Using thermomechanical extrusion, waxy maize starch and 4% (w/w) lipid were formed into strips. The lipids used were free fatty acids (mostly linoleic) and antioxidant stripped sunflower oil (either previously oxidised or fresh, with and without copper ions). By altering their water content it was possible to store, at the same temperature, sample strips in the glassy and rubbery states. Lipid oxidation was monitored by determining hexanal in the headspace above all stored samples. The molecular weights of the amylopectin in the samples of extruded products (dissolved in dimethylsulfoxide, precipitated and redissolved using a pressure cell) were determined by asymmetric flow-field-flow fractionation, analytical ultracentrifugation and light scattering. The initial rate of hexanal generation was higher in samples stored in the glassy state compared with those in the rubbery state. Waxy maize and extruded samples, at the start of the storage period and after 42 days, were used to establish the molecular weight of the amylopectin. A significant fall (a decrease of 40%) in molecular weight was found on storage of samples containing sunflower oil and copper ions and those containing free fatty acids, irrespective of the method used for molecular weight determination.  相似文献

The morphology of tongue in straw-coloured fruit bat from tropical forests was evaluated in relation to frugivorous diets and in comparison with other species that consumes other food types. Gross, stereomicroscopy, scanning electron microscope and histological methods were used. The tongue was relatively long with round tip, which closely fitted into oral cavity. Five types of mechanical papillae included crown-like and trifid filiform papillae. Also bulky, cone-shaped papillae and long conical papillae were identified. These mechanical types also showed variations in shape, size and number of processes of papillae. Transitional forms of these mechanical papillae were present. Fungiform papillae with taste pores were interposed amongst filiform types in apex and body; three ovoid-shaped vallate papillae were in triangular arrangement on root and displayed taste pores. Some bulky, cone-shaped papillae surrounded the vallate papillae. Histologically, mechanical filiform types showed highly keratinized stratified squamous epithelium and dense connective tissue core with secondary papillae. Taste buds appeared in fungiform and vallate papillae. Neutral and acidic secretions were identified in lingual glands of root. The presence of prominent filamentous processes of filiform papillae and conical papillae of the tongue in conjunction with gustatory papillae ensures adaptation to copious fruit diets. The gross morphometric and histometric parameters of the tongue did not differ remarkably from previous values obtained for some fruit bats with comparable weight. This investigation showed similarities with fruit bats such as large flying fox and Egyptian fruit bat and reflect common diet and feeding habits but varied from insectivorous and nectivorous bats.  相似文献

1. This study was undertaken to examine the effect of feeding glycine (Gly)-fortified low protein (LP) diets on the growth performance, duodenal morphology and caecal microbial populations of broiler chickens raised under unheated, cyclic or constant heat stress environmental conditions.

2. From d 1 to 21 (starter phase), an equivalent number of birds were fed either a normal protein (NP) diet or a LP diet fortified with Gly. From d 22 to 42 (grower phase), an equivalent number of birds from each starter diet were distributed to one of the following dietary groups: (i) an NP diet during the starter and grower phases (NPNP), (ii) an NP diet during the starter phase and a LP diet during the grower phase (NPLP), (iii) an LP diet during the starter phase and an NP diet during the grower phase (LPNP) or (iv) LP diets during both phases (LPLP).

3. Commencing from d 22, an equivalent number of birds from each dietary group were exposed to (i) 23 ± 1°C throughout (unheated), (ii) 34 ± 1°C for 7 h each day from 10:00 to 17:00 (cyclic heat) or (iii) 34 ± 1°C throughout (constant heat).

4. Feeding the LP diet during the starter phase resulted in feed intake (FI), weight gain (WG), feed conversion ratios (FCR) and energy efficiency ratios (EER) similar to those for the NP diet. The birds fed the LP diet had a significantly higher protein efficiency ratio (PER) compared with the birds fed the NP diet.

5. During the grower phase, there were significant diet × temperature interactions for F, WG, FCR, PER, EER, villus height, crypt depth and caecal Clostridia. The birds fed the NPLP and LPLP diets had lower FI, WG and EER, higher FCR, shorter villus height and crypt depth and higher caecal Clostridia compared with the birds fed LPNP and NPNP diets under constant heat stress. However, feeding birds the NPLP and LPLP diets resulted in FI, WG, EER, FCR, morphology parameters and caecal Clostridia equivalent to the birds fed LPNP and NPNP diets, as well as improved PER, under unheated and cyclic heat stress conditions.

6. In conclusion, our results indicate that Gly-fortified LP diets can be fed to broilers under normal and acute heat stress environmental conditions without any adverse effects on performance. However, the use of such LP diets can be detrimental to broilers under chronic heat stress conditions.  相似文献

Staphylococcus aureus is a commensal and pathogenic bacterium with impact on public health and livestock industry. The study investigated nasal carriage, antibiotic resistance, and molecular characterization of S. aureus in pigs and pig workers. Nasal swabs from 300 backyard-raised pigs and 101 pig workers were used for the study. Resulting isolates were confirmed using MALDI-TOF MS, tested for antibiotic resistance, and three different multiplex PCRs were used to detect enterotoxin, mecA, spaA, scn, and pvl genes. spa typing was used to annotate the isolates into MLST clonal complexes (CC). Structured questionnaire was used to access possible risk factors for S. aureus carriage. The prevalence of S. aureus in pigs and pig workers were 5.3 and 12.9%, respectively. The isolates were resistant to beta-lactams (97%), tetracycline (62%), sulfonamide (52%), aminoglycoside (20.6%), fluoroquinolone (24%), and mupirocin (3.4%). Twenty seven (93%) of the isolates carried scn, 7(24%) pvl, and 12 (41%) enterotoxin genes, respectively. Questionnaire survey showed medical-related occupation of household members was associated (p?<?0.5) with S. aureus carriage. This study suggests the presence of human multidrug resistant strains of S. aureus, high carriage of pvl, and enterotoxin genes, and CC5, CC15, and CC152 were the CC-groups shared among pigs and pig workers.  相似文献

Oligodendrocyte and myelin‐related studies have been pivotal in understanding disruption of central nervous system (CNS) myelin through injury, toxicological, pathological degeneration or genetic intervention. The African giant rat (AGR) has been postulated as an indigenous wild‐type model within the African context. This work thus describes oligodendrocyte morphologies and myelin components of the developing African giant rat brain using histological, immunohistochemical and ultrastructural techniques. Five types, precursor‐progenitor oligodendrocytes, pre‐oligodendrocytes, immature oligodendrocytes, mature non‐myelinating oligodendrocytes and mature myelinating oligodendrocytes, were identified. The first four types were observed in neonates while juvenile and adult AGR had predominantly mature myelinating oligodendrocytes with evidence of myelin sheath deposition. All cell types identified showed positive CNPase‐positive immunosignalling across all age groups. This suggests CNPase as a suitable, sensitive and reliable biomarker for studying CNS neurodegenerative/demyelinating disorders in the AGR. This baseline study has given detailed insight into the morphology of oligodendrocytes and myelin in the AGR. It may be useful for anatomical studies and detection of alterations in neurocellular profile of oligodendrocytes and myelin in the AGR in real‐life or in experimental models.  相似文献

This study aimed to investigate effects on growth and development and alterations in the nutrient profiles for Arabidopsis thaliana seedlings following uranium exposure. Seventeen-day-old Arabidopsis thaliana seedlings, grown in hydroponics, were exposed to 0, 0.1, 1, 10 and 100 μM uranium for one, three and seven days. Fresh weight of leaves and roots decreased after exposure to 100 μM uranium for one and three days and after exposure to 1, 10 and 100 μM uranium for seven days. Anthocyanous-colored leaves and stunted yellow roots were observed after exposure to 100 μM uranium. The uranium content of the roots highly increased but the root-to-shoot transfer of uranium was limited. Uranium exposure resulted in disturbed macro- and micronutrient profiles, especially following exposure to 100 μM. This study indicates that elevated uranium concentrations can cause important morphological and physiological effects and disturb the nutrient profiles in Arabidopsis thaliana seedlings.  相似文献

A 56-day feeding trial was conducted to investigate the effect of dietary supplementation of dried ginger meal (DGM) on the growth performance, nutrient digestibility, serum parameters, gut morphology, and microflora of growing helmeted guinea fowl (Numidia meleagris). One hundred sixty 28-day-old male keets were assigned to four dietary treatments. There were 40 birds per treatment replicated four times with 10 keets each. The experimental diets consisted of the basal diet (control), ginger-supplemented diets containing 20, 40, and 60 g/kg DGM, respectively. Guinea fowls fed diet supplemented with DGM had higher (P <0.05) final body weight, body weight gain and lower (P <0.05) feed intake. Optimum weight gain was obtained with supplementation level of 36.15-g DGM (R 2?=?0.923). Guinea fowls fed diet supplemented with 40 g/kg DGM recorded the highest (P <0.05) coefficient of total tract apparent digestibility of dry matter, ether extract and longest (P <0.05) duodenal and ileal villus heights. The crypt depth of the duodenum and ileum reduced (P <0.05) with increasing level of dietary supplementation of DGM. Dietary supplementation of DGM resulted in increased (P <0.05) total serum protein, serum albumin and low (P <0.05) serum cholesterol concentration. Ileum content of birds fed diet supplemented with 40 g/kg DGM recorded the highest (P <0.05) lactobacillus count. In conclusion, dietary inclusion of 40-g DGM per kilogram diet is hereby recommended for improved growth, apparent nutrient digestibility, gut morphology, serum chemistry, and stimulation of balanced intestinal microflora in growing guinea fowl.  相似文献

The use of feathers in veterinary clinical practice simplifies the sampling of avian genomic DNA, especially when blood extraction is difficult because of the age or the size of the bird. A rapid and accurate protocol was used to isolate high-quality genomic DNA from feathers. The technique includes a lysis step of the feather quill, which differs in temperature and time of incubation depending on the feather size. Purification of genomic DNA is performed with phenol: chloroform: isoamyl alcohol extraction and ethanol precipitation. This protocol consistently provided significant amounts of high-quality genomic DNA from more than 800 birds belonging to 120 different species. Genomic DNA isolated with this method was used for Southern blotting and also in several polymerase chain reaction systems devoted to sex determination and paternity testing.  相似文献

Swamps, marshes, hills, mountains and uncultivated marginal lands are always in danger of erosion, drought and low structural stability and compaction. Generally, marginal lands do not guarantee sustainable cultivation without external inputs. However, Nigerian farmers continue to crop the marginal lands because of increasing population growth and the use of agricultural lands for housing, roads, industry and other activities.
