Pigments are important contributors to the appearance and healthful properties of both avocado fruits and the oils extracted from these fruits. This study determined carotenoid and chlorophyll pigment concentrations in the skin and three sections of the flesh (outer dark green, middle pale green, and inner yellow flesh-nearest the seed) and anthocyanin concentrations in the skin of Hass avocado during ripening at 20 degrees C. Pigments were extracted from frozen tissue with acetone and measured using high-performance liquid chromatography. Pigments were also measured in the oil extracted from freeze-dried tissue sections by an accelerated solvent extraction system using hexane. Carotenoids and chlorophylls identified in the skin, flesh, and oil were lutein, alpha-carotene, beta-carotene, neoxanthin, violaxanthin, zeaxanthin, antheraxanthin, chlorophylls a and b, and pheophytins a and b with the highest concentrations of all pigments in the skin. Chlorophyllides a and b were identified in the skin and flesh tissues only. As the fruit ripened and softened, the skin changed from green to purple/black, corresponding to changes in skin hue angle, and a concomitant increase in cyanidin 3-O-glucoside and the loss of chlorophyllide a. In flesh tissue, chroma and lightness values decreased with ripening, with no changes in hue angle. The levels of carotenoids and chlorophylls did not change significantly during ripening. As fruit ripened, the total chlorophyll level in the oil from the flesh sections remained constant but declined in the oil extracted from the skin.  相似文献

It is shown that potentially persistent transformation products can be formed from the herbicides bromoxynil (3,5-dibromo-4-hydroxybenzonitrile) and ioxynil (3,5-diiodo-4-hydroxybenzonitrile), and possible leaching to groundwater is discussed. A similar process to the formation of BAM (2,6-dichlorobenzamide) from the herbicide dichlobenil (2,6-dichlorobenzonitrile) can be anticipated as bromoxynil and ioxynil are analogues of dichlobenil and they are degraded by the enzymes nitrilase, nitrile hydratase and amidase. A biodegradation study using cultured Variovorax sp. DSM 11402, a species commonly found in soil, demonstrated that ioxynil and bromoxynil were fully transformed into their corresponding amides in 2-5 days. These amides were not further degraded within 18 days, and formation of other degradation products was not observed. These results are in agreement with biodegradation experiments with dichlobenil. In soil, dichlobenil is transformed into its only observed degradation product BAM, which is persistent and mobile, and has been found in 19% of 5000 samples of Danish groundwater. Variovorax sp. is known to degrade the non-halogenated analogue benzamide, suggesting that degradation of the three amides may be hindered by the halogenated substituents (meta-Br; meta-I; ortho-Cl). This hypothesis is supported by QSAR modelling of fundamental properties. Using a new optimised liquid chromatography-tandem mass spectrometry (LC-MS/MS) method, the sorption and desorption properties of bromoxynil and ioxynil were characterised in sandy topsoil at four concentration levels. The estimated sorption coefficient K(d) was 1.4 L kg(-1) for bromoxynil and 5.4 L kg(-1) for ioxynil, indicating weak to moderate sorption to topsoil. Desorption of the herbicides showed that they were strongly and irreversible bound to the soil (K(des) > K(d)). The amount of herbicide desorbed depended on the initial concentration level. At low levels, K(des) values were higher, indicating stronger binding than at higher levels. The isocratic LC-MS/MS method developed for simultaneous detection of bromoxynil, ioxynil and their main degradation products is described. Using negative electrospray ionisation (ESI-), the detection limits were 0.4-1.0 microg L(-1), with relative standard deviations of 4-10% (n = 10) using direct injection without clean-up steps. Background

The objective of this study was to investigate the association between average daily gain and the number of Lawsonia intracellularis bacteria in faeces of growing pigs with different levels of diarrhoea.

Methods

A longitudinal field study (n = 150 pigs) was performed in a Danish herd from day 29 to 47 post weaning. Every third day all pigs were weighed, subjected to a clinical examination and faecal samples were obtained. Faecal samples were subjected to dry matter determination and absolute quantification by PCR for L. intracellularis and porcine circovirus type 2 (PCV2). Association between average daily gain, faecal dry matter content, numbers of L. intracellularis bacteria and PCV2 genome copies in faeces was investigated in a multilevel mixed-effects linear model.

Results

Increasing numbers of L. intracellularis log10 bacteria/g faeces were significantly associated with decreasing average daily gain (P < 0.001). The association was decreasing with increasing faecal dry matter content (P < 0.01). The number of PCV2 log10 copies/g faeces was not significantly associated with average daily gain of the pigs (P > 0.5).

Conclusion

The results suggest a potential application of a PCR quantifying L. intracellularis in growing pigs. Faecal dry matter content must be taken into consideration in interpretation of such test results.  相似文献

Oligodendrogliomas are the second most common malignant brain tumor in adults and exhibit characteristic losses of chromosomes 1p and 19q. To identify the molecular genetic basis for this alteration, we performed exomic sequencing of seven tumors. Among other changes, we found that the CIC gene (homolog of the Drosophila gene capicua) on chromosome 19q was somatically mutated in six cases and that the FUBP1 gene [encoding far-upstream element (FUSE) binding protein] on chromosome 1p was somatically mutated in two tumors. Examination of 27 additional oligodendrogliomas revealed 12 and 3 more tumors with mutations of CIC and FUBP1, respectively, 58% of which were predicted to result in truncations of the encoded proteins. These results suggest a critical role for these genes in the biology and pathology of oligodendrocytes.  相似文献

Drought is one of the important factors limiting crop production in arid and semi-arid regions. Four drought resistance criteria, consisting of canopy temperature, stomatal resistance, transpiring area and rate of water loss by excised-leaves were examined in two experiments conducted in optimum and stress moisture conditions in 1995. A randomized complete block design with four replications and six genotypes was used for each experiment. Under optimum moisture conditions, there was a significant difference in canopy temperature at midday between drought resistant and drought sensitive genotypes. A significant linear relationship was also obtained between this criterion and yield reduction ratio as determined by Yr = 1-(Ys/Yp) (Ys and Yp = yield under stress and non-stress conditions, respectively), at the ear emergence stage. The only significant difference between cultivars in respect to stomatal resistance in stress conditions was that of adaxial leaf surface in ear emergence stage. No significant linear relationship was obtained between transpiring area and yield reduction ratio. Cultivars differed in respect to rate of water loss (RWL) and initial water content (IWC) in ear emergence and grain filling in stress environment only. Ultimately, three criteria namely canopy temperature, stomatal resistance and RWL, in explicated conditions were recognized as benificial drought resistance indicators. Scaling up the hatchery production of juvenile sandfish Holothuria scabra is constrained by limited hatchery space and the associated high operational costs. To shorten the hatchery rearing phase, ocean nursery systems like floating hapa nets have been used with good prospects but with limitations during rough sea conditions. In this study, the potential of bottom‐set trays (0.14 m2) as an alternative ocean nursery system for early sandfish juveniles (0.5 ± 0.1 cm) was evaluated. The effects of stocking density and presence of artificial substrates (AS) on the growth and survival were determined in a 60‐day field experiment. Average length and growth rates at lower stocking density treatment (100 individuals tray?1) were significantly higher (1.45 ± 0.22 cm; 0.03 ± 0.01 cm day?1) than at higher stocking density treatments (400 and 500 individuals tray?1) 0.95 ± 0.06 cm; 0.03 ± 0.004 cm day?1) with or without AS (< .05) respectively. The coefficient of variation in length (CV) at high stocking densities were significantly higher than at low densities (< .05) and growth rate was strongly negatively correlated with density. Survival was significantly higher (55% ± 9%) in trays with AS across all stocking density treatments than in trays without AS (34% ± 2%). Results suggest that AS may have reduced intra‐ and interspecific interactions, resulting to significantly lower growth variations and higher survival. The bottom‐set tray with AS can be a practical alternative ocean nursery unit for rearing early sandfish juveniles particularly when the sea surface condition is rough. With improved design and density management, survival and growth may be further enhanced.  相似文献

Low concentrations of a disulfide reducing agent were added to unheated and heated (80 °C for 30 min) skim milk, with and without added whey protein. The reduction of the β-lactoglobulin and κ-casein disulfide bonds was monitored over time using electrophoresis. The distribution of the proteins between the colloidal and serum phases was also investigated. κ-Casein disulfide bonds were reduced in preference to those of β-lactoglobulin in both unheated and heated skim milk (with or without added whey protein). In addition, in heated skim milk, while the serum κ-casein was reduced more readily than the colloidal κ-casein, the distribution of κ-casein between the two phases was not affected.  相似文献

During the past 20 years, plant ingredients have taken over as the main constituents in feed for Atlantic salmon. This has changed the nutrient composition of the feeds, the bioavailability of nutrients and perhaps nutrient metabolism. Plant‐based diets also contain more anti‐nutrients. The EU‐funded project ARRAINA re‐evaluated recommendations for micronutrient supplementation to Atlantic salmon feeds, and the present study compared a diet supplemented with micronutrients according to NRC (2011) (control diet, 100% NP (nutrient package)) with a diet supplemented according to the new recommendations (New NP). Tissue concentrations of pyridoxine, pantothenic acid, niacin, vitamin C, Zn and Se were significantly higher; and Cu was lower in Atlantic salmon fed the diet with the New NP compared to the control fish. The New NP also gave a near significant effect on growth, decreased muscle firmness and increased muscle cell size, and it affected metabolism of nitrogen‐containing metabolites in the muscle. While we cannot be certain which micronutrient(s) gave these effects, the B vitamins are probable candidates, since they are mediators of intermediary metabolism and have been shown to influence some of the affected metabolites.  相似文献

Considering a potential application of selected biochemically activated insect hormonogen substances (juvenogens) against pest termite species, we aimed this study to describe the metabolism of these compounds by termites and soil bacteria and to evaluate the potential impact of their metabolites on the environment. Radiolabelled derivatives of the juvenogens cis- and trans-isomers of ethyl N-{2-[4-(2-butanoyloxycyclohexyl)methyl]phenoxy}ethyl carbamate were metabolized by the termite Reticulitermes flavipes and the bacteria Bacillus simplex and Bacillus sp., strain 05 (GenBank EU399813) giving rise to different numbers of metabolites. The trans-isomer of the juvenogen was metabolized by both Bacillus species into its parent synthetic structure, ethyl trans-N-{2-[4-(2-hydroxycyclohexyl)methyl]phenoxy}ethyl carbamate, while the cis-isomer was metabolized into further products. Both racemic juvenogens were metabolized bytermites, affording mainly the parent juvenoids. In terms of ecotoxicity, the trans-juvenogen shows a significantly lower toxicity than the cis-juvenogen. In contrast, the toxicity of the cis-juvenoid (main degradation product of cis-juvenogen) is higher than the toxicity of trans-juvenoid (main degradation product of trans- juvenogen). The precursors of the two juvenogens cis- and trans-2-(4-hydroxybenzyl)cyclohexanol were also tested but exhibited a low toxicity. The results demonstrate that bacteria can metabolize the juvenogen in liquid media culture and have implications for the development of a strategy for bioremediation of soil. Moreover, the products of the biodegradation exhibited low toxicity. Both juvenogens have a high juvenilizing effect, cause low mortality and are stable within a period of two weeks.  相似文献
