ObjectiveTo evaluate the physiological effect and response to noxious stimulation at five concentrations of MS-222 in koi (Cyprinus carpio).Study designProspective experimental study.AnimalsTwenty-one healthy adult unknown sex koi fish weighing mean 450 ± SD 120 g.MethodsEach fish was exposed to five different concentrations of MS-222 (50, 70, 110, 150 and 190 mg L?1) in a random sequence during the same anaesthetic event. For each concentration of MS-222, vital functions such as heart rate (HR) (via Doppler) and opercular rate (OpR) were recorded after a standardized induction period. Response to two noxious stimuli in the form of haemostat clamp pressure applied on the tail and the lip was evaluated, and blood was drawn to measure biochemical and blood gas values.ResultsDecrease in response to noxious stimulation with an increase of MS-222 concentration both for the lip (p = 0.0027) and the tail (p < 0.0001) stimulus was observed. Biochemical values were unaffected by the concentration of MS-222 with the exception of lactate concentration which was weakly correlated with the duration of anaesthesia (r = 0.31, p < 0.001) and the number of times the fish was clamped or bled prior to sampling (r = 0.23, p < 0.001). Opercular rate decreased with the increase in anaesthetic concentration, and HR was not affected.Conclusions and clinical relevanceOur results indicated a decrease in response to stimulus and a decrease in OpR that were associated with increased concentrations of MS-222. This may assist in establishing anaesthetic protocols using MS-222 in fish and supports the use of supramaximal pressure stimuli to teleost fish under variable MS-222 concentrations as a model for future studies.  相似文献

The limited field of view (FOV) associated with single-resolution very-large-scale aerial (VLSA) imagery requires users to balance FOV and resolution needs. This balance varies by the specific questions being asked of the data. Here, we tested a FOV-resolution question by comparing ground cover measured in the field with the use of point-intercept transects with similar data measured from 50-mm-per-pixel (mmpp) VLSA imagery of the same locations. Particular care was given to spatial control of ground and aerial sample points from which observations were made, yet percent cover estimates were very different between methods. An error budget was used to calculate error of location and error of quantification. These results indicated location error (43.5%) played a substantial role with significant quantification error (21.6%) also present. We conclude that 1) although the georectification accuracy achieved in this project was actually quite good, the level of accuracy required to match ground and aerial sample points represents an unrealistic expectation with currently available positioning technologies, 2) 50-mmpp VLSA imagery is not adequate for accurate species identification or cover assessments of plant functional groups, and 3) the balance between resolution and FOV needs is best addressed by using multiple cameras to acquire nested imagery at multiple VLSA resolutions simultaneously. We recommend ground cover be measured from 1-mmpp imagery and that the imagery be nested in lower-resolution, larger FOV images simultaneously acquired.  相似文献

This study aimed to evaluate the effects of certain embryo transfer parameters on the pregnancy rate after equine embryo transfer of the Brazilian Jumper Horse breed. The size, embryonic development stage, embryo quality, and synchronization of ovulation between the donor (n = 120) and recipient (n = 420) were evaluated in 396 embryos. Embryo recovery was performed on Day 6-9 after ovulation (Day 0 = day of ovulation). The recipient mares were chosen on the day of embryo recovery, and the transfers were performed that same day. The embryo size (diameter including envelopes; n = 396) ranged from 150 to 3000 μm; 67.1% measured between 400 and 1199 μm. The embryo size (400-1199 μm vs. ≤399 μm); stage of development (n = 396; blastocyst and expanded blastocyst versus compact morula and early blastocyst); quality (n = 396; grade 1 [excellent]), 2 [good], or 3 [poor]); and synchronization of ovulation between the donor and recipient (0, 1, 2, 3, and 4 days versus −1, 5, and 6 days, respectively) all affected pregnancy rate (P < .05). The pregnancy rate did not differ significantly among transfers performed on Days 0, 1, 2, 3, and 4. In conclusion, embryos measuring 400-1199 μm produced higher pregnancy rates in recipients than embryos measuring 150-399 μm, and blastocysts and expanded blastocysts produced pregnancy more efficiently than morulae and early blastocysts. The embryo quality also affected the pregnancy rate. Synchronization of donor and recipient ovulation to Days 0-4 improved the efficiency of embryo transplant.  相似文献

Stereotypies are considered a cause and symptom of impaired welfare, and have been associated with suboptimal husbandry systems. The aim of this study was to estimate the prevalence of stereotypic and other abnormal behaviors in Chilean thoroughbreds by direct observation, and examine their associations with biological characteristics and management practices. Seven hundred forty-three racehorses were observed directly, every 5 minutes during 1 hour before and after feeding, to identify behavioral disorders. Although the exact mechanism(s) by which estradiol (E2) enhances muscle growth in a number of species, including humans and cattle, is not known, E2 treatment has been shown to stimulate proliferation of cultured bovine satellite cells (BSCs). This is particularly significant because satellite cells are the source of nuclei needed to support postnatal muscle fiber hypertrophy and are thus crucial in determining the rate and extent of muscle growth. The objective of this study was to assess the role of estrogen receptor-α (ESR1) and the type 1 insulin-like growth factor receptor (IGFR1) in E2-stimulated proliferation of cultured BSCs. To accomplish this, we have used small interfering RNA (siRNA) to silence expression of ESR1 or IGFR1 and assessed the effects on E2-stimulated proliferation in BSC cultures. In BSCs treated with nonspecific siRNA, E2 significantly (P < 0.05) stimulates proliferation under conditions in which neither IGF-1 nor IGF-2 expression is increased; however, treatment of ESR1- or IGFR1-silenced cells with E2 does not significantly stimulate proliferation. These results indicate that both ESR1 and IGFR1 are required for E2 to stimulate proliferation in BSC cultures. The fact that this occurs under culture conditions in which neither IGF-1 nor IGF-2 mRNA expression is increased strongly suggests that E2 activates IGFR1 via a mechanism that does not involve increased IGF-1 or IGF-2 binding to the receptor.  相似文献

Evidence mounts for the influence of climate variability on temporal trends in the phenology of many organisms including various species of fish. Accordingly, we examined variation in adult Atlantic salmon Salmo salar run timing in thirteen Newfoundland and Labrador rivers where returns were monitored at fishways or fish‐counting fences. Run timing varied significantly among rivers with the median date of return differing by up to 5 weeks. Duration of runs was generally short with most adults returning over a period of three to 5 weeks. A mixed model analysis incorporating a first‐order autoregressive error structure was used to generalise changes in run timing among all monitored rivers. Results indicated that the median date of return has advanced by almost 12 days over a 35‐year interval from 1978 to 2012, while several individual rivers have advanced by almost 21 days. The influence of climate on median timing was evident when the simultaneous effects of both climate and salmon abundance were controlled. We found earlier runs associated with overall warmer climate conditions on the Newfoundland and Labrador Shelf. Results contrast with those from the north‐east Atlantic where Atlantic salmon are returning later in some rivers coincident with warming climate conditions.  相似文献
