Qualitative and quantitative analyses of phenolic compounds were carried out on quince fruit samples from seven different geographical origins in Portugal. For each origin, both pulp and peel were analyzed by reversed-phase HPLC-DAD and HPLC-DAD/MS.The results revealed differences between the phenolic profiles of pulps and peels in all studied cases. The pulps contained mainly caffeoylquinic acids (3-, 4-, and 5-O-caffeoylquinic acids and 3,5-dicaffeoylquinic acid) and one quercetin glycoside, rutin (in low amount). The peels presented the same caffeoylquinic acids and several flavonol glycosides: quercetin 3-galactoside, kaempferol 3-glucoside, kaempferol 3-rutinoside, and several unidentified compounds (probably kaempferol glycoside and quercetin and kaempferol glycosides acylated with p-coumaric acid). Microalgae are known as a producer of proteins and lipids, but also of valuable compounds for human health benefits (e.g., polyunsaturated fatty acids (PUFAs); minerals, vitamins, or other compounds). The overall objective of this research was to prospect novel products, such as nutraceuticals from microalgae, for application in human health, particularly for metabolic diseases. Chlorella vulgaris and Chlorococcum amblystomatis were grown autotrophically, and C. vulgaris was additionally grown heterotrophically. Microalgae biomass was extracted using organic solvents (dichloromethane, ethanol, ethanol with ultrasound-assisted extraction). Those extracts were evaluated for their bioactivities, toxicity, and metabolite profile. Some of the extracts reduced the neutral lipid content using the zebrafish larvae fat metabolism assay, reduced lipid accumulation in fatty-acid-overloaded HepG2 liver cells, or decreased the LPS-induced inflammation reaction in RAW264.7 macrophages. Toxicity was not observed in the MTT assay in vitro or by the appearance of lethality or malformations in zebrafish larvae in vivo. Differences in metabolite profiles of microalgae extracts obtained by UPLC-LC-MS/MS and GNPS analyses revealed unique compounds in the active extracts, whose majority did not have a match in mass spectrometry databases and could be potentially novel compounds. In conclusion, microalgae extracts demonstrated anti-obesity, anti-steatosis, and anti-inflammatory activities and could be valuable resources for developing future nutraceuticals. In particular, the ultrasound-assisted ethanolic extract of the heterotrophic C. vulgaris significantly enhanced the anti-obesity activity and demonstrated that the alteration of culture conditions is a valuable approach to increase the production of high-value compounds. 相似文献

Soybean rust (SBR), caused by Phakopsora pachyrhizi, is the most important yield-damaging fungal disease of soybean due to severe reduction in healthy leaf area and acceleration of leaf fall. In experimental research, SBR severity is estimated visually aided/trained by a standard area diagram (SAD) developed and validated during the mid-2000s (Old SAD). In this study, we propose a new SAD set for SBR with six true-colour diagrams following linear increments (c. 15% increments) amended with four additional diagrams at low (<10%) severities, totalling 10 diagrams (0.2%, 1%, 3%, 5%, 10%, 25%, 40%, 55%, 70%, and 84%). For evaluation, 37 raters were split into two groups. Each assessed severity in a 50-image sample (0.25%–84%), first unaided and then using either the Old SAD or the New SAD. Accuracy, precision, and reliability of estimates improved significantly relative to unaided estimates only when aided by the New SAD (accuracy >0.95). Low precision (<0.78) and a trend of underestimation with an increase in severity were the main issues with the Old SAD, which did not differ from unaided estimates. Simulation to evaluate the impact of the errors by different methods on hypothesis tests, showed that the new SAD was more powerful for detecting the smallest difference in mean control (e.g., 70% vs. 65% disease reduction) than the Old SAD; the latter required a 2-fold increase in sample size to achieve the same power. There is a need to improve some SADs, taking advantage of new knowledge and technology to increase accuracy of the estimates, and to optimize both resource use efficiency and management decisions. 相似文献

Detrital carbon accumulation accounts for most of an ecosystem's capacity to store organic carbon because the carbon contained as plant detritus exceeds that stored in living plants by about threefold. A comparative analysis of the mass and turnover of detrital carbon in ecosystems demonstrates that these properties are strongly related to the turnover rate of the dominant primary producers and are poorly related to ecosystem primary production. These results contribute to an understanding of the factors that control carbon storage in ecosystems and the role of carbon storage in the global carbon budget. 相似文献

We evaluated recruitment of larvae of catarina scallop, Argopecten ventricosus, in the area of Puerto Peñasco, NE Gulf of California. We moored artificial collectors in six sites from June 2007 to August 2008 and replaced them every 2 months. We used monthly (July 2002–September 2011) sea surface temperature (SST, °C) and surface chlorophyll‐a concentration (SSChl, mg m?3) Aqua/MODIS satellite data to describe seasonal environmental behaviour study area. Also, we recorded bottom temperature at each site every 4 h, and every 2 months measured sea surface salinity, temperature and dissolved oxygen. We used a repeated measures anova to evaluate differences in the number of recruited spat between main factors, and analysed the presence of multimodal spat shell size frequency distributions. Overall, spat recruitment was negatively correlated with seawater temperature and showed higher spat recruitment abundances throughout winter, which is the season with the highest surface chlorophyll a concentration. We estimated multimodal shell size frequency distributions characterized by more than one modal size. The natural collection of A. ventricosus spat on artificial collectors in the area can be successfully performed over a protracted period (November–December to May–June). Our results extend the area where collection of A. ventricosus spat can be successful. 相似文献

The lion's paw scallop, Nodipecten subnodosus (Sowerby) has considerable aquacultural potential due to its fast growth and large adductor muscle. Prior investigations throughout northwestern Mexico's littoral have reported highly variable growth rates; furthermore, no studies are available of the environment on growth and gametogenesis in this species under culture conditions. This investigation assesses the effect of food availability and temperature on the growth and gametogenesis of N. subnodosus in a suspended culture system at Guerrero Negro lagoon, Mexico. After 1 year of cultivation, N. subnodosus reached 69.13 mm in shell height (SH) (0.196 mm day?1, 14 months old). Two significant growth spurts were observed: over the two first months of culture (August and September 2001, mean growth rate 0.4 mm day?1) and in September 2002 (0.3 mm day?1), both related to high temperatures and chlorophyll a concentrations. The onset of gametogenesis occurred in April 2002, with an increase in temperature (10‐month‐old scallops, 54.5 mm SH). The first spawning occurred in October and November (86.2 and 93 mm SH), with peak temperatures. These results, together with the analysis of previous reports, indicate that N. subnodosus has a higher preference for temperate areas; therefore, the Guerrero Negro lagoon appears to be a suitable site to culture this species. 相似文献

A study was undertaken to determine the effect of various dietary carbohydrate‐to‐lipid ratios on growth performance, whole‐body composition and tissue lipid content in Senegalese sole (Solea senegalensis) juveniles. Data on the dietary regulation of key hepatic enzymes of the lipogenic and glycolytic pathways (glucose‐6‐phosphate dehydrogenase, G6PD; malic enzyme, ME; fatty acid synthetase, FAS; pyruvate kinase, PK and glucokinase, GK) were also generated. Four isonitrogenous (crude protein: 52% dry matter (DM)) diets were formulated to contain one of two lipid levels (11% and 21% DM). Within each dietary lipid level, the nature of the carbohydrate fraction (raw or extruded peas) was varied. Triplicate groups of 54 sole (initial body weight: 23.6±1.2 g) were grown in recirculated seawater over 67 days. Fish were fed using automated feeders. At the end of the study, whole‐body, liver, viscera and muscle samples were withdrawn for analyses. During the experimental period, the mean fish weight about doubled in all treatments. No significant differences were found in growth performance (ranging from 1.1% to 1.4% body weight day?1) among dietary treatments. High‐fat diets increased whole‐body fat content. Similarly, daily fat gain ranged from 0.54 to 0.78 g kg?1 day?1 and highest values were found in fish fed high‐lipid diets. Dietary treatments also affected tissue lipid content (liver, viscera and muscle), with highest values in fish fed high‐fat diets. The nature of dietary carbohydrates had little influence on performance criteria, but affected tissue lipid deposition. The activities of G6PD, ME and FAS were depressed by elevated levels of dietary lipid, confirming the inhibitory effect of dietary fats on lipid biosynthesis. At both dietary lipid levels, ME and FAS activities were little affected by dietary carbohydrate. Activities of PK and GK were not affected by the starch level of the diets. In Senegalese sole juveniles, the lipogenic pathway is more susceptible to modulation by dietary means (particularly through lipid intake) than the glycolytic pathway. 相似文献

The aim of this study was to determine whether the sexual activity of local male Creole goats in subtropical Mexico can be induced during the non-breeding season by a long-day treatment followed by insertion of two melatonin implants. The experiment was carried out in the Laguna region in the State of Coahuila, Mexico (26 degrees N). Fourteen male goats were allocated to two balanced groups (n = 7 each) according to body and testicular weights. Males were kept together in two separate groups and fed lucerne hay for ad libitum intake and 300 g of commercial concentrate and had free access to water and mineral blocks. The control group remained in open sheds under natural photoperiod and ambient temperature conditions. The experimental group was placed in a light-proof building and exposed to 2.5 mo of long days (16 h of light/d) from November 1 to January 15. On January 16, each male received two s.c. melatonin implants and was exposed to natural photoperiodic changes in an open shed. In the control group, testicular weight exhibited seasonal variations; the highest value occurred on May 30 (146 +/- 10 g). Treated males reached maximum testicular weight earlier (March 15; 147 +/- 11 g), and sperm quality from January to March was higher than that observed in the control group (P < 0.05). Treatment caused an increase in LH pulse frequency (2.0 +/- 0.5 vs 0.3 +/- 0.2 pulse/8 h in February, 4.6 +/- 1.1 vs 0.1 +/- 0.1 pulse/8 h in March; experimental vs control group, respectively). In the control group, plasma testosterone remained low until mid-June and increased thereafter to remain elevated until the end of the study. In the experimental group, elevated plasma testosterone was observed from February to April and from July to November. Treating male goats in subtropical latitudes with artificial long days and melatonin can induce an intense sexual activity during the natural nonbreeding season. 相似文献
