β-Cryptoxanthin (β-cry), an antioxidant abundant in citrus fruits, plays an important role in the prevention and treatment of certain diseases, especially cancers. In the present study, to increase the content of β-cry in citrus flavedo, the effects of blue (470 nm) and red (660 nm) light-emitting diode (LED) lights on the accumulation of carotenoids and expression of genes related to carotenoid biosynthesis were investigated in the flavedo of Satsuma mandarin. The results showed that accumulation of β-cry was induced by red light, while it was not affected by blue light. The accumulation of β-cry under red light was attributed to simultaneous increases in the expression of CitPSY, CitPDS, CitZDS, CitLCYb1, CitLCYb2, CitHYb, and CitZEP. The results presented herein might provide new strategies to enhance the commercial and nutritional value of citrus fruits.  相似文献

To establish blood and biochemical references for the endangered Kiso horse, blood samples were collected from 111 adult Kiso horses, 74.5% of the existing breed. The samples were analyzed for 23 hematological and biochemical parameters to determine their means and standard deviations (SD). We compared the mean ± 2SD with the reference values cited in one of the most commonly used veterinary textbooks in Japan. The hematology of Kiso horses is characterized by lower erythrocyte count and hematocrit and hemoglobin levels. In addition, their serum biochemistry showed lower levels of aspartate transaminase, alkaline phosphatase, and γ-glutamyl transferase. Whether these propensities are attributed to breed-specific factors or are acquired factors remains unclear. Nevertheless, this study provides useful diagnostic indices for the endangered Kiso horse.  相似文献

The objective was to determine the effect of glucose supplementation on development (to the blastocyst stage) of in vitro matured (IVM) porcine oocytes that were either in vitro fertilized (IVF) or electrically activated (EA). Embryos were incubated for 46 or 58 h post insemination (hpi) in an NCSU37-based medium containing 0.17 mM sodium pyruvate and 2.73 mM sodium lactate (IVC-PyrLac), and then transferred to an NCSU37-based medium containing 5.55 mM glucose (IVC-Glu) and cultured until Days 6 (Day 0 = day of EA or IVF). The proportions of oocytes that had formed full blastocysts by Day 6 following transfer to IVC-glu at 46 hpi was 23.5 and 41.2% in the IVF and EA groups respectively; these were lower (P<0.001) than the proportions of oocytes that formed full blastocysts after transfer at 58 hpi (60.3 and 78.7%). However, there was no significant difference in total cell number (at Day 6) between embryos transferred at 46 vs 58 hpi. We inferred that in vitro-derived pig embryos can efficiently use glucose as an energy source starting at approximately 58 hpi; exposure to glucose at that time enhanced development to the blastocyst stage as well as blastocyst quality.  相似文献

The clinical utility of the urine albumin/creatinine ratio (UAC) using a simplified analyzer for estimation of proteinuria was studied in cats and dogs. Measurement results for diluted feline and canine albumin standard solutions showed linearity. Although conversion formulas (y=1.28x+1.04 and y=1.67x+10.47 for cats and dogs, respectively) were necessary, urine albumin concentrations could be determined in both animals. In cats and dogs with proteinuria, the UAC changed parallel with the urine protein/ creatinine ratio (UPC), and the Log UAC and Log UPC were significantly correlated (r=0.803 (p<0.01) in cats, r=0.801 (p<0.01) in dogs). The UAC using an UAC analyzer could be used clinically as one of the basic in-hospital laboratory tests for estimation of proteinuria in cats and dogs.  相似文献

A 7‐year‐old neutered female domestic shorthaired cat born in Poland and then moved to Japan presented to the local clinic with recent onset of convulsive cluster seizures and status epilepticus. Magnetic resonance imaging revealed bilateral swelling of the hippocampus with T2 hyperintensity and contrast enhancing image, suggesting hippocampal necrosis. The cat completely recovered after treatment with antiepileptic drugs (AED) and administration of prednisolone (1 mg/kg PO q24h for 4 days and tapered). However, cluster seizures reoccurred and developed into status epilepticus despite increasing doses of AED. Although the convulsions were resolved by other AEDs, stupor and renal failure developed, and the cat was euthanized. Pathological findings were consistent with hippocampal necrosis. Immunological analysis for leucine‐rich glioma inactivated 1 (LGI1) autoantibodies was negative, but antibodies against DCC (deleted in colorectal carcinoma) known as netrin‐1 receptor were found. This report describes a case of feline autoimmune limbic encephalitis and hippocampal necrosis that were presumably associated with DCC autoantibodies.  相似文献

Canine prostate cancer (cPCa) is a malignant neoplasm with no effective therapy. The BRAF V595E mutation, corresponding to the human BRAF V600E mutation, is found frequently in cPCa. Activating BRAF mutations are recognized as oncogenic drivers, and blockade of MAPK/ERK phosphorylation may be an effective therapeutic target against BRAF-mutated tumours. The aim of this study was to establish a novel cPCa cell line and to clarify the antitumor effects of MEK inhibitors on cPCa in vitro and in vivo. We established the novel CHP-2 cPCa cell line that was derived from the prostatic tissue of a cPCa patient. Sequencing of the canine BRAF gene in two cPCa cell lines revealed the presence of the BRAF V595E mutation. MEK inhibitors (trametinib, cobimetinib and mirdametinib) strongly suppressed cell proliferation in vitro, and trametinib showed the highest efficacy against cPCa cells with minimal cytotoxicity to non-cancer COPK cells. Furthermore, we orally administered 0.3 or 1.0 mg/kg trametinib to CHP-2 xenografted mice and examined its antitumor effects in vivo. Trametinib reduced tumour volume, decreased phosphorylated ERK levels, and lowered Ki-67 expression in xenografts in a dose-dependent manner. Although no clear adverse events were observed with administration, trametinib-treated xenografts showed osteogenesis that was independent of dosage. Our results indicate that trametinib induces cell cycle arrest by inhibiting ERK activation, resulting in cPCa tumour regression in a dose-dependent manner. MEK inhibitors, in addition to BRAF inhibitors, may be a targeted agent option for cPCa with the BRAF V595E mutation.  相似文献

ABSTRACT

Due to a decrease in phosphorus (P) and potassium (K) mining, manure is incinerated to concentrate P and K in ash. To understand the alternative use of manure-derived ash as P and K sources, laboratory and greenhouse experiments were conducted to determine the relationship between extractability and P and K uptake in cattle manure ash (CMA) and that between CMA application and a grass tetany hazard. The results showed that more P was extracted with 2% citric acid (90% of the total P) than with 2% formic acid (72–84% of the total P). Ninety-one percent of the total K was soluble in water. A greenhouse pot experiment was conducted to test P and K availability to Guinea grass (Megathyrsus maximus). Cattle manure ash or calcium dihydrogen phosphate (CF) was incorporated into sandy soil at 10, 20, and 50 g P2O5 m?2. Two combinations of CMA and CF were tested at 20 g P2O5 m?2. Potassium rates followed K content in CMA applied at different rates of P equivalent to 19, 38, or 96 g K2O m?2. In four harvests, there was no significant difference in the total yields between CMA and CF treatments. The total P uptake was significantly lower in the CMA treatment than in the CF treatment, while it was not in the combined CMA and CF treatments. The P uptake in response to different extraction methods indicated that the extraction of P by 2% formic acid without sonication is recommended to predict P availability in CMA. The potassium uptake from CMA application was comparable to that from the KCl application, and excessive K occurred at 38 and 96 g K2O m?2. The grass tetany hazard ratio higher than 2.2 was observed at the beginning period at the lowest application rates of CMA and CF. In conclusion, the combination use of CMA and CF was better than the single use of CMA. Moreover, CMA would be an available K source, but the grass tetany hazard still needs to be considered in application rates and pretreatments.  相似文献

The relay intercropping system for cultivating direct-seeding rice and winter cereal is a low-cost method particularly for double cropping, because it eliminates the steps of raising seedlings, paddling, and transplanting. However, in this system, the seedling establishment rate (SER) of rice is low and unstable. The objective of this study was to identify the factors affecting SER to highlight the ways to improve SER. Experiments were conducted in experimental fields in Tsukuba City, Ibaraki Prefecture in 2015 and 2016. To determine the time of rice seed death and to calculate seed survival rates, ‘Akidawara’ seeds were embedded in soil (depth: 3 cm) and dug up after defined periods. We then analyzed the correlation between SER and meteorological factors at two sites (Ibaraki Prefecture and Gunma Prefecture) over the same two years. Based on mean air temperature (MT), we divided the period from seeding to June 20 (when irrigation had been initiated in almost all the fields) into four phases. In Phase 1, the number of days with rainfall (≥5 mm) and soil-wetting days (water potential ≥ ?100 kPa) were significantly and negatively correlated with SER. We found that most of the seed deaths occurred after germination, and the germination rates were presumably affected by water absorption during the low temperature phase. Further investigations are needed to understand the occurrences during the seedling emergence period. This study contributes to a better understanding of the factors affecting variations in the SER of direct-seeded rice grown in the relay-intercropping system.

Cigarette smoking is one of the major risk factors of bladder cancer in humans. To date, however, there is no experimental evidence for the effects of inhalation exposure to mainstream cigarette smoke on bladder carcinogenesis. The purpose of the present study was to evaluate the effect of inhalation of mainstream cigarette smoke on mouse bladder carcinogenesis using a cigarette smoke inhalation exposure system. Six-week-old male C57BL mice were administered 0.025% N-butyl-N-(4-hydroxybutyl) nitrosamine (BBN) in their drinking water for 8 weeks and then divided into 2 groups and exposed to 0 or 300 mg/m3 wet total particulate matter mainstream cigarette smoke for 2 h per day, five times per week, for 22 weeks. The incidences of bladder tumors (papilloma and urothelial carcinoma) tended to increase in the cigarette smoke-exposed group (25.0%) compared with the controls (15.8%), albeit without a statistically significant difference. We also evaluated mRNA expression levels of cytochrome P450 (cyp) enzymes and proliferating cell nuclear antigen (PCNA) in the bladder epithelium. Expression of cyp1a1 was significantly increased in the cigarette smoke-exposed group. Cigarette smoke exposure did not have a significant effect on the expression of cyp1a2, cyp 1b1, cyp 2a4, cyp 2b10, cyp 2e1, or PCNA. In conclusion, limited exposure to mainstream cigarette smoke for 22 weeks, caused a significant increase in cyp1a1 expression. This increase coupled with the nonsignificant increase in bladder tumors suggests that a longer period of exposure is required to clarify the effects of cigarette smoke on bladder carcinogenesis.  相似文献
