Canine histiocytic sarcoma is an aggressive, fatal neoplastic disease with a poor prognosis. Lomustine is generally accepted as the first‐line systemic therapy, although this compound does not provide complete regression. Therefore, research into a novel approach against canine histiocytic sarcoma is needed. However, anti‐tumour effects of oncolytic therapy using reovirus against histiocytic sarcoma are unknown. Here, we showed that reovirus has oncolytic activity in canine histiocytic sarcoma cell lines in vitro and in vivo. We found that reovirus can replicate and induce caspase‐dependent apoptosis in canine histiocytic sarcoma cell lines. A single intra‐tumoural injection of reovirus completely suppressed the growth of subcutaneously grafted tumours in NOD/SCID mice. Additionally, we demonstrated that susceptibility to reovirus‐induced cell death was attributable to the extent of expression of type I interferons induced by reovirus infection in vitro. In conclusion, oncolytic reovirus appears to be an effective treatment option for histiocytic sarcoma, and therefore warrants further investigation in early clinical trials. 相似文献

A generalised linear model was used to predict climatically suitable habitat for the golden bowerbird (Prionodura newtonia De Vis), an endemic species of the Wet Tropics of North Queensland. Mean temperature of the coldest quarter and mean precipitation of the wettest quarter were found to be the best habitat predictors. For independent validation data, accuracy was 67% (Kappa statistic, n=30). Past, and future habitats suitable to the golden bowerbird were mapped by altering the temperature and precipitation input to the model. In the current climate, total potential habitat is estimated to be 1564 km2, occurring as a number of separate patches with distinct bowerbird subpopulations. Past habitat was most limiting, 406 km2, during the Holocene climatic optimum (c. 5000-3600 YBP, +2 °C and +50% rainfall relative to today). With future warming, assuming a 10% decrease in rainfall, potential habitat is reduced to 582 km2 (1 °C warming), 163 km2 (2 °C warming) and 37 km2 (3 °C warming). Thus, global warming in the coming decades is likely to be a significant threat to the survival of this and similar upland and highland species in the tropics. 相似文献

Theory predicts the water hexamer to be the smallest water cluster with a three-dimensional hydrogen-bonding network as its minimum energy structure. There are several possible low-energy isomers, and calculations with different methods and basis sets assign them different relative stabilities. Previous experimental work has provided evidence for the cage, book, and cyclic isomers, but no experiment has identified multiple coexisting structures. Here, we report that broadband rotational spectroscopy in a pulsed supersonic expansion unambiguously identifies all three isomers; we determined their oxygen framework structures by means of oxygen-18-substituted water (H(2)(18)O). Relative isomer populations at different expansion conditions establish that the cage isomer is the minimum energy structure. Rotational spectra consistent with predicted heptamer and nonamer structures have also been identified. 相似文献

Edwardsiella ictaluri was consistently isolated from the spleens, livers, and head kidneys of diseased Nile tilapia Oreochromis niloticus from a farm experiencing mortality events in several culture ponds. We describe the first published outbreak of E. ictaluri-induced edwardsiellosis in Nile tilapia. Pure cultures of the isolated bacteria were characterized both biochemically and molecularly. Biochemical analysis was performed using the API-20E and RapID One systems, and antimicrobial susceptibility was determined by the broth microdilution method. Molecular analysis involved sequencing of the 16S rRNA gene, species-specific real-time polymerase chain reaction (PCR), and PCR-mediated genomic fingerprinting (rep-PCR). Pairwise sequence analysis of the 16S rRNA gene identified the case isolates to be a 100% match to E. ictaluri cultured from channel catfish in the southeastern United States. However, rep-PCR analysis identified the case isolates to be genetically different from representative strains isolated from disease outbreaks in cultured channel catfish in Mississippi. Infectivity challenges (intraperitoneal injection and immersion) demonstrated that a representative E. ictaluri strain isolated from tilapia was pathogenic to naive tilapia, reproducing clinical signs and mortality, thereby establishing Koch's postulates. 相似文献

Plant-soil variation related to perennial-plant resource islands (coppices) interspersed with relatively bare interspaces is a major source of heterogeneity in desert rangelands. Our objective was to determine how native and exotic grasses vary on coppice mounds and interspaces (microsites) in unburned and burned sites and underlying factors that contribute to the variation in sagebrush-steppe rangelands of the Idaho National Lab, where interspaces typically have abiotic crusts. We asked how the exotic cheatgrass (Bromus tectorum L.) and native bluebunch wheatgrass (Pseudoroegneria spicata [Pursh] A. Löve) were distributed among the microsites and measured their abundances in three replicate wildfires and nearby unburned areas. We conducted a common-garden study in which soil cores from each burned microsite type were planted with seed of either species to determine microsite effects on establishment and growth of native and exotic grasses. We assessed soil physical properties in the common-garden study to determine the intrinsic properties of each microsite surface and the retention of microsite soil differences following transfer of soils to the garden, to plant growth, and to wetting/drying cycles. In the field study, only bluebunch wheatgrass density was greater on coppice mounds than interspaces, in both unburned and burned areas. In the common-garden experiment, there were microsite differences in soil physical properties, particularly in crust hardness and its relationship to moisture, but soil properties were unaffected by plant growth. Also in the experiment, both species had equal densities yet greater dry mass production on coppice-mound soils compared to interspace soils, suggesting microsite differences in growth but not establishment (likely related to crust weakening resulting from watering). Coppice-interspace patterning and specifically native-herb recovery on coppices is likely important for postfire resistance of this rangeland to cheatgrass. 相似文献

Francisella noatunensis subsp. orientalis (syn. F. asiatica) (Fno) is an emergent fish pathogen that causes acute to chronic disease in a wide variety of freshwater, brackish and marine fish. Due to the emergent nature of this bacterium, established protocols to measure antimicrobial susceptibility are lacking. In this project we compare three different methods to examine the antimicrobial susceptibility (Etest, broth microdilution and disk diffusion) of 10 different isolates of Fno from two different fish species and four different geographic outbreaks from 2006 to 2010. PCR mediated genomic fingerprinting (rep-PCR) performed on the different isolates confirmed genetic homogeneity amongst the different isolates. The in vitro susceptibility data presented here provides important baseline data for future research monitoring the development of antibiotic resistance among Fno isolates as well as provides invaluable data for the development of potential therapeutics. 相似文献

OBJECTIVE: To evaluate outcome of small ruminants after tube cystostomy without urethral flushing. STUDY DESIGN: Retrospective clinical study. ANIMALS: Small ruminants (n=63; 50 goats, 13 sheep). METHODS: Medical records (January 1996-October 2003) for all small ruminants undergoing tube cystostomy were analyzed. Signalment, laboratory data, intraoperative findings, urolith type, and postoperative outcome were compared. Long-term follow-up was obtained at 6 and > or= 12 months postoperatively. RESULTS: Most animals were castrated males. Pygmy goats were overrepresented. Mean time to normal urination after tube cystostomy was 11 days, and mean hospitalization was 14 days. Seventy-six percent of animals were successfully treated with tube cystostomy (90% of those discharged from hospital). Short- and long-term survival was good for those animals with follow-up data. Reobstruction occurred in <20% of animals at 6 and 12 months. Goats, castrated males, and animals with normal serum potassium, no fluid in the abdomen, or no urethral process amputation were more likely to survive. Stone type, bladder appearance, and age at castration were not associated with outcome. CONCLUSION: Surgical tube cystostomy without urethral flushing is an effective technique for resolution of obstructive urolithiasis in small ruminants. CLINICAL RELEVANCE: Goats, small ruminants with an intact urethral process, absence of abdominal fluid, and serum potassium concentration <5.2 mEq/L at admission were all associated with survival after tube cystostomy. Castrated males may also be more likely to survive. These factors should be considered in management of small ruminants with urolithiasis. 相似文献

Episodic acidification of surface waters has been observed in the Great Smoky Mountains National Park, similar to other forested watersheds with base-poor bedrock in the eastern US receiving acids from atmospheric deposition. Three remote, forested, high-elevation streams were selected in the Little Pigeon River watershed for study; two of which brook trout have extirpated, and believed to have resulted from severe acidity during stormflows. This research characterized stream chemistry during episodes in order to better understand potential factors that contribute to rapid drops in pH and acid neutralizing capacity (ANC) during stormflows. Autosamplers initialized by sondes, collected samples during storm events for analysis of pH, ANC, cations, and anions over a 15-month period. ANC and pH depressions, and increased concentrations in sulfate, nitrate, and organic acids were observed for all storms at each study site. ANC contribution analysis indicated sulfate was the strongest contributor to ANC depressions, but nitrate, cation dilution, and organic acids were also significant in some cases. Acidic deposition appears to be the primary source of episodic acidification, supported also by the finding that larger stormflows preceded by long, dry periods resulted in significantly larger pH depressions. It appears stream acidification episodes may be driven by acid deposition. However, this study documents the variability of several ion contributors to observed stormflow ANC depressions illustrating the spatial and temporal complexity of watershed processes that influence this phenomenon. 相似文献
