The c-Met proto-oncogene is the receptor for hepatocyte growth factor (HGF), which is a member of the tyrosine kinase family. Activation of the HGF/c-Met signal pathway leads to cell proliferation, motility, regeneration, and morphogenesis. In this study, the complete nucleotide sequence of complementary DNA (cDNA) of canine c-Met was cloned, and its distribution was determined in tissues. The canine c-Met cDNA clone had an open reading frame of 4419 bp that encoded a putative polypeptide of 1383 amino acids. The c-Met mRNA was expressed in a variety of canine tissues including peripheral blood mononuclear cells (PBMC), bone marrow, liver, kidney, lung, stomach, uterus, testis, thymus, lymph node, small intestine, colon, adrenal gland, thyroid gland, heart, muscle, skin, pancreas, ovary, prostate, spleen, fat, cerebrum, and cerebellum. In addition, the c-Met mRNA expression in normal and regenerated liver was examined. The levels of the mRNA increased 2-fold in regenerated liver compared to that found in normal liver, indicating that c-Met is involved in various functions including remodeling of canine hepatocytes.  相似文献

A pot experiment was conducted to investigate whether the shoot cadmium (Cd) concentration in 11 rice and 10 soybean cultivars varied among 4 soils with different levels of Cd contamination. Significant differences in shoot Cd concentration were found among rice or soybean cultivars grown in the 4 soils. The ranking of the rice cultivars for the shoot Cd concentration varied considerably among the soils. On the other hand, the soybean cultivars were ranked similarly in terms of shoot Cd concentration in the 4 soils. Significant and positive correlations were found between the Cd and Zn concentrations and between the Cd and Mn concentrations in the shoot of rice cultivars, when they were grown in 2 soils with relatively moderate levels of Cd contamination. The shoot Cd concentration in the soybean cultivars, however, was not correlated with the concentrations determined for any of the metals (Zn, Mn, Cu, and Fe) across the 4 soils. Significant and positive correlations between the concentrations of Cd in younger shoots and mature seeds were detected among the soybean cultivars in 2 soils used, unlike among the rice cultivars, indicating that it may be difficult to evaluate the genotypic variation in seed Cd concentration using relatively younger shoots in the case of rice. These results revealed that genotypic differences in shoot Cd concentration in rice or soybean are variable or invariable among soils, respectively.  相似文献

Activities of the oxygen radical-scavenging enzymes were determined in leaves of ‘Kyomidori’ pepper (Capsicum annuum L.) and ‘Senryo’ eggplant (Solanum melongena L.) seedlings under two photoperiod treatments; 12 h light/12 h dark (12/12 h) or with continuous light (24/0 h) for six days. Growing eggplant under continuous light resulted in leaf chlorosis after four days and a sharp decline in the chlorophyll content. In eggplants grown under 24/0 h photoperiod, the activities of antioxidative enzymes; superoxide dismutase (SOD), catalase (CAT) and peroxidase (POD) rapidly increased from day two. This was an indication of photooxidative damage. In contrast, leaf chlorosis did not occur in pepper plants grown under continuous light, while the chlorophyll content was even higher in the 24/0 h than in the 12/12 h photoperiod. Continuous light did not enhance the activities of SOD and POD in leaves of pepper. However, the CAT activity increased faster and was significantly higher in pepper than in eggplants grown under continuous illumination. Pepper plants did not appear to suffer from photooxidative damage and thus grew normally under continuous illumination.  相似文献

Striga gesnerioides is a root parasitic weed of economic significance to cowpea (Vigna unguiculata) crops in Western Africa. Seeds of the parasite germinate in response to cowpea root exudates. Germination stimulants for the seeds were isolated from the hydroponic culture filtrate of cowpea, and their structures were unambiguously determined as (-)-(3aR,4R,8bR,2'R)-ent-2'-epi-orobanchol and (+)-(3aR,4R,8bR,2'R)-ent-2'-epi-orobanchyl acetate, on the basis of mass, CD, and (1)H NMR spectra; optical rotatory power; and chromatographic behavior on HPLC. The alcohol was first isolated and identified from the cowpea root exudates, and the acetate may be the same compound that had been previously isolated from the exudates and designated as alectrol. Identity of the stimulants produced by cowpea to those produced by red clover (Trifolium pratense) was confirmed.  相似文献

Strigolactones are highly potent germination stimulants for seeds of the parasitic weeds Striga and Orobanche spp. 4-Hydroxy-GR24 and 4-acetoxy-GR24 were prepared and their abilities to induce seed germination of Striga gesnerioides evaluated. Optically active (8bR,2'R)-isomers induced germination, although the racemic diastereomers were inactive. In contrast, the stereoisomer of GR24 with the same configuration induced negligible germination. Some stereoisomers of GR24 and its analogues acted as effective antagonists for induction of seed germination by cowpea root exudates. These results suggest that both an oxygenated substituent at C-4 and the configuration of the tricyclic lactone and the D-ring are essential structural requirements for induction of germination in S. gesnerioides seeds.  相似文献

Oxidative stress has been recognized as an important factor in the pathophysiology of preeclampsia. It has been reported that the expression of xanthine oxidase (XO) in the cytotrophoblast and plasma hydrogen peroxide (H2O2) level are significantly higher in preeclamptics than in control women. The aim of this study was to clarify the biological influence of reactive oxygen species (ROS) produced by XO on extravillous trophoblast (EVT) cells. TCL1 cells, a human immortalized EVT cell line, were incubated with xanthine and XO (X/XO). We then measured the cell number, urate level of the culture media and the apoptotic cell ratio. Similar experiments were performed with additional administration of allopurinol, catalase, L-NAME or D-NAME, and with administration of H2O2 in substitution for X/XO. We assessed the effects of H2O2 on invasion ability, tube-like formation and protein expression of HIF1A and ITGAV of TCL1. Finally, the apoptotic cell ratio using primary cultured trophoblasts was measured following exposure to H2O2. X/XO decreased the relative cell number and increased the urate level and apoptotic cell ratio significantly. Elevation of the urate level and apoptotic cell ratio was attenuated by allopurinol and catalase, respectively. L-NAME and D-NAME had no influence on these effects. H2O2 also decreased the relative cell number. Pretreatment with H2O2 significantly inhibited the invasion ability, tube-like formation and HIF1A and ITGAV of TCL1. H2O2 also induced apoptosis in primary cultured trophoblasts. In conclusion, ROS produced by XO induced apoptosis and affected EVT function including invasion and differentiation.  相似文献

Heavy metal contamination of agricultural soil and countermeasures in Japan   总被引:1,自引:0,他引:1  
Many heavy metals exist in minute amounts in natural agricultural soil. However, when their amounts exceed a certain level due to pollutants brought from outside, soil contamination occurs and agricultural products become contaminated. There have been many cases in Japan of heavy metal contamination originating from old mines and smelters, and soil contamination of agricultural land has become a social issue. In particular, cadmium (Cd) is one of the most harmful heavy metals. If agricultural products absorb an excessive amount of Cd, they may adversely affect people’s health, and therefore allowable concentrations are regulated by law. If agricultural land has become contaminated with Cd, measures for minimizing the absorption of Cd by agricultural crops are necessary; these include: (1) soil dressing, (2) water management (paddy field), (3) chemical cleaning of soil, (4) phytoextraction, and (5) use of different varieties and rootstock. Other heavy metals such as arsenic, lead, copper, zinc, and mercury also sometimes cause contamination of agricultural soil.  相似文献
