The recovery potential of stream acidification from years of acidic deposition is dependent on biogeochemical processes and varies among different acid-sensitive regions. Studies that investigate long-term trends and seasonal variability of stream chemistry in the context of atmospheric deposition and watershed setting provide crucial assessments on governing biogeochemical processes. In this study, water chemistries were investigated in Noland Divide watershed (NDW), a high-elevation watershed in the Great Smoky Mountains National Park (GRSM) of the southern Appalachian region. Monitoring data from 1991 to 2007 for deposition and stream water chemistries were statistically analyzed for long-term trends and seasonal patterns by using Seasonal Kendall Tau tests. Precipitation declined over this study period, where throughfall (TF) declined significantly by 5.76?cm?year?1. Precipitation patterns play a key role in the fate and transport of acid pollutants. On a monthly volume-weighted basis, pH of TF and wet deposition, and stream water did not significantly change over time remaining around 4.3, 4.7, and 5.8, respectively. Per NDW area, TF SO4 2- flux declined 356.16?eq?year?1 and SO4 2- concentrations did not change significantly over time. Stream SO4 2- remained about 30???eq L?1 exhibiting no long-term trends or seasonal patterns. SO4 2- retention was generally greater during drier months. TF monthly volume-weighted NH4 + and NO3 - concentrations significantly increased by 0.80???eq L?1?year?1 and 1.24???eq L?1?year?1, respectively. TF NH4 + fluxes increased by 95.76?eq?year?1. Most of NH4 + was retained in the watershed, and NO3 - retention was much lower than NH4 +. Stream monthly volume-weighted NO3 - concentrations and fluxes significantly declined by 0.56???eq L?1?year?1 and 139.56?eq?year?1, respectively. Overall, in NDW, inorganic nitrogen was exported before 1999 and retained since then, presumably from forest regrowth after Frazer fir die-off in the 1970s from balsam wooly adelgid infestation. Stream export of NO3 - was greater during winter than summer months. During the period from 1999 to 2007, stream base cations did not exhibit significant changes, apparently regulated by soil supply. Statistical models predicting stream pH, ANC, SO4 2-, and NO3 - concentrations were largely correlated with stream discharge and number of dry days between precipitation events and SO4 2- deposition. Dependent on precipitation, governing biogeochemical processes in NDW appear to be SO4 2- adsorption, nitrification, and NO3 - forest uptake. This study provided essential information to aid the GRSM management for developing predictive models of the future water quality and potential impacts from climate change.  相似文献

Climate change is predicted to cause upward shifts in forest tree distributions, which will require seedling recruitment beyond current forest boundaries. However, predicting the likelihood of successful plant establishment beyond current species' ranges under changing climate is complicated by the interaction of genetic and environmental controls on seedling establishment. To determine how genetics and climate may interact to affect seedling establishment, we transplanted recently germinated seedlings from high- and low-elevation provenances (HI and LO, respectively) of Pinus flexilis in common gardens arrayed along an elevation and canopy gradient from subalpine forest into the alpine zone and examined differences in physiology and morphology between provenances and among sites. Plant dry mass, projected leaf area and shoot:root ratios were 12-40% greater in LO compared with HI seedlings at each elevation. There were no significant changes in these variables among sites except for decreased dry mass of LO seedlings in the alpine site. Photosynthesis, carbon balance (photosynthesis/respiration) and conductance increased >2× with elevation for both provenances, and were 35-77% greater in LO seedlings compared with HI seedlings. There were no differences in dark-adapted chlorophyll fluorescence (Fv/Fm) among sites or between provenances. Our results suggest that for P. flexilis seedlings, provenances selected for above-ground growth may outperform those selected for stress resistance in the absence of harsh climatic conditions, even well above the species' range limits in the alpine zone. This indicates that forest genetics may be important to understanding and managing species' range adjustments due to climate change.  相似文献

Recent studies have identified a phenomenon in which ciliated protozoa engulf Salmonella and the intra-protozoal environment hyperactivates virulence gene expression and provides a venue for conjugal transfer of antibiotic resistance plasmids. The former observation is relegated to Salmonella bearing the SGI1 multiresistance integron while the latter phenomenon appears to be a more generalized event for recipient Salmonella. Our previous studies have assessed virulence gene hyperexpression only with protozoa from the bovine rumen while conjugal transfer has been demonstrated in rumen protozoa from cattle and goats. The present study examined virulence gene hyperexpression for Salmonella exposed to rumen protozoa obtained from cattle, sheep, goats, or two African ruminants (giraffe and bongo). Conjugal transfer was also assessed in these protozoa using Salmonella as the recipient. Virulence gene hyperexpression was only observed following exposure to the rumen protozoa from cattle and sheep while elevated virulence was also observed in these animals. Conjugal transfer events were, however, observed in all protozoa evaluated. It therefore appears that the protozoa-based hypervirulence is not universal to all ruminants while conjugal transfer is more ubiquitous.  相似文献

Limited information is available on the transmission and spread of influenza virus in pig populations with differing immune statuses. In this study we assessed differences in transmission patterns and quantified the spread of a triple reassortant H1N1 influenza virus in naïve and vaccinated pig populations by estimating the reproduction ratio (R) of infection (i.e. the number of secondary infections caused by an infectious individual) using a deterministic Susceptible-Infectious-Recovered (SIR) model, fitted on experimental data. One hundred and ten pigs were distributed in ten isolated rooms as follows: (i) non-vaccinated (NV), (ii) vaccinated with a heterologous vaccine (HE), and (iii) vaccinated with a homologous inactivated vaccine (HO). The study was run with multiple replicates and for each replicate, an infected non-vaccinated pig was placed with 10 contact pigs for two weeks and transmission of influenza evaluated daily by analyzing individual nasal swabs by RT-PCR. A statistically significant difference between R estimates was observed between vaccinated and non-vaccinated pigs (p < 0.05). A statistically significant reduction in transmission was observed in the vaccinated groups where R (95%CI) was 1 (0.39-2.09) and 0 for the HE and the HO groups respectively, compared to an Ro value of 10.66 (6.57-16.46) in NV pigs (p < 0.05). Transmission in the HE group was delayed and variable when compared to the NV group and transmission could not be detected in the HO group. Results from this study indicate that influenza vaccines can be used to decrease susceptibility to influenza infection and decrease influenza transmission.  相似文献

Fire danger rating systems commonly ignore fine scale, topographically-induced weather variations. These variations will likely create heterogeneous, landscape-scale fire danger conditions that have never been examined in detail. We modeled the evolution of fuel moistures and the Energy Release Component (ERC) from the US National Fire Danger Rating System across the 2009 fire season using very high resolution (30 m) surface air temperature, humidity and snow ablation date models developed from a network of inexpensive weather sensors. Snow ablation date occurred as much as 28 days later on North-facing slopes than on South-facing slopes at upper elevations. South-facing slopes were hotter and drier than North-facing slopes but slope position, in addition to aspect, was also important because nocturnal air temperatures were coolest and humidity was highest in valley bottoms. These factors created heterogeneous fuel moistures and fire danger across the study area. In the late season (August and September), nocturnal cold air drainage and high relative humidity fostered fuel moisture recovery in valley bottoms, where fuel moistures and ERC values were 30% and 45% higher and lower, respectively at peak fire danger (September 29th). Dry fuel moistures and relatively high ERC values persisted on low elevation, South-facing slopes. The driest conditions were observed 100-200 m above the valley floor where mid-slope thermal belts frequently developed above areas of cold air pooling. We suggest that a complete understanding of these variations may help improve fire management decision making.  相似文献

Canine histiocytic sarcoma is an aggressive, fatal neoplastic disease with a poor prognosis. Lomustine is generally accepted as the first‐line systemic therapy, although this compound does not provide complete regression. Therefore, research into a novel approach against canine histiocytic sarcoma is needed. However, anti‐tumour effects of oncolytic therapy using reovirus against histiocytic sarcoma are unknown. Here, we showed that reovirus has oncolytic activity in canine histiocytic sarcoma cell lines in vitro and in vivo. We found that reovirus can replicate and induce caspase‐dependent apoptosis in canine histiocytic sarcoma cell lines. A single intra‐tumoural injection of reovirus completely suppressed the growth of subcutaneously grafted tumours in NOD/SCID mice. Additionally, we demonstrated that susceptibility to reovirus‐induced cell death was attributable to the extent of expression of type I interferons induced by reovirus infection in vitro. In conclusion, oncolytic reovirus appears to be an effective treatment option for histiocytic sarcoma, and therefore warrants further investigation in early clinical trials.  相似文献

Although CTL are important for control of lentiviruses, including equine infectious anemia virus (EIAV), it is not known if CTL can limit lentiviral replication in the absence of CD4 help and neutralizing antibody. Adoptive transfer of EIAV-specific CTL clones into severe combined immunodeficient (SCID) foals could resolve this issue, but it is not known whether exogenous IL-2 administration is sufficient to support the engraftment and proliferation of CTL clones infused into immunodeficient horses. To address this question we adoptively transferred EIAV Rev-specific CTL clones into four EIAV-challenged SCID foals, concurrent with low-dose aldesleukin (180,000U/m2), a modified recombinant human IL-2 (rhuIL-2) product. The dose was calculated based on the specific activity on equine PBMC in vitro, and resulted in plasma concentrations considered sufficient to saturate high affinity IL-2 receptors in humans. Despite specific activity on equine PBMC that was equivalent to recombinant equine IL-2 and another form of rhuIL-2, aldesleukin did not support the engraftment and expansion of infused CTL clones, and control of viral load and clinical disease did not occur. It was concluded that survival of Rev-specific CTL clones infused into EIAV-challenged SCID foals was not enhanced by aldesleukin at the doses used in this study, and that in vitro specific activity did not correlate with in vivo efficacy. Successful adoptive immunotherapy with CTL clones in immunodeficient horses will likely require higher doses of rhuIL-2, co-infusion of CD4+ T lymphocytes, or administration of equine IL-2.  相似文献

Ecological restoration is predicated on our abilities to discern plant taxa. Taxonomic identification is a first step in ensuring that plants are appropriately adapted to the site. An example of the need to identify taxonomic differences comes from big sagebrush (Artemisia tridentata). This species is composed of three predominant subspecies occupying distinct environmental niches, but overlap and hybridization are common in ecotones. Restoration of A. tridentata largely occurs using wildland collected seed, but there is uncertainty in the identification of subspecies or mix of subspecies from seed collections. Laboratory techniques that can determine subspecies composition would be desirable to ensure that subspecies match the restoration site environment. In this study, we use spectrophotometry to quantify chemical differences in the water-soluble compound, coumarin. Ultraviolet (UV) absorbance of A. tridentata subsp. vaseyana showed distinct differences among A.t. tridentata and wyomingensis. No UV absorbance differences were detected between A.t. tridentata and wyomingensis. Analyses of samples from > 600 plants growing in two common gardens showed that UV absorbance was unaffected by environment. Moreover, plant tissues (leaves and seed chaff) explained only a small amount of the variance. UV fluorescence of water-eluted plant tissue has been used for many years to indicate A.t. vaseyana; however, interpretation has been subjective. Use of spectrophotometry to acquire UV absorbance provides empirical results that can be used in seed testing laboratories using the seed chaff present with the seed to certify A. tridentata subspecies composition. On the basis of our methods, UV absorbance values < 2.7 would indicate A.t. vaseyana and values > 3.1 would indicate either A.t. tridentata or wyomingensis. UV absorbance values between 2.7 and 3.1 would indicate a mixture of A.t. vaseyana and the other two subspecies.  相似文献

Context

Reestablishing foundational plant species through aerial seeding is an essential yet challenging step for restoring the vast semiarid landscapes impacted by plant invasions and wildfire-regime shifts. A key component of the challenge stems from landscape variability and its effects on plant recovery.

Objectives

We assessed landscape correlates, thresholds, and tipping points for sagebrush presence from fine-scale sampling across a large, heterogeneous area burned the previous year, where we were able to quantify soil surface features that are typically occluded yet can strongly affect recovery patterns.

Methods

Hypothesis testing and binary-decision trees were used to evaluate factors affecting initial sagebrush establishment, using 2171 field plots (totaling?~?2,000,000 m2 sampled) over a 113,000-ha region.

Results

Sagebrush established in 50% of plots where it was seeded, a?>?12-fold greater establishment frequency than in unseeded areas. Sagebrush establishment was enhanced in threshold-like ways by elevation (>?1200 m ASL), topographic features that alter heatload and soil water, and by soil-surface features such as “fertile islands” that bore the imprint of pre-fire sagebrush. Sagebrush occupancy had a negative, linear relationship with exotic-annual grass cover and parabolic relationship with perennial bunchgrasses (optimal at 40% cover).

Conclusions

Our approach revealed interactive, ecological relationships such as novel soil-surface effects on first year establishment of sagebrush across the burned landscape, and identified “hot spots” for recovery. The approach could be expanded across sites and years to provide the information needed to explain past seeding successes or failures, and in designing treatments at the landscape scale.
