Somatic cell nuclear transfer allows trans-acting factors present in the mammalian oocyte to reprogram somatic cell nuclei to an undifferentiated state. We show that four factors (OCT4, SOX2, NANOG, and LIN28) are sufficient to reprogram human somatic cells to pluripotent stem cells that exhibit the essential characteristics of embryonic stem (ES) cells. These induced pluripotent human stem cells have normal karyotypes, express telomerase activity, express cell surface markers and genes that characterize human ES cells, and maintain the developmental potential to differentiate into advanced derivatives of all three primary germ layers. Such induced pluripotent human cell lines should be useful in the production of new disease models and in drug development, as well as for applications in transplantation medicine, once technical limitations (for example, mutation through viral integration) are eliminated. 相似文献

Among the brain tumors, glioma is the most common. In general, different biochemical mechanisms, involving nicotinic acetylcholine receptors (nAChRs) and the arachidonic acid cascade are involved in oncogenesis. Although the engagement of the latter in survival and proliferation of rat C6 glioma has been shown, there are practically no data about the presence and the role of nAChRs in C6 cells. In this work we studied the effects of nAChR antagonists, marine snail α-conotoxins and snake α-cobratoxin, on the survival and proliferation of C6 glioma cells. The effects of the lipoxygenase and cyclooxygenase inhibitors either alone or together with α-conotoxins and α-cobratoxin were studied in parallel. It was found that α-conotoxins and α-cobratoxin promoted the proliferation of C6 glioma cells, while nicotine had practically no effect at concentrations below 1 µL/mL. Nordihydroguaiaretic acid, a nonspecific lipoxygenase inhibitor, and baicalein, a 12-lipoxygenase inhibitor, exerted antiproliferative and cytotoxic effects on C6 cells. nAChR inhibitors weaken this effect after 24 h cultivation but produced no effects at longer times. Quantitative real-time polymerase chain reaction showed that mRNA for α4, α7, β2 and β4 subunits of nAChR were expressed in C6 glioma cells. Dietary protein and energy utilization were compared in three Pangasiid species of high economical interest for fish farming in South‐East Asia. Feeding experiments were performed during 4 weeks on Pangasius bocourti, P. djambal and P. hypophthalmus fingerlings, 6.68, 7.69 and 6.41 g mean mass, respectively. A high‐protein diet containing mostly fishmeal was designed to provide proteins without energy limitation. Growth response, variation in body composition and nutrient utilization were compared for fish receiving variable amounts of diet corresponding to increasing protein supply, 0, 5, 15, 25, 35 and 45 g kg?1 day?1. The highest specific growth rate was observed for P. bocourti (6.0% day?1), followed by P. djambal (5.6) and P. hypothalamus (4.1). Pangasius djambal exhibited the highest protein deposition rate, whereas P. bocourti was characterized by a high fat deposition rate even with a low feeding rate. The availability of iron is known to exert a controlling influence on biological productivity in surface waters over large areas of the ocean and may have been an important factor in the variation of the concentration of atmospheric carbon dioxide over glacial cycles. The effect of iron in the Southern Ocean is particularly important because of its large area and abundant nitrate, yet iron-enhanced growth of phytoplankton may be differentially expressed between waters with high silicic acid in the south and low silicic acid in the north, where diatom growth may be limited by both silicic acid and iron. Two mesoscale experiments, designed to investigate the effects of iron enrichment in regions with high and low concentrations of silicic acid, were performed in the Southern Ocean. These experiments demonstrate iron's pivotal role in controlling carbon uptake and regulating atmospheric partial pressure of carbon dioxide. 相似文献

Fucoidans from brown macroalgae are sulfated fucose-rich polysaccharides, that have several beneficial biological activities, including anti-inflammatory and anti-tumor effects. Controlled enzymatic depolymerization of the fucoidan backbone can help produce homogeneous, defined fucoidan products for structure-function research and pharmaceutical uses. However, only a few endo-fucoidanases have been described. This article reports the genome-based discovery, recombinant expression in Escherichia coli, stabilization, and functional characterization of a new bacterial endo-α-(1,4)-fucoidanase, Fhf1, from Formosa haliotis. Fhf1 catalyzes the cleavage of α-(1,4)-glycosidic linkages in fucoidans built of alternating α-(1,3)-/α-(1,4)-linked l-fucopyranosyl sulfated at C2. The native Fhf1 is 1120 amino acids long and belongs to glycoside hydrolase (GH) family 107. Deletion of the signal peptide and a 470 amino acid long C-terminal stretch led to the recombinant expression of a robust, minimized enzyme, Fhf1Δ470 (71 kDa). Fhf1Δ470 has optimal activity at pH 8, 37–40 °C, can tolerate up to 500 mM NaCl, and requires the presence of divalent cations, either Ca2+, Mn2+, Zn2+ or Ni2+, for maximal activity. This new enzyme has the potential to serve the need for controlled enzymatic fucoidan depolymerization to produce bioactive sulfated fucoidan oligomers. 相似文献

Maize prolamin (zein), together with starch, hydroxypropyl methylcellulose, sugar, salt, yeast and water can form wheat-like cohesive, extensible, viscoelastic dough when mixed above room temperature (e.g. 40 °C). This dough is capable of holding gas. However, it is excessively extensible, and when used for hearth-type rolls, it tends to become flat. Bench-scale defatting of zein with chloroform at room temperature significantly (P < 0.05) improved specific volume (4.5 ml/g vs. 3.3 ml/g) and shape of the rolls (width-to-height 2.0 vs. 3.9). The total lipid content determined by accelerated solvent extraction (100 °C, 69 bar, chloroform), however, only decreased from 8.0 to 6.6% due to this bench-scale defatting. Staining experiments with Naphthol Blue Black suggested that bench-scale defatting removed surface lipids from the zein particles, and thus facilitated their aggregation. Aggregation experiments with zein and water at 40 °C, and laser scanning confocal microscopy with zein-starch dough confirmed that zein particles aggregated more easily when surface-defatted. Dynamic oscillatory temperature sweeps demonstrated that surface-defatting lowered the temperature at which protein cross-linking occurred by 2 °C. This research can help to produce superior gluten-free bread and could also possibly contribute to the better understanding of wheat dough. 相似文献

Nitrogen (N) fertilizer represents a significant cost for the grower and may also have environmental impacts through nitrate leaching and N2O (a greenhouse gas) emissions associated with denitrification. The objectives of this study were to analyze the genetic variability in N-use efficiency (grain dry matter (DM) yield per unit N available from soil and fertilizer; NUE) in winter wheat and identify traits for improved NUE for application in breeding. Fourteen UK and French cultivars and two French advanced breeding lines were tested in a 2 year/four site network comprising different locations in France and in the UK. Detailed growth analysis was conducted at anthesis and harvest in experiments including DM and N partitioning. Senescence of either the flag leaf or the whole leaf canopy was assessed from a visual score every 3-4 days from anthesis to complete canopy senescence. The senescence score was fitted against thermal time using a five parameters monomolecular-logistic equation allowing the estimation of the timing of the onset and the rate of post-anthesis senescence. In each experiment, grain yield was reduced under low N (LN), with an average reduction of 2.2 t ha−1 (29%). Significant N × genotype level interaction was observed for NUE. Crop N uptake at harvest on average was reduced from 227 kg N ha−1 under high N (HN) to 109 kg N ha−1 under LN conditions while N-utilization efficiency (grain DM yield per unit crop N uptake at harvest; NUtE) increased from 34.0 to 52.1 kg DM kg−1 N. Overall genetic variability in NUE under LN related mainly to differences in NUtE rather than N-uptake efficiency (crop N uptake at harvest per unit N available from soil and fertilizer; NUpE). However, at one site there was also a positive correlation between NUpE and NUE at LN in both years. Moreover, across the 2 year/four site network, the N × genotype effect for NUpE partly explained the N × genotype effect for grain yield and NUE. Averaging across the 16 genotypes, the timing of onset of senescence explained 86% of the variation in NUtE amongst site-season-N treatment combinations. The linear regression of onset of senescence on NutE amongst genoytpes was not significant under HN, but at three of the four sites was significant under LN explaining 32-70% of the phenotypic variation amongst genotypes in NutE. Onset of senescence amongst genotypes was negatively correlated with the efficiency with which above-ground N at anthesis was remobilized to the grain under LN. It is concluded that delaying the onset of post-anthesis senescence may be an important trait for increasing grain yield of wheat grown under low N supply. 相似文献
