Measurement of fresh tomato fruit overall quality, and particularly lycopene content, is challenging in the context of high-volume production. An experiment was conducted to simultaneously measure various quality parameters of tomato in a nondestructive manner using vis-NIR reflectance spectroscopy and chemometrics. The sampling set included different cultivars that are obtainable from both retailers' shelves and two greenhouse producers. Results indicate that lycopene content was accurately predicted [r(2) = 0.98; root mean square error of cross-validation (RMSECV) = 3.15 mg/kg], along with color variables such as Hunter a (r(2) = 0.98), L, and b (r(2) = 0.92). Tomato color index (TCI) was better predicted (r(2) = 0.96) than the a/ b ratio (r(2) = 0.89). Firmness prediction, with an r(2) of 0.75, is comparable to what is reported in the literature for other fruits and may have a practical interest. Prediction of internal quality such as pH, soluble solids, titratable acidity, and electrical conductivity was less accurate, partly due to a low variability of these parameters among samples. Predictions were robust with regard to cultivars, except for pink variety tomato. The 400-1000 nm range gave results almost as accurate as the 400-1500 nm range.  相似文献

Coagulase-negative staphylococci (CNS) have become the predominant pathogens causing bovine mastitis in many countries. CNS infections are associated with damage to milk secretory tissue of the mammary gland by increased connective tissue stroma, moderate increases of somatic cells count in milk and significant production decreases. These consequences impose serious economic losses for the farmers and the dairy industry. Routine veterinary laboratories do not usually identify CNS at the species level. Thereby, the aims of this study were to identify the most common staphylococcal pathogens involved in bovine mastitis using PCR-restriction fragment length polymorphism (RFLP) analysis of a partial groEL gene sequence and to compare our results with the identification carried out by the conventional method. A total of 54 isolates of Staphylococcus, involved in bovine mastitis, were analyzed by this method. The size and number of the fragments obtained by either AluI or HindIII/PvuII digestions made possible to form clear patterns differentiating, among the isolates, 11 of the most common species of animal staphylococcal pathogens. Most of the isolates clustered together with the reference strain of Staphylococcus chromogenes (28) and the type strain of Staphylococcus epidermidis (8). Besides, some isolates clustered together with the type strain of Staphylococcus aureus (5). All patterns were confirmed by the conventional biochemical method, showing concordant results. Thus, the PCR-RFLP of the groEL gene constitutes a reliable and reproducible molecular method for identification of CNS species responsible for bovine mastitis.  相似文献

Samples from moribund lumpfish were collected in a marine hatchery in Scotland in 2015. Black nodules were noted on the skin, and gills and fungal hyphae were extensively distributed in musculature and internal organs. Multifocal chronic inflammatory lesions displaced structures in all affected organs. Mortalities commenced on completion of spawning in May and were evenly distributed over the second year in the temperature range 11–15°C. The main systemic infection causing agent was initially identified based on morphological characteristics as an Exophiala species. Ribosomal DNA (rDNA) ITS regions of the isolates were subsequently sequenced confirming the isolates belonged to Exophiala genus. All isolates fell in a single phylogenetic cluster, which is represented by Exophiala angulospora. Fish were treated with either formalin or Bronopol or a combination of both, but there was no effect on the pattern or numbers of mortalities. Isolates were also tested against three different concentrations of Latrunculin A, Amphotericin B and Itraconazole with no success. It is of utmost importance to increase the knowledge on pathogen–host interactions to successfully develop sustainable control methods.  相似文献

Ouro Negro (Honduras 35) is a highly productive Mesoamerican black seeded bean cultivar that possesses a major dominant gene conferring resistance to anthracnose (causal organism Colletotrichum lindemuthianum). In this work the anthracnose resistance gene present in Ouro Negro was characterized by studying allelic relationships to the following previously characterized anthracnose resistance genes (cultivars): Co-1 (MDRK), Co-1 2 (Kaboon), Co-1 3 (Perry Marrow), Co-2 (Cornell 49-242), Co-3 (Mexico 222), Co-4 (TO), Co-4 2 (SEL 1308), Co-5 (SEL1360), Co-6 (AB 136), and the resistance genes present in PI 207262 and Widusa. In addition, we determined the resistance spectrum of Ouro Negro in relation to 19 pathotypes of C. lindemuthianum. The allelism tests confirmed that the dominant anthracnose resistance gene present in Ouro Negro is positioned at a locus distinct from those with which it was compared. We propose that this new gene be named Co-10. The inoculation of Ouro Negro with the 19 pathotypes of C. lindemuthianum demonstrated that Co-10 confers resistance to pathotypes 23, 64, 67, 73, 81, 83, 87, 89, 95, 102, 117, 119, 343, 453, 1033, 1545 and 1600. The identification of Co-10 is an important contribution to bean breeding programs that are in constant need of new sources of resistance to anthracnose. Mast cell tumor (MCT) is one of the most prevalent neoplasms that affect skin and soft tissue in dogs. Because mast cell tumors present a great variety of clinical appearance and behavior, their treatment becomes a challenge. Trichostatin A (TSA), an antifungal antibiotic, has shown inhibitory effects on the proliferation and induction of apoptosis in various types of cancer cells. In order to evaluate the potential of trichostatin A as a therapeutic drug, cells of grade 3 MCT were cultured and treated with concentrations of 1 nM to 400 nM of TSA. MTT assay and trypan blue exclusion assays were performed to estimate cell growth and cell viability, and cell cycle analysis was evaluated. TSA treatment showed a reduction in numbers of viable cells and an increase of cell death by apoptosis. The cell cycle analysis showed an increase of hypodiploid cells and a reduction of G0/G1 and G2/M –phases. According to these results, trichostatin A may be an interesting potential chemotherapeutic agent for the treatment of canine MCT.  相似文献
