Single vitamin ingredients and blends in premixes are widely used in the food and supplement industries and are predominantly in powder form. To meet label claims and/or determine appropriate overages, it is important to characterize the stability of these ingredients. Although moisture is a known promoter of instability in powder blends, the combined effects of storage relative humidity (RH), formulation, and deliquescence on the stability of these systems are not well-characterized. The objective of this study was to determine the effect of deliquescence on the stability of vitamins B 1, B 6, and C and their mixtures. Deliquescence points (RH 0s) for all formulations were determined by moisture sorption analysis. Single, binary, ternary, and quaternary mixtures of thiamin HCl, pyridoxine HCl, sodium ascorbate, and fructose were stored in RH-controlled environmental chambers between 43 and 98% RH at 22 degrees C for up to 12 weeks. Vitamin stability was determined by high-performance liquid chromatography (HPLC). Formulation and storage RH significantly affected vitamin stability. Thiamin and ascorbate degradation were significantly promoted above the RH 0, while pyridoxine was least affected by storage RH. The deliquescence lowering phenomenon enhanced moisture sorption of blends at RHs below the RH 0s. Ascorbate enhanced thiamin degradation. Therefore, formulation, storage conditions, and the relation of these to deliquescence points may affect the shelf life, quality, and functionality of vitamin blends and should be considered in product development, processing, storage, and use.  相似文献

Triticum urartu is a wild diploid wheat that has been identified as the donor of the A genome in polyploid wheats. The gliadins of this species were analysed by acid-PAGE in accessions from Armenia, Iran, Iraq, Lebanon, former-Soviet Union and Turkey. Up to twenty-six different alleles for each locus (Gli-A u 1 and Gli-A u 2) implicated in the synthesis of these proteins, were found. The genetic diversity was high, although a great part of this diversity is in danger of erosion given that the distribution of the combinations is not random. Consequently, loss of these accessions could mean loss of the allelic variants. These results provide new basic knowledge regarding the genetic variability of the seed storage proteins synthesised by the A u genome, as well as their potential to obtain novel germplasm for the improvement of wheat quality in breeding programmes.  相似文献

The current experiments correlate the disposition kinetics of albendazole (ABZ) following its intravenous (i.v.) and intraruminal (i.r.) administrations to Moniezia spp.-infected sheep, with the pattern of drug/metabolite uptake by tapeworms collected from treated animals. The ex vivo uptake pattern of ABZ and albendazole sulphoxide (ABZSO) by the same cestode parasite was also investigated. Naturally infected (Moniezia spp.) Corriedale lambs were treated with ABZ by either i.v. (Group A, n = 15) or i.r. (Group B, n = 15) administration at 7.5 mg/kg. Plasma and abomasal fluid samples were obtained over a 120-h period. Two animals per group were killed at 0.5, 1, 2, 4 and 6 h post-treatment; parasite material (tapeworms), bile and intestinal fluid samples were recovered. Furthermore, Moniezia spp. tapeworms obtained from sheep killed at the local abattoir were incubated with either ABZ or ABZSO for different time periods in a Kreb's Ringer Tris buffer (ex vivo experiments). Samples were analysed by high performance liquid chromatography for ABZ, ABZSO and albendazole sulphone (ABZSO2). ABZ plasma concentrations decreased rapidly and were not detectable beyond 10 h following i.v. administration. ABZSO and ABZSO2 were the metabolites recovered in plasma after both treatments. ABZ and its metabolites were extensively distributed to the digestive tract, mainly into the abomasal fluid, after the i.v. and i.r. administrations. The parent drug and its active ABZSO metabolite were recovered in tapeworms collected from both i.v. and i.r. treated lambs. However, the availability of both ABZ and ABZSO was higher in parasite material recovered from i.v. treated animals. The uptake of ABZ by the cestode parasite, both in vivo and ex vivo, was significantly greater than that of its sulphoxide metabolite, which agrees with the higher lipophilicity of the parent drug.  相似文献

Intra-specific diversity within Moraxella bovis was investigated analysing DNA fingerprints, outer membrane proteins (OMP) and lipopolysaccharides (LPS) profiles. Three collection strains and 57 isolates of M. bovis, collected during 3 years from cattle with infectious bovine keratoconjunctivitis (IBK) symptoms, from diverse geographical locations of Argentina, were examined. The LPS and OMP profiles were studied through SDS–PAGE analysis and genotype was determined by PCR-DNA fingerprinting. Genotyping identified five DNA types while analysis of LPS and OMP profiles identified three rough LPS types and three OMP types among the 60 isolates of M. bovis including the three collection strains. None of the three methods employed to assess diversity was discriminating when used alone because the degree of heterogeneity in each group of surface structures was limited, but when data of each typing method were combined, 15 distinct subgroups were determined. This subgrouping was clearly able to differentiate isolates of the same genotype. These typing methods appear to be useful to assess different aspects of the disease such as the diversity within a population of M. bovis associated to epidemic conditions, track the causal agent in an outbreak of the disease, monitoring vaccination programs and studies on virulence.  相似文献

A survey of the genetic diversity among the major cuban rice cultivars was conducted using isozyme, RAPD and AFLP markers. Polymorphisms were detected for esterases, peroxidases, alcohol dehydrogenases and polyphenoloxidases systems; 21 RAPD primers and four AFLP primer combinations. Heterozygosity arithmetic mean value (Hav(p)), the effective multiplex ratio (EMR) and the marker index (MI), were calculated for isozyme, RAPD and AFLP markers. The mean value of genetic similarity among the different varieties was 0.92 for isozyme, 0.73 for RAPD and 0.58 for AFLP analyses. Thus, AFLP were able to detect polymorphisms with higher efficiency than RAPD (+15%) and isozyme (+34%). Data from the isozyme, RAPD and AFLP analyses were used to compute matrices of genetic similarities. The efficiency of the UPGMA for the estimation of genetic relatedness among varieties was supported by cophenetic correlation coefficients. The resulting values indicated that the distortion level for the estimated similarities was minimal. The correlation coefficients obtained by the Mantel matrix correspondence test, which was used to compare the cophenetic matrices for the different markers, showed that estimated values of genetic relationship given for isozyme and RAPD markers (r = 0.89), as well as for AFLP and RAPD markers (r = 0.82) were properly related. However, AFLP and isozyme data showed only moderate correlation (r = 0.63). Although the genetic variability found among the different cultivars was low, both RAPD and AFLP markers proved to be efficient tools in assessing the genetic diversity of rice genotypes. Hexaploid tritordeum is an amphiploid derived from the cross between Hordeum chilense and durum wheat. This amphiploid has shown potential for bread making, which has been associated to the prolamins from H. chilense. The role of each prolamin subunit on the gluten strength in tritordeum has been evaluated. Advanced progenies from two hexaploid tritordeum crosses were analysed for prolamins composition and gluten strength. Six loci were found for the prolamins synthesised at the Hch genome, which showed significant effects on gluten strength. Although these tritordeum lines represent only a small proportion of the genetic variability available in the development programme for this new crop, a certain degree of variation for prolamins composition was detected. In fact, up to three allelic variants have been detected for some loci, which have shown different effect on gluten strength in tritordeum. Each of these six loci appeared on the same linkage group that corresponded to chromosome 1Hch. The search of new variants for these loci could be useful for tritordeum quality breeding and, using tritordeum as a bridge species, this genetic variability could be introgressed into bread wheat. Background  

Aliphatic molecules containing free carboxyl groups are important intermediates in many metabolic and signalling reactions, however, they accumulate to low levels in tissues and are not efficiently ionized by electrospray ionization (ESI) compared to more polar substances. Quantification of aliphatic molecules becomes therefore difficult when small amounts of tissue are available for analysis. Traditional methods for analysis of these molecules require purification or enrichment steps, which are onerous when multiple samples need to be analyzed. In contrast to aliphatic molecules, more polar substances containing free carboxyl groups such as some phytohormones are efficiently ionized by ESI and suitable for analysis by LC-MS/MS. Thus, the development of a method with which aliphatic and polar molecules -which their unmodified forms differ dramatically in their efficiencies of ionization by ESI- can be simultaneously detected with similar sensitivities would substantially simplify the analysis of complex biological matrices.  相似文献

The rapid growth of tilapia culture has stimulated the expansion of tilapia feed production and a search for novel protein sources to replace fish meal. Vegetable or plant sources are promising alternatives and legumes are both naturally abundant and high in protein content. A study was carried out to evaluate the effects of replacing the protein from fish meal with peanut ( Arachis hypogaea ) leaf meal (PLM) in diets for male tilapia ( Oreochromis niloticus , initial body weight 75.3 g). Four isonitrogenous (35% CP) and isocaloric (18.834 kJ g−1) diets were prepared to include 0% (CON), 10% (PLM10), 20% (PLM20) and 30% (PLM30) of PLM protein. Average fish weights at the end of the 126-day experiment were not statistically different among the fish fed CON, PLM10 and PLM20 diets. The PLM30 diet produced the poorest growth performance. Organic matter and protein contents of fish were similar in the CON, PLM10 and PLM20 diets. Carcass chemical composition showed a decrease in body fat content as PLM replacement levels increased. The high survival ratio in all dietary groups (>97%) suggests that PLM can be used in O. niloticus feeds for long periods without affecting fish growth performance or health.  相似文献

Chia (Salvia hispanica L.) plant is native from southern Mexico and northern Guatemala. Their seeds are a rich source of bioactive compounds which protect consumers against chronic diseases. Germination improves functionality of the seeds due to the increase in the bioactive compounds and associated antioxidant activity. The purpose of this study was to obtain functional flour from germinated chia seeds under optimized conditions with increased antioxidant activity, phenolic compounds, GABA, essential amino acids, and dietary fiber with respect to un-germinated chia seeds. The effect of germination temperature and time (GT = 20–35 °C, Gt = 10–300 h) on protein, lipid, and total phenolic contents (PC, LC, TPC, respectively), and antioxidant activity (AoxA) was analyzed by response surface methodology as optimization tool. Chia seeds were germinated inside plastic trays with absorbent paper moisturized with 50 mL of 100 ppm sodium hypochlorite dissolution. The sprouts were dried (50 °C/8 h) and ground to obtain germinated chia flours (GCF). The prediction models developed for PC, LC, TPC, and AoxA showed high coefficients of determination, demonstrating their adequacy to explain the variations in experimental data. The highest values of PC, LC, TPC, and AoxA were obtained at two different optimal conditions (GT = 21 °C/Gt = 157 h; GT = 33 °C/Gt = 126 h). Optimized germinated chia flours (OGCF) had higher PC, TPC, AoxA, GABA, essential amino acids, calculated protein efficiency ratio (C-PER), and total dietary fiber (TDF) than un-germinated chia seed flour. The OGCF could be utilized as a natural source of proteins, dietary fiber, GABA, and antioxidants in the development of new functional beverages and foods.  相似文献
