The significance of Mn and Fe for the growth of a coastal marine diatom Thalassiosira weissflogii was investigated by performing culture experiments containing macronutrients with either Mn or Fe, or both. Only the addition of both Mn and Fe induced the highest growth rates and maximal cell yields. Maximal growth was maintained in continuous culture media, which were repeatedly prepared by an inoculation of pre-culture and the addition of both Mn and Fe to the control culture medium containing macronutrients. In particular, it was found that the full growth recovery in Mn-sufficient medium (without added Fe) is accomplished by the addition of Fe even after several days’ incubation. On the contrary, there was no sufficient growth recovery by the addition of Mn after a long incubation time in Fe-sufficient medium but without additional Mn. These results suggest that T. weissflogii in Mn-sufficient waters retains the ability for full physiological recovery for a long time, probably resulting from the decrease in the oxidative stress of phytoplankton by the production of antioxidant enzyme Mn superoxide dismutase during a long incubation period.  相似文献

Neighboring genes are often coordinately expressed within cis-regulatory modules, but evidence that nonparalogous genes share functions in mammals is lacking. Here, we report that mutation of either TMEM138 or TMEM216 causes a phenotypically indistinguishable human ciliopathy, Joubert syndrome. Despite a lack of sequence homology, the genes are aligned in a head-to-tail configuration and joined by chromosomal rearrangement at the amphibian-to-reptile evolutionary transition. Expression of the two genes is mediated by a conserved regulatory element in the noncoding intergenic region. Coordinated expression is important for their interdependent cellular role in vesicular transport to primary cilia. Hence, during vertebrate evolution of genes involved in ciliogenesis, nonparalogous genes were arranged to a functional gene cluster with shared regulatory elements.  相似文献

The determination of the fine thermal structure of the solar corona is fundamental to constraining the coronal heating mechanisms. The Hinode X-ray Telescope collected images of the solar corona in different passbands, thus providing temperature diagnostics through energy ratios. By combining different filters to optimize the signal-to-noise ratio, we observed a coronal active region in five filters, revealing a highly thermally structured corona: very fine structures in the core of the region and on a larger scale further away. We observed continuous thermal distribution along the coronal loops, as well as entangled structures, and variations of thermal structuring along the line of sight.  相似文献

The velocity changes of ultrasonic shear waves propagating transversely to the applied stress direction in wood were investigated. The wave oscillation directions were parallel and normal to the uniaxially applied stress direction. The velocities of the shear waves for both oscillations decreased as the compressive load increased, and increased as the tensile load increased. The velocity of the normally oscillated shear wave showed smaller change against the stress applied than that of the parallel oscillated wave. The initial birefringence due to the orthotropy of wood was observed without any stress. Velocity changes in the two principally oscillated shear waves were proportional to the stress within the stress range tested. The acoustoelastic birefringence effect was obtained from the velocity difference between the two shear waves. The relative difference between the two velocities (called acoustic anisotropy) was given as a function of the applied stress. The acoustoelastic birefringence constants were obtained from the relationships between the acoustic anisotropy and the applied stress.  相似文献

Legumes establish symbiosis with nitrogen-fixing rhizobia through root nodules to acquire nitrogen. Legumes control nodule number through systemic (autoregulation of nodulation) as well as local regulation. Moreover, plants defend themselves against bacteria and other pathogens through the induction of localized (localized acquired resistance) and systemic (SAR, systemic acquired resistance; ISR, induced systemic resistance) responses. Herein, we show that the number of root nodules is suppressed by programmed cell death (PCD), and is simultaneously controlled by SAR and ISR in soybean (Glycine max [L.] Merr.). The wild-type soybean cultivar Williams 82 showed markedly fewer root nodule primordia and PCD symptoms, including accelerated DNA degradation, enhanced generation of reactive oxygen species (visualized by 3,3′-diaminobenzidine staining), and excessive cell death (detected on staining with trypan blue) compared to the hypernodulation mutant NOD1-3. These results suggest that PCD suppresses the formation of root nodules in wild-type soybean. In addition, microarray and gene ontology analyses showed that essential components of hypersensitive response (HR) or disease resistance, such as resistance (R) genes, mitogen-activated protein kinase cascade, SAR, salicylic acid, jasmonic acid, ethylene, etc., were activated in wild-type plants. These analyses corroborate the above findings, demonstrating that the suppression of root nodule formation by PCD is accompanied by HR, and is simultaneously controlled by SAR and ISR in soybean. These findings provide new insight into the control of nodulation to balance nutritional requirements and energy status in legumes.  相似文献

Carnation (Dianthus caryophyllus L.) belongs to the family Caryophyllaceae in the order Caryophyllales. Plants in this order do not accumulate carotenoids in petals. To understand how carotenoid accumulation is controlled in carnation petals, we analysed the expression of genes related to carotenoid accumulation. Petals at an early stage of development accumulated small amounts of carotenoids. As petals matured, their carotenoid content decreased to extremely low levels. In contrast, carnation leaves contained substantial amounts of carotenoids that are essential for photosynthesis. Most of the carotenogenic genes were expressed in petals at levels similar to those in leaves, and the expression levels of these genes increased during petal development. Genes encoding phytoene synthase and lycopene ε‐cyclase were exceptions. Their expression levels in petals were very low compared with those in leaves. Expression of the gene encoding carotenoid cleavage dioxygenase 4 was detected in neither leaves nor petals. These data suggest that the low levels of carotenoids in carnation petals are caused not by enzymatic degradation but rather by low rates of carotenoid biosynthesis.  相似文献

The salmonid pituitary produces two chemically distinct gonadotropins (GTHI and GTHII). Ultrastructural characteristics of GTHI- and GTHII-producing cells were studied in the trout pituitary with electronmicroscopic immunocytochemistry using antisera against salmon GTHIβ- and IIβ-subunits. In pituitaries from vitellogenic fish, GTHI-cells were characterized by numerous dilated cisternae of the granular endoplasmic reticulum (GER) and a small number of Iβ-positive granules (diameter, 100–300 nm), whereas GTHIIβ-immunoreactivity was found on granules (diameter, 200–400 nm) and large globules (diameter, 500–4000 nm) in apparently different cells (GTHII-cells). Distinct cellular distributions of GTHI and GTHII were maintained during gametogenesis, although morphological characteristics of GTHI- and GTHII-cells overlapped each other due to changes in number and size of the granules, globules and cisternae of the GER. Interestingly, the globules in the GTHI-cells were immunonegative for GTHIβ, although in the GTHII-cells they were always stained with GTHIIβ-antiserum. These results confirm that GTHIβ and GTHIIβ are synthesized in distinctly different cell-types in the salmonid pituitary and indicate that morphological characteristics cannot be used to distinguish these two cell-types.
Résumé L'hypophyse des salmonidés produit deux gonadotropines (GTHI et GTHII) chimiquement distinctes. Les caractéristiques ultrastructurales des cellules produisant la GTHI et la GTHII ont été étudiées par immunocytochimie en microscopie électronique en utilisant des anticorps dirigés contre les sousunités GTHIβ et GTHIIβ de saumon. Chez les poissons en vitellogenèse, les cellules à GTHI se caractèrisent par un réticulum endoplasmique granulaire (REG) contenant de nombreuses citernes dilatées et un petit nombre de granules positives à la GTHIβ (diamètre 100–300 nm) tandis qu'une immunoréactivité à la GTHII était trouvée sur des granules (diamètre 200–4000 nm) et de grands globules (diamètre 500–4000 nm) dans des cellules apparemment différentes (cellules à GTHII). Des distributions cellulaires distinctes de la GTHI et la GTHII se sont maintenues à des stades plus avancées de la gamétogenèse, bien que les caractéristiques morphologiques des cellules à GTHI et GTHII se recoupent suite à des changements dans le nombre et la taiile des granules, des globules et des citernes du REG. II est à remarquer que les globules prśents dans les cellules à GTHI étaient immunonégatifs à la GTHIβ alors que dans les cellules à GTHII ils étaient toujours marqués par l'anticorps contre la GTHIIβ. Ces résultats confirment que, dans l'hypophyse des salmonidés, les GTHIβ et GTHIIβ sont synthetisées dans des types cellulaires différents et indiquent que des caractéristiques morphologiques ne peuvent être utilisées pour distinguer ces deux types cellulaires.

The animal model is a powerful and fundamental tool in the field of biochemical research including toxicology, carcinogenesis, cancer therapeutics and prevention. In the carcinogenesis animal model system, numerous examples of preneoplastic lesions have been isolated and investigated from various perspectives. This may indicate that several options of endpoints to evaluate carcinogenesis effect or therapeutic outcome are presently available; however, classification of preneoplastic lesions has become complicated. For instance, these lesions include aberrant crypt foci (ACF), dysplastic ACF, flat ACF, β-catenin accumulated crypts, and mucin-depleted foci. These lesions have been induced by commonly used chemical carcinogens such as azoxymethane (AOM), 1,2-dimethylhydrazine (DMH), methylnitrosourea (MUN), or 2-amino-1-methyl-6-phenylimidazo[4,5-b]pyridine (PhIP). Investigators can choose any procedures or methods to examine colonic preneoplastic lesions according to their interests and the objectives of their experiments. Based on topographical, histopathological, and biological features of colon cancer preneoplastic lesions in the animal model, we summarize and discuss the character and implications of these lesions.  相似文献

Western blotting was performed to analyze Neospora caninum tachyzoite antigens recognized by mouse IgG at different stages of infection including recrudescence. At the early stage of infection, a 36-38 kDa antigen was clearly recognized by the mouse antisera. After day 48 postinoculation, the signal of the 36-38 kDa antigen gradually weakened. Meanwhile, a 43 kDa antigen was intensely and continuously recognized from 48 to 125 days postinoculation. This 43 kDa antigen was clearly detectable with the antisera from the mice under immunosuppression. Sera from naturally infected cattle strongly reacted with the 43 kDa antigen. Therefore, the 43 kDa antigen may be useful for immunological reactions to detect infected animals except in the early stage of the infection.  相似文献

An 8-year and 6-month-old female Maltese dog showed a stoop with rigidity of her cervix and back. Neurologic examination showed loss of proprioception, and deficiency of pain response. Postmortem examination revealed the neoplastic mass replacing the central area in the cervical spinal cord at the level from 4th to 5th segments. Histologically, the mass was composed of neoplastic ependymal cells. The neoplastic cells showed marked atypism, and occasionally formed ependymal rosettes. Based on the morphologic features, the tumor was diagnosed as anaplastic ependymoma. Immunohistochemistry showed that the neoplastic cells were negative for glial fibrillary acid protein, and slightly positive for vimentin and cytokeratin.  相似文献
