Fungal secondary compounds play an important role for springtail food choice and fitness. Little is known, however, on the role of olfactory cues for Collembola foraging behaviour and whether Collembola can olfactorily perceive volatiles associated with fungal secondary metabolite pathways. We investigated the ability of three species of Collembola (Folsomia candida, Heteromurus nitidus and Supraphorura furcifera) to use olfactory cues for discriminating between fungi of different phylogenetic affiliation (Aspergillus nidulans, Cladosporium cladosporioides, Ascomycota; Laccaria bicolor, Basidiomycota) and toxicity using fungal strains of A. nidulans with reduced secondary metabolite production. Furthermore, we studied if olfactory cues from hyphae injured by grazing affect the foraging behaviour of Collembola. We hypothesized that (i) Collembola are able to olfactorily perceive and respond to fungal species/strains with different secondary metabolite pathways and that (ii) Collembola are able to sense fungal mycelia injured by grazing. Each of the Collembola species studied preferentially oriented their foraging towards fungal strains of A. nidulans with suppressed secondary metabolites, and in particular towards the mutant where the global regulator for secondary metabolites (ΔlaeA) has been silenced. Two of the three Collembola species (H. nitidus and S. furcifera) sensed olfactory cues of previously grazed fungi and moved towards ungrazed fungi, however, the response of S. furcifera was restricted to fungi extensively exposed to grazing (5 days) suggesting that the response varies between Collembola species.Overall, the results support the first and second hypothesis indicating that Collembola (1) are able to olfactorily differentiate fungi of different toxicity and move towards more palatable fungi, and (2) showed a lesser preference for fungi previously exposed to grazing.  相似文献

The fatty acid composition of rotifer phospholipids is largely dependent on that of their food, indicating that the ingested lipids are hydrolysed in the gut, resorbed, metabolized and incorporated into body phospholipids. It is stable and does not change considerably after 1 or 2 days of starvation.Considerable differences were found in the fatty acid composition of rotifers fed baker's yeast (Saccharomyces cerevisiae), Chlorella or Isochrysis. Rotifers fed on baker's yeast for several generations contained considerable amounts of PUFA (1.6% of 22:6 ω3) although this yeast contains mainly 16:1 and 18:1 acids and is completely devoid of 16:3, C:20 and C:22 unsaturated and saturated fatty acids. This observation suggests that fatty acid synthesis and elongation had occurred. De novo synthesis of PUFA was further suggested by experiments in which 1-14C acetate was incorporated into radioactive acids and was verified by chemical degradation. Upon decarboxylation, approximately 10% of the total radioactivity in the PUFA was recovered as 14CO2, indicating that all the uneven C-atoms must contain 14C. Oxidative cleavage of all of the double bonds of PUFA yielded 14C-labelled propionic and hexanoic acid from the methyl end of the molecule. However, the rate of synthesis of these acids is rather low. Hence, in order to supply large amounts of PUFA to marine fish larvae, rotifers must be fed on PUFA-rich food.  相似文献

The study was performed on 16 Holstein‐Friesian calves divided into two groups of eight animals each. The first group was composed of calves whose mothers did not receive selenium supplements (Se0). The second group consisted of calves whose mothers were administered intramuscular injections of a selenium and vitamin E supplement containing 0.5 of sodium selenite/ml and 50 mg of tocopherol acetate/ml in a single dose of 30 ml (Se30) ml, 10 days before the expected parturition date (10 ± 2 days). The calves were fed 2.5 L of the mother's colostrum administered by stomach tube 2 hr after birth and another 2 L 6 hr after birth. Blood from all calves was collected 7 times from external jugular vein (day 0–before colostrum administration and on the 2nd, 3rd, 4th, 7th, 14th and 21st days of life) for analyses of selenium, ceruloplasmin, transferrin, lactoferrin, immunoglobulin G (IgG) concentrations and gamma‐glutamyl transferase (GGT) and lysozyme activity. Selenium concentration was significantly higher in calves whose mothers received selenium supplements than in the offspring of non‐supplemented cows until 72 hr after birth (p ≤ .05). Lysozyme and GGTP activity and IgG concentration were significantly higher in the S30 group during the entire experiment (p ≤ .05). Supplementation of selenium to the mothers did not influence the ceruloplasmin, lactoferrin and transferrin levels in calves. A single injection of a selenium supplement administered to cows during late pregnancy increases selenium levels in calves and enhances passive transfer from the mother to the offspring.  相似文献

Polyphenol-rich apple extracts have been reported to suppress human colon cancer cell growth in vitro. The protein kinase C (PKC) is among the signaling elements known to play an important role in colon carcinogenesis. In the present study, we investigated whether apple polyphenols affect PKC activity and induce apoptosis in the human colon carcinoma cell line HT29. A polyphenol-rich apple juice extract (AE02) was shown to inhibit cytosolic PKC activity in a cell-free system. In contrast, incubation of HT29 cells for 1 or 3 h with AE02 up to 2 mg/mL did not affect the cytosolic PKC activity. After prolonged incubation (24 h), cytosolic PKC activity was modulated, albeit a u-shaped curve of effectiveness was observed, with an initial inhibitory effect followed by the recurrence and even induction of enzyme activity. Concomitantly, in the cytosol, a significant decrease of the protein levels of PKCalpha, PKCbetaII, and PKCgamma together with a significant increase of a proapoptotic PKCdelta fragment was observed. However, the effects on the protein levels of these PKC isoforms in the cytosol were not associated with translocation between the different cellular compartments but might instead result from the onset of apoptosis. Indeed, the treatment with AE02 was shown to induce apoptosis by the activation of caspase-3, DNA fragmentation, and cleavage of poly(ADP ribose) polymerase. So far, identified and available constituents of the apple extract did not contribute substantially to the observed effects on PKC and apoptosis induction. In summary, apple polyphenols were found to inhibit PKC activity in a cell-free system. However, our results indicate that within intact cells PKC does not represent the primary target of apple polyphenols but appears to be affected in the course of apoptosis induction.  相似文献

The phenolic composition of white grapes is important since browning reactions may dramatically compromise the overall quality of musts and wines. Only few scientific contributions are available, which investigated how fertilization can influence this class of compounds. Thus, the aim of this work was to examine the effects of NPK soil fertilization coupled with soil or foliar applications of Mg, Fe, and Zn on the concentrations of K, Mg, Fe, and Zn of grape berries and leaf petioles of potted Rebula grapevines (Vitis vinifera L.), also revealing the change of hydroxycinnamic acids in grape juice. The results obtained over the three‐year study (2009–2011) showed that NPK fertilization positively affected K and Zn concentrations of petioles and grape berries, and negatively Mg. In addition, K (synergistic) and Mg (antagonistic) had an influence on Zn uptake. Our findings suggest that the supply of NPK was profitable for a reduction of trans‐caftaric acid in grape juice, while only few differences were observed with further application of nutrients.  相似文献

The abundance and distribution of leptocephalus larvae of the European eel (Anguilla anguilla, Anguillidae) were examined using ten historic and recent Sargasso Sea expeditions that were selected on the basis of having the largest number of sampling stations and highest catches. The surveys cover the period 1920–2014. Station data were recalculated to the same unit of larval density per unit area, and the irregular station positions were transformed to a regular spatial grid to allow calculation of comparable measures of abundance of the youngest (0+) leptocephalus cohort. The result is that the mean and maximum densities of 0+ leptocephali after 2007 on average have decreased by 70%–80% from the densities during the period before the drastic decrease in glass eel recruitment, which started in the 1980s. This is of the same magnitude as the change in spawning stock, if the total continental commercial landings are used as a proxy. In the same period, the glass eel recruitment in Europe has decreased by more than 95%. The conclusion is that a major cause for the recruitment decrease may be an increased leptocephalus mortality during the oceanic phase or a large geographic shift in glass eel arrival. Combining the survey data, the spatial distribution of 0+ leptocephali was concentrated south of the northernmost front in the Subtropical Convergence Zone, but high densities were also found far south of the front in the western part of the distribution area and leptocephali were present also north of the average frontal position.  相似文献

Delphinidin has been found to possess DNA strand-breaking properties in cell culture. In the present study, we demonstrated that the extent of DNA damage by delphinidin is not affected by the expression of tyrosyl-DNA-phosphodiesterase 1, indicating that the induction of DNA strand breaks is not predominantly topoisomerase-mediated. However, the DNA-damaging properties of delphinidin were decreased by the addition of catalase to the cell culture medium, counteracting delphinidin-mediated hydrogen peroxide formation. Under these conditions, delphinidin showed clearly antioxidative properties in HT29 cells, preventing menadione-induced oxidative DNA damage. In contrast, in the absence of catalase, delphinidin lacked antioxidative properties. In conclusion, delphinidin acted as an effective antioxidant within intact cells if the formation of hydrogen peroxide was prevented. In the absence of catalase, the accumulated hydrogen peroxide appears to play a substantial role for the observed DNA-damaging properties of delphinidin and the apparent lack of antioxidative properties of this anthocyanidin.  相似文献

Recent findings of a potential human carcinogen, acrylamide, in foods have focused research on the possible mechanisms of formation. We present a mechanism for the formation of acrylamide from the reaction of the amino acid asparagine and a carbonyl-containing compound at typical cooking temperatures. The mechanism involves formation of a Schiff base followed by decarboxylation and elimination of either ammonia or a substituted imine under heat to yield acrylamide. Isotope substitution studies and mass spectrometric analysis of heated model systems confirm the presence of key reaction intermediates. Further confirmation of this mechanism is accomplished through selective removal of asparagine with asparaginase that results in a reduced level of acrylamide in a selected heated food.  相似文献
