16 t/ha while the limited irrigated regime produce 11 t/ha and the no-postplant irrigation regime produced 7 t/ha of dry matter. The fiber quality results indicated significant (0.05 level) differences only in 50% span length and micronaire, with the 2.5% span length, uniformity index, elongation and strength indicating no difference.Cotton lint yield was found to be directly related to total evapotranspiration although the relationship was slightly non-linear while dry matter yield was found to be linearly related to evapotranspiration. Interfacially active block copolymer amphiphiles have been synthesized and their self-assembly into micelles in supercritical carbon dioxide (CO2) has been demonstrated with small-angle neutron scattering (SANS). These materials establish the design criteria for molecularly engineered surfactants that can stabilize and disperse otherwise insoluble matter into a CO2 continuous phase. Polystyrene-b-poly(1,1-dihydroperfluorooctyl acrylate) copolymers self-assembled into polydisperse core-shell-type micelles as a result of the disparate solubility characteristics of the different block segments in CO2. These nonionic surfactants for CO2 were shown by SANS to be capable of emulsifying up to 20 percent by weight of a CO2-insoluble hydrocarbon into CO2. This result demonstrates the efficacy of surfactant-modified CO2 in reducing the large volumes of organic and halogenated solvent waste streams released into our environment by solvent-intensive manufacturing and process industries.  相似文献

Efforts to solve the epidemiologic puzzle of AIDS in Africa are complicated by the presence of multiple human retroviruses. Simple serologic tests that unambiguously distinguish among infections by these retroviruses are essential. To that end, a partially conserved immunoreactive epitope was identified in the transmembrane glycoproteins of human immunodeficiency viruses (HIV) types 1 and 2. Synthetic peptides derived from these conserved domains were used in sensitive and specific immunoassays that detect antibodies in sera from patients infected with HIV-1 or HIV-2. By making single amino acid substitutions in the HIV-1 peptide, it was possible to demonstrate HIV-1 strain-specific antibody responses to this epitope. Such custom-designed peptides synthesized from this domain are likely to detect newly discovered HIV types, define infection with specific HIV strains, and allow detection of group-common antibodies.  相似文献

This paper describes the field evaluation of a serological test and a new in vitro assay for cell-mediated reactivity for the diagnosis of bovine tuberculosis. The use of a Mycobacterium bovis-specific antigen (MPB-70) in an ELISA to test the serological response to tuberculosis infection resulted in a specificity of 96.4% and a sensitivity of 18.1%. The most favourable results were obtained with the interferon gamma (IFN-gamma) assay which had a sensitivity of 81.8% and a specificity of 99.1%. Respective figures for the single intradermal tuberculin test were 68.1% and 96.7%. The use of MPB-70 as the antigen in the IFN-gamma assay reduced the sensitivity of this assay, without producing any useful increase in specificity. The IFN-gamma assay was also demonstrated to be a practical diagnostic test for use with large groups of cattle.  相似文献

Regulation of Ras-GAP and the neurofibromatosis-1 gene product by eicosanoids   总被引:10,自引:0,他引:10  
Ras-GAP (GTPase activating protein) is a regulatory protein that stimulates the intrinsic guanosine triphosphatase (GTPase) activity of the proto-oncogene product p21ras. A domain of the neurofibromatosis gene product (NF1) that has sequence similarity to the catalytic domain of Ras-GAP and to yeast IRA gene products also has a specific stimulatory activity toward p21ras GTPase. Arachidonic acid and phosphatidic acid inactivate GAP, but no agents have been identified that stimulate GAP and thereby switch p21ras off. With the use of recombinant Ha-c-Ras and Ras-GAP, NF1, and GAP catalytic domains, it was found that prostaglandins PGF2 alpha and PGA2 stimulated Ras-GAP and that prostacyclin PGI2 inhibited Ras-GAP. The stimulatory effect of PGF2 alpha was saturable and structure-specific and competed with the inhibitory effect of arachidonic acid. Arachidonic acid also inhibited the catalytic activity of NF1, but prostaglandins were not stimulatory. These results suggest a mechanism for the allosteric control of Ras function through the modulation of arachidonate metabolism.  相似文献
