Quinoa offers a promising alternative for staple food, considering its outstanding nutritional value and tolerance to abiotic stresses. To develop breeding programmes in quinoa, a reliable crossing method for increasing the genetic variation is required. In the following study, we aimed to develop segregating populations in quinoa. We tested the efficiency of three different crossing methods (hand emasculation, warm water emasculation and no emasculation). Moreover we developed a two-stage selection strategy based on morphological traits and molecular markers for the selection of hybrid plants. We reported hand emasculation to be the most efficient crossing method, followed by warm water emasculation and no emasculation. Our results demonstrated that crosses in quinoa can be successfully performed, despite its complicated flower structure and high self-pollination rate. Additionally, we developed 30 segregating populations from crosses between accessions of different origins with varying phylogenetic relationship, which offers a promising perspective for quinoa breeding programmes in the future.  相似文献

Genetic diversity was evaluated by RAPD markers and morpho-agronomic characters for a total of 42 accessions of Barberton daisy (Gerbera jamesonii) consisting of 29 commercial and 13 wild accessions. A total of 74 polymorphic bands were obtained employing a set of 12 primer pairs. The average genetic similarity coefficient for the 42 accessions, evaluated by Jaccard index was 0.55 ranging from 0.28 to 1.00. The genetic structure found among Barberton daisy accessions was evaluated by hierarchic classification analyses and UPGMA modeling, revealing six clusters of genotypes where two of them include the wild accessions and the remaining four including commercial material, except for wild genotype number 9. Shannon (H′) index was calculated using the molecular markers to investigate the genetic variation among the Gerbera accessions and showed higher values for the commercial cluster in comparison to the values obtained for the individuals from the non-commercial cluster, namely 0.34 versus 0.27, respectively. Therefore, both calculated indices (Jaccard and Shannon) indicated the presence of higher genetic variation among commercial accessions in comparison to the cluster representing non-commercial accessions, suggesting that genetic breeding programs may focus on commercial accessions to recombine interesting genotypes with commercially important and marketing-desired characteristics.  相似文献

Apparent digestibility coefficients (ADCs) provide estimates of nutrient availability in feedstuffs and can be used as a tool for selecting ingredients to compound cost‐effective diets for fish. Extrusion process is one factor that could influence ADC values. Thus, a digestibility trial was undertaken to evaluate ADC of protein, energy, and dry matter, and apparent amino acid availability (AAAA) in five cereal grain products/by‐products (corn, wheat middlings, broken rice, rice bran, and sorghum) in compounded diets for Nile tilapia. Chromic oxide was used as an external inert marker. The digestibility values of energy and dry matter in ingredients, in decreasing order, were highest for broken rice (95.34 and 96.45%), sorghum (82.37 and 87.29%), corn (67.34 and 82.21%), rice bran (57.58 and 55.59%), and wheat middlings (48.94 and 45.88%). Protein digestibility value was higher for corn (72.86%) and the lowest value was observed for sorghum (56.77%), while broken rice, wheat middlings, and rice bran presented values ranging from 63.01 to 66.88%. AAAA values were higher for corn (70.05%) and the lowest methionine and cystine availability values were recorded for wheat middlings and broken rice, respectively. Broken rice appears as one of the most indicated ingredient for using in Nile tilapia diets because of its highest values of nutrient digestibility when compared to the other energetic sources. The use of this information should aid more precise and economic diet formulation for Nile tilapia.  相似文献

Soybean molasses was evaluated as a partial replacement for sugarcane molasses as a carbon source for biofloc development in the superintensive culture of Pacific white shrimp (Litopenaeus vannamei). A 50‐day study was conducted with juvenile (3.2 g) shrimp stocked in 16 800 L tanks at a stocking density of 250 shrimp m?3. Control of total ammonia concentration was performed by the addition of combined mixtures of soybean and sugarcane molasses to the culture water. Three different molasses treatments were evaluated using different soybean‐to‐sugarcane molasses ratios: 15–85%, 38–62% and 60–40% respectively. The control group was treated only with sugarcane molasses. Water quality, chlorophyll a concentration, heterotrophic bacterial load, Vibrio spp. concentration and zootechnical indexes were all evaluated. Total ammonia concentration was controlled by heterotrophic and chemotrophic pathways. Biofloc formation, as quantified by measuring the total suspended solids, was not altered. The Vibrio spp. concentration showed a significant reduction in treatments with soybean‐to‐sugarcane molasses ratios of 38–62% and 60–40%. All combined mixtures of soybean and sugarcane molasses could maintain water quality and productivity in the superintensive culture of L. vannamei using the biofloc system. Thus, the potential use of a residue from agroindustry as a carbon source in a biofloc culture is demonstrated.  相似文献

Portable X-ray fluorescence (pXRF) spectrometry and magnetic susceptibility (MS) via magnetometer have been increasingly used with terrain variables for digital soil mapping. However, this methodology is still emerging in many countries with tropical soils. The objective of this study was to use proximal soil sensor data associated with terrain variables at varying spatial resolutions to predict soil classes using the Random Forest (RF) algorithm. The study was conducted on a 316-ha area featuring highly variable soil classes and complex soil-landscape relationships in Minas Gerais State, Brazil. The overall accuracy and Kappa index were evaluated using soils that were classified at 118 sites, with 90 being used for modeling and 28 for validation. Digital elevation models (DEMs) were created at 5-, 10-, 20-, and 30-m resolutions using contour lines from two sources. The resulting DEMs were processed to generate 12 terrain variables. Total Fe, Ti, and SiO2 contents were obtained using pXRF, with MS determined via a magnetometer. Soil class prediction was performed using the RF algorithm. The quality of the soil maps improved when using only the five most important covariates and combining proximal sensor data with terrain variables at different spatial resolutions. The finest spatial resolution did not always provide the most accurate maps. The high soil complexity in the area prevented highly accurate predictions. The most important variables influencing the soil mapping were MS, Fe, and Ti. Proximal sensor data associated with terrain information were successfully used to map Brazilian soils at variable spatial resolutions.  相似文献

The potential of postharvest dip treatments with fludioxonil (FLU) (a synthetic analogue of the bacterial metabolite of pyrrolnitrin), in controlling postharvest decay caused by Penicillium digitatum and Penicillium italicum of citrus fruit was investigated in comparison with the conventional fungicide imazalil (IMZ). The ultrastructural changes of fruit epicuticular wax was investigated as a function of water dip temperature, and the possible role of these changes was related to residue accumulation under FLU treatment. Residues retained by fruit were determined as a function of fungicide concentration, dip temperature, and fruit storage conditions. Scanning electron microscopy analysis revealed that fruit dipping in water at 30 or 40 degrees C did not cause differences in cuticular wax's ultrastructure in comparison to control fruit, while treatments at 50, 55, or 60 degrees C caused the disappearance of wax platelets, resulting in relatively homogeneous skin surface, due to partial "melting" of epicuticular wax. Residues of FLU in fruit treated at 20 or 50 degrees C were significantly correlated with the doses of fungicide applied. When equal amounts of fungicide were employed, the residue concentrations were notably higher (from 2.6- to 4-fold) in fruit treated at 50 degrees C than in fruit treated at 20 degrees C. The dissipation rate of FLU in "Salustiana" and "Tarocco" oranges was lower in fruit subjected to treatment at 50 degrees C. The minimal FLU concentration for almost complete decay control in artificially wounded fruit during 7-d storage at 20 degrees C was 400 mg/L active ingredient (ai) in fruit treated at 20 degrees C and 100 mg/L ai in fruit treated at 50 degrees C. Results on nonwounded Tarocco oranges subjected to 3 weeks of simulated quarantine conditions at 1 degrees C, plus 6 weeks of standard storage at 8 degrees C and an additional two weeks of simulated marketing period (SMP) at 20 degrees C revealed that almost complete decay control with FLU applications of 100 mg/L at 50 degrees C and 400 mg/L at 20 degrees C resulted in ca. 0.8 mg/kg FLU fruit residues, in agreement with results on wounded citrus fruit. When equal concentrations and temperatures were applied, FLU treatments were as effective as IMZ. In vitro trials showed a low sensitivity to FLU against P. digitatum and P. italicum isolates. MIC values for the complete inhibition of mycelium growth were >or=100 microg/mL, while ED(50) values ranged from 0.1 to 1 microg/mL for P. digitatum and from 1 to >100 microg/mL for P. italicum. The latter result suggests that care should be taken to avoid exclusive application of FLU in a sustainable program for management of fruit decay. However, integrating fungicide application and hot water dip may reduce the possibility of selecting fungicide-resistant populations of the pathogen, by increasing the effectiveness of the treatment.  相似文献

Summary Aluminum toxicity due to the cation Al+3 is a major factor limiting yields in acid soils. Wide genetic variability to aluminum tolerance is found in oat genotypes. The objectives of this study were to determine the number of genes controlling aluminum tolerance in oats and to verify if any detrimental effects were present of the aluminum tolerance genes on grain yield and grain quality in Al+3free soils. Aluminum tolerance was estimated as the average regrowth of the main root after exposure to toxic levels of Al+3 in a hydroponic solution under controlled conditions. The number of genes controlling that trait was estimated from the distribution of the average root regrowth frequencies in a population of 333 recombinant inbred lines (RIL's) in generations F5:6 and F5:7. The effects on grain yield and grain quality were assessed in a subpopulation of 162 RIL's chosen based on their aluminum tolerance response. Aluminum tolerance in the evaluated population was controlled by one dominant major gene with the tolerant genotypes carying Al a Al a and the sensitive ones al a al a alleles. Cv. Star Ruby grapefruit (Citrus paradisi Macf.) were subjected to a 3-min dip in water at room temperature (20 degrees C) or at 50 degrees C with or without 25, 50, or 100 mg/L azoxystrobin (AZX). Then, the fruits were subjected to cold quarantine at 2 degrees C and 90-95% relative humidity (RH) for 3 weeks and then stored for 5 weeks at 8 degrees C and approximately 85% RH and for another 2 weeks at 20 degrees C and 80% RH to simulate a 2-week marketing period (SMP). No AZX residues were detected in the albedo and pulp following treatments at 20 or 50 degrees C, the total amount of residues being recovered from the flavedo tissue. There was a relationship between the AZX uptake in fruit and the amount of fungicide employed at 20 or 50 degrees C. When AZX was applied to the fruit at 25 mg/L at 20 degrees C, the residue level averaged 0.11 mg/kg (active ingredient, whole fruit basis). This residue concentration increased by 50 and 75% when the application rate increased from 25 to 50 or 100 mg/L, respectively. A similar pattern of accumulation was detected in fruit subjected to treatments at 50 degrees C. However, treatments at 50 degrees C produced residue levels higher than the treatments at 20 degrees C, with increases ranging from 63 to 84%, for the same concentration. Storage conditions did not affect the amount of AZX residues in the fruit. Treatment at 50 or 100 mg/L at 20 degrees C reduced the incidence of moderate to severe chilling injury (CI). Water dips at 50 degrees C reduced the incidence and severity of CI to a very low extent, with no additional advantages when hot water was used in combination with AZX. Treatments with 50 or 100 mg/L of AZX at 20 degrees C produced beneficial effects in decay control similar to those of 25 mg/L AZX at 50 degrees C or hot water alone. Better results were achieved with 50 or 100 AZX at 50 degrees C, providing complete control of decay during cold storage and with negligible decay after SMP. It was concluded that when AZX was applied at 50 degrees C, low doses of fungicide and minimal residue levels in fruit were required to control the postharvest decay of grapefruit. This treatment does not impair fruit quality and offers very interesting prospects for large scale application, due to the reduced potential toxicity of AZX to nontarget organisms and to the environment.  相似文献

Emmer wheat (Triticum dicoccon Schrank, 2n = 4x = 28) consists in a hulled wheat; its cultivation has been drastically reduced during the last century as a consequence of its low yield. Recently, its agronomic and nutritive values, as well as the increase of popularity of organic agriculture, have led to a renewed interest making its cultivation economically viable in the marginal lands with an increase of the cultivated areas. In Italy, it mainly survives in few marginal lands of central and southern Italy, where local varieties, adapted to the natural environment from where they originate, are used; moreover, some selected lines have also been developed. In the present work, agro-morphological and qualitative traits, together with molecular analyses of 20 emmer accessions consisting of Italian landraces, breeding lines, and cultivars, were performed. The field experiments were conducted for two consecutive years (2001/2002–2002/2003) in two locations: Viterbo in central Italy, and Foggia in south Italy. The analyzed emmer wheat accessions showed a good amount of genetic variability for both evaluated agro-morphological and molecular traits. This study illustrates an increase in earliness, GY, TW, TKW, and YI going from landraces, breeding lines to cultivars, while the variability does not change proportionally.  相似文献
