The complete nucleotide sequences of L3 and S7 segments of Ibaraki virus encoding for the major inner capsid proteins, VP3 and VP7

The complete nucleotide sequences of the genes encoding two of the major inner capsid proteins of Ibaraki virus (IBAV), belonging to epizootic hemorrhagic disease virus serotype 2 (EHDV-2) were determined. The L3 RNA segment is 2768 nucleotides in length which encodes VP3 polypeptides of 899 amino acid residues (M.W. 103 kDa). The S7 RNA segment, which encodes the VP7 core protein, is 1162 nucleotides in length and encodes 349 amino acids (M.W. 38 kDa). These RNA segments had the characteristic consensus motifs of Orbivirus RNA segments in termini, namely 5'-GUUAAA... and ...ACUUAC-3'. The comparison of the IBAV L3 and S7 sequences with those of other two EHDV-2 isolates revealed the higher homologies of 93% and 92% against EHDV-2 Australia isolate (EHDV-2AUS) and lower homologies of 80% and 81% against EHDV-2 North America isolate, respectively. The phylogenetic analysis based on L3 and S7 genes also indicated close relationships between IBAV and EHDV-2AUS. KEY WORDS: dsRNA gene, lbaraki virus, inner capsid, VP3, VP7.     

Phylogenetic analysis of Trypanosomatina (Protozoa: Kinetoplastida) based on minicircle conserved regions

Phylogenetic relationships within the suborder Trypanosomatina were inferred from the kinetoplast DNA minicircle conserved region sequences. Trees built using distance-matrix (Neighbor-Joining) and maximum parsimony methods showed that the minicircle conserved regions (CRs) provide a sensitive and specific molecular marker suitable for phylogenetic analyses of subspecies and strains of trypanosomatid flagellates, as testified by the subdivision of the genus Leishmania into the subgenera Leishmania. Viannia and Sauroleishmania. However, since Phytomonas and monogenetic parasites of insects represent the earliest diverging groups, the CRs do not seem to be useful for inference of relationships among major lineages of the order Kinetoplastida.     

Parvovirus infections in dogs and cats: problems of immunization

The general problems of immunization against parvoviroses are only understandable when knowing the basic principles of epidemiology, pathogenesis and immunology of those virus infections. The main difficulties with regard to vaccinations are 1. efficacy of homologous respectively heterologous vaccines, 2. indications for live resp. inactivated vaccines, 3. combination with other vaccines, 4. bridging of the "immunological gap" (refractory phase) of puppies, 5. mode of application and timing of vaccinations (schedule of vaccinations), 6. failure in vaccinated animals, 7. complications originating from vaccinations, 8. passive immunization.     

True hermaphroditism in a horned goat with 60XX/60XY chimerism

Anatomical, behavioral, histological, endocrinological, and cytogenetic characteristics were determined in a horned intersex goat. Histology of the gonads confirmed that the goat was a true hermaphrodite. Cytogenetic analysis showed that it was a chimera (60XX/60XY). Use of laparoscopy allowed determination of characteristics of the internal gonads.     

Evaluation of an oscillometric blood pressure monitor for use in anesthetized cats

OBJECTIVE: To determine accuracy of an oscillometric blood pressure monitor used over a wide range of pressures in anesthetized cats. DESIGN: Prospective study. ANIMALS: 6 healthy cats. PROCEDURE: 4 female cats and 2 male cats that weighed 2.7 to 4.5 kg (5.9 to 9.9 lb) and were 2 to 8 years old were anesthetized. Blood pressure was measured directly with an arterial catheter placed in the right femoral artery and indirectly from the left antebrachium by use of an oscillometric monitor. A series of diastolic arterial pressure (DAP), mean arterial pressure (MAP), and systolic arterial pressure (SAP) measurements were obtained during hypotension, normotension, and hypertension. Values obtained indirectly and directly were compared. RESULTS: The oscillometric monitor was accurate for DAP and MAP throughout the entire pressure range and met the standards of the Association for the Advancement of Medical Instrumentation (mean +/- SD difference from values obtained directly, < or = 5 +/- 8 mm Hg). The SAP was increasingly underestimated with increasing overall pressure; mean differences from direct measurements were -5.2, -12.1, and -17.7 mm Hg during hypo-, normo-, and hypertension, respectively. Standard deviations for SAP were all < or = 8 mm Hg. The monitor gave readings during all attempts. The direct blood pressure recording system appeared to perform well with neither under- nor overdamping. CONCLUSIONS AND CLINICAL RELEVANCE: Except for a minor underestimation of SAP during normo- and hypertension, the oscillometric monitor yielded reliable and easily obtainable blood pressure measurements in anesthetized cats.     

Einfluss Weicher Röntgenstrahlen und Mechanischer Effekte auf das Keimen von Kartoffeln

Zusammenfassung Um eine schonende Keimhemmung bei Kartoffeln der SorteBona zu erzielen, wurden Versuche mit sehr weichen R?ntgenstrahlen von 12 keV Energie durchgeführt. Sogar mit einer Bestrahlungsdosis von 10.000 bis 15.000 r wird — bei der hier angewandten weichen R?ntgenstrahlung — eine über 22 Wochen andauernde Keimhemmung erreicht ohne Verschlechterung des ?usseren Ansehens und Geschmacks. Der Schwund der Knollen wurde einzeln bis 5.000 r gemessen und mit anwachsender Dosis bis zu 15.000 r eine starke Verminderung des Schwundes erzielt. Au?erdem wurde nachgewiesen, da? durch langstündiges, vorsichtiges Rollen in der beschriebenen Versuchsanordung ebenfalls eine Keimhemmung zu erreichen ist. Es wird versucht, diese Erscheinung biologisch und rechnerisch zu deuten. Aus praktischen Gründen w?re es wünschenswert, den neuen, erw?hnten Effekt weiter zu untersuchen.

Summary In order to bring about sparing sprout inhibition of theBona variety of potatoes experiments were undertaken with very weak X-rays of an energy of 12 keV. A radiation dose of only 10.000 to 15.000 r (with the weak X-radiation employed in this instance) was sufficient to inhibit sprouting for over 22 weeks without adversely affecting the appearance and flavour. The tuber shrinkage was individually measured up to 5.000 r and a marked reduction of shrinkage obtained with an increasing dose up to 15.000 r. It was also shown that sprouting may be inhibited by protracted, careful rolling in the described order of experiments. An attempt is made to explain this phenomenon biologically and analytically. From practical considerations it would be desirable to make a further study of the novel effect referred to.

Résumé Pour réaliser de fa?on délicate une inhibition de la germination des pommes de terre de la variétéBona, nous avons effectué des essais de traitement par les rayons X très mous d'une énergie de 12 keV. Même l'irradition à une dose de 10.000 à 15.000 r sufit déjà — dans le cas des rayons très mous utilisés dans ces essais — pour inhiber la germination pendant 22 semaines au moins sans nuire à l'aspect ni au go?t des tubercules. La perte de poids des tubercules fut mesurée isolément jusqu'à 5000 r. Les doses croissantes jusqu'à 15.000 r réduisaient fortement la perte de poids. Il a également été constaté que dans les conditions de l'essai, un roulement très lent et doux des tubercules permet aussi de retarder la germination. Nous avons tenté d'expliquer ce phénomène au point de vue biologique et statistique. Pour des raisons pratiques, il serait désirable que l'étude de cet effet nouvellement observé soit approfondie.
