ABSTRACT: Mycoplasma haemofelis is a pathogenic feline hemoplasma. Despite its importance, little is known about its metabolic pathways or mechanism of pathogenicity due to it being uncultivatable. The recently sequenced M. haemofelis str. Langford 1 genome was analysed and compared to those of other available hemoplasma genomes.Analysis showed that in hemoplasmas genes involved in carbohydrate metabolism are limited to enzymes of the glycolytic pathway, with glucose appearing to be the sole energy source. The majority of the pentose phosphate pathway enzymes that catalyze the de novo synthesis of ribonucleotides were absent, as were cell division protein FtsZ and chaperonins GroEL/ES. Uncharacterized protein paralogs containing putative surface expression motifs, comprised 62% of M. haemofelis and 19% of Mycoplasma suis genome coverage respectively, the majority of which were present in a small number of unstructured islands. Limited mass spectrometry and immunoblot data matched a number of characterized proteins and uncharacterized paralogs, confirming their expression and immunogenicity in vivo.These data have allowed further characterization of these important pathogens, including their limited metabolic capabilities, which may contribute to their uncultivatable status. The Fort Valley Experiment Station (now Fort Valley Experimental Forest) has contributed many long-term studies to forest research. This paper focuses on a “Methods of Cutting” study initiated in 1913 on the Coulter Ranch Unit of Fort Valley and how that long-term study yielded important ecological and management lessons. We quantified the historical and contemporary forest patterns at this ponderosa pine-Gambel oak site, which was harvested using three different harvesting systems in 1913 (seed tree, group selection, and light selection) and was partially excluded from livestock browsing in 1919. Using nine historically stem-mapped permanent plots for the following three stand structural scenarios: 1913 pre-harvest (modeled), post-harvest (actual), and 2003-2006 (actual) conditions, we examined the short- and long-term consequences of harvest and livestock grazing land-use and stand dynamics. We assessed changes in spatial pattern under each harvesting system and in each structural scenario, and lastly, we examined spatial and temporal tree recruitment patterns as observed in the contemporary (2003-2006) conditions. The seed tree harvests effectively converted the spatial patterns from aggregated to random and left few trees, while the group selection and light selection had varying effects, but consistently exaggerated the spatial patchiness of the stand. By 2003-2006, all plots were aggregated at all scales and were one large patch of predominately small trees. Sites that were harvested, but excluded from livestock browsing had 40% more trees in 2003-2006. Contemporary recruitment patterns were significantly aggregated under all harvesting systems, but were most strongly aggregated if the site received a group selection or light selection cut. For group and light selection, pine seedlings initially established in stump patches created by harvesting and then proceeded to fill-in the remaining area, with recruitment rarely found under the residual pine or oak trees. Long-term data sets, such as these established by the Fort Valley Experimental Forest in 1913, are essential for quantifying the impact of historical land-use practices on contemporary forest composition and structure. Ignoring land-use legacies may lead to the misinterpretation of stand dynamics and development, and therefore should be explicitly quantified and incorporated into future management and restoration activities. 相似文献

OBJECTIVE: To determine the effects of ketamine hydrochloride on hemodynamic and immunologic alterations associated with experimentally induced endotoxemia in dogs. ANIMALS: 9 mixed-breed dogs. PROCEDURES: In a crossover study, dogs were randomly allocated to receive ketamine (0.5 mg/kg, IV, followed by IV infusion at a rate of 0.12 mg/kg/h for 2.5 hours) or control solution (saline [0.9% NaCl] solution, 0.25 mL, IV, followed by IV infusion at a rate of 0.5 mL/h for 2.5 hours). Onset of infusion was time 0. At 30 minutes, lipopolysaccharide (LPS; 1 microg/kg, IV) was administered. Heart rate (HR), systolic arterial blood pressure (SAP), plasma tumor necrosis factor (TNF)-alpha activity, and a CBC were evaluated. RESULTS: Mean SAP was significantly reduced in dogs administered ketamine or saline solution at 2 and 2.5 hours, compared with values at time 0. However, there was no significant difference between treatments. At 1, 2, and 2.5 hours, dogs administered ketamine had a significantly lower HR than dogs administered saline solution. Although plasma TNF-alpha activity significantly increased, compared with values at time 0 for both groups, ketamine-treated dogs had significantly lower peak plasma TNF-alpha activity 1.5 hours after LPS administration. All dogs had significant leukopenia and neutropenia after LPS administration, with no differences detected between ketamine and saline solution treatments. CONCLUSIONS AND CLINICAL RELEVANCE: Administration of a subanesthetic dose of ketamine had immunomodulating effects in dogs with experimentally induced endotoxemia (namely, blunting of plasma TNF-alpha activity). Recent studies have identified the novel, host adapted Cryptosporidium bovis and the deer-like genotype in dairy cattle from farms in the United States, China, India and Europe. This novel species and genotype appear to be more prevalent in older, post-weaned dairy cattle than previously thought. However, little information is available on their prevalence in beef cow-calf operations. In the present study, we determined the prevalence of Cryptosporidium species in 98 calves (6-8 months old) and 114 cows (>2 years old) in seven beef cow-calf herds in western North Dakota. DNA was extracted from fecal samples and Cryptosporidium spp. were identified by amplification of the 18S rRNA gene followed by sequencing or RFLP analysis. All seven herds tested positive for Cryptosporidium. Overall, 43/212 (20.3%) animals were positive. Only five of these positives were from cows. C. bovis, the deer-like genotype and C. andersoni were identified in 9.4, 6.6 and 1.4% of animals sampled, respectively. C. parvum was not identified in any of the positive samples. C. bovis, the deer-like genotype and C. andersoni were detected in 6/7, 5/7 and 2/7 herds, respectively. C. bovis and the deer-like genotype were primarily detected in calves, while C. andersoni was only detected in cows. Six isolates could not be typed. These results show a relatively high prevalence of C. bovis and the deer-like genotype in 6-8-month-old beef calves compared to cows older than 2 years in the seven herds studied. 相似文献

Context
Habitat complexity in rivers is linked to dynamic fluvial conditions acting at various spatial scales. On regulated rivers in the western United States, tributaries are regions of high energy and disturbance, providing important resource inputs for riparian ecosystems.Objectives
This study investigated spatial patterns and extents of tributary influence on riparian habitat complexity in the near channel zone along regulated reaches of the Colorado (>?200 km) and Dolores Rivers (~?300 km) in the western United States. Because tributary confluences are regions of increased dynamism, we hypothesized that: (1) geomorphic and land cover complexity would be greatest close to tributary junctions and decrease with distance from tributaries; and (2) patterns in complexity would vary across different sized spatial units.Methods
Using a combination of remote sensing and spatial analysis, we classified fluvial features and land cover classes to investigate patterns longitudinally at 10-, 25-, and 100-m spatial units in the near channel zone of two regulated rivers.Results
Using change point analysis and randomization tests, we detected shifts in riparian habitat complexity closer to tributary junctions. Patterns varied across 10-, 25-, and 100-m spatial units in the near channel zone, with significance (p?≤?0.05) recorded for 10- and 25-m spatial units.Conclusions
Tributary junctions deliver critical resource inputs on regulated systems, providing for increased geomorphic and land cover diversity upstream and downstream of tributaries. We found that patterns of response were non-linear and discontinuous, varying across spatial units and potentially influenced by the degree of mainstem flow regulation.707.

The goal of this prospective study was to determine the effect of hemoclip use on the size of radiation treatment fields based on a 3-cm margin around a surgical incision alone (field setup 1) vs. a 3-cm margin around the surgical incision plus hemoclips (field setup 2). Forty-seven dogs that underwent surgical resection of a total of 55 soft tissue masses had surgical hemoclips placed at the time of surgery and orthogonal radiographs made immediately postoperatively. Radiation treatment field simulation was done and field areas measured. Additional determinations included number of hemoclips outside of the radiation treatment field based on a margin around the incision alone, hemoclip distance from the incision, and association between incision length and greatest distance of hemoclips from the incision. There was a significant difference in radiation treatment field size using information regarding the location of hemoclips in conjunction with the surgical scar compared with the surgical scar alone for truncal (P = 0.0003) vs. extremity tumors (P = 0.087). In simulating radiation treatment fields hemoclips were located outside of field setup 1 for the majority of tumors (79%) resected from the trunk but only in a minority of tumors (10.7%) resected from extremity sites. The findings from this study suggest that surgical hemoclips have potential utility in simulation of radiation treatment fields in the postoperative setting. 相似文献

Observational studies of cattle production systems usually find that cattle from conventional dairies harbor a higher prevalence of antimicrobial resistant (AMR) enteric bacteria compared to organic dairies or beef-cow operations; given that dairies usually use more antimicrobials, this result is not unexpected. Experimental studies have usually verified that application of antimicrobials leads to at least a transient expansion of AMR bacterial populations in treated cattle. Nevertheless, on dairy farms the majority of antibiotics are used to treat mastitis and yet AMR remains relatively low in mastitis pathogens. Other studies have shown no correlation between antimicrobial use and prevalence of AMR bacteria including documented cases where the prevalence of AMR bacteria is non-responsive to antimicrobial applications or remains relatively high in the absence of antimicrobial use or any other obvious selective pressures. Thus, there are multi-factorial events and pressures that influence AMR bacterial populations in cattle production systems. We introduce a heuristic model that illustrates how repeated antimicrobial selection pressure can increase the probability of genetic linkage between AMR genes and niche- or growth-specific fitness traits. This linkage allows persistence of AMR bacteria at the herd level because subpopulations of AMR bacteria are able to reside long-term within the host animals even in the absence of antimicrobial selection pressure. This model highlights the need for multiple approaches to manage herd health so that the total amount of antimicrobials is limited in a manner that meets animal welfare and public health needs while reducing costs for producers and consumers over the long-term. 相似文献

Equine follicle stimulating hormone (FSH) and luteinizing hormone (LH) were measured in the serum obtained from pregnant mares, bled daily, from up to 17 days before parturition until the first ovulation after parturition. LH was at baseline levels until 2 days before ovulation when it started, and continued, to rise until after ovulation. FSH surges occurred at approximately 24 and 14 days before this post-partum ovulation (12 and 2 days before parturition), thus showing a similar pattern to the cyclic mare, consistent with the hypothesis that 2 surges at these times prepare and prime follicles for subsequent ovulation. The high plasma oestradiol levels that occurred during and immediately before these FSH surges did not show a negative feedback, or the inverse relationship between FSH and oestradiol which occurs in the cyclic mare, and in other species. 相似文献
