The valorization of extracts from Brazilian vetiver (Vetiveria zizanioides (L.) Nash ex Small) roots was studied. This study took into account the extraction method, the chemical composition of the extracts, their sensorial characteristics, and the possibility of chemical transformations of the product. The performed extraction methods were hydrodistillation and extraction with supercritical carbon dioxide. Some pretreatment methods were tested on the vetiver roots and evaluated in terms of extraction yield, process time, chemical composition, and sensorial properties. Supercritical carbon dioxide extraction resulted in high yield (3.2%) in significantly less time than the other methods. The chemical compositions of the extracts obtained by the different methods were also compared to those of commercial vetiver oils from other sources, showing that Brazilian samples had a greater acid amount. An extraction in basic medium from Brazilian vetiver oil was done to remove its main acid (zizanoic acid), which was chemically transformed into an alcohol (khusimol) of desirable sensorial properties. Sensory evaluation indicated that the Brazilian volatile oil without acid could be used in perfumery and the extract obtained with supercritical carbon dioxide could have application in food.  相似文献

Tomato spotted wilt virus (TSWV) is an economically important virus of flue-cured tobacco. Activation of systemic acquired resistance (SAR) by acibenzolar-S-methyl (ASM) in flue-cured tobacco was studied under greenhouse conditions by challenge inoculation with a severe isolate of TSWV. ASM restricted virus replication and movement, and as a result reduced systemic infection. Activation of resistance was observed within 2 days after treatment with ASM and a high level of resistance was observed at 5 days onward. Expression of the pathogenesis-related (PR) protein gene, PR-3, and different classes of PR proteins such as PR-1, PR-3, and PR-5 were detected at 2 days post-ASM treatment which inversely correlated with the reduction in the number of local lesions caused by TSWV. Tobacco plants treated with increased quantities of ASM (0.25, 0.5, 1.0, 2.0, and 4.0 g a.i./7,000 plants) showed increased levels of SAR as indicated by the reduction of both local and systemic infections by TSWV. The highest level of resistance was at 4 g a.i., but this rate of ASM also caused phytotoxicity resulting in temporary foliar spotting and stunting of plants. An inverse correlation between the TSWV reduction and phytotoxicity was observed with the increase of ASM concentration. ASM at the rate of 1 to 2 g a.i./7,000 plants activated a high level of resistance and minimized the phytotoxicity. Use of gibberellic acid in combination with ASM reduced the stunting caused by ASM. Present findings together with previous field experiments demonstrate that ASM is a potential option for management of TSWV in flue-cured tobacco.  相似文献

In Mexico, natural populations of the opossum pipefish, Microphys brachyurus, are under pressure by anthropogenic alteration of their habitat and unregulated fishing as this species is traded as an aquarium fish, where its survival is low due to inadequate culture practices. The development of culturing techniques has been a successful strategy to protect many exploited species through providing an alternative source to wild exploitation. The aim of this study was to examine the effect on growth, condition, and survival of pipefish cultured for 6 wk at temperatures of 26, 28, and 30 C. Pipefish were fed enriched Artemia nauplii maintaining a ration rate of 14% body weight per day (dry weight Artemia : wet weight fish). The pipefish cultured at 26 and 28 C were longer, heavier, grew faster, and presented better condition than those at 30 C. The survival of pipefish cultured at 26 C was greater (83%) than at 30 C (30%) but not different than at 28 C (50%). This study is the first to report the acceptance of Artemia nauplii by juveniles of the species and that a temperature of 30 C is not suitable for juvenile M. brachyurus culture.  相似文献

During tibial plateau leveling osteotomy (TPLO) the saw blade produces frictional heat. The purpose of this study was to evaluate and compare heat generated by two TPLO blade designs (Slocum Enterprises [SE] and New Generation Devices [NDG]), with or without irrigation, on cadaveric canine tibias. Thirty-six paired tibias were used to continuously measure bone temperatures during osteotomy through both cortices (i.e., the cis and trans cortices). Each pair was assigned to either an irrigation or nonirrigation group during osteotomy, and each tibia within a pair was osteotomized using a different saw blade design. Saw blade temperatures were recorded and temperatures were compared for all combinations of blade type, cortex, and irrigation. In the cis cortex group, the SE blade generated more bone heat than the NGD blade (P=0.0258). Significant differences in temperature generation between saw blade types were seen only when the osteotomy site was not irrigated (P=0.0156). For all variables measured, bone and saw blade temperature generation was lower with irrigation (P<0.05). None of the osteotomies performed with either saw blade produced a critical duration of damaging temperature ranges in this study. Although saw blade design and irrigation influence heat generation during the TPLO, the potential for bone thermal damage during TPLO is low. The use of the NGD blade with irrigation is recommended.  相似文献

Edwardsiella ictaluri was consistently isolated from the spleens, livers, and head kidneys of diseased Nile tilapia Oreochromis niloticus from a farm experiencing mortality events in several culture ponds. We describe the first published outbreak of E. ictaluri-induced edwardsiellosis in Nile tilapia. Pure cultures of the isolated bacteria were characterized both biochemically and molecularly. Biochemical analysis was performed using the API-20E and RapID One systems, and antimicrobial susceptibility was determined by the broth microdilution method. Molecular analysis involved sequencing of the 16S rRNA gene, species-specific real-time polymerase chain reaction (PCR), and PCR-mediated genomic fingerprinting (rep-PCR). Pairwise sequence analysis of the 16S rRNA gene identified the case isolates to be a 100% match to E. ictaluri cultured from channel catfish in the southeastern United States. However, rep-PCR analysis identified the case isolates to be genetically different from representative strains isolated from disease outbreaks in cultured channel catfish in Mississippi. Infectivity challenges (intraperitoneal injection and immersion) demonstrated that a representative E. ictaluri strain isolated from tilapia was pathogenic to naive tilapia, reproducing clinical signs and mortality, thereby establishing Koch's postulates.  相似文献

Catchment characteristics and disturbances control the conditions and processes found in stream ecosystems. We examined nutrient cycling linkages between riparian soils and adjacent streams and the impacts of the removal of ungulate grazing on these ecosystems and processes at six grazing exclosure sites in the Valles Caldera National Preserve, NM, USA. The exclusion of native and domestic ungulate grazers for 3 yr significantly increased the riparian aboveground biomass of standing vegetation (273 ± 155 in grazed vs. 400 ± 178 g . m-2 in exclosures) and litter (58 ± 75 in grazed vs. 110 ± 76 g . m-2 in exclosures) (P = 0.003 and 0.006, respectively). Except for an increase in total soil phosphorous (P) at three of the six sites, soil nutrient values were minimally affected by grazing after five growing seasons. Within the six sites studied, no connection was found between 0–15-cm depth soils, which were P-limited based on stoichiometric ratios, and stream nutrient availability or limitation, which were nitrogen limited. Stream geomorphology was not significantly altered by 5 yr of grazing exclusion. The elimination of grazing suppressed instream nutrient processing with significantly longer NH4 uptake lengths (P = 0.003). These results suggest the exclusion of ungulate grazing impacts terrestrial characteristics (increased standing vegetative biomass) that are linked to ecosystem services provided by adjacent aquatic ecosystems (reduced N-uptake). Management plans should carefully balance the positive effect of grazing on stream nutrient processing and retention reported here with the well-documented grazing-related loss of other ecosystem services such as decreased fish and aquatic invertebrate habitat and effects on water-quality parameters such as turbidity and water temperature.  相似文献

The relationship between factors influencing the in vitro and in vivo drug solubilization is highly dependent upon the physiology of the individual animal species and the physico-chemical properties of the drug. The solubilization of a drug in an aqueous medium is controlled by the strength of the interaction between a molecule of the active pharmaceutical ingredient (API) and the molecules of the solvent. In this regard, the stronger this interaction, the greater the likelihood that the drug will go into solution. Counteracting this solubilization process is the strength of the affinity of the solute for itself, or how tightly bound the compound is to its own solid state form. The stronger affinity of a solute for itself, the more difficult it will be for that molecule to go into solubility. However, solubility is not a universal value. Rather, it should be considered from the perspective of the interactions between the API in its solid form and the solution conditions such as: pH, the nature of the primary solvent, the presence of co-solvents, the presence of solubilizing additives, the ionic strength of the medium, incubation time, and temperature. Each of these factors needs to be considered when evaluating potential interspecies differences in drug solubility.  相似文献

This study addresses the quantification of the Amazon River sediment budget which has been assessed by looking at data from a suspended sediment discharge monitoring network and remote sensing estimates derived from MODIS spaceborne sensor. Surface suspended sediment concentration has been sampled every 10 days since 1995 (390 samples available) by the international HYBAM program at the Óbidos station which happens to be the last gauged station of the Amazon River before the Atlantic Ocean. Remote sensing reflectance is derived from continuous time series of 554 MODIS images available since 2000 and calibrated with the HYBAM field measurements. Discharge shows a weak correlation with the suspended sediment concentration during the annual hydrological cycle, preventing us from computing sediment discharge directly from the water discharge. Accordingly, river sediment discharge is assessed by multiplying daily water discharge measurements by the suspended sediment concentration averaged on a monthly basis. Comparisons of annual sediment discharge assessed using both field and satellite datasets show a very good agreement with a mean difference lower than 1%. Both field and satellite-derived estimates of the sediment concentration of the Amazon River are combined to get an uninterrupted monthly average suspended sediment discharge from 1995 to 2007. Unlike the water discharge which exhibits a steady trend over the same period at Óbidos, the 12-year suspended sediment discharge increases by about 20% since 1995, significant at the 99% level. In particular, the inter-annual variability is much more significant in the sediment discharge than in the river discharge.  相似文献

The relationship between phenolic compounds and maize pith resistance to Fusarium graminearum, the causal agent of Gibberella stalk rot, was investigated. The phenolic acid profiles in the stalks of six maize inbred lines of varying susceptibility were evaluated from silking to grain maturity. Four different fractions of phenolic compounds were extracted from inoculated and non-inoculated (control) pith tissues: insoluble cell-wall-bound, free, soluble ester-bound, and soluble glycoside-bound phenolics. Analysis by HPLC revealed that p-coumaric acid and ferulic acid were the most abundant compounds in the soluble and cell-wall-bound fractions. The quantity of free, glycoside-bound, and ester-bound phenolics in the pith was lower than the level required for the inhibition of Fusarium growth or mycotoxins production; however, significant negative correlations between diferulic acid contents in the cell walls and disease severity ratings 4 days after inoculation were found. The results indicated that future studies should focus on the levels of diferulic acids during the early infection process. Diferulates may play a role in genotypic resistance of maize to Gibberella stalk rot as preformed barriers to infection.  相似文献

The transition of DNA secondary structure from an analogous B to Z conformation modulates the dielectric environment of the single-walled carbon nanotube (SWNT) around which it is adsorbed. The SWNT band-gap fluorescence undergoes a red shift when an encapsulating 30-nucleotide oligomer is exposed to counter ions that screen the charged backbone. The transition is thermodynamically identical for DNA on and off the nanotube, except that the propagation length of the former is shorter by five-sixths. The magnitude of the energy shift is described by using an effective medium model and the DNA geometry on the nanotube sidewall. We demonstrate the detection of the B-Z change in whole blood, tissue, and from within living mammalian cells.  相似文献
