Avian pathogenic Escherichia coli (APEC) are often found in poultry and are responsible for a set of diseases, commonly referred to as avian colibacillosis. One of the important virulence factors is adhesion to different epithelial surfaces, which is mediated by pili. P pili are thought to play a role by means of their PapG adhesin, which occurs in three molecular variants: PapGI, PapGII and PapGIII. This study is the first to determine and analyse the distribution of the different papG alleles in APEC. Our results show a significant predominance of the papGII allele above all other alleles or allele combinations. No statistically significant associations could be found between papG allele distribution and the type of bird, organ of isolation and O serogroup. Finally, the papGII and papGIII sequences showed high homology with mammalian (including human) source papG sequences.
Foot-and-mouth disease virus (FMDV) is an aphthovirus of the family Picornaviridae and the etiological agent of the economically most important animal disease. As a typical picornavirus, FMD virions are nonenveloped particles of icosahedral symmetry and its genome is a single stranded RNA of about 8500 nucleotides and of positive polarity. FMDV RNA is infectious and it replicates via a complementary, minus strand RNA. FMDV RNA replication is error-prone so that viral populations consist of mutant spectra (quasispecies) rather than a defined genomic sequence. Therefore FMDV in nature is genetically and antigenically diverse. This poses important challenges for the diagnosis, prevention and control of FMD. A deeper understanding of FMDV population complexity and evolution has suggested requirements for a new generation of anti-FMD vaccines. This is relevant to the current debate on the adequacy of non-vaccination versus vaccination policies for the control of FMD.
OBJECTIVE: To evaluate use of a pasteurized tumoral autograft prepared from the resected primary bone neoplasm for limb sparing in a dog with distal radial osteosarcoma (OSA). STUDY DESIGN: Clinical case report. ANIMALS: A 9-year-old male Maremma shepherd dog. METHODS: After right distal radial OSA removal, the tumoral autograft was pasteurized. The excised bone segment was placed in a sterile watertight box containing sterile saline solution preheated to 65 degrees C in a water bath. The box was kept immersed in the water bath at 65 degrees C for 40 minutes to kill the tumor cells. The autograft was then fixed in the host with a plate and screws based on standard AO/ASIF technique for carpal arthrodesis. Three doses of cisplatin (70 mg/m(2) intravenously) were administered, 3 weeks apart; the initial dose was administered the day after surgery. RESULTS: The autograft was incorporated in a manner comparable to an allograft, and after 708 days, the metallic implants were removed. A 1-month activity restriction as well as spoon splint to protect the leg from a full loading were used thereafter. Limb function was fair to good, and the dog remains disease free after 56 months. CONCLUSIONS: A pasteurized autograft consisting of the resected primary bone neoplasm is a valid alternative to a cortical bone allograft for limb sparing in dogs with appendicular OSA in terms of feasibility and pattern of healing. CLINICAL RELEVANCE: This procedure can be an alternative method of limb sparing when difficulties are encountered in establishing and maintaining a canine bone allograft bank.
The development of the nematode Spinitectus inermis (Zeder, 1800), a parasite of the stomach of eels, Anguilla anguilla (L.) in Europe, was experimentally studied. Mayfly nymphs Caenis macrura, Ecdyonurus dispar, Heptagenia sulphurea, Potamanthus luteus and Seratella ignita from Portugal and the Czech Republic were found to serve as experimental intermediate hosts. After ingestion of the nematode eggs by the mayfly nymphs, the toothed first-stage larvae were released and penetrated into the body cavity of the intermediate host. There they moulted twice (on day 4 and 6 post infection [p.i.] at water temperatures of 20-25 degrees C), attaining the third infective stage. The definitive host, A. anguilla, undoubtedly acquires infection by feeding on mayfly nymphs harbouring infective-stage larvae. In an experimentally infected eel, the fourth-stage larva undergoing the third moult was observed 28 days p.i. at water temperature of 20 degrees C. The larval stages, including moulting forms, are described and illustrated. The prepatent period of S. inermis is estimated to be about two months.
An experiment of selection for ovulation rate was carried out. Animals were derived from a synthetic line first selected 12 generations for litter size, then 10 generations for uterine capacity. Selection was relaxed for 6 generations. Selection was based on the phenotypic value of ovulation rate with a selection pressure on does of 30%. Males were selected from litters of does with the highest ovulation rate. Males were selected within sire families in order to reduce inbreeding. Ovulation rate was measured in the second gestation by a laparoscopy, 12 days after mating. Each generation had about 80 females and 20 males. Results of three generations of selection were analyzed using Bayesian methods. Marginal posterior distributions of all unknowns were estimated by Gibbs sampling. Heritabilities of ovulation rate (OR), number of implanted embryos (IE), litter size (LS), embryo survival (ES), fetal survival (FS), and prenatal survival (PS) were 0.44, 0.32, 0.11, 0.26, 0.35, and 0.14, respectively. Genetic correlation between OR and LS was 0.56, indicating that selection for ovulation rate can augment litter size. Response to selection for OR was 1.80 ova. Correlated responses in IE and LS were 1.44 and 0.49, respectively. Selection for ovulation rate may be an alternative to improve litter size.
Agreement among pathologists interpreting histologic specimens is an area of interest within human pathology, but little work in this area has been reported in the veterinary literature. Agreement among pathologists evaluating routine histologic sections of amputated digits from cats and dogs submitted to multiple diagnostic centers was examined. Histologic sections from surgical specimens were reviewed in a blinded fashion by two pathologists, and a comparison to the original diagnosis, as stated in the diagnostic report, was recorded. A total of 513 cases were reviewed, and complete agreement was reached in 409 (79.7%). Of the 104 instances of disagreement, 77 (74.0%) were considered to be of clinical significance. The diagnosis of keratoacanthoma was disagreed with in 19 of 21 diagnoses (90.4%). No other individual diagnosis was similarly disputed. The overall level of disagreement is large and is similar to that reported in human pathology and suggests that further study of this issue would be useful in veterinary pathology.
The effects of origin of seed potatoes and the cropping history on the phenotypic structure of Phytophthora infestans populations was studied in northern Hessia, central Germany, from 2000 to 2002. Populations originating from fields with
a history of potato cropping with only short or no rotation (old fields) were compared with populations from new fields, i.e.,
where no potatoes had been grown for at least 30 years and seed potatoes were either imported from breeders or produced on-farm
(certified). The main goal was to determine the importance of seed potato infection in the establishment of new P. infestans populations. Isolates were characterized for mating type, virulences and rep- (repetitive extragenic palindromic) PCR fingerprints.
Among a total of 639 isolates sampled from 31 sites, mating types A1 and A2 co-existed in all three years in 60–92% of the
sites. Over all three years, 53 pathotypes were detected in a subsample of 272 isolates. Isolates originating from the new
fields had significantly higher frequencies of the virulences v1, v2, v3, v6 and v7, indicating general effects of seed introduction
into a new region. Thirty-six fingerprints were detected in a subsample of 281 isolates of which 22 were unique while four
occurred in all three years and in many sites. Pathogen populations from potato fields that were grown from seed tubers of
geographically different origin differed significantly based on χ2 tests. While the Nei genetic distances were less than .1 among the local populations, distances to the US lineages US-1,
US-6, US-7 and US-8 ranged from .22 to .47; however, the bootstrap values were not significant. Populations from old fields
were more diverse and 14 of the 22 rep-PCR types occurred there among 132 isolates tested in comparison to six in the new
fields (n = 140 isolates) and two among six isolates from volunteers. The results also suggest that both sexual and asexual reproduction
play a role.
