A comparison of gene content and genome architecture of Trypanosoma brucei, Trypanosoma cruzi, and Leishmania major, three related pathogens with different life cycles and disease pathology, revealed a conserved core proteome of about 6200 genes in large syntenic polycistronic gene clusters. Many species-specific genes, especially large surface antigen families, occur at nonsyntenic chromosome-internal and subtelomeric regions. Retroelements, structural RNAs, and gene family expansion are often associated with syntenic discontinuities that-along with gene divergence, acquisition and loss, and rearrangement within the syntenic regions-have shaped the genomes of each parasite. Contrary to recent reports, our analyses reveal no evidence that these species are descended from an ancestor that contained a photosynthetic endosymbiont. 相似文献

Chemical reaction outcomes are often rationalized on the basis of independent analyses of steric and electronic effects. We applied three-dimensional free energy relationships correlating steric and electronic effects to design and optimize a ligand class for the enantioselective Nozaki-Hiyama-Kishi propargylation of ketones. Seven-transmembrane receptor (7TMR) signaling is transduced by second messengers such as diacylglycerol (DAG) generated in response to the heterotrimeric guanine nucleotide-binding protein Gq and is terminated by receptor desensitization and degradation of the second messengers. We show that beta-arrestins coordinate both processes for the Gq-coupled M1 muscarinic receptor. beta-Arrestins physically interact with diacylglycerol kinases (DGKs), enzymes that degrade DAG. Moreover, beta-arrestins are essential for conversion of DAG to phosphatidic acid after agonist stimulation, and this activity requires recruitment of the beta-arrestin-DGK complex to activated 7TMRs. The dual function of beta-arrestins, limiting production of diacylglycerol (by receptor desensitization) while enhancing its rate of degradation, is analogous to their ability to recruit adenosine 3',5'-monophosphate phosphodiesterases to Gs-coupled beta2-adrenergic receptors. Thus, beta-arrestins can serve similar regulatory functions for disparate classes of 7TMRs through structurally dissimilar enzymes that degrade chemically distinct second messengers. 相似文献

We have observed both superluminal and ultraslow light propagation in an alexandrite crystal at room temperature. Group velocities as slow as 91 meters per second to as fast as -800 meters per second were measured and attributed to the influence of coherent population oscillations involving chromium ions in either mirror or inversion sites within the crystal lattice. Namely, ions in mirror sites are inversely saturable and cause superluminal light propagation, whereas ions in inversion sites experience conventional saturable absorption and produce slow light. Final fruit diameter is the prime determinant of sweet cherry fruit value. Previous research has shown that mesocarp cell size accounts predominantly for variability in final fruit size, within a genotype. Our research program evaluated the potential to improve sweet cherry fruit size/weight with growth regulators to affect cell division and/or cell expansion stages. In the current study we screened 8 plant growth regulators (PGRs), including cytokinins, gibberellins, and auxins, and their combinations for their ability to increase ‘Bing’ fruit weight. Each PGR was mixed in lanolin paste and applied to fruit pedicels at 9 or 30 days after full bloom (DAFB), to coincide with estimated peak in cell division and cell expansion activity, respectively. Several cytokinins applied 30 DAFB improved fruit weight significantly (ca. +15%) with N-(2-Chloro-4-pyridyl)-N′-phenylurea (CPPU) and 6-(3-hydroxybenzylamino) purine (mt-Topolin) at 100 mg l−1 being the most effective. Gibberellins, applied alone, improved fruit size and delayed fruit maturation and exocarp coloration. GA3 at 200 mg l−1 applied at 9 DAFB was the most effective and improved final fruit weight by 15%. Fifty-six percent of the fruit from this treatment were ≥9 g compared to 15% of similar weight fruit from untreated limbs. Both GA3 and GA4/7 treatments applied 9 DAFB increased fruit radial expansion. 4-Chlorophenoxyacetic acid, a synthetic auxin, also stimulated higher fruit growth rates at stage I and stage II, and fruit color development, but did not improve final fruit size. 相似文献

Goat browsing can be used as an alternative brush management option for redberry (Juniperus pinchotii Sudw.) and ashe (Juniperus asheii Buch) juniper instead of more expensive and invasive brush control methods, assuming consumption of juniper does not adversely affect the marketability of offspring. Some wildlife species reportedly retain juniper flavor when consumed. We determined if juniper consumption affected meat quality or flavoring of Boer-cross kid carcasses. Twenty recently weaned, Boer-cross wethers were randomly assigned to one of four treatments with treatments fed different amounts of juniper (0%, 10%, 20%, 30% juniper in the diet). All goats were fed juniper for 28 d at the Angelo State University (ASU) Management, Instruction, and Research Center. All goats were also fed a feedlot ration to meet maintenance requirements (2% body weight). Juniper intake varied (P < 0.05) between all treatments (0%, 10%, 20%, 30%) primarily because treatments were fed different amounts of juniper. Following a 28-d trial, goats were harvested at the ASU Food Safety and Product Development Laboratory. Carcass characteristics including live weight, hot carcass weight, dressing percentage, loineye area, body wall fat thickness, and leg circumference were similar (P > 0.05) among treatments. Sensory characteristics including tenderness, juiciness, flavor intensity, off-flavor, and overall acceptability were also similar (P > 0.05) among treatments. Unfortunate bovine fatalities occurring after ingestion of mold-damaged sweetpotatoes preclude the use of the culled tubers in livestock feed. In cattle, mold-damaged sweetpotatoes induce an acute respiratory distress syndrome resulting in asphyxiation. Because of this potential toxicity and the general abundance of culled sweetpotatoes, the detoxification efficacy of ensiling was explored since it is an easy and economically viable technique often applied to preserve livestock feed. Sweetpotato slices with or without mold damage were stored either frozen (to represent unfermented samples) or fermented for 6 weeks at room temperature. Following fermentation, organic extracts were generated for administration to mice. Thirty hours following administration of the extracts, mice were evaluated for gross and microscopic lesions affecting the lungs, liver, and kidneys. Fermentation of 6 weeks duration was observed to inadequately eliminate the lung, liver, and kidney toxicity caused by mold-damaged sweetpotatoes. In fact, fermentation exacerbated the hepatotoxicity of mold-damaged sweetpotatoes. This is also the first demonstration that sweetpotato regions lacking visible mold damage can induce lung and kidney injury, which, however, is preventable by fermentation. 相似文献

The Gila Monster (Heloderma suspectum) is a large, venomous lizard protected throughout its distribution in the southwestern United States and northwestern Mexico. Rapid urban growth in key areas of its range and increased encounters with humans prompted us to investigate translocation as a conservation tool with “nuisance” Gila Monsters. Twenty-five Gila Monsters reported as nuisances by residents in the northeastern Phoenix Metropolitan Area were translocated from 0 to 25,000 m from their point of capture. Subjects (N=18) translocated less than 1000 m returned to their original site of capture within 2-30 days; none of those (N=7) translocated more than 1000 m successfully returned, they exhibited high daily rates of speed, and were deprived the use of familiar refuges. We conclude that small distance translocations within suitable habitats are ineffective in removing Gila Monsters from areas deemed unsuitable. Moreover, individuals moved significantly greater distances are unlikely to remain at a translocation site, and may experience a variety of costs (e.g., predation risk) associated with high rates of movement. 相似文献
