(1) Background: G protein-coupled inward-rectifier potassium (GIRK) channels, especially neuronal GIRK1/2 channels, have been the focus of intense research interest for developing drugs against brain diseases. In this context, venom peptides that selectively activate GIRK channels can be seen as a new source for drug development. Here, we report on the identification and electrophysiological characterization of a novel activator of GIRK1/2 channels, AsKC11, found in the venom of the sea anemone Anemonia sulcata. (2) Methods: AsKC11 was purified from the sea anemone venom by reverse-phase chromatography and the sequence was identified by mass spectrometry. Using the two-electrode voltage-clamp technique, the activity of AsKC11 on GIRK1/2 channels was studied and its selectivity for other potassium channels was investigated. (3) Results: AsKC11, a Kunitz peptide found in the venom of A. sulcata, is the first peptide shown to directly activate neuronal GIRK1/2 channels independent from Gi/o protein activity, without affecting the inward-rectifier potassium channel (IRK1) and with only a minor effect on KV1.6 channels. Thus, AsKC11 is a novel activator of GIRK channels resulting in larger K+ currents because of an increased chord conductance. (4) Conclusions: These discoveries provide new insights into a novel class of GIRK activators.  相似文献

This study investigated the acute toxicity (LC50-24 hr) effects of the essential oil of Cymbopogon citratus for adult Argulus sp. and Dolops discoidalis, before and during oviposition. In vitro acute toxicity (LC50-24 hr) was tested using 20, 40, 60, 80, 100, 120, 140 and 160 μg/L of C. citratus essential oil, and two control groups (one with cultivation tank water and one with cultivation tank water + alcohol) were used. Specimens of Argulus sp. and D. discoidalis submitted to acute toxicity were evaluated using histological procedures. The major chemical compounds of C. citratus essential oil were geranial (47.5%), neral (35.6%) and myrcene (6.7%). The LC50-24 hr for Argulus sp. adults was 67.97 μg/L, while for D. discoidalis it was 59.55 µg/L. In the oviposition of both species of argulids, maximum mortality began with treatments of 140 μg/L, while the LC50-24 hr for Argulus sp. and D. discoidalis was 83.98 μg/L and 82.48 μg/L, respectively. In both argulid species exposed to C. citratus essential oil, morphological alterations were observed only in the eyes, and they occurred in the ommatidium and rhabdomeres and were dependent on the concentration of C. citratus essential oil and the parasite species.  相似文献

Fish farming has grown rapidly in Brazil over the last two decades, due in part to the availability of water and species with potential for cultivation, such as Colossoma macropomum. This farmed fish, however, can be affected by parasitic infections with high mortality rates (>35%). The main objective of the present study was to describe the first occurrence of the Trypanosoma species in Colossoma macropomum, which occurred in fish cultured in the Brazilian Amazon region, as well as to morphologically characterize the trypomastigotes and the impact of these hemoparasites on the body condition of the hosts. The trypanosomes found in the blood of Colossoma macropomum were morphologically characterized by the size of the trypomastigote form, indicating the presence of only one morphotype. Of the 39 hosts examined from one fish farming, 41.0% were infected by Trypanosoma sp., with low levels of infection (mean blood density of 3.7/μL). The condition factor of the asymptomatic hosts was not affected by hemoparasitism. There was no correlation between the abundance of Trypanosoma sp. and the condition factor and size of the hosts. Finally, our understanding of host-parasite interactions and the detection of emerging diseases are fundamental for aquaculture.  相似文献

Soils from central Brazil have been intensively used over the last decades because of the rapid conversion of savannas ( Cerrado ) into corn/soybean fields. The objective of this work is to study modifications in the physical properties of soils in the Rio Verde watershed, as a function of the land use time for agriculture, determined from classification of Landsat satellite images between 1980 and 2010. Soil samples were collected at surface (0–20 cm) and subsurface (20–40 cm) horizons for the different classes of land use time (<10, 10–20, 20–30, and >30 years). The following physical properties were measured: bulk density (BD), air permeability (Ka), penetration resistance (PR), microporosity (MI), macroporosity (MA), and total porosity (TP). Results showed a strong expansion with time of agriculture that occupied 35·3% (1980), 37·4% (1990), 51·3% (2000), and 60·9% (2010) of the watershed area. When properties were compared with those from the reference areas (preserved soils under native vegetation), significant differences were observed for all the physical attributes of soils for a land use time higher than 20 years. Overall, BD and PR increased with land use time, and the opposite was verified for Ka, MA, and TP. Some physical properties presented values (e.g., 1·54 g cm−3 for BD and 0.06 cm3 cm−3 for MA) close to the critical ones reported to affect crop development, but they were not still impacting on local soybean yield. Ornithobacterium rhinotracheale (ORT) has been associated with respiratory disease, increased mortality, retarded growth, and decreased egg production in chickens and turkeys. Surveillance of exposure to ORT infection in the field has shown that prevalence of the infection is higher during winter months. The ability of ORT to remain viable in the poultry litter was studied at different temperatures over time. Presterilized poultry litter was inoculated with 10(11) colony-forming units of ORT and kept at -12 C, 4 C, 22 C, 37 C, and 42 C. Reisolation and titration of ORT from litter was attempted at intervals. Results indicate that ORT survived for 1 day at 37 C, 6 days at 22 C, 40 days at 4 C, and at least 150 days at -12 C. ORT did not survive 24 hr at 42 C. The survival of ORT at lower temperatures may be associated with the higher incidence of ORT infection in poultry during winter months.  相似文献

Ornithobacterium rhinotracheale (ORT) is a bacterium responsible for a respiratory disease in turkeys and chickens and has been identified as one of the emerging respiratory bacterial pathogens. The clinical signs and lesions caused by ORT are very similar to those caused by other respiratory infectious agents; therefore, an accurate diagnostic test is necessary to identify the infection. In this study, we have investigated the use of outer membrane proteins of ORT in an indirect enzyme-linked immunosorbent assay (ELISA) to detect the exposure to ORT infection. Outer membrane proteins of ORT were extracted and used as an antigen in ELISA to detect infection in turkeys exposed to different serotypes of ORT. The ELISA results were compared with the conventional serum plate agglutination test. The agglutination test detected specific antibodies for ORT in 65% of experimentally infected turkeys during the first 2 wk of infection. The ELISA detected up to 100% of infected birds for 8 wk postinfection. The results suggest that ELISA is able to detect the exposure to ORT in later stages of the infection and this assay can be used in serologic surveillance of ORT infection for poultry in the field.  相似文献
