Brucellosis not only represents an important health restraint on livestock but also causes high economic losses in many developing countries worldwide. Despite considerable efforts made for the control of brucellosis, the disease is still spreading in many regions (such as the Middle East) where it represents one of the most important health hazards impacting both animals and humans. The present review aims to investigate the efficacy of veterinary control programs regarding brucellosis, with a special focus on current prevention, control, and eradication approaches. The reasons for unsuccessful control programs such as the absence of highly effective vaccines and non-certified bulls are also debated, to understand why the prevalence of brucellosis in livestock is not decreasing in many areas despite considerable efforts taken to date. The importance of governmental and regional investment in brucellosis control remains one of the main limiting factors owing to the limited budget allocated to tackle this disease. In this context, one health concept has generated novel comprehensive approaches with multiple economic implications across the livestock industry and public health. However, the implementation of such global preventive strategies appears to be a key issue for many endemic and low-income countries. According to the collected data, epidemiological contexts including management and trade systems along with well-defined agro-ecological zones should be evaluated in brucellosis endemic countries to improve milk production and to enhance the sustainability of the livestock sector at both national and regional levels.  相似文献

The enzyme rhodanese (EC 2.8.1.1) is an ubiquitous enzyme which is present in all living organisms, from bacteria to man. It is speculated that this enzyme plays a central role in cyanide detoxification. However, its wide tissue distribution suggests this enzyme might perform other functions beside cyanide detoxification. Although the distribution of rhodanese in different tissues of human and domestic animals has been studied, little is known about the pattern of distribution and physiological roles of this enzyme in the cat. The purpose of this investigation was to determine the enzyme levels and compare the distribution of this enzyme in different tissues of the cat. A selection of tissue samples was assayed for rhodanese activity. The protein content of tissue extracts and enzymatic activities were calculated as units per gram tissue and units per milligram protein of the tissue. Results showed that in terms of units per milligram protein of the tissue (specific activity of the enzyme), colon and rectum mucosal layers and testis were the richest sources of the enzyme followed by ovary, mucosal layer of jejunum and liver. With respect to units/gram tissue, liver followed by testis, colon and rectum mucosal layers, ovary and mucosa of jejunum exhibited highest activities. The results of this study will allow one to speculate on the involvement of rhodanese in several biochemical and physiological functions in different tissues and organs of this species.  相似文献

Diazinon (DZN) as an organophosphate pesticide may cause oxidative stress in different tissues. Antioxidants increase tissue protection from oxidative stress. The aim of the present study was to investigate prophylactic and therapeutic effects of vitamin C against oxidative stress caused by DZN in various tissues of male Wistar rats. Thirty rats were divided into five groups: control group received corn oil as DZN solvent, DZN group received 100 mg/kg of DZN, C group received 200 mg/kg of vitamin C, C‐DZN and DZN‐C groups received vitamin C before and after DZN injection. Plasma and various tissues were prepared and evaluated for measurement of the biochemical parameters and oxidative stress biomarkers. Results showed that acute administration of DZN significantly increased superoxide dismutase and glutathione‐S‐transferase activities and malondialdehyde level in all tissues, catalase (CAT) activity in liver, kidney and heart and some plasma biochemical indices, while it decreased cholinesterase and lactate dehydrogenase activities and glutathione content in all tissues. CAT activity in erythrocytes, brain and spleen was decreased in DZN‐exposed rats compared with the control group. Administration of vitamins C in both prophylactic and therapeutic groups ameliorated in these parameters, although all these tests in tissues did not return to the normal level. These data suggest that oxidative stress is an essential mechanism involved in DZN‐induced adversity effect, as evidenced by the altered activity of antioxidant enzymes, depleted GSH content and the enhanced membrane lipid peroxidation. Both the prophylactic and therapeutic treatments of rats to vitamin C have beneficial effects against oxidative stress and cholinergic hyperactivity induced by DZN in tissues especially in the brain tissue through free radical scavenging.  相似文献

Background:Mesenchymal stem cells enhance tissue repair through paracrine effects following transplantation. The versican protein is one of the important factors contributing to this repair mechanism. Using MSC conditioned medium for cultivating monocytes may increase versican protein production and could be a good alternative for transplantation of MSCs. This study investigates the effect of culture medium conditioned by human MSCs on the expression of the versican gene in PBMCs under hypoxia-mimetic and normoxic conditions. Methods:The conditioned media used were derived from either adipose tissue or from WJ. Flow cytometry for surface markers (CD105, CD73, and CD90) was used to confirm MSCs. The PBMCs were isolated and cultured with the culture media of the MSC derived from either the adipose tissue or WJ. Desferrioxamine and cobalt chloride (200 and 300 µM final concentrations, respectively) were added to monocytes media to induce hypoxia-mimetic conditions. Western blotting was applied to detect HIF-1α protein and confirm hypoxia-mimetic conditions in PBMC. Versican gene expression was assessed in PBMC using RT-PCR. Western blotting showed that the expression of HIF-1α in PBMC increased significantly (p < 0.01). Results:RT-PCR results demonstrated that the expression of the versican and VEGF genes in PBMC increased significantly (p < 0.01) in hypoxia-mimetic conditions as compared to normoxia. Conclusion:Based on the findings in the present study, the secreted factors of MSCs can be replaced by direct use of MSCs to improve damaged tissues.Key Words: Adipose tissue, Hypoxia, Mesenchymal stem cells, Wharton’s jelly  相似文献

This study investigates the effects of Coriander (C. sativum) on the growth, antioxidant, and immune‐associated genes and serum and mucosal immune parameters in zebrafish (D. rerio). The experimental fish were treated with 0 [control], 5, 10 and 20 g/kg coriander supplemented diets for 8 weeks. The results revealed that coriander remarkably increased mucosal immune parameters (the total Immunoglobulin, protease and lysozyme activity). The highest levels of the measured parameters were noticed in 20 g/kg CP treatment. Also, the zebrafish fed 20 g/kg coriander powder showed significantly higher expression gene for lysozyme and TNF‐alpha. The same results were noticed in case of IL‐1B gene expression. In case of sod gene expression there was no significant difference between treatments. However, regarding cat gene expression, significant difference was noticed between 20 g/kg CP treatment and other groups. In addition, no significant changes were observed between coriander fed zebrafish and control treatments regarding GH gene expression level, although IGF‐I remarkable up‐regulate in 20 g/kg coriander powder treatment compared other groups. In conclusion, it can be proposed that dietary Coriander powder can improve mucosal immune parameters and immune and antioxidant genes expression.  相似文献

OBJECTIVE: To establish the aetiology and define the main clinical features of a syndrome characterised by severe feed refusal, death of piglets and reduced milk production in pigs and dairy cattle. DESIGN: Clinical, pathological, toxicological and epidemiological examination of clinical cases in 10 piggeries and 4 dairies, located between 50 and 150 km south-west of Rockhampton. RESULTS: All cases were associated with the feeding of sorghum grain infected with sorghum ergot (Claviceps africana). There was mild to severe feed refusal when the sorghum was first offered. Sows fed ergot before farrowing had shrunken udders, produced no colostrum, and displayed signs of oestrus. All of their piglets died, apparently from starvation: necropsy of a few piglets showed that they were born alive and walked, but had ingested no milk. Sows fed the grain after farrowing had severe reductions in milk production despite aggressive sucking by piglets, leading to very poor growth of piglets. There were no signs of infectious disease. Ergot in sorghum samples ranged from 1 to 31% ergot sclerotes by weight. Total alkaloid concentrations in mixed feeds ranged from 5 to 40 mg/kg, with dihydroergosine accounting for approximately 90%. At the same time, in the same districts, there were reports of feed refusal and reduced milk production from 4 dairy farms. Grain samples from these farms contained up to 17% C africana ergot sclerotia. CONCLUSION: Agalactia and feed refusal are classical signs of poisoning by rye ergot (C purpurea), but this is the first time that sorghum ergot has been associated with a similar syndrome.  相似文献
